The Fission Yeast Gene Encoding Monothiol Glutaredoxin 5 Is Regulated by Nitrosative and Osmotic Stresses

  • Glutaredoxin (Grx) is a small, heat-stable redox protein acting as a multi-functional glutathione (GSH)-dependent disulfide oxidoreductase. We have cloned the monothiol Grx5 gene from the genomic DNA of the fission yeast Schizosaccharomyces pombe. It has 1,904 bp, with one intron, and encodes a putative protein of 146 amino acids with a molecular mass of 16.5 kDa. Recombinant Grx5 produced functional Grx in S. pombe cells. NO-generating sodium nitroprusside (SNP, 1.0 and 2.0 mM) and potassium chloride (KCl, 0.2 and 0.5 M) increased the synthesis of ${\beta}$-galactosidase from a Grx5-lacZ fusion gene, and transcription of Grx5 was also enhanced by SNP and KCl. Synthesis of ${\beta}$-galactosidase from the Grx5-lacZ fusion was lower in Pap1-negative TP108-3C cells than in wild type KP1 cells, and when Pap1 was overproduced in KP1 cells, the level of ${\beta}$-galactosidase increased. We also found that Pap1 is involved in the induction of Grx5 by SNP and KCl. S. pombe Grx5 may play a crucial role in responses to nitrosative and osmotic stresses.

Tethered DNA shear dynamics in the flow gradient plane: application to double tethering

  • We examine the wall contact of a $3\;{\mu}m$ tethered DNA chain's free end under shear with a focus on developing schemes for double-tethering in the application of making scaffolds for molecular wires. At this scale our results are found to be highly dependent on small length scale rigidity. Chain-end-wall contact frequency, mean fractional extension deficit upon contact, and standard deviation in extension upon contact are examined for scaling with dimensionless flow strength, Wi. Predictions made using a one dimensional approximation to the Smoluchowski equation for a dumbbell and three dimensional dumbbell simulations produce extension deficit, standard deviation, and frequency scaling exponents of -1/3, -1/3, and 2/3, respectively whereas more fine-grained Kratky-Porod (KP) simulations produce scaling exponents of -0.48, -0.42, and 0.76. The contact frequency scaling of 2/3 is derived from the known results regarding cyclic dynamics Analytical scaling predictions are in agreement with those previously proposed for ${\lambda}-DNA$. [Ladoux and Doyle, 2000, Doyle et al., 2000]. Our results suggest that the differences between the dumbbell and the KP model are associated with the addition of chain discretization and the correct bending potential in the latter. These scaling results will aide future exploration in double tethering of DNA to a surface.

Distribution Characteristics of Geologic Age and Rock Type of Bedrocks at the National Wood Culture Heritage Site by GIS (GIS에 의한 국가지정 목조문화재 기반암류의 지질시대별 및 암층별 분포특성)

  • The purpose of the work was carried out to contribute the factors related to geologic realm in the disaster stability evaluation items of the national wood culture heritages. Among the total heritages, the study targets mainly include 304 cases interpreted as a rock type in the geologic map of the bedrocks with GIS interpretation. The cases show the geologic ages, geologic provinces and rock types as the following distribution characteristics. In geologic ages, they are decreasing in the orders of Jurassic, Cretaceous, Quaternary, Precambrian, Age-unknown Cambro-Ordovician Carboniferous and Tertiary. Among the ages, the former fours occupy 285 cases (93.8%) of the targets, which show most of the wood culture heritages. In geologic provinces classified into 15, they are decreasing in the orders of Daebo intrusives, alluvium, Gyeongsang supergroup, Bulgugsa intrusives, Yeongnam massif, and Gyeonggi massif which occupy of predominant distribution 271 cases (89.1%) of them. In rock types of 52, those of 6, which are Jgr, Qa, Kp, Krt+Kav+Kav1+Kav2, Kbgr and GC2, occupy total 182 cases (59.9%) showing distinctly dominant trends from the rest of 46.

Structural Damage Detection through System Identification (시스템 동정을 통한 구조물의 결함 탐지)

  • This paper presents an experimental investigation of a recently developed Kronecker Product (KP) method to determine the type, location, and intensity of structural damage from an identified state-space model of the system. Although this inverse problem appears to be highly nonlinear, the system mass, stiffness, and damping matrices are identified through a series of transformations, and with the aid of the Kronecker product, only linear operations are involved in the process. Since a state-space model can be identified directly from input-output data, an initial finite element model and/or model updating are not required. The test structure is a two-degree-of-freedom torsional system in which mass and stiffness are arbitrarily adjustable to simulate various conditions of structural damage. This simple apparatus demonstrates the capability of the damage detection method by not only identifying the location and the extent of the damage, but also differentiating the nature of the damage. The potential applicability of the KP method for structural damage identification is confirmed by laboratory test.

