Institutional Development of the U.S. House: Reforms in Legislative Rules (미국 의회 의사규칙의 역사적 진화와 이론적 쟁점)

  • This paper describes the changes in legislative rules in the U.S. House of Representatives and discusses the roles of legislative rules in public policy making. I argue that much of reform experiences in the U.S. House provide good references by which the National Assembly of the Republic of Korea may follow for its reforms. For this, I summarize primary reforms of legislative rules in the U.S. House and discuss dynamics in power distribution between committees and party leadership. These reforms are then reviewed on the basis of political stability, protection of minority rights, and legislative deliberation.

A Nucleolar Protein, MoRRP8 Is Required for Development and Pathogenicity in the Rice Blast Fungus

  • The nucleolus is the largest, membrane-less organelle within the nucleus of eukaryotic cell that plays a critical role in rRNA transcription and assembly of ribosomes. Recently, the nucleolus has been shown to be implicated in an array of processes including the formation of signal recognition particles and response to cellular stress. Such diverse functions of nucleolus are mediated by nucleolar proteins. In this study, we characterized a gene coding a putative protein containing a nucleolar localization sequence (NoLS) in the rice blast fungus, Magnaporthe oryzae. Phylogenetic and domain analysis suggested that the protein is orthologous to Rrp8 in Saccharomyces cerevisiae. MoRRP8-GFP (translational fusion of MoRRP8 with green fluorescence protein) co-localizes with a nucleolar marker protein, MoNOP1 fused to red fluorescence protein (RFP), indicating that MoRRP8 is a nucleolar protein. Deletion of the MoRRP8 gene caused a reduction in vegetative growth and impinged largely on asexual sporulation. Although the asexual spores of DMorrp8 were morphologically indistinguishable from those of wild-type, they showed delay in germination and reduction in appressorium formation. Our pathogenicity assay revealed that the MoRRP8 is required for full virulence and growth within host plants. Taken together, these results suggest that nucleolar processes mediated by MoRRP8 is pivotal for fungal development and pathogenesis.

A Study on the Prority Channel Acquisition of Purchase Information in E-Commerce in South Korea

  • Purpose: The purpose of this paper is to investigate the consumption behavior of consumers more closely and efficient purchasing channels of domestic e-commerce buyers by specifically analyzing the relationship between demographics and channels that are used first to acquire purchasing information during e-commerce. Research design, data and methodology: Korean Media Panel Survey performs 17 cities and provinces nationwide, and 4,537 panel households (all household members aged 6 or older) are surveyed, and a household visit interview is conducted. Correspondence analysis, one of the popular multivariate techniques, is exploited to explore the association between priority channel for acquisition of information and demographics. Results: The findings show that the considered demographics are closely associated with the priority channel for acquisition of purchase information. In particular, 'searching for portal sites' are closely relevant to 'more than 5 million won', 'device & machine control assembly', 'graduate school or higher', and '40-59', while 'searching for online open market' is linked with college graduate and '20-39'. Conclusions: The substantial contribution of this work is that by analyzing the association between demographics and priority channel for acquisition of purchase information in e-commerce in South Korea, we can discern the segmentation standard factor for e-commerce market and advance the subdivided market.

Computer - Aided Korean Wood Identification (COMPUTER를 이용(利用)한 한국산(韓國産) 목재(木材)의 식별(識別)에 관(關)한 연구(硏究))

