버섯을 이용한 젤리 제조 및 품질특성에 관한 연구 (Study on Preparation and Quality of Jellies using Mushrooms)

  • 영지, 표고, 눈꽃동충하초 그리고 번데기동충하초를 이용한 버섯젤리의 제조와 제품의 색도, 물성 및 기호도를 비교 조사하였다. 버섯 젤리 제조를 위한 추출액의 적정혼합비율을 선발한 결과, 영지버섯 젤리는 버섯추출액 85%, 대추추출액 10%, 황기추출액 5%를 혼합했을 때, 표고버섯 젤리는 버섯추출액 80%, 대추추출액 10%, 감초추출액 5%, 오미자추출액 5%를 혼합했을 때, 눈꽃동충하초와 번데기동충하초 젤리는 버섯추출액 05%, 대추추출액 10%, 감초추출액 5%를 혼합했을 때 가장 우수하였다. 젤화제 종류별로 버섯 추출액에 대한 응고 효과는 모든 버섯에 대해 carrageenan이 가장 효과적이었다 버섯 젤리의 색도는 carrageenan 첨가량에 따라 큰 차이가 없었고, hardness, gumminess, chewiness는 증가하는 경향이었다. 관능은 젤화가 완전히 이루어지면서 hardness가 낮은 carrageenan 0.6% 첨가가 가장 우수하였다.

An Improved Total RNA Extraction Method for White Jelly Mushroom Tremella fuciformis Rich in Polysaccharides

  • An improved method for extracting high quality and quantity RNA from a jelly mushroom and a dimorphic fungus-Tremella fuciformis which is especially rich in polysaccharides, is described. RNA was extracted from T. fuciformis mycelium M1332 and its parental monokaryotic yeast-like cells Y13 and Y32. The A260/280 and A260/230 ratios were both approximately 2, and the RNA integrity number was larger than 8.9. The yields of RNA were between 108 and $213{\mu}g/g$ fresh wt. Downstream molecular applications including reverse transcriptional PCR and quantitative real-time PCR were also performed. This protocol is reliable and may be widely applicable for total RNA extraction from other jelly mushrooms or filamentous fungi rich in polysaccharides.

Current Taxonomical Status of Korean Auricularia Species

  • Auricularia (Auriculariaceae, Basidiomycota) is well-known jelly fungi, and most species are cultivated or utilized medicinally around the world. A total of 127 Auricularia specimens were collected during Korean mushroom forays by the Korea National Arboretum from 2008 to 2020. In this investigation, fungal barcode analysis and morphological examinations of 33 specimens were conducted and five Korean Auricularia species were identified. Of the species identified, A. americana, A. heimuer, and A. villosula were new to Korea, while A. auricula-judae which was previously identified in Korea, was not found. Herein, we discuss the Korean Auricularia species and provide a taxonomic key for their morphological identification.

  • Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. This study was carried out to obtain the basic information for mycelial culture conditions of Auricularia auriculajudae. According to colony diameter and mycelial density, the media for suitable mycelial growth were PDA and MCM. The optimum temperature for mycelial growth was $25{\sim}30^{\circ}C$. Carbon and nitrogen sources were mannose and malt extract, respectively. The optimum C/N ratio was in the range of 10 to 1 with 2% glucose. Other minor components for the optimal growth were thiamine-HCl and biotin as vitamins, succinic acid and lactic acid as organic acids, and $MgSO_4{\cdot}7H_2O$ and $KH_2PO_4$ as mineral salts.

Quality Characteristics of Low-salt Chicken Sausage Supplemented with a Winter Mushroom Powder

  • Chicken meat is a low-fat and high-protein food and consumption of chicken meat has been increasing globally. Various food ingredients are widely added for their specific purpose to processed chicken meat. Nonetheless, concerns about the association between high sodium intake and various diseases as well as negative perceptions of artificial additives are increasing. Therefore, in meat products, it is necessary to reduce the amount of salt and to replace artificial additives with natural ingredients. Our aim was to investigate the quality characteristics of low-salt chicken sausages manufactured with the addition of a winter mushroom powder. Sausages was manufactured with sodium pyrophosphate (0.3%) or winter mushroom powder (0%, 0.5% and 1.0%) to ground chicken breast. As a result of addition of the winter mushroom powder to low-salt chicken sausages, pH of the meat batter increased, and the proportion of jelly and melted fat exuded from sausages was reduced. The texture of sausages was softened and lipid oxidation in sausages was inhibited by the winter mushroom powder. This powder did not negatively affect the color and sensory properties of the sausages. According to the results of this study, the winter mushroom powder can serve as a natural ingredient to improve quality of low-salt chicken sausages.

