  • In order to evaluate the availability of lacZ as a reporter gene for producing transgenic mud loach, foreign DNA, bacterial \beta-galactosidase$ gene (lacZ) was microinjected into mud loach eggs and its insertion and expression were examined. X-gal based histochemical assay, fluorimetric analysis of \beta-galactosidase$ with 4-methylumbelliferyl-$\beta$-D-galactoside (MUG) and molecular biological examination using polymerase chain reaction (PCR), dot blot, southern blot and sequence analysis of PCR products were carried out to analyze both microinjected group and non-injected controls. The results are disccussed in this paper.

An Overturn disease of Cultured Mud Loach (Misgurnus mizolepis) (미꾸라지 (Misgurnus mizolepis)의 뒤집힘병에 관한 연구)

  • In late summer of 2001, a new disease "overturn disease" occurred among mud loach (Misgurnus mizolepis) cultured in a farm Chonbuk Province. The fish exhibited a lethargic, erratic and spiral swimming at the water surface. Affected fish showed swollen abdomen with accumulation of air bubbles in the intestinal canal. The bubbles were much larger in the anterior part than in the posterior part. Hematological and histopathological examinations were done. No significant differences were found in RBC count, Ht value and Hb concentration between the diseased and the healthy fish. Gill filaments showed proximal hyperplasia of the respiratory epithelia and fusing of the gill lamella. Hemorrhage and necrosis occurred in the intestinal epithelia and within the lamina propria, and the mucosal epithelia were separated from the muscularis. Hepatocytes underwent atrophy. In the fish experimentally tied between the annal fin and the anus by a thread to confirm accumulation of air bubbles within intestine, accumulation of air bubbles was confirmed. This disease in mud loach appeared to be induced by circulatory disturbances in the intestinal wall owing to accumulation of air bubbles in the alimentary canal after fed high protein diet throughout all culturing period.

  • High Temperature-cooking conditions of cultured fishes(loach, crucian carp, bastard halibut, and jacopever) were optimized by response surface methodology(RSM), and plastein products were prepared using enzymatic hydrolysis. Four models were proposed with regard to effects of time(t), temperature(T), and water/fish meat (w/f) ratio on the amount of 0.3M TCA soluble fractions. The model coefficients were ranged from p<0.0001 for jacopever to p<0.0433 for bastared halibut. Cooking conditions for 60% hydrolysis were optimized at 1) 14$0^{\circ}C$ except for crucian carp(136$^{\circ}C$); 2) 10.08 hours(loach), 7.25 hours(crucian carp), 9.85 hours(ba-stard harlibut), and 9.37 hours(iacopever); 3) 1:1(w/f) ratio except for the crucian carp(1.1:1). When protein hydrolyzates were employed for the plastein synthesis, optimum plastein-reaction conditions were determined to be pH 9.0 with chymotrypsin for the loach and crucian carp hydrolyzates, pH 9.0 with papain for the bastard halibut hydrolyzate, and pH 11.0 with trypsin for the jacopever hydrolyzate. Plastein reaction could be performed in water at concentration up to 20%(w/f).

Induction of Tetraploid Cyprinid Loach, Misgurnus anguillicaudatus (4 배체 미꾸리(Misgurnus anguillicaudatus)의 유도)

  • A tetraploid cyprinid loach, Misgurnus anguillicaudatus was induced by applying heat shock. The efficiency of inductions changed with the time of initiation of treatment and the duration of treatment. The highest incidence of tetraploid was obtained at the treatment of $41^{\circ}C$ for 3 min, 28 min after fertilization. Ploidy level was estimated by kariological examination and flow cytometric analysis. The tetraploid cyprinid loach had 4n = 100 chromosomes, while the diploid had 2n = 50. The erythrocytic volume of the tetraploid was 1.65 times larger than that of the diploid. The DNA content of the tetraploid and diploid showed 6.547 pg/cell and 3.067 pg/cell, respectively.

