DNA Topoisomerases I and II Inhibitory Activity of Constituents Isolated from Juglans mandshurica

  • Nine diarylheptanoids (1-9), one triterpene (10), one sesquiterpenoid (11), one naphthoquinone (12), four tetralones (13-16), one naphthalene carboxylic acid glucoside (17) and six naphthalenyl glycosides (18-23) were isolated from the roots of Juglans mandshurica Maximowicz (Juglandaceae), and their structures determined from the chemical and spectral data. Here, we report the inhibitory effects, on the DNA topoisomerases I and II activities, of all these compounds. Compounds 10 and 23 showed more potent inhibitory effects, on the DNA topoisomerases I and II (94.0 and 86.0% inhibitions at the concentration of 5 $\mu$ g/mL, respectively), than the positive control compounds, camptothecin and etoposide.

Anti-Oxidative and Anti-Neuroinflammatory Effect of Ethanol Extracts from Walnuts's (Juglans regia L.) Shell

  • In this study, antioxidant and anti-neuroinflammatory of ethanol extracts from walnuts's (Juglans regia L.) shell were investigated in vitro. Radical-scavenging activities of the walnuts's shell ethanol extracts (WSE) were examined by using ABTS radicals and ${\alpha},{\alpha}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radicals assay. In the ABTS and DPPH radical scavenging activity, $RC_{50}$ of WSE were measured as 15.74 and $40.13{\mu}g/mL$, respectively. Also, to evaluate the anti-neuroinflammatory effects of WSE in lipopolysaccharide (LPS)-stimulated BV-2 microglial cells. The production of proinflammatory cytokines NO were examined by LPS in BV-2 cell. BV-2 cells activated with LPS were treated with various doses (10, 25, 50, $100{\mu}g/mL$) of WSE. Supernatants were analyzed for the production of NO using Griess reagent. WSE up to $10{\mu}g/mL$ still required to inhibit NO induced by LPS. These results showed that walnuts's (Juglans regia L.) shell can be used as an easily accessible source of natural anti-neuroinflammatory and natural antioxidants.

  • As described previously. we reported twenty two compounds including one naphthoquinone. eight diarylheptanoids, two tetralone. one sesquiterpenoid. one diarylheptanoid glucoside, two tetralone glucosides. one naphthalene carboxylic acid glucoside and six naphthalenyl glycosides were isolated from the roots of Juglans mandshurica (17-22). Here we report that al of these compounds and a known triterpene are tested for the inhibitory effects against DNA topoisomerases I and II activities. (omitted)

Secondary Metabolites with Anti-complementary Activity from the Stem Barks of Juglans mandshurica Maxim

  • Juglans mandshurica is a fast growing hard species, which is a tree in family of Juglandaceae and has a wide distribution in China, Korea and eastern Russia. Plant materials from J. mandshurica have extensively been used in folk medicines to prevent or cure gastric, esophageal, lung and cardiac cancer. As one chain of our searching for anticomplementary agents from natural sources, two epimeric ellagitannins, [2,3-O-4,4',5,5',6,6',-hexahydroxydiphenoyl (HHDP))-(${\alpha},{\beta}$)-D-glucose] (I) and pedunculagin (II) were purified from 70% acetone extracts of the stem barks of J. mandshurica by Thin Layer Chromatography and Sephadex LH-20 column chromatography approaches. The chemical structures of the isolated compounds were characterized by MS, NMR, and a careful comparation with published literatures. The epimeric ellagitannins I and II exhibited inhibitory properties against a classical pathway of complementary system with 50 % inhibitory concentrations ($IC_{50}$) values of 65.3 and $47.7{\mu}M$, respectively, comparing with riliroside ($IC_{50}=104{\mu}M$) and rosmarinic acid ($IC_{50}=182{\mu}M$), which were used as positive controls. Thus, the work indicated both the two secondary metabolites possess excellent inhibitory activity and might be developed as potential anticomplementary chemicals.

