Depigmentation Activity of Barley, Unpolished Rice, Job's-tear (보리·현미·율무의 미백효과에 관한 연구)

  • Objectives: The aim of this study was to investigate the depigmentation effects of Barley, Unpolished rice, Job's-tear. Metbods: We investigated that the extracts of Barley, Unpolished rice, Job's-tear inhibit activity of tyrosinase, the enzyme which convert ts 3-(3,4-dihydroxyphenyl) alanine to dopachrom in the biosynthetic process of melanin, the UV absorbance of the extracts in the UV-A region and UV-B region was measured by UV scanning, the effect of extracts on cell viability and melanin production in cultured B16 mouse melanoma cells were measured, and cytoprotective effects of extracts on PC12 cells injured by hydrogen peroxide measured by MTT assay. Results: The extracts of Barley, Unpolished rice, Job's-tear inhibited activity of tyrosinase in low density. The Barley, Job's-tear extracts not only showed inhibitory effects on melanin production in cultured B16 mouse melanoma cells, but also exhibited cytoprotective effects on PC12 cells injured by hydrogen peroxide in low density. Unpolished rice extract showed inhibitory effect on melanin production in cultured B16 mouse melanoma cells, but did not showed cytoprotective effect Barley, Unpolished rice, Job's-tear extracts did not showed an absorbance effect in the UV-A region and UV-8 region. Conclusions: These results suggest that Barley, Unpolished rice, Job's-tear inhibit melanin biosynthesis which is involved in hyperpigmentation and could be used as a whitening agent.

Characteristics of Job′s tear gruel by various mixing ratio, particle size and soaking time of Job′s tear and rice flour (쌀가루 및 율무가루의 배합비율, 입자크기 및 수침시간에 따른 율무죽의 특성)

  • The Job's tear gruels were prepared by various mixing ratios, particle size and soaking times of Job's tear and rice flours, and the physical and sensory properties of these gruel samples were measured by using a Brabender visco-amylograph and sensory evaluation. Also, the changes in the pH and viscosity of the gruel samples stored for 14 days at 4$\^{C}$ were measured as the parameters of gruel quality. The results showed that the peak viscosity was decreased by increasing the ratio of Job's tear flours. The values of peak viscosity and consistency of rice and Job's tear flours increased as the particle size decreased. As the soaking time of rice and Job's tear flours increased, the initial pasting temperature decreased. In sensory characteristics, the nutty taste was increased as the Job's tear flour levels increased. Overall acceptability of Job's tear gruel was the highest in the one of rice and Job's tear flours at 50:50 ratio. During 14 days of storage, the pH and viscosity of Job's tear gruels were increased till the sixth day, and then decreased slightly.

  • Numerous investigators have studied various activities of natural products and have found that they have not only nutritional effects, but also beneficial properties to cure various diseases and to maintain good health. Job's Tear(Yul-Moo) is a grass crop that has long been used in traditional medicine and as a nourishing food. Although its mechanism of action remains unclear, Job's Tear has been reported to exhibit anti-inflammatory, stomachic, anti-allergic, and anti-spastic effects and has been used in China for the treatment of warts, rheumatism, and neuralgia. Previous results in our laboratory demonstrated that the ethanol extract and the water extract of Job's Tear exerted an immune regulatory function on mice cells in vitro. The present study was performed to investigate the ex vivo effect of Job's Tear on immune function. Seven to eight weeks old mice(Balb/c) were fed chow diet ad libitum and water extract of Job's Tear was administered orally every other day for four weeks at two different concentrations(50 and 500mg/kg B.W.). Splenocytes proliferation with mitogen stimulation with Con A and LPS was enhanced at 50 mg/kg B.W. of Job's Tear compared to those of the control group. The results of this ex vivo study showed that proliferation of splenocytes and macrophage activation were seen in the mice orally administrated 50 mg/kg B.W. of Job's Tear water extracts. In conclusion, this study suggests that Job's Tear extracts may enhance immune function by regulating splenocyte proliferation and the cytokine prodution capacity of activated macrophages in mice.

  • The present study examined the ex vivo effect of Job's tear on immune function. Seven to eight week old mice(Balb/c) were fed a chow diet ad libitum two different concentrations (50 and 500 mg/kg BW) of water extract of Job's tear were orally administ every other day for two weeks. The results indicated that macrophage activation had occurred in the mice receiving 50 mg/kg B. W. of Job's tear water extract. Overall, using a mouse model, this study demonstrated that Job's tear extract may enhance immune function by regulating the $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ and IL-10 cytokine production capacity of activated macrophages in mice. This study may suggest that supplementation of Job's tear water extracts may enhance the immune function by regulating the enhancing the cytokine production by activated macrophage ex vivo.

