The detection and distribution of Aeromonas in water supplies were investigated by using the USEPA Method 1605. Water samples were collected from the Han River, finished waters and tap waters supplied from Water Treatment Plants in Seoul monthly from July 2002 to December 2003. Aeromonas species in each water sample were quantified based on the development of yellow colonies on the surface of membrane filter using a selective medium (Ampicillin-Dextrin Agar with Vancomycin). The Quality Control (QC) for this study met the acceptance criteria of Method 1605. The concentrations of Aeromonas species in surface water samples ranged from $1.0{\times}10^{0}\;to\;9.8{\times}10^{3}\;CFU/ml$. Aeromonas species were found only in one tap water sample with concentration of 1 CFU/500 ml. No Aeromonas species were found in any finished water samples. Aeromonas species detected here were identified as A. salmonicida(51%), A. caviae(4.7%), A. schubertti(3.4%), A. sobria(3.8%), A. hydrophila(2.1%), and A. ichithiosmia(0.4%). A. salmonicida was the dominant species, which is of no significance to human health. Chlorine resistance of A. salmonicida was evaluated and as a result, 99.99% of A. salmonicida decreased after 30 seconds exposure at residual free chlorine 0.2 mg/L. These suggest that the waters supplied in Seoul may be safe against the pathogenic agent Aeromonas.
To evaluate the bacteriological water quality in Yongnam-Gwangdo, located in western Jinhae Bay, seawater samples were analyzed using sanitary indicator bacteria at 57 sampling stations. According to survey results from January 2007 to December 2009, the range of the geometric mean and the estimated 90th percentile for coliforms and fecal coliforms in the samples were <1.8-16.5 and 1.8-246.8 MPN/100 mL and <1.8-7.1 and 1.8-74.8 MPN/100 mL, respectively. The samples, including those taken from stations located in Wonmunman, Gwangdo, and Dangdong, showed high levels of microbial contamination caused by the climate and weather patterns in the marine environment. The bacteriological water quality in the area met Korean criteria for a designated shellfish growing area for export and National Shellfish Sanitation Program criteria for an approved shellfish growing area, except at station #49. A total of 24 direct pollution sources were discharged into the shellfish growing area. The radius of impact was calculated for each pollution source to assess the effect on the shellfish growing area. The calculated radius of impact for most of the pollution sources was below 300 m. However, the radius of impact for the combined pollution sources in Kyeonnaeryang was 93-1973 m. There were significant differences between the calculated closed sea area and actual monitoring results. The closed sea area values calculated from the fecal coliform load in drainage water tended to be higher than the actual monitoring results. Tidal currents and environmental factors such as salinity, water temperature, sunlight, and microbiological factors affect the survival of fecal indicator bacteria in seawater.