  • In order to identify an unknown wood sample native to Korea. the softwood databases(KSWCHUN; Korean SoftWood CHUN) and the hardwood databases(KHWCHUN; Korean HardWood CHUN) had been built. and the new computer searching programs(IDINEX; IDentification INformation EXpress) has been written in Turbo Pascal(V.5.0) and in Macro Assembly(V.5.0). The characters of the data were based on the 74 features of softwood and on the 148 features of hardwood which are a part of new "IAWA list of microscopic features for hardwood identification" published in 1989. For the purpose of this investigation the wood anatomical nature of 25 species of softwood(13 genera of 5 families) and of 112 species of hardwood(57 genera of 31 families) were observed under a scanning electron microscope and light microscope. and a lot of literature used. The IDINEX programs are based on edge-punched card keys. with several improvements. The maximum number of features in the IDINEX is 229. but that is fixed for a given database. Large numbers of taxa are handled efficiently and new taxa easily added. A search may be based on sequence numbers of features. Comparisons are made sequentially by feature and taxon using the entire suite of features specified to produce the list of possible matching taxa. The results are followings. (1) The databases of Korean wood and the searching programs(IDINEX) had been built. (2) The databases of Korean wood could be an information to search an unknown wood. (3) The databases would be valuable. for the new features, which were not mentioned in Korean wood up to the present. were observed in details. (4) The ultrastructures of the cell walls(warty layer) and crystals observed under a scanning electron microscope will be helpful to search an unknown wood in particular. (5) The searching process is more quick and accurate than the others. 6) We can obtain the information on the differences of a species from the other and search an unknown wood using probability. in IDINEX, (7) The IDINEX will be utilized to identify and classify an animal life, vegetable world, mineral kingdom, and so on.

Pepper Blight Disease Inhibition Metagenome Clone Screening Using Soil Metagenome Library (토양 Metagenome Library로부터 고추역병 저해 클론 탐색)

  • We have purified Phytophthora capsici alpha and beta tubulin from Escherchia coli BL21(DE3). The recombinant alpha and beta tubulins were assembled into microtubule in vitro with specific conditions. The metagenome library was isolated from soil in the Mt. Yeo-Ki, Suwon, Korea and manufactured with the method mentioned in experiment contents for in vitro screening of microtubule assembly screening. FRET effect was used for microtubule assembly inhibitor screening with metagenome library. We got 2 metagenome clones from in vitro screening, and these 2 hit clones showed P. capsici growth inhibition activity on the growing pepper plants. These results suggest that new development of potent inhibitor for pepper blight disease and new approach to prevention of pepper blight disease.

Electrochemical Characteristics of Electrode by Various Preparation Methods for Alkaline Membrane Fuel Cell (알칼리막 연료전지용 전극의 제조방법에 따른 전기화학적 특성 분석)

  • Catalyst poisoning by ionomers in membrane electrode assemblies of alkaline membrane fuel cells has been reported recently. We tried to improve the membrane electrode assembly's performance by controlling the solvent's ratio during electrode manufacturing. 4 Different mixing ratios of N-Methyl-2-pyrrolidone (NMP) and ethylene glycol (EG) gave four different cathode electrodes with platinum and Fuma-Tech ionomers. The electrode with higher EG improved polarization performance by about 36% compared to the NMP-based commercial ionomer. The dependence of the ionomer's dispersibility on the solvent seems responsible for the difference, which means that the non-uniform distribution of ionomers improves the performance of the electrode. High-frequency resistance, internal resistance corrected polarization curve, Tafel slope, mass activity, and impedance spectroscopy characterized the electrode. We can find that the existence of poor solvent improves cathode electrode performance. It seems to be the result of reduced poisoning of the catalyst according to the particle size distribution of the ionomer.

  • We performed integration and standardization of the omics data related agriculture. To do this, we requires progressed computational methods and bioinformatics infrastructures for integration, standardization, mining, and analysis. It makes easier biological knowledge to find. we potentialize registration a row and processed data in NABIC (National Agricultural Biotechnology Information Center) and its processed analysis results were offered related researchers. And we also provided various analysis pipelines, NGS analysis (Reference assembly, RNA-seq), GWAS, Microbial community analysis. In addition, the our system was carried out based on the design and build the quality assurance in management omics information system and constructed the infrastructure for utilization of omics analyze system. We carried out major improvement quality of omics information system. First is Improvement quality of registration category for omics based information. Second is data processing and development platform for web UI about related omics data. Third is development of proprietary management information for omics registration database. Forth is management and development of the statistics module producers about omics data. Last is Improvement the standard upload/ download module for Large omics Registration information.

