Antioxidative and Antiaging Effects of Jeju Native Plant Extracts(II) (제주 자생 식물 추출물의 항산화 및 항노화에 관한 연구(II))

  • In the previous study, we reported the antioxidative and cellular protective effects of Jeju native plant extracts. In this study, we investigated the anti-oxidative, anti-wrinkle and whitening effects of new 37 plant extracts collected from self-growing plants in Jeju island. Their anti-oxidant activities were measured by the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging assay and reactive oxygen species(ROS) scavenging assay in $Fe^{3+}-EDTA/H_2O_2$ system. The cytoprotective properties of 37 plant extracts were assessed in the rose-bengal sensitized photohemolysis of human erythrocytes. The inhibitory effect of 37 plant extracts on tyrosinase were investigated to assess their whitening efficacy. Finally, their anti-elastase activities were measured to predict the anti-wrinkle efficacy in the human skin. The results showed that the extracts of Myrica rubra stem bark and Securinega suffruticosa have the free radical scavenging activity($FSC_{50}:\;5,\;8{\mu}g/mL$, respectively), and the extracts of Quercus acutissima leaf and Securinega suffruticosa stem bark have the prominent ROS scavenging activity($OSC_{50}:\;0.009{\mu}g/mL$). Photohemolysis of erythrocytes in the presence of rose-bengal as a sensitizer was inhibited by the extracts of Securinega suffruticosa stem bark and Salix koreensis stem(${\tau}_{50}$, 895 min, 640 min at 50 ${\mu}g/mL$, respectively. Myrica rubra stem bark extract(77.8% at 200 ${\mu}g/mL$) and Salix koreensis stem extract(76.2% at 200 ${\mu}g/mL$) also have the inhibitory effect on tyrosinase and elastase activities, respectively. These results indicated that the stem park of Myrica rubra, Securinega suffruticosa, and Camellia japonica, the stem of Salix koreensis, and the leaf of Quercus aqutissima and Camellia japonica could have e benefitial effects when they are added as ingredients in cosmetics.

Stand Water Balance and Stream Water Quality in Small Forested Watershed Yangpyong Gyeonggido (경기도(京畿道) 양평지역(陽平地域) 산림(山林) 소류역(小流域)의 수수지(水收支)와 계류수(溪流水)의 수질특성(水質特性))

  • This study was carried out to investigate the characteristics of water quality variations by stand water balance in YangPyong-Gun Gejung-Lee small forest watershed. Water quantity. pH, $Cl^-$, $NO_3{^-}$, $SO_4{^{2-}}$, $Na^+$, $NH_4{^+}$, $K^+$, $Mg^{2+}$, $Ca^{2+}$ were monitored in open rainfall for one unit storm and long-term stream water in small forest watershed from January. 1998 to December. 1999. The results were summarized as follows: The runoff rate was 46.4% in 1998 and 52.2% in 1999. The average pH values of rainfall were 4.8 to 6.2 and those of stream water were 6.4 to 7.1 in small forest watershed. Total amount of input anion and cation values (kg/ha) in rainfall were $SO_4{^{2-}}>NO_3{^-}>Ca^{2+}>NH_4{^+}>Cl^->Na^+>K^+>Mg^{2+}$ and in stream water were $NO_3{^-}>Ca^{2+}>SO_4{^{2-}}>Na^+>Cl^->K^+>Mg^{2+}>NH_4{^+}$ in the order, respectively. The dissolved $NH_4{^+}$ was stored 5.29kg/ha and output of the other contents were more flow than input in small forest watershed.

Plant Growth Promotion of Calystegia soldanella and Ischaemum anthephoroides by the Strain Penicillium citrinum KACC43900 (Penicillium citrinum KACC43900에 의한 갯메꽃과 갯쇠보리의 생장촉진활성)