Background Surgical reconstruction of chronic wounds is often infeasible due to infection, comorbidities, or poor viability of local tissues. The aim of this study was to describe the authors' technique for improving the regenerative and antimicrobial potential of a combination of modified nanofat and platelet-rich plasma (PRP) in nonhealing infected wounds. Methods Fourteen patients met the inclusion criteria. Fat tissue was harvested from the lower abdomen following infiltration of a solution of 1,000 mL of NaCl solution, 225 mg of ropivacaine, and 1 mg of epinephrine. Aspiration was performed using a 3-mm cannula with 1-mm holes. The obtained solution was decanted and mechanically emulsified, but was not filtered. Non-activated leukocyte-rich PRP (naLR-PRP) was added to the solution before injection. Patients underwent three sessions of injection of 8-mL naLR-PRP performed at 2-week intervals. Results Thirteen of 14 patients completed the follow-up. Complete healing was achieved in seven patients (53.8%). Four patients (30.8%) showed improvement, with a mean ulcer width reduction of 57.5%±13.8%. Clinical improvements in perilesional skin quality were reported in all patients, with reduced erythema, increased thickness, and increased pliability. An overall wound depth reduction of 76.6%±40.8% was found. Pain was fully alleviated in all patients who underwent re-epithelization. A mean pain reduction of 42%±33.3% (as indicated by visual analog scale score) was found in non-re-epithelized patients at a 3-month follow-up. Conclusions The discussed technique facilitated improvement of both the regenerative and the antimicrobial potential of fat grafting. It proved effective in surgically-untreatable infected chronic wounds unresponsive to conventional therapies.
This interlaboratory study was designed to evaluate protozoan test methods in water and to predict the major causes of deviation of the methods. Each of four laboratories with previous experience of protozoa analysis in water participated, and met the initial performance criteria of EPA 1623 method provided. The protozoan analysis procedure consists of filtrations, concentration, immunomagnetic separation, dyeing (staining) and counting with observation. We tested three different filtration equipments: capsule filter for 10 L of surface water, and high volume (HV) capsule filter and membrane filter for 100 L of finished water. When the recovery of each step of the procedure was evaluated with EasySeed, the commercial stock of each 100 Cryptosporidium and Giardia, immunomagnetic separation and filtration step were the most crucial steps affecting the stability of the recovery, especially for Cryptosporidium. There was no significant difference of recovery among the filtration methods. Recovery of protozoa from source water were evaluated with spiked EasySeed as matrix tests. The recoveries of Giardia increased significantly in the matrix tests compared those in the deionized water. We also applied red stained mixture stocks of Cryptosporidium and Giardia called ColorSeed as internal standards of water sample tests. The recoveries of both EasySeed and ColorSeed in samples tested were within the range of the criteria, however, the Giardia recoveries using ColorSeed decreased significantly. Further optimization study with ColorSeed will be necessary, considering the convenience of using the internal standard without additional sample analysis. The significant factors of the recovery variation were identified as the differences of laboratories as well as water quality and type of the stock for spiking. The results of this study emphasize the importance of the quality assurance program for protozoan analysis lab in water.

The hair nourisher products are used for prevention of hair loss and classified as quasi-drug in Korea. As concerns about hair loss has been increased, the demand for hair nourisher products has been growing. It is difficult to analyze their main ingredients because they contain various ingredients including natural plant extracts, vitamins, preservatives and exfoliators. The purpose of this study was to develop and validate simultaneous analytical methods of active ingredients in hair nourisher products such as nicotinamide, tocopherol, salicylic acid, dexpanthenol and benzyl nicotinate by HPLC. The active ingredients were separated on a $C_{18}$ column by using acetonitrile/phosphate buffer as a mobile phase, and detected at UV 220, 270 and 300 nm. The calibration curve showed linearity in the range of $12.5{\sim}800\;{\mu}g/mL$ and the recoveries were 97.3 ~ 103.5 % (RSD 0.9 ~ 2.8 %) in liquid matrix and 101.9 ~ 115.9 % (RSD 0.7 ~ 7.7 %) in shampoo matrix. Validated method was applied to hair nourisher products obtained from distribution market. Fortunately, all samples met their criteria. This study might be expected to provide the method for determining active ingredients in hair nourisher products and lead to promote a rapid market entry.

Silkworms have traditionally been used to produce silk and textiles. However, steamed and freeze-dried mature silkworm larval powder (SMSP) contain large amounts of amino acids, vitamins, and essential minerals. In this study, we investigated the potential of SMSP as a hangover-relieving agent. Thirty individuals who met the selection criteria and exclusion criteria were included in the study and subsequently underwent a double-blind, randomized, placebo-controlled, cross-design human application test. Importantly, the test product containing SMSP (CKDHC) was proven to alleviate hangovers through a significant reduction in the plasma concentration of acetaldehyde in the context of an alcohol-induced hangover model. In particular, from 0.5 h after SMSP intake, the blood acetaldehyde concentration (mg/L), area under the time curve (AUC; indicating the degree of bioabsorption of blood acetaldehyde), and the highest blood acetaldehyde concentration (Cmax) were reduced. Altogether, these results suggest that the test product (CKDHC) exhibits an accelerated hangover-relieving effect.