Isoform-specific response of two GAPDH paralogs during bacterial challenge and metal exposure in mud loach (Misgurnus mizolepis: Cypriniformes) kidney and spleen

  • Gene expression of two glyceraldehyde-3-phosphate dehydrogenase (GAPDH) paralogs was examined during Edwardsiella tarda challenge and heavy metal exposures in mud loach (Misgurnus mizolepis; Cypriniformes) kidney and spleen. Transcription of the two mud loach GAPDH paralogs (mlGAPDH-1 and mlGAPDH-2) was significantly modulated by these stimulatory challenges in an isoform-dependent manner. Based on the real-time RT-PCR analysis, the mlGAPDH-2 transcripts were more preferentially induced by E. tarda challenge, whereas the mlGAPDH-1 transcripts were proven to show more inducibility in response to heavy metal exposure using Cd, Cu, Mn and Zn at $5{\mu}M$. Their isoform-specific response patterns were closely in accordance with the TF binding profiles in promoter and intron-1 of the two mlGAPDH isoforms, in which the mlGAPDH-2 has more binding sites for immune-related transcription factors than mlGAPDH-1 while the mlGAPDH-1 possesses exclusively metal responsive elements in its intron. Collectively, the mlGAPDHs are potentially involved in cellular pathways independent of glycolysis and the two GAPDH paralogs might undergo functional diversification or subfunctionalization at least at the transcription level.

  • On September in 2010, myxosporidian parasites were found on the fins of Chinese muddy loach, Misgurnus mizolepis, Several cysts of approximately 2 mm diameter were observed on the fins. Numerous spores were found in the cyst. The total length of the spore was 13.0 to 15.8 ${\mu}m$, and the width of spore was 6.0 to 7.5 ${\mu}m$. As the results, the myxosporidians were finally identified as Thelohanellus misgurni (Kudo, 1919) on the basis of the presence of characteristic cysts and size of spores.

MMTS, a New Subfamily of Tc1-like Transposons

  • A novel Tc1-like transposable element has been identified as a new DNA transposon in the mud loach, Misgurnus mizolepis. The M. mizolepis Tc1-like transposon (MMTS) is comprised of inverted terminal repeats and a single gene that codes Tc1-like transposase. The deduced amino acid sequence of the transposase-encoding region of MMTS transposon contains motifs including DDE motif, which was previously recognized in other Tc1-like transposons. However, putative MMTS transposase has only 34-37% identity with well-known Tc1, PPTN, and S elements at the amino acid level. In dot-hybridization analysis used to measure the copy numbers of the MMTS transposon in genomes of the mud loach, it was shown that the MMTS transposon is present at about $3.36{\times}10^4$ copies per $2{\times}10^9$ bp, and accounts for approximately 0.027% of the mud loach genome. Here, we also describe novel MMTS-like transposons from the genomes of carp-like fishes, flatfish species, and cichlid fishes, which bear conserved inverted repeats flanking an apparently intact transposase gene. Additionally, BLAST searches and phylogenetic analysis indicated that MMTS-like transposons evolved uniquely in fishes, and comprise a new subfamily of Tc1-like transposons, with only modest similarity to Drosophila melanogaster (foldback element FB4, HB2, HB1), Xenopus laevis, Xenopus tropicalis, and Anopheles gambiae (Frisky).

Distillers' Dried Grain as a Replacement for Plant-derived Sources in the Diet of Juvenile Muddy Loach Misgurnus anguillicaudatus (배합사료 내 식물성 원료 대체원으로 막걸리 부산물이 미꾸리(Misgurnus anguillicaudatus) 치어의 성장 및 체조성에 미치는 영향)

