In vitro and in vivo antibacterial activities of root extract of tissue cultured Pluchea indica (L.) Less.

  • The methanolic root extract of tissue cultured Pluchea indica (L.) Less. was tested for its antibacterial potentiality against 102 different strains of bacteria belonging to both Gram positive and Gram negative groups. The bacteria could be arranged according to their decreasing order of sensitivity as follows: Staphylococcus aureus, Vibrio cholerae, Bacillus spp. Vibrio parahaemolyticus, shigellae, Salmonella spp., and Escherichia coli. The extract was found to be bacteriostatic in nature against Salmonella typhimurium NCTC 74. When administered to Swiss strain of white mice at the doses of 0.5 and 1.0 mg/kg body weight, the extract could significantly protect the animals challenged with 50 MLD of S. typhimurium NCTC 74. According to the chi-square test, the in vivo data is highly significant (P < 0.001).

  • Antimicrobial activity of various extracts of Xanthium strumarium L. was tested against 25 strains of bacteria, yeast and fungus. The crude ethylacetate extract exhibited strong growth inhibition to the tested strains with the exception of partial Gram-negative bacteria. The property of antimicrobial compound was very stable under heat treatment at $120^{\circ}C$, but it was unstable in acid (pH 3.0) and alkali (pH 10.0) treatment. The antimicrobial compounds were purified by boiling water extraction, ethylacetate extraction, charcoal column chromatography, silica gel column chro- matography and reverse phase HPLC. The purified compound A and B were detected in a single peak (each above 98% purity) through the HPLC analysis. The compound A and B showed a strong growth inhibition against Gram-negative and positive bacteria in the agar diffusion method. When tested by the FDA method using the esterase, compound A mainly inhibited the growth of bacteria and compound B showed the growth inhibition of both bacteria and yeasts.

  • To investigate the effect on the antimicrobial activity against S.aureus ATCC 6538, E.coli KCTC 1039 and cell toxic level against transformed mouse fibroblast L929 in formula added with various concentrations of UVB blockers commonly used in cosmetic products, these experiments were carried out by preservative efficacy testing methods and in vitro cytotoxicity methods. The results obtained were as follow ; 1) Octyl Dimethyl PABA had a broad antibacterial spectrum against the Gram (+) and the Gram(-) bacteria at 5.84 % concentration, but not Octyl Methoxycinnamate. 2) Antibacterial activity was decreased in a combined UVB blocker system of squalane base. Especially, Octyl Dimethyl PABA was inactivated by Octyl Methoxycinnamate at 5.84% concentration to a large extents , but not 4-Methylbenzylidene Camphor. 3) Within in vitro cytotoxicity by use of mouse fibroblast L929 on UV-B blockers, NR assay was more excellent than MTT assay on quantitative

The Effects of Antibacterial Activity of Exopolysaccharide Isolated from Tibetan Mushroom Culture against Foodborne Pathogenic Bacteria: A Preliminary Study

  • This study aimed to evaluate the effects of antibacterial activity of Tibetan mushroom exopolysaccharide against foodborne pathogenic bacteria Staphylococcus aureus 305, Listeria monocytogenes ATCC19114, Escherichia coli O157:H7 ATCC42894 and Escherichia coli O55. The yield of exopolysaccharide isolated from Tibetan mushroom culture was 620 mg/L. The antibacterial activity of exopolysaccharide against foodborne pathogenic bacteria exhibited 15 mm and 12 mm clear zone against S. aureus 305 and L. monocytogenes ATCC 19114, respectively. However, no clear zone was observed against E. coli O157:H7 ATCC 42894 and E. coli O55. In conclusion, exopolysaccharide isolated from Tibetan mushroom culture have the antibacterial activity only against Gram-positive foodborne pathogenic bacteria.