Improvement of Skin Photoaging by Polysaccharide Extract Derived from Tremella fuciformis (White Jelly Mushroom)

  • Chronic ultraviolet (UV) radiation causes photoaging, which represents skin damage, disrupts skin barrier function, and promotes wrinkle formation. We investigated that the polysaccharide extract of an edible basidiomycetous white jelly mushroom, Tremella fuciformis, (TF-Glucan®) exhibited statistically photoprotective activity by inhibiting matrix metalloproteases (MMPs) and increasing collagen synthesis, and an anti-inflammatory activity by inhibiting nitric oxide and pro-inflammatory cytokines at the concentrations of less than 1000 ㎍/ml, which is not cytotoxic (p < 0.05). Additionally, TF-Glucan® increased the expression of involucrin and filaggrin to prevent the disruption of UVB-induced barrier function (p < 0.05). TF-Glucan® was assessed as a safe material by the human primary skin irritation (1, 3, 5%), human repeated insult patch test (no sensitization at 5%), 3T3 NRU phototoxicity assay (no phototoxicity, PIF < 2, MPE < 0.1), eye irritation test test by BCOP (no category, IVIS ≤ 3) and local lymph node assay (negative at 10, 25, 50%) for identifying potential skin sensitizing. These results suggest that TF-Glucan® may be useful as an anti-photoaging ingredient for developing cosmeceuticals.

국내에서 발견된 Hypoxylon truncatum sensu Miller의 형태 및 유전적 특징 (First Note of Hypoxylon truncatum sensu Miller in Korea)

  • 국내에서 처음으로 Hypoxylon truncatum을 채집하여 형태 및 유전적 특징을 관찰하였다. 채집된 균주의 형태적 특징은 문헌에 기록된 외국의 기존 균주의 특정과 일치하였으며 각기 다른 두 형태의 stromata를 지니고 있었다. 대상균주로부터 DNA를 추출한 후 ITS 부분에 대한 PCR-RFLPs를 이용하여 분석을 하였다. 그 결과, 총 6종의 제한효소로부터 유래된 절편들 중 대상균주간에는 서로 공통되는 절편이 관찰되지 않았다. 이러한 결과를 볼 때 다른 형태의 stromata를 지니고 있는 균주간에는 상당한 유전적 거리가 있다고 할 수 있다. 그러므로 다른 형태의 stromata를 지니고 있는 H. truncatum 균주는 다른 종일 것이라 사료된다. 또한 H. truncatum은 백목이 버섯의 인공재배에 있어서 아주 중요한 역할을 할 수 있으므로 본 균주를 사용하기 전에, 이 균에 대한 정확한 분류학적 개념 확립은 백목이 버섯 인공재배를 위한 정확한 균주 선발에 큰 도움이 되리라 생각된다.

잎새버섯(Grifola frondosa) 추출물 및 잎새버섯과 흰목이 (Tremella fuciformis) 혼합추출물의 급성독성시험 (Acute toxicity evaluation of maitake mushroom extract and the mixture of maitake and white jelly mushroom extract)

  • 본 연구에서는 혈당강하 소재로서 가능성이 확인된 GFPC와 잎새버섯 및 흰목이 추출 혼합물의 안전성을 평가하기 위하여 수행되었었다. 마우스에 대한 급성경구독성을 평가하기 위해 체중 kg 당 최고 5g의 각 시험물질을 마우스 위에 직접 투여하여 48시간 동안 관찰 한 결과 모든 실험군에서 사망예가 관찰되지 않아 LD50 은 5g/kg B.W. 이상으로 계산되었으며, 임상 증상이나 체중에서도 유의할만한 소견이 관찰되지 않았다. 따라서 본 실험의 GFPC와 잎새버섯 및 흰목이 추출 혼합물은 마우스에 있어서 단기 급성 경구독성이나 부작용을 유발하지 않는 안전한 식품소재로 평가되었다. 결론적으로 GFPC와 잎새버섯 및 흰목이 추출 혼합물은 급성 독성이나 부작용을 유발하지 않아 임상사용의 가능성을 제시하여 주었다.

톱밥재배에 의한 목이의 자실체 생산 (Fruit-body Production of Auricularia auricula-judae by Sawdust Cultivation)

  • Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. Present experiments were conducted to determine the possibility of artificial cultivation with oak sawdust of A. auricula-judae. The duration of mycelial growth and days of pinhead formation on oak sawdust bag (1.4 kg) were 29~32 days and 13~17 days, respectively. The yield of fresh fruit-body was 275~350 g.

Cytotoxic Effects of Extracts from Tremella fuciformis Strain FB001 on the Human Colon Adenocarcinoma Cell Line DLD-l

  • Cytotoxic effects of extracts from Tremella fuciformis strain FB001 were evaluated on the DLD-1 human colon adenocarcinoma cell line and the content of polyphenolic compounds in the extracts were analyzed. Hexane, chloroform, and ethyl acetate subfractions (experimental setting I) exhibited cytotoxic effects on the human colon adenocarcinoma DLD-1 cell line with $IC_{50}$ values of 350, 400, and 450 ppm, respectively. When T. fuciformis was extracted sequentially with ether, ethyl acetate, chloroform, and ethanol (experimental setting II), the ether extract demonstrated potent cytotoxicity with an $IC_{50}$ value of 150 ppm, followed by ethyl acetate and chloroform fractions. If the first extraction solvent was chloroform instead of ether (experimental setting III), exposure of the cell line to chloroform, ethyl acetate, and ether extracts at 1,000 ppm led to cell death. High levels of phenolic compounds were estimated for all hydrophobic extracts, which exhibited cytotoxic effects. We propose that this useful information gives additional support to our understanding of the biology and utility of this particular mushroom.
