Gintonin, a Panax ginseng-derived LPA receptor ligand, attenuates kainic acid-induced seizures and neuronal cell death in the hippocampus via anti-inflammatory and anti-oxidant activities

  • Background: Gintonin (GT), a Panax ginseng-derived lysophosphatidic acid receptor (LPAR) ligand, has positive effects in cultured or animal models for Parkinson's disease, Huntington's disease, and so on. However, the potential therapeutic value of GT in treating epilepsy has not yet been reported. Methods: Effects of GT on epileptic seizure (seizure) in kainic acid [KA, 55mg/kg, intraperitoneal (i.p.)]-induced model of mice, excitotoxic (hippocampal) cell death in KA [0.2 ㎍, intracerebroventricular (i.c.v.)]-induced model of mice, and levels of proinflammatory mediators in lipopolysaccharide (LPS)-induced BV2 cells were investigated. Results: An i.p. injection of KA into mice produced typical seizure. However, it was significantly alleviated by oral administration of GT in a dose-dependent manner. An i.c.v. injection of KA produced typical hippocampal cell death, whereas it was significantly ameliorated by administration of GT, which was related to reduced levels of neuroglial (microglia and astrocyte) activation and proinflammatory cytokines/enzymes expression as well as increased level of the Nrf2-antioxidant response via the upregulation of LPAR 1/3 in the hippocampus. However, these positive effects of GT were neutralized by an i.p. injection of Ki16425, an antagonist of LPA1-3. GT also reduced protein expression level of inducible nitric-oxide synthase, a representative proinflammatory enzyme, in LPS-induced BV2 cells. Treatment with conditioned medium clearly reduced cultured HT-22 cell death. Conclusion: Taken together, these results suggest that GT may suppress KA-induced seizures and excitotoxic events in the hippocampus through its anti-inflammatory and antioxidant activities by activating LPA signaling. Thus, GT has a therapeutic potential to treat epilepsy.

Protective Effect of Glycyrrhiza New Domestic Variety on the Acute Pancreatitis (국내 감초 신품종의 급성 췌장염 보호 효과)

  • Glycyrrhiza korshinskyi Grig. (Gk), which is a new variety of Glycyrrhiza, was developed by the National Institute of Horticultural and Herbal Science in Korea Rural Development Administration. It is improved the limitations of traditional Glycrrhiza species such as G. uralensis Fisch., G. glabra L., and G. inflata Batal. and has been reported various pharmacological effects. The aim of this study is to investigate the protective effect of Gk on the acute pancreatitis, because which has not been revealed yet. AP was induced via intraperitoneal injection of cerulein (50 ㎍/kg) hourly for 7 times in C57BL/6 mice. Gk water extract (20 or 50 mg/kg) or saline was administrated via oral gavage 1 h before the first injection of cerulein. The mice were sacrificed at 5 h after the final injection of cerulein. The inflammatory cell infiltration in pancreatic tissue was decreased by the administration of Gk water extract compared to the AP group. Also, serum amylase and lipase levels were decreased by the administration of Gk water extract compared to the AP group. These results suggest that Gk have protective effect on the acute pancreatitis.

The Study on the Analgesic Effect and its Serotonergic Mechanism of Electroacupuncture in the Rat Model of Collagen-induced Arthritis (Collagen 유발(誘發) 관절염(關節炎) 동물모델에 대(對)한 전침자극(電鍼刺戟)의 진통효과(鎭痛效果) 및 그 기전(機轉)에 관(關)한 연구(硏究) - serotonergic receptor(5-HT1, 5-HT2)와 관련(關聯)된 기전연구(機轉硏究) -)

  • Objective: To investigate the analgesic effect and its serotonergic mechanism, especially related with 5-HTI and 5-HT2 receptor, of electroacupuncture(EA) in the rat model of collagen-induced arthritis(CIA). Methods : Immunization of male Sprague-Dawley rats with bovine type II (C II) collagen emulsified in Freund's incomplete adjuvant, followed by a booster injection 14 days later, leads to development of arthritis in more than 70% of rats by 21 days postinjection. After three weeks of first immunization, EA stimulation(2 Hz, 0.07 mA, 0.3 ms) was delivered into Jogsamni($ST_{36}$) for 30 minutes. Analgesic effect was evaluated by tail flick latency(TFL). We compared the analgesic effect of EA with TFLs between pretreatment of normal saline and pretreatment of spiroxatrine (5-HT1 receptor antagonist, 1mg/kg, intraperitoneal) and spiperone (5-HT2 receptor antagonist, 1 mg/kg, intraperitoneal) in CIA. Results : 1. TFLs were gradually decreased in CIA as increasing severity of arthritis. 2. Jogsamni($ST_{36}$) EA stimulation in CIA increased TFLs and the effect lasted for 60 minutes. 3. Increased TFLs with Jogsamni($ST_{36}$) EA stimulation were inhibited by pretreatment of spiroxatrine and spiperone in CIA. Conclusions : Jogsamni($ST_{36}$) EA showed analgesic effects in CIA The analgesic effects of Jogsamni($ST_{36}$) EA were inhibited by spiroxatrine and spiperone pretreatment. These observations suggest that 5-HT1 and 5-HT2 serotonergic receptor, which involve the release of serotonin neurotransmitter, play an important roles in analgesic mechanism of EA stimulation.