Differential Subcellular Localization of Ribosomal Protein L7 Paralogs in Saccharomyces cerevisiae

  • In Saccharomyces cerevisiae, ribosomal protein L7, one of the ~46 ribosomal proteins of the 60S subunit, is encoded by paralogous RPL7A and RPL7B genes. The amino acid sequence identity between RPl7a and RPl7b is 97 percent; they differ by only 5 amino acid residues. Interestingly, despite the high sequence homology, Rpl7b is detected in both the cytoplasm and the nucleolus, whereas Rpl7a is detected exclusively in the cytoplasm. A site-directed mutagenesis experiment revealed that the change in the amino acid sequence of Rpl7b does not influence its subcellular localization. In addition, introns of RPL7A and RPL7B did not affect the subcellular localization of Rpl7a and Rpl7b. Remarkably, Rpl7b was detected exclusively in the cytoplasm in rpl7a knockout mutant, and overexpression of Rpl7a resulted in its accumulation in the nucleolus, indicating that the subcellular localization of Rpl7a and Rpl7b is influenced by the intracellular level of Rpl7a. Rpl7b showed a wide range of localization patterns, from exclusively cytoplasmic to exclusively nucleolar, in knockout mutants for some rRNA-processing factors, nuclear pore proteins, and large ribosomal subunit assembly factors. Rpl7a, however, was detected exclusively in the cytoplasm in these mutants. Taken together, these results suggest that although Rpl7a and Rpl7b are paralogous and functionally replaceable with each other, their precise physiological roles may not be identical.

Regulation of ADAMTS-2 by 1,25-Dihydroxyvitamin D3 in Osteoblastic Cells

  • Biosynthetic processing of fibrillar procollagens is essential for producing mature collagen monomers that polymerize into fibrils by a self-assembly process. The metalloproteinase ADAMTS-2 is the major enzyme that processes the N-propeptide of type I procollagen in the skin and also of type II and type III procollagens. Mutations in the ADAMTS-2 gene cause dermatospraxis in animals and Ehlers-Danlos syndrome VIIC in humans, both of which are characterized by the accumulation of type I pN-collagen and the formation of abnormal collagen fibrils in the skin. Despite its importance in procollagen processing, little is known about the regulation of ADAMTS-2 expression. Here, we demonstrate that ADAMTS-2 can be regulated by 1,25-dihydroxyvitamin D3, an inducer of type I procollagen synthesis. This steroid hormone induced ADAMTS-2 mRNA ${\sim}3-fold$ in MG-63 human osteosarcoma cells and MC3T3-E1 murine osteoblastic cells. This induction was dose- and time-dependent in MG-63 cells. In contrast, secreted ADAMTS-2 protein was increased only 1.4-fold with 1,25-dihydroxyvitamin D3. Finally, 1,25-dihydroxyvitamin D3 in the presence of ascorbate increased levels of secreted ADAMTS-2 1.9-fold over ascorbate treatment alone, which did not appreciably change ADAMTS-2 expression. These data indicate that the regulation of ADAMTS-2 is coupled with the synthesis of type I procollagen through 1,25-dihydroxyvitamin D3 signaling and may involve translational or posttranslational control.

DEVELOPMENT OF AN OPTIMIZATION TECHNIQUE OF A WARM SHRINK FITTING PROCESS FOR AN AUTOMOTIVE TRANSMISSION PARTS

  • A fitting process carried out in the automobile transmission assembly line is classified into three classes; heat fitting, press fitting, and their combined fitting. Heat fitting is a method that applies heat in the outer diameter of a gear to a suitable range under the tempering temperature and assembles the gear and the shaft made larger than the inner radius of the gear. Its stress depends on the yield strength of a gear. Press fitting is a method that generally squeezes gear toward that of a shaft at room temperature by a press. Another method heats warmly gear and safely squeezes it toward that of a shaft. A warm shrink fitting process for an automobile transmission part is now gradually increased, but the parts (shaft/gear) assembled by the process produced dimensional change in both outer diameter and profile of the gear so that it may cause noise and vibration between gears. In order to solve these problems, we need an analysis of a warm shrink fitting process in which design parameters such as contact pressure according to fitting interference between outer diameter of a shaft and inner diameter of a gear, fitting temperature, and profile tolerance of gear are involved. In this study, an closed form equation to predict the contact pressure and fitting load was proposed in order to develop an optimization technique of a warm shrink fitting process and verified its reliability through the experimental results measured in the field and FEM, thermal-structural coupled field analysis. Actual loads measured in the field have a good agreement with the results obtained from theoretical and finite element analysis and also the expanded amounts of the outer diameters of the gears have a good agreement with the results.