  • We designed a 16-week feeding experiment to test the effects of dietary supplementation with distillers' dried grain (DDG) on the growth, feed utilization, and body composition of juvenile muddy loach Misgurnus anguillicaudatus. Seven isonitrogenous and isocaloric diets were formulated to contain 0% DDG (Control); 10%, 20%, and 30 % DDG from rice (diets RM10, RM20 and RM30); and 10%, 20%, and 30% DDG from rice and wheat flour (diets RWM10, RWM20 and RWM30), respectively. Three replicate groups of juvenile muddy loach averaging $1.5{\pm}0.10$ g were fed one of the diets to satiation twice daily. Survival of juvenile muddy loach fed the RWM20 and RWM30 diets was higher than that of the control group (P<0.05), while weight gain tended to decrease when fish were fed diets containing DDG. Weight gain of fish fed the RWM10 diet was greater than that of fish fed other diets, but the RM30 diet resulted in less weight gain than did the control (P<0.05). The feed and protein efficiency ratios of fish fed the RM10, RM20, RM30, RWM10, RWM20 and RWM30 diets were all similar to the control (P>0.05). Neither daily feed intake nor proximate composition of the whole body was affected by dietary DDG (P>0.05). The compositions of isoleucine, leucine, lysine, threonine, and valine were all elevated in fish fed the RM30, RWM10, RWM20 and RWM30 diets relative to those of other groups (P<0.05). In contrast, methionine + cystine and phenylalanine + tyrosine were all lower in fish fed the RM30, RWM10, RWM20 and RWM30 diets, as compared to those of other groups (P<0.05). Our results suggest that DDG is suitable as a partial replacement for soybean meal and wheat flour, and could be used at a rate of up to 20% for rice, or 30% for rice and wheat flour, for optimum growth performance of juvenile muddy loach.

Toxicological effects of pesticides on loach in rice paddy (벼 재배 논 서식 미꾸리에 대한 농약의 영향)

  • A toxicological study of the pesticides for rice paddy to loach was conducted with iprobenfos 17% GR, diazinon 3 % GR, and butachlor 5 % GR to establish the field test method and their toxicological effects on loach under the actual field conditions. The type C of cage was more effective in the sense of a little stress from the cage. The nets for shading and for preventing the birds were necessary to maintain the water temperature and to prevent the predators. The cumulative mortality of loach exposed to iprobenfos 17% GR, diazinon 3% GR and butachlor 5% GR were 10, 55 and 22%, respectively, during 7 days of exposure. The averaged concentration of the pesticides in paddy water 2 days after application were 1.67, 0.22 and 0.26 mg/L, and mortalities were 7, 50 and 17%, respectively, representing the similar results with acute toxicity $(48h-LC_{50})$.

Effects of Biocontrol Agents on Weed Control, Abundance of Aquatic Animals and Insects, and Yield in Paddy Rice Fields (생물자원의 논 잡초 생물학적 방제 효과와 그에 따른 수서동물과 곤충 종수 및 벼 수량 변화)

  • Golden apple snails (Pomacea canaliculat) (GAS) are widely used for weed control in rice cultivation. However, concerns on the ecological risk of invasive GAS species are increasing. This study aimed to evaluate the overall impacts of GAS on weed control, abundance of aquatic animals and insects, and rice yield, in comparison with that from other biocontrol agents such as loach (Misgurnus mizolepis) and catfish (Silurus asotus), which are alternatives for biological weed control in rice paddy cultivation. Field experiments included five treatments; control, herbicide, GAS, loach, and catfish. During the rice growth, weed appearance and biological abundance were monitored, and at harvest, the rice yield was determined. Weed control efficiency was the highest for GAS treatment (100%), followed by that for herbicide (95.8%), loach (57.5%), and catfish treatments (31.7%). Insect abundance was considerably decreased in GAS treatment due to heavy weed removal, which affects the habitat of aquatic animals and insects. The amount of rice yield (unit: kg 10 a-1) was in the order of GAS (798.9) > loach (708.1) = herbicide (700.7) > catfish (629.4) > control (496.0). Therefore, considering the weed control efficiency, biological abudance, and rice yield, loach could be a potential alternative for biological weed control in organic rice farming. However, the activity of the biocontrol agents are susceptible to environmental conditions; therefore, further studies under different conditions are essential to confirm these findings.
