To find serotonin(5-hydroxytryptamine, 5-HT)-antagonizing activities in traditional herbal drugs, crude extracts from 66 kinds of traditional herbal drugs were randomly screened for inhibitory effects on 5-hydroxytryptophan(HTP)-induced diarrhea in mice. Intraperitoneal injection of 5-HTP(2.5 mg/kg) induced diarrhea in 92% of mice, when observed from 10 to 15 min after injection. Crude extracts(2 g/kg) from 66 kinds of traditional herbal drugs were orally pretreated for 1 h before 5-HTP injection. Of the 66 herbal drugs screened, Ephedrae Herba(麻黃), Cimicifugae Rhizoma(升麻), Anisi stellati Fructus(八角茴香), Aurantii Fructus(枳實), Polygalae Radix(遠志) showed the most potent inhibiting activities against 5-HTP(2.5 mg/kg)-induced diarrhea in mice. There are at least 3 possible mechanisms that would be responsible for the inhibitory effect of crude extracts on 5-HTP-induced diarrhea; 1) crude extract-induced inhibition of the activity of aromatic aminoacid decarboxylase catalyzing the conversion of 5-HTP to 5-HT, 2) crude extract-induced blockade of 5-HT receptor(s) in the gastrointestinal tract responsible for 5-HTP-induced diarrhea, 3) crude extract-induced inhibition of gastrointestinal activity, irrespective of 5-HT system. The exact mechanisms and molecules, responsible for the inhibitory effect of crude extracts on 5-HTP-induced diarrhea remain to be clarified.
In order to search for protein tyrosine phosphatase 1B (PTP1B) inhibitors as therapy of type 2 diabetes and obesity from Korean traditional prescriptions, we selected 58 traditional prescriptions based on a review of the Korean traditional medicine books. The hot water extracts of Korean traditional prescriptions were screened for the inhibitory activity against PTP1B. Among the tested extracts, water extracts of Jakgamhwangsinbu-tang, Seonbanghwalmyung-eum, and Takreeonjoong-tang showed relatively good inhibitory activity against PTP1B at the concentration of $30{\mu}g/ml$. Additionally, we evaluated PTP1B inhibitory effect for each herbal ingredient and composition in Jakgamhwangsinbu-tang (芍甘黃辛附湯). Of the tested ingredients from this herbal medicine, water extracts of Paeoniae Radix rubra and Rhei Rhizoma, and ethanol extracts of Paeoniae Radix alba, Rhei Rhizoma, Asiasari Radix, and Aconiti Tuber showed good PTP1B inhibitory effect. Herbal compositions composed of these active herbal ingredients exhibited significant activity for PTP1B inhibition over 70% at $7.5{\mu}g/ml$.

Trichomonicidal activity of thirty methanolic herbal extracts used in traditional medicine in Korea was evaluated. Trichomonas vaginalis was used as experimental model, and anti-Trichomonas activity was determined over cultures of the parasite in TYM Diamond medium. Six methanolic extracts such as Acanthopanacis Cortex, Agrimoniae Herba, Pulsatillae Radix, Sanguisorbae Radix, Sophorae Radix, and Torilidis Fructus showed more than 50% trichomonicidal activity at the concentration of 200 g/ml. These extracts were further fractionated into n-butanol soluble and aqueous phases. Except for Acanthopanacis Cortex, all of n-butanol soluble phases showed potent trichomonicidal activity, while none of aqueous phases exhibited trichomonicidal activity.