Effects of Green Pepper (Capsicum annuum var.) on Antioxidant Activity and Induction of Apoptosis in Human Breast Cancer Cell Lines (품종별 청고추의 항산화 효과 및 유방암 세포주에서의 세포 사멸 연구)

  • This study investigated flavonoid, total phenol, total flavonoid content, antioxidant and antiproliferative activity on human breast cancer cells (MCF-7, MDA-MB-231). Four varieties of Korean green peppers (KP: kkuri pepper, PP: phut pepper, CP: cheongyang pepper, OP: ohi pepper) and one foreign green pepper (JP: jalapeno) were used. The contents of luteolin, quercetin and apigenin, which are abundant flavonoids in green pepper, were the highest in KP. Also, the contents of total phenol, and total flavonoids were the highest in KP, followed by CP, JP, PP, and OP (KP: total phenol $13.29{\pm}0.45$ mg GAE/g D.W., total flavonoid $7.02{\pm}0.13$ mg QE/g D.W. In DPPH ABTS radical-scavenging activity, KP showed the most potent antioxidant activity. In the result of viability in human breast cancer cells, KP had the highest antiproliferative effect. These results suggest that green peppers have significant antioxidant activity and can be a possible candidate for treatment of breast cancer.

  • In this paper, we proposed a PID tuning algorithm by the fuzzy set theory to improve the performance of the PID controller. The new tuning algorithm for the PID controller has the initial value of parameter Kp, $\tau_{I}$, $\tau_{D}$. by the Ziegler-Nichols formula that uses the ultimate gain and ultimate period from a relay tuning experiment. We will get the error and the error rate of plant output corresponding to the initial value of parameter and fnd the new proportion gain(Kp) and the integral time ($\tau_{I}$) from fuzzy tuner by the error and error rate of plant oueut as a membership function of fuzzy theory. This fuzzy auto tuning algorithm for PID controller considerably reduced the overshoot and rise time as compared to any other PID controller tuning algorithms. And in real parametric uncertainty systems, it constitutes an appreciable improvement of performance. The significant property of this algorithm is shown by simulation

Growth and Characterization of Lithium Potassium Phthalate (LiKP) Single Crystals for Third Order Nonlinear Optical Applications

  • Single crystals of lithium potassium phthalate (LiKP) were successfully grown from aqueous solution by solvent evaporation technique. The grown crystals were characterized by single crystal X-ray diffraction. The lithium potassium phthalate $C_{16}\;H_{12}\;K\;Li_3\;O_{11}$ belongs to triclinic system with the following unit-cell dimensions at 298(2) K;$a=7.405(5){\AA}$;$b=9.878(5){\AA}$;$c=13.396(5){\AA}$;${\alpha}=71.778(5)^{\circ}$;${\beta}=87.300(5)^{\circ}$;${\gamma}=85.405(5)^{\circ}$; having a space group P1. Mass spectrometric analysis provides the molecular weight of the compound and possible ways of fragmentations occurs in the compound. Thermal stability of the crystal was also studied by both simultaneous TGA/DTA analyses. The UV-Vis-NIR spectrum shows a good transparency in the whole of Visible and as well as in the near IR range. Third order nonlinear optical studies have also been studied by Z-scan technique. Nonlinear absorption and nonlinear refractive index were found out and the third order bulk susceptibility of compound was also estimated. The results have been discussed in detail.

  • Noise predictive maximum likelihood(NPML) detector embeds noise prediction/whitening process in branch metric calculation of Viterbi detector and improves the reliability of branch metric computation. Therefore, PRML detector with a noise predictor achieves some performance improvement and has an advantage of low complexity. This thesis random sequences are applied to linear channel. In perpendicular magnetic recording density KP=2.5, NP(121)ML and NP(1221)ML detection system which is based on a noise predictive PR-equalized signal are evaluated by the Performance through a computing simulation. Therefore, NPML systems are implemented and are verified by VHDL.

  • This paper proposes a scheme of auto-tuning fuzzy PI controller and input voltage feed forward to control the output voltage of a three-phase Z-source inverter (ZSI). The proposed scheme adjusts the ts (Kp and Ki) in real time in order to find the most suitable Kp and Ki for PI controller and to simplify the controller design. The proposed scheme is verified the validity by experiment and co-simulation in PSIM and MATLAB/SIMULINK both load step change and input DC voltage variation in Z-source inverter, and has compared with the conventional PID control scheme. The experiment results involve of three-phase output voltage, Z-network capacitor voltage and dc-link peak voltage value. By those analysis and comparison, the availability of the proposed method in output voltage transient response quality improving has been verified. Compared with conventional PID method, the proposed method showed a more effective and robust control performance for coping with the severe disturbance conditions.

Accuracy Analysis of Predicted CODE GIM in the Korean Peninsula

  • One recent notable method for real-time elimination of ionospheric errors in geodetic applications is the Predicted Global Ionosphere Map (PGIM). This study analyzes the level of accuracy achievable when applying the PGIM provided by the Center for Orbit Determination of Europe (CODE) to the Korean Peninsula region. First, an examination of the types and lead times of PGIMs provided by the International GNSS Service (IGS) Analysis Center revealed that CODE's two-day prediction model, C2PG, is available approximately eight hours before midnight. This suggests higher real-time usability compared to the one-day prediction model, C1PG. When evaluating the accuracy of PGIM by assuming the final output of the Global Ionosphere Map (GIM) as a reference, it was found that on days with low solar activity, the error is within ~2 TECU, and on days with high solar activity, the error reaches ~3 TECU. A comparison of the errors introduced when using PGIM and three solar activity indices-Kp index, F10.7, and sunspot number-revealed that F10.7 exhibits a relatively high correlation coefficient compared to Kp-index and sunspot number, confirming the effectiveness of the prediction model.