Identification of Uncharacterized Anti-microbial Peptides Derived from the European Honeybee (꿀벌 Apis mellifera에서 유래 한 특성화 되지 않은 항균성 펩티드의 동정)

  • The European honeybee (Apis mellifera L.) has multiple anti-microbial peptides, but many were unknown and demands for their characterization have increased. This study therefore focused on identifying novel anti-microbial peptides (AMPs) from A. mellifera L. To obtain high-throughput transcriptome data of the honeybee, we implemented next-generation sequencing (NGS), isolating novel AMPs from total RNA, and generated 15,314 peptide sequences, including 44 known, using Illumina HiSeq 2500 technology. The uncharacterized peptides were identified based on specific features of possible AMPs predicted in the sequencing analysis. AMP5, one such uncharacterized peptide, was expressed in the epidermis, body fat, and venom gland of the honeybee. We chemically synthesized this peptide and tested its anti-bacterial activity against Gram-negative Escherichia coli (KACC 10005) and Gram-positive Bacillus thuringiensis (KACC 10168) by anti-microbial assay. AMP5 exhibited anti-bacterial activity against E. coli (MIC50=22.04±0.66 μM) but not against B. thuringiensis. When worker bees were injected with E. coli, AMP5 was up-regulated in the body fat. This study therefore identified AMP5 in adult European honeybees and confirmed its anti-bacterial activity against Gram-negative E. coli.

Phytochemicals and antioxidant capacity of some tropical edible plants

  • Objective: To find biological functions such as antibacterial and antioxidant activities in several tropical plants and to investigate the possibility of antibiotic substitute agents to prevent and treat diseases caused by pathogenic bacteria. Methods: Plants such as Poncirus trifoliata fruit (Makrut), Zingiber officinale Rosc (Khing), Areca catechu L. (Mak), Solanum melongena L. I (Makkhuayao), and Solanum melongena L. II (Makhurapro) were extracted by methanol, n-hexane, chloroform, ethyl acetate, butanol and water. The free radical scavenging activities were measured using 2-diphenyl-2-picryl hydrazyl photometric assay. Antibacterial activities with a minimum inhibitory concentration (MIC) were observed by agar diffusion assay against pathogenic strains of Escherichia coli, Burkholderia sp., Haemopilus somnus, Haemopilus parasuis, Clostridium perfringens, and Pantoea agglomerans. Results: Poncirus trifoliata fruit methanol extract showed antibacterial activities against gram-negative and gram-positive pathogens. Additionally, this showed the strongest antibacterial activity against Burkholderia sp. and Haemopilus somnus with MIC $131{\mu}g/mL$, respectively. Areca catechu L. water extract showed antibacterial activities against Burkholderia sp., Haemopilus somnus, and Haemopilus parasuis. The MIC value for Haemopilus parasuis was $105{\mu}g/mL$ in this. Antioxidant activity of Zingiber officinale Rosc n-hexane extract showed 2.23 mg/mL effective concentration 50% ($EC_{50}$) value was the highest activity among tropical plants extracts. Total polyphenol content in Zingiber officinale Rosc methanol extract was $48.4{\mu}g/mL$ and flavonoid content was $22.1{\mu}g/mL$ showed the highest values among tested plants extracts. Conclusion: Taken together, these results suggest that tropical plants used in this study may have a potential benefit as an alternative antibiotics agent through their antibacterial and antioxidant activities.

Antimicrobial Characteristics of Different Parts of Guava against Food-Borne Bacteria (식중독 세균에 대한 구아바 부위별 추출물의 항균 특성)

  • Guava (Psidium guajava L.) contains high amount of vitamins and minerals, and its leaves have been reported to be very effective on reducing blood pressure. In this study, antimicrobial characteristics of extracts from four different parts of guava (fruit, branch, leaf, and seed) with four different solvents (methanol, ethanol, acetone, and water) were evaluated. Four targeted food-borne microorganisms were selected; two Gram negatives (Escherichia coli O157:H7 and Salmonella Typhimurium) and two Gram positives (Staphylococcus aureus and Listeria monocytogenes). By the paper disc method, guava extracts showed stronger clear hollow zone against Gram positive bacteria than Gram negatives in nutrient broth agar plate. Especially, extracts of branches and leaves showed significant antibacterial activity. Guava extracts also showed significant inhibition activity on the growth of Gram positive food-borne bacteria in nutrient broth. For example, S. aureus did not grow at all at 200 ppm of acetone extracts of guava branch and leaf. In the case of L. monocytogenes, the same concentration of acetone extracts of guava branch and leaf inhibited the growth 33.5% and 55.4%, respectively, at 32 hrs of incubation time. The results indicated that extracts of guava branch and leaf showed significant antibacterial activities against food-borne Gram positive microorganisms, and that guava branches, the byproducts of guava, might be a valuable resource for antibacterial materials.