Microstroma juglandis에 의한 호두나무 흰곰팡이병 발생 (Occurrence of Downy Leaf Spot on Juglans regia Caused by Microstroma juglandis in Korea)

  • 2011년 6월에 김천과 무안의 호두나무 재배지에서 잎에 노균병 병징과 유사한 증상이 관찰되었다. 병든 잎의 뒷면에는 다각형의 병반에 흰색의 곰팡이가 눈꽃처럼 형성되었고, 상응하는 잎의 앞면에는 퇴록 부분이 생겼다. 병세가 진전되면서 병든 조직은 붕괴되었고 나중에는 괴사되었다. 이 병의 원인균은 형태적 및 배양적 특징으로 보아 Microstroma juglandis로 동정되었다. 또한 균주의 ITS rDNA 염기서열을 GenBank에 등록된 M. juglandis의 자료와 비교하여 100% 일치함을 확인하였다. 호두나무 묘목에 병원성 검정을 실시하여 코흐의 원칙을 만족시켰다. 이 병은 지금까지 북미, 유럽, 오세아니아 그리고 서아시아(인도, 파키스탄, 터키)에서 보고되었다. 이 연구를 통하여 한국을 포함한 동아시아에서는 이 병을 처음으로 보고하며, 병명은 '호두나무 흰곰팡이병'으로 제안한다.

북미산 흑호두나무 Bi-Sn 용융합금 복합체의 제조와 특성 (Manufacturing and Properties of Bi-Sn Impregnated Wood Composites of Juglans nigra)

  • The metalized wood composites with natural grain of imported Juglans nigra, which was impregnated with low melting alloy were manufactured and evaluated in this study. And the proper manufacturing conditions was also investigated in this study. The low melting alloy with bismuth (Bi) and tin (Sn) which are harmless to humans, was applied to this new composites. The composites showed not only no defects of discoloration, delamination, swelling, and cracking, because of high dimensional stability and low thickness swelling, but also much improved performance such as high bending strength, high hardness, abrasion resistance, high thermal conductivity as floor materials. This study also suggested the proper impregnating condition, such as 10 minutes of the preliminary vacuum time, $186^{\circ}C$ of the heating temperature and 10 minutes of the maintaining pressure time at the pressure of $30kgf/cm^2$. This metalized wood composites showed 7 times higher density than control, great increase in bending strength from $131.8N/mm^2$ to $192.3N/mm^2$, and great increase in hardness from $18.2N/mm^2$ to $90.4N/mm^2$. The composites demonstrated not only high emissivity of 91%, high shilding effectiveness of 92.59∼99.99%, high fire resistance but also great decrease in abrasion depth, water absorption and thickness swelling.

국산 가래나무 간벌재활용을 위한 금속주입목재의 제조 및 특성 (Properties and Manufacturing of Low Melting Alloy Impregnated Wood Composites for using Domestic Thinned Logs of Juglans mandshurica)

The produced composites showed 9 times higher density for small specimen, 6.6 times for actual size sample and great increase in bending strength from 102.05N/$mm^2$ to 189.47N/$mm^2$ for small size sample and to 205.4N/$mm^2$ for actual size sample, also great increase in hardness from 15.1N/$mm^2$ to 73.38N/$mm^2$ for small size sample and 64.87N/$mm^2$ for actual size sample. And the composites demonstrated great decrease in abrasion depth and in water absorption.

Apoptosis-Inducing Activity of Galloylglucoses from Juglans mandshurica in Human Promyeloid Leukemic HL-60 Cells

  • Two galloyl monosaccharides, 1,2,6-trigalloylglucose (1, TRgG) and 1,2,3,6- tetragalloylglucose (2, TEgG), were isolated from the stem-bark of Juglans mandshurica. Two galloylglucoses showed cytotoxic effects on human promyelocytic leukemia HL-60 cells. In order to elucidate their mechanism of action, we have investigated the flow cytometric analysis after Annexin V-FITC and PI staining, caspase-3 activity, and internucleosomal DNA fragmentation in HL-60 cells. HL-60 cells treated with both compounds 1 and 2 at 150 and $100\;{\mu}M$, respectively, led to a morphological features of apoptosis, such as plasma membrane blebbing and cell shrinkage. TRgG (1) and TEgG (2) increased the percentage of $FITC^+\;and\;FITC^+PI^+$ cells in flow cytometry after Annexin V-FITC and PI staining. The increase of apoptotic cells was preceded by the activation of caspase-3 reported to play a central role in apoptotic process and inducing internucleosomal DNA fragmentation. TEgG (2) showed to have stronger apoptosis inducing activity in HL-60 cell lines as compared with TRgG (1).