Proliferation of mice spenocytes and antibody production to sheep red blood cells(SRBC) using hemolytic plague forming cell assay were used to indicate the immune activity. Splenocytes proliferation of Job's Tear with mitogen stimulation such as Con A and LPS was enhanced at 50 mg/kg B.W. concentrations compared to those of control group. In case of antibody production to sheep red blood cells, the number of antibody- secreting cells was increased by administration of 50mg/kg B.W. concentration in mice immunized as a T-dependent antigen. From the present study, Job's Tear water extracts may be suggested to stimulate the mice immune response by enhancing the splenocytes proliferation and the number of plague forming cells.

Effects of Extruded Job s-tear Cereal on Lipid Metabolism in High Fat Fed Rats (압출성형된 율무 시리얼 식이가 고지방섭취 흰쥐의 지질대사에 미치는 영향)

  • Coronary heart disease (CHD) is caused by elevated blood lipids and the death rate from CHD is continuously increasing in recent years. In Korea, death rate from CHD is more than 30%. Recently, it has been reported that Job's tear decreaseds blood cholesterol level. However, most of the reports used Job's tear itself. Therefore, we have examined the effect of extruded Job's-tear cereal on blood and liver lipid levels in rats fed with a high fat diet for 5 weeks. The energy intake and body weight gain were higher in Job's tear cereal group (JFD) than in high fat fed group(HFD). Blood triglyceride concentration was significantly reduced in JFD group (p<0.05). And Atherogenic Index was decreased about 20% in JFD suggesting favorable effect of Job's-tear cereal on hyperlipidemia. Liver lipids levels were not affected by Job's-tear cereal.

  • This study was attempted to observe the effect of Job's tear diet on change of body weight and tissue in rat. The changes of body weight were higher in Job's tear non-diet group(A) and seemed to lower in Job's tear diet group(c) as expected. The weight of each organ appeared to heavy more C group than A,Bgroup in Brain, Spleen. Lungs and Kidney. Liver was heavier in Job's tear to diet one day among six day group(B) and ovary was lighter in C group, The levels of each organ TBA-value were higher in Brain, Liver of A group and showed to higher Spleen, Lungs, Ovary of C group TBA-value of Kidney was revealed to lower more C group than A,B group.

The Effects of Job's Tear by Oral Administration on Steroid Hormone Changes in Rats (율무쌀의 경구투여가 흰쥐의 지용성 Steroid Hormone 변화에 미치는 영향)

  • This study was attempted to report the effect of Does for Job's tear on steroid hormone changes in rats. Experimental object was one male rat and four female rats in one group. The experimental diet keeped pace with basal diet and Does of Job's tear. A group was basal diet and basal diet & Does for Job's tea was B group(1/day), C group(2/day). D group(3/day), F group(1 day among 3 day, 3/day), G group (1day among 5 day. 3/day) and E group was made diet & Job's tear(3/day). The reduce of weight was appeared to higher in D group and revealed to higher was F, C, B, G and A group. The changes of steroid hormone were showed to significant difference in A group and indicated to significant difference were G, F, B, C, D and E group.

Determination of the Optimal Mixture Ratio for Extrudates of Job's-tear and Wheat Flour by Mixture Design Analysis (혼합실험계획법에 의한 율무와 밀가루의 압출성형 최적 배합 조건 설정)

  • The aim of this study was to identify the optimal ingredient-mixing ratio of Job's-tear and wheat flour subjected to extrusion. A modified distance-based mixture design analysis was chosen for the experimental design. The results showed that the bending failure force, the a-value, and the water absorbtion index (WAI) value had no correlation with the mixture ratio of Job's-tear and wheat flour. However, the diameter of the extrudates, the L- and b-values, and the water solubility index (WSI) value were all found to be correlated to the mixture ratio. While the first three values were determined using a linear model, the WSI value was determined using a quadratic model. The diameter and color had an inverse correlation to the mixture ratio: the higher the ratio of Job's-tear to wheat flour, the lower the diameter and darker the color. The L- and b-values were found to change in accordance with the mixture ratio of Job's-tear to wheat flour. The lowest WSI value was obtained at a 40:60 ratio of Job's-tear to wheat.

The Quality Characteristics of Jeolpyon with Different Amounts of Job's Tears Flour (율무가루 첨가량이 절편의 품질특성에 미치는 영향)

  • The aim of this study was to determine the optimum addition amount of Job's tears flour, in place of rice flour, in the preparation of Yulmoo Jeolpyon (Job's tears rice cake). The moisture contents of the Jeolpyon samples with added Job's tears flour were 43-45%. With increasing additions of Job's tears flour, the L-value decreased, whereas the a- and b-values increased. In the mechanical evaluation of the Job's tears Jeolpyon, hardness decreased with increasing amounts of Job's tears flour. Cohesiveness, springiness, gumminess and chewiness were lower in the Jeolpyon samples with added Job's tears flour with more than 0% Job's tears flour, respectively. The adhesiveness did not differ significantly with the addition of Job's tears flour. In the sensory evaluation, overall acceptability was highest at the 20% level of Job's tear flour. In conclusion the Jeolpyon made with 20% Job's tear flour to rice flour was found to be the best formulation, in terms of its overall acceptability and textural qualities.