Bactericidal Effect of Electrolyzed Activated Water Prepared at Different Water Temperatures on Gram-Positive and Gram-Negative Bacteria (전해수 생성온도에 따른 그람양성균과 그람음성균의 살균 효과)

  • Electrolyzed activated water (EAW) has been reported to exhibit strong bactericidal effects on foodborne microorganisms. However, the disinfection efficacy of EAW is affected by factors such as water source and hardness. This study investigated bactericidal effects of EAW against three gram-positive (Bacillus cereus, Listeria monocytogenes, and Staphylococcus aureus) and three gram-negative (Cronobacter sakazakii, Escherichia coli O157:H7, and Salmonella Enteritidis) foodborne pathogens. Six strains were treated with EAW prepared at different water temperatures (4, 22, and $40^{\circ}C$) for 15 min, and D-values were generated. The results show that the lowest D-values for Lis. monocytogenes by EAW produced at $4^{\circ}C$ and $40^{\circ}C$ were 6.60 and 1.57 min, respectively. The lowest D-value for Sal. Enteritidis by EAW produced at $22^{\circ}C$ was 2.92 min. D-values of all strains treated by EAW produced at $40^{\circ}C$ decreased significantly compared to those treated by EAW produced at $4^{\circ}C$ (P<0.05). These results demonstrate that applying EAW produced at warm temperature is more effective for reducing foodborne pathogens for food safety.

The consideration about exact set-up with stereotactic radiosurgery for lung cancer. (폐암 환자의 전신 정위적 방사선 수술시 정확한 SET UP에 대한 고찰)

  • Purpose : What confirm a patient's set-up precisely is an important factor in stereotactic radiosurgery Especially, the tumor is moved by respiration in case of lung cancer. So it is difficult to confirm a exact location by L-gram or EPID. I will verify a exact patient's set-up about this sort of problem by verification system(exactrac 3.0) Materials and Methods : The patient that had lung cancer operated on stereotactic radiosurgery is composed of 6 people. The 5 patients use an ABC tool and 1 patient doesn't use it. I got such a patient's L-gram and EPID image by Body frame(elekta, sweden), compared Ant. image with Lat. one, and then confirmed a set-up. I fused DRR image of CT and X-ray image of Verification system(exactrac 3.0) 3 dimensional, analyzed the coordinate value(vertical, longitudinal, lateral), and then confirmed a difference of existing method. Results : In case of L-gram and EPID, we judge an exact of the patient's set-up subjectively, and on we could treat the patient with radiation. As a result of using Verification system(exactrac 3.0), coordinate value(vertical, longitudinal, lateral) of patient's set-up was comprised within 5mm. We could estimate a difference of the coordinate value visually and objectively. Consequently, Verification system(exactrac 3.0) was useful in judging an exact patient's set-up. Conclusion : In case of Verification system(exactrac 3.0), we can confirm an exact patient's set-up at any time whenever, However, there are several kinds of the demerit. First, it is a complex process of confirmation than the existing process. Second, thickness of CT scan slice is within 3mm. The last, X-ray image has to have shown itself clearly. If we solve this problem. stereotactic radiosurgery will be useful in treating patients why we can confirm an exact patient's positioning easily.

Inhibition Effect of the Harmful Food-Born Microorganisms on Germination Condition of Acorn Pollen (도토리 화분의 발아 조건에 따른 식품유해균 억제효과)

  • This study investigated the antimicrobial effect of germinated acorn pollen solution on harmful food-borne microorganisms. The antimicrobial activity when 8% (w/v) acorn pollen in 10% (w/v) sugar solution was extracted at $30^{\circ}C$ for 4 days. The minimal inhibitory concentration of this germinated acorn pollen solution was $40\;{\mu}L/mL$ for Gram-positive bacteria and $30\;{\mu}L/mL$ for Gram-negative bacteria. Acetic and lactic acids were present at high levels in germinated acom pollen solution. As pollen germination releases heat, the antimicrobial activities are heat-stable. The activities are tolerant of low pH. In summary, acorn pollen germination solution showed active antibiosis and should be developed as a natural preservative material.