  • Coastal sand-dune plants can survive very effectively with the help of various microbes, especially ecto- and endomycorrihizae. Penicillium citrinum KACC43900 was screened according to growth promoting activity on sand-dune plants Calystegia soldanella and Ischaemum anthephoroides. In this study, coastal sand-dune plants were treated with a fungal culture filtrate of endophytic fungus P. citrinum KACC43900 to confirm the promotion of plant growth. C. soldanella and I. anthephoroides were used as representative coastal sand-dune plants, and their shoot length, plant length, and dry weight were analyzed. The shoot lengths of control C. soldanella and I. anthephoroides were 8.60 cm and 8.12 cm, and those of samples treated with fungal culture filtrates of C. soldanella and I. anthephoroides for 20 days were 16.30 cm and 10.56 cm, respectively. The plant lengths of control C. soldanella and I. anthephoroides were 14.90 cm and 14.80 cm, and those of samples treated with fungal culture filtrates of C. soldanella and I. anthephoroides for 20 days were 24.06 cm and 17.06 cm, respectively. The dry weight of C. soldanella and I. anthephoroides treated with fungal culture filtrates of C. soldanella and I. anthephoroides for 20 days were 0.163 g and 0.032 g, respectively. It was shown that the growth of shoots in C. soldanella and I. anthephoroides was promoted, 89.54% and 29.60%, by culture filtrate of P. citrinum KACC43900.

Relation between ERCC1 Expression in Sputum and Survival after Cisplatin-Based Chemotherapy in Patients with Non-Small Cell Lung Cancer (비소세포 폐암환자의 객담 내 ERCC1 발현정도와 cisplatin 복합화학요법 후 치료반응)

  • Background : Excision repair cross complementing gene 1 (ERCC1) not only has a protective role against carcinogens, but plays an important role in cisplatin-resistance via the repair of cisplatin-DNA adducts. This study investigated the association between the ERCC1 expression levels in sputum and survival after cisplatin-based chemotherapy in patients with inoperable non-small cell lung cancer (NSCLC). Methods : Using the sputum collected from 67 inoperable (stage IIIa-IV) NSCLC patients treated with either taxanes (33 cases) or gemcitabine (34 cases) plus cisplatin, the relative expression levels of ERCC1 and the expression of the tumor specific antigen, MAGE, were examined by the quantitative RT-PCR and RT-PCR, respectively. The response and survival were compared with the relative level of ERCC1 or MAGE expression and the treatment modality. Results : In the sputum, ERCC1 and MAGE was detected in 74.6% and 40.2% of patients, respectively. Using the median ERCC1 level, the patients were classified as having high or low ERCC1 expression. The median overall survival (MST) was significantly longer in patients with a high ERCC1 expression level than those with a low expression level (84 weeks vs. 44 weeks respectively, P=0.017). In the taxene-based treatment group, the MST was longer than the gemcitabine group (79 weeks vs. 47 weeks, respectively, P=0.03). The levels of ERCC1 were significantly higher in patients who were MAGE-positive (P=0.003). In the MAGE-negative patients, the MST was longer in the high ERCC1 group (103 weeks vs. 43 weeks, P=0.008), but not in the MAGE-positive patients (62 weeks vs. 44 weeks, P=0.348). Conclusion : ERCC1 expression in the sputum can be a prognostic factor for survival after chemotherapy in patients with inoperable NSCLC.

Change of Antioxidant Activities in Preparing Freeze Dried Wild Vegetable Block for the Long-term Storage (장기저장을 위해 제조한 동결건조 산채 블록의 항산화 활성 변화)

  • The antioxidant activities of water extracts from wild vegetables such as Ligularia fischeri (GC), Capsicum annuum L. (GCY), Aster scaber (CNM), Petasites japonicus S. et Z. Max (MYD), Ipomoea batatas L. (Lam) (GGM) were evaluated and compared with water extracts from freeze dried block. The antioxidant properties of water extracts from wild vegetables and their freeze dried block were evaluated using different antioxidant tests; 2.2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging, hydroxyl radical scavenging and nitrite scavenging activities. The water extracts from wild vegetables were found to have a higher total phenolic content than water extracts from freeze dried block. Total phenolic contents of water extracts from GC, GCY, CNM, MYD, and GGM were $471.66{\pm}3.52\;{\mu}g/mg,\;141.33{\pm}2.51\;{\mu}g/mg,\;177.33{\pm}2.88\;{\mu}g/mg,\;238.66{\pm}9.50\;{\mu}g/mg\;and\;122.67{\pm}3.51\;{\mu}g/mg$, respectively. At the concentrations of 1000 ppm, water extracts from GC, GCY, CNM, and GGM showed higher activities than water extracts from their freeze dried block on DPPH radical scavenger activity. The activity of water extracts from CNM, GC, GCY, MYD, and GGM were 90.9%, 89.9%, 76.6%, 71.1%, and 57.4%, respectively. When 10000 ppm of GC, GCY, CNM, MYD, and GGM water extracts tested for hydroxyl radical scavenging activity, activities were increased by 38.8%, 33.4%, 35.9%, 34.3%, and 33.8%, respectively and a similar effect was found with water extracts from GCY, CNM, and GGM freeze dried block at 10000 ppm concentration. However, the water extracts from GC and MYD was slightly more effective than freeze dried block extracts. The water extracts from wild vegetables and their freeze dried block had effective DPPH radical scavenger activity and hydroxyl radical scavenging activity at all tested concentrations. Nitrite scavenging activity of GC water extract significantly increased in a dose-dependent manner and the extract had higher nitrite scavenging activity than extracts freeze dried block extracts. We found that freeze dried block maintained antioxidant activities of the wild vegetables.