Background: The present study evaluated the clinical implications of adjuvant chemotherapy according to the mismatch repair (MMR) status and clinicopathologic features of patients with intermediate- and high-risk stage II colon cancer (CC). Methods: This study retrospectively reviewed 5,774 patients who were diagnosed with CC and underwent curative surgical resection at Kyungpook National University Chilgok Hospital. The patients were enrolled according to the following criteria: (1) pathologically diagnosed with primary CC; (2) stage II CC classified based on the 7th edition of the American Joint Committee on Cancer staging system; (3) intermediate- and high-risk features; and (4) available test results for MMR status. A total of 286 patients met these criteria and were included in the study. Results: Among the 286 patients, 54 (18.9%) were identified as microsatellite instability-high (MSI-H) or deficient MMR (dMMR). Although all the patients identified as MSI-H/dMMR showed better survival outcomes, T4 tumors and adjuvant chemotherapy were identified as independent prognostic factors for survival. For the intermediate-risk patients identified as MSI-low (MSI-L)/microsatellite stable (MSS) or proficient MMR (pMMR), adjuvant chemotherapy exhibited a significantly better disease-free survival (DFS) but had no impact on overall survival (OS). Oxaliplatin-containing regimens showed no association with DFS or OS. Adjuvant chemotherapy was not associated with DFS in intermediate-risk patients identified as MSI-H/dMMR. Conclusion: The current study found that the use of adjuvant chemotherapy was correlated with better DFS in MSI-L/MSS or pMMR intermediate-risk stage II CC patients.

This study aims to analyze the summer human thermal environment in Jeju City's outdoor parking lots by measuring microclimate data and comparing pavement and vegetation albedoes and elements through computer simulations. In measured cases, results due to albedo showed no significance, but there was a significant difference between sunny and shaded areas by trees. The sunny area had a PET (physiological equivalent temperature) in the 'very hot' level, while the shaded area exhibited a 2-step lower 'warm' level. UTCI (universal thermal climate index) also showed that the sunny area was in the 'very strong heat stress' level, whereas the shaded area was 1-step lower in the 'strong heat stress' level, confirming the role of trees in reducing incoming solar radiant energy. Simulation results, using the measured albedoes, closely resembled the measured results. Regarding vegetation, scenarios with a wide canopy, high leaf density, and narrow planting spacing were effective in mitigating the human thermal environment, and the differences due to tree height varied across scenarios. The scenario with the lowest PET value was H9W9L3D8 (tree height 9m, canopy width 9m, leaf area index 3.0, planting spacing 8m), indicating a 0.7-step decrease compared to the current landscaping scenario. Thus, it was confirmed that, among landscaping elements, trees have a significant impact on the summer human thermal environment compared to ground pavement.
This study, estimates the predicted no effect concentration (PNEC) for the protection of organisms in aquatic and soil environments, considering the mode of action of Perfluorooctanesulfonic acid (PFOS). PNECs were derived using the species sensitivity distribution (SSD) approach to estimate the hazardous concentration for 5% of species (HC5), with applying assessment factors. Chronic toxicity data on PFOS were collected through the USEPA's ECOTOX database and literature reviews, and classified by toxicity endpoints. PNECs were then derived for each of seven toxicity endpoints that met the criteria for SSD fitting. For aquatic organisms, the PNEC for PFOS, based on all available chronic toxicity data, was determined to be 0.53 ㎍/L. The PNECs for development, genetics, enzymes, growth, reproduction, population, and biochemical biomarkers were 0.28, 0.43, 0.83, 0.90, 2.17, 111.17, and 3.53 ㎍/L, respectively. The lowest PNEC was observed when the toxic endpoint was set as development, which is considered to be due to the mode of action of PFOS, known to cause developmental toxicity by disrupting the endocrine system of organisms. For soil organisms, toxicity data were insufficient to estimate PNECs for individual endpoints, so all available data were used to estimate a PNEC of 0.75 mg/kg. Estimating PNECs that consider the mode of action of contaminants is expected to reduce the likelihood of underestimating protection levels for environmental contaminants. Additionally, this study highlights the need for ecotoxicological assessments for individual toxicity endpoints of emerging contaminants, including Per- and polyfluoroalkyl substances, in soil environments.