This study evaluated the effects of two Korean traditional plant extracts (Diospyros kaki THUNB. leaf; DK, and Nelumbo nucifera leaf; NN) on the fermentation, functional and sensory properties of herbal yogurts. Compared to control fermentation, all plant extracts increased acidification rate and reduced the time to complete fermentation (pH 4.5). Supplementation of plant extracts and storage time were found to influence the characteristics of the yogurts, contributing to increased viability of starter culture and phenolic compounds. In particular, the increase in the counts of Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus was highest (2.95 and 1.14 Log CFU/mL respectively) in DK yogurt. Furthermore, supplementation of the plant extracts significantly influenced to increase the antioxidant activity and water holding capacity and to produce volatile compounds. The higher antioxidant activity and water holding capacity were observed in NN yogurt than DK yogurt. Moreover, all of the sensory characteristics were altered by the addition of plant extracts. Addition of plant extracts increased the scores related to flavor, taste, and texture from plain yogurt without a plant extract, as a result of volatile compounds analysis. Thus, the overall preference was increased by plant extracts. Consequently, supplementation of DK and NN extracts in yogurt enhanced the antioxidant activity and physical property, moreover increased the acceptability of yogurt. These findings demonstrate the possibility of using plant extracts as a functional ingredient in the manufacture of herbal yogurt.
Enhanced formation and accumulation of advanced glycation end products (AGEs) have been implicated as a major pathogenesis process leading to diabetic complications, normal aging, atherosclerosis and Alzheimer's disease. In our ongoing project to discover novel treatments for diabetic complications from natural sources, we have investigated on the inhibitory activity of 67 ethanol extracts from 57 Korean herbal medicines against the formation of AGEs in vitro. Of these, 22 extracts were found to have a significant AGEs inhibitory activity ($IC_{50}$<50 ${\mu}g$/ml) compared with aminoguanidine ($IC_{50}$=75.98 ${\mu}g$/ml). Particularly, 6 extracts from 3 herbal medicines, Castanea crenata (flower, leaf, bark-twig), Acer tatarium subsp. ginnala (fruit) and Sapium japonicum (leaf, twig) showed (approximately 8-17 fold) stronger inhibitory activity than that of aminoguanidine.
Many medicinal plants have been used for the treatment of edema, jaundice, and gonorrhea in traditional Oriental medicine. This screening study was designed to search the positive inotropic effects of herbal extracts in beating rabbit atria. Aquarius extracts of twenty six herbs were examined in atrial mechanical dynamics such as pulse pressure and stroke volume and atrial natriuretic peptide (ANP), one of the main hormones involved in the regulation of the body fluid and blood pressure homeostasis in perfused beating rabbit atria. Sophora flavescens Ait., Rheum officinale Baill., Acorus gramineus Sol., Chelidonium majus L., Pulsatilla koreana Nakai., Reynoutria japonica Houtt., Euphorbia lathyris L., Pyrrosia lingua (Thunb.) Farwell, Poncirus trifoliata Rafin., Anemarrhena asphodeloides Bunge, Kochia scoparia Schrad. significantly increased stroke volume and pulse pressure. However, those herbal extracts were not induced ANP secretion. We clarified the eleven herbal extracts for the positive inotropic effect independent of ANP secretion in beating rabbit atria. Thus these results provide a beneficial data for the treatment of the impairment of body fluid and blood pressure in traditional Oriental medicine.
Objective: Nowadays the combined use of herbal extracts and western medicines has been prevalent, but concern about its risk is also increasing. Even though the importance of clinical trials is well recognized, there have been only a few studies on the combined use of herbal extracts and western medicines. This study was aimed to examine which herbal extracts and antiplatelets or anticoagulants are most commonly prescribed together to inpatient ischemicstroke patients and investigate the combined prescription rate. Methods: We investigated the most frequently prescribed herbal extracts from two different sources. First, we chose herbal medicine extracts from 2008 Traditional Korean Medicine Utilization Status. Then, among patients who were admitted to Kyung-Hee Oriental Medical Center diagnosed with ischemic stroke, we found patients who were administered with these herbal medicine extracts and among these patients, we investigated how many were concomitantly administered with antiplatelets or anticoagulants. Second, we chose other herbal medicine extracts that were most often prescribed within Kyung-Hee Oriental Medical Center and found patients who were administered with these herbal medicine extracts, then investigated how many of them were concomitantly administered with specific antiplatelets or anticoagulants. Results: The most commonly prescribed herbal medicine extracts among ischemic stroke patients were Ojeok-san, Bojungikgi-tang, Sochungryong-tang and Samsoeum. About 46 to 69 percent were concomitantly administered with either specific antiplatelets or anticoagulants. Conclusions: The most often prescribed herbal medicine extracts in inpatient ischemic stroke patients are Ojeok-san, Bojungikgi-tang, Sochungryong-tang, and Samsoeum. Among patients who were prescribed with Ojeok-san, Bojungikgi-tang, Sochungryong-tang, Samsoeum, patients who were concomitantly administered with either antiplatelets or anticoagulants were about 46% to 69%.

To investigate the antioxidant capacity of traditional Korean medicines, water extracts from 57 species were tested on their antioxidant activity using radical scavenging effects against ABTSㆍ+. Of which, Rhus javanica, Caesalpinia sappan, Rosa rugosa, Spatholobus suberectus, and Magnolia obovata showed strong antioxidant capacities at 10㎍ concentration. Therefore, antioxidant capacities of 5 traditional medicine extracts in the different concentration (1㎍, 5㎍, and 10㎍) were determined. The 5 traditional medicine extracts was detected in antioxidant capacity dose dependently. R. javanica was showed the highest antioxidant capacity, the antioxidant activity at 1 ㎍ of herbal extract being 0.85 mM TE. At the same time, the antiperoxidation effects of these 5 medicines were determined. Lipid peroxidation in brain homogenates induced by NADPH and ADP-Fe/sup 2+/ was strong inhibited by S, suberectus, R. rugosa, and M. obovata. Extract of C. sappan was showed the highest inhibition against lipid peroxidation compared with 4 herbal extracts in vitro. These traditional medicines are a potent antioxidant capacity and antiperoxidation activity, further investigation into the in vivo antioxidant therapeutic potential for treatment of human disorders such as Alzheimer's disease.
Water extracts of 21 traditional Korean herbal drugs were prepared, and a dose range of 100 mg/kg to 400 mg/kg was administered orally into mice once a day for five days. Changes of serum enzyme activities of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), blood urea nitrogen (BUN), alkaline phosphatase, body weight changes and histo pathological examination of various organs were investigated. Water extract of Ephedra Herba caused severe body weight loss at a dose of 100 mg/kg and death from a dose level of 200 mg/kg by oral administration. Angelica koreanae Radix and Anthrisci Radix showed a slight body weight loss and damages to liver and kidney.
Objective : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro. Methods : herbal medicines(80% ethanol or water extracts) were screened for their inhibitory activities against elastase. Results : Rheum undulatum (final concentrstion 1 mg/ml) appeared over 30% of inhibition of elastase activity. so we are investigated anti wrinkle effects of Rheum undulatum look through MMP-1 inhibition activity. also Extracts of Rheum undulatum showed higher anti tyrosinase activity than arbutin (final concentration 1 mg/ml). Conclusion : These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.