Inhibition effects of extracts and its solvent fractions isolated from Limonium tetragonum on growth of human cancer cells (염생식물 갯질경 [Limonium tetragonum]의 추출물이 암세포성장에 미치는 효과)

  • In this study, crude extracts of the halophyte Limonium tetragonum and their solvent fractions were evaluated on anticancer activity in AGS and HT-29 human cancer cells using MTT assay. Each of the crude extracts (MeOH and $CH_2Cl_2$) of Limonium tetragonum showed a significant inhibitory effect on the growth of human cancer cells. The combined crude extracts of MeOH and $CH_2Cl_2$ were partitioned between $CH_2Cl_2$ and water. The organic layer was further partitioned between 85% aq. MeOH and n-hexane, and then the aqueous layer was fractionated with n-BuOH and $H_2O$, successively. Growth inhibition effects of crude extracts and their solvent fractions from Limonium tetragonum increased in a dose-dependent manner. Among them, 85% aq. MeOH, n-hexane and n-BuOH fractions revealed very good inhibition effects on the growth of human cancer cells. These results suggest that we can isolate active compounds from Limonium tetragonum to show much more strong anticancer activity.

Effect of Temperature, Relative Humidity on Germination and Development of Powdery Mildew (Leveillula taurica) on Pepper and Its Inoculation Method (고추 흰가루병균(Leveillula taurica)의 발아특성 및 접종법)

  • Pepper powdery mildew, Leveillula taurica is one of the most important pathogens of pepper in the greenhouses and fields in Korea and is becoming a worldwide disease. These experiments were carried out to investigate the optimal germination, disease development conditions, disease incidence and effective inoculation methods of pepper powdery mildew. The incidence of powdery mildew was investigated throughout the country based on the major pepper growing areas in 2009. The average rate of infected leaves ranged from 15.3% to 81.8% in greenhouses and fields. Powdery mildew incidences were more severe in greenhouses than those of fields. The optimal temperature for conidial germination was $25^{\circ}C$ and showed the highest germination at 6 hs after treatment. The range of temperature for germination was $10^{\circ}C$ to $35^{\circ}C$. Temperature of greater than $30^{\circ}C$ and below $20^{\circ}C$ affected the abnormal germination and germ tube elongation. The optimal relative humidity for germination and germ tube elongation was 85% and germination increased as relative humidity increased. Disease development started 8 days after inoculation and showed the highest disease severity at 15 days in greenhouse and field regardless of varieties. Among three different inoculation methods tested the spore dropping method was better than touching with infected leaves and spraying with spore suspension. However, the method has limitation in mass inoculation due to the amount of time consumed.

Reduction of the bacteria from fecal contaminated diapers through washing and heating (분변오염 기저귀에서 분리한 플라스틱과 펄프의 미생물 저감화)

  • Three different methods (simple washing of plastic and pulp sample, washing after direct heating of the diapers, and the heating after washing of plastic and pulp sample) were carried out to decrease total coliforms and heterotrophic plate count (HPC) in the diaper's plastic and pulp. Plastic and pulp samples were obtained from diaper by treatment with 10% $CaCl_2$ and 4% sea salt water, dilution with 1,000 ml tap water, and draining by using sieves. Three times washing was the most appropriate for the reduction of microorganisms in plastic and pulp. By three times washing, the number of total coliforms in the plastic and pulp samples showed 92.8% and 99.8% of decrease, respectively, and the number of HPC showed 97.3% of decrease in the plastic and 98.5% of decrease in the pulp. The washing after direct heating of the fecal contaminated diapers was not effective because HPC in the plastic and pulp samples were still detected about 2-3 log CFU/g in the plastic and 1-2 log CFU/g in the pulp, respectively, even after heating at $60^{\circ}C$, $80^{\circ}C$, $100^{\circ}C$ for 12 h. Meanwhile, total coliforms and HPC were completely sterilized at $80^{\circ}C$ for 4 h by heating after washing of plastic and pulp samples, suggesting that this method was the most appropriate method for the reduction of microorganisms in plastic and pulp obtained from fecal contaminated diapers.

The Seasonal Pattern Assessment Questionnaire in Relation to the Temperament and Character Inventory of Personality in Korean High School Students (SPAQ에 의한 계절성과 TCI에 의해 평가된 기질과 성격과의 관련성 - 한국 고등학생을 대상으로 -)

  • Objectives : Recently, many authors investigated the relation of temperament and character to seasonality and the genetic factors in seasonality and personality. In this study, we investigated the relation between personality structure obtained from the psychobiological Temperament and Character Inventory (TCI) and self-reported seasonal variations in mood and behavior measured by the Seasonal Pattern Assessment Questionnaire (SPAQ). Methods : 600 high-school students who live in Seoul of Korea, were participated in this study. Subjects were surveyed with the Korean translation of the SPAQ of Rosenthal, an instrument for investigating mood and behavioral changes with respect to the seasons of the you. Korean version of the TCI of Cloninger was also used for assessment of personality. Results : Female students were significantly higher scores on harm avoidance, reward dependence, and cooperativeness than male students who had significant higher scores on persistence and self·transcendence than the formers. GSS were showed positive correlation with harm avoidance and self-transcendence, and negative correlation with cooperativeness and self-directedness. By employing logistic regressions, the regression coefficients of SAD were significantly negative in SD, and positive in ST. Also, the regression coefficients of S-SAD were significantly negative in CO, and positive in ST. Those with GSS at least 11 were significantly higher mean ST values compared to the remaining individuals, and those with worst in winter were significantly lower mean CO values compared to the remaining individuals. Those who considered the seasonality as a problem at least to a moderate degree were associated with high HA and ST. Conclusion : We add the evidence that the seasonality is related to the temperament and character. Although there have been many reports about the seasonality in relation to the temperament and character in foreign countries, there has been very limited report from Korea before. We hope that this result would be a helpful in the clinical field in Korea.

Survey on Quality of Hospice.Palliative Care Programs in Korea (한국 호스피스.완화의료기관 실태 조사)

  • Purpose : The purpose of this study was to evaluate the present status of hospice palliative care programs in Korea as a basic database for standardization of hospice palliative care. Method : The data was collected from July to October, 2001. The instrument used for this study was the questionnaires which was consisted of the general characteristics of organization, recipient of service, manpower, contents of service, financial conditions and facilities. Sixty-four hospice palliative care programs answered the questionnaires, confirmed by telephone. Results : They were 40 hospital-based hospice palliative care programs and 24 nonmedical hospice palliative care programs. 11 Hospital-based hospice palliative programs have isolated unit or hospital affiliated free standing hospice. 6 Non-hospital hospice palliative programs have a free standing hospice. Major subjects of hospice palliative program were terminal cancer patients but patients with non-terminal illness were also included. Only 24 of 64 hospice palliative programs had all of the essential professionals : physicians, nurses, social workers, and clergies. Home hospice palliative care programs have a referral system in hospital based (89.7%) and nonmedical programs (73.7%). 24hr hospice are were provided in 26 hospital-based (65.0%) and 9nonmedical programs (37.5%). There were rooms for family in half of hospital-based programs. 73.9% of hospice palliative care programs have financial problems. 62.0% of Hospice palliative care programs need financial support from government. Conclusion : 64 Hospice palliative care programs provided hospice palliative services but had many problems in manpower, quality of care and facility. For improving the quality of terminal patients' life and promoting the cost effectiveness of health care resources, it is necessary to consider the standardization and institutionalization of hospice palliative care.