Analgesic effects of eucalyptus essential oil in mice

  • Background: The use of aroma oils dates back to at least 3000 B.C., where it was applied to mummify corpses and treat the wounds of soldiers. Since the 1920s, the term "aromatherapy" has been used for fragrance therapy with essential oils. The purpose of this study was to determine whether the essential oil of Eucalyptus (EOE) affects pain pathways in various pain conditions and motor coordination. Methods: Mice were subjected to inhalation or intraperitoneal injection of EOE, and its analgesic effects were assessed by conducting formalin, thermal plantar, and acetic acid tests; the effects of EOE on motor coordination were evaluated using a rotarod test. To determine the analgesic mechanism, 5'-guanidinonaltrindole (${\kappa}$-opioid antagonist, 0.3 mg/kg), naltrindole (${\delta}$-opioid antagonist, 5 mg/kg), glibenclamide (${\delta}$-opioid antagonist, 2 mg/kg), and naloxone (${\mu}$-opioid antagonist, 4, 8, 12 mg/kg) were injected intraperitoneally. Results: EOE showed an analgesic effect against visceral pain caused by acetic acid (EOE, 45 mg/kg); however, no analgesic effect was observed against thermal nociceptive pain. Moreover, it was demonstrated that EOE did not have an effect on motor coordination. In addition, an anti-inflammatory effect was observed during the formalin test. Conclusions: EOE, which is associated with the ${\mu}$-opioid pain pathway, showed potential effects against somatic, inflammatory, and visceral pain and could be a potential therapeutic agent for pain.

The Effect of Low-Intensity Treadmill Exercise on Blood Levels of Glucose, Insulin and Lipids in Streptozotocin-Induced Diabetic Rats (저강도 treadmill 운동이 streptozotocin 유도 당뇨쥐의 혈당, 인슐린 및 지질 농도에 미치는 영향)

  • Exercise is beneficial to the diabetic patients and streptozotocin-induced diabetic rat has been used for the study of exercise effect. The purpose of this study was to establish the optimal condition of induction of hyperglycemic diabetic rat using streptozotocin and to examine the preventive effect of treadmill exercise on the diabetic rat before and after streptozotocin injection. Intraperitoneal injection of increasing amount of streptozotocin up to 40 mg/kg dose-responsively induce hyperglycemic diabetic rat and inversely reduced the blood insulin level. Body weight was also gradually reduced with the increasing amount of streptozotocin. Control and diabetic rats exercised for 4 weeks before streptozotocin injection. The exercise was performed in the treadmill for 25 minutes a day and 5 times a week with low intensity (0 degree tilt, 15 m/min velocity). Following streptozotocin injection, the blood glucose level was measured every week and the rat was sacrificed after 4 weeks to measure the concentration of insulin and blood lipids. The blood levels of glucose and insulin was significantly reduced with exercise before streptozotocin injection, while those were not changed after streptozotocin injection. The levels of blood lipids such as total cholesterol, HDL-cholesterol, LDL-cholesterol and triglyceride were close to normal control rats. From this study, researchers found the optimal condition of preparation of streptozotocin-induced hyperglycemic diabetic rat, and the mild treadmill exercise has beneficial effect on preventing hyperglycemia and hyperlipidemia. Thus, even low intensive running prevent not only diabetes but also diabetic vascular complications.

Effect of Medicinal Herb Composites on Antioxidative and Cognition-Enhancing Activities in Rats (생약복합물이 흰쥐의 체내에서 항산화 및 인지개선활성에 미치는 영향)

  • The purpose of this experiment was designed to investigate the effects of medicinal herbs (MH) extracts on dementia induced by trimethyltin chloride (TMT) in rats. Six-week-old male Sprague-Dawley rats were randomly divided into five groups; normal group (group 1), control group (group 2), MH extracts group (250, 500 mg/kg) (group 3, group 4) and positive control group (tacrine group, group 5). In the control group to induce dementia, a 2.5 mg/kg of TMT intraperitoneal injection was used for 14 days (1 per day) in the rats. In the MH extracts group 250 mg/kg and 500 mg/kg of MH extracts were medicated in an oral inoculation for 20 days (1 per day). After 30 minutes, a 2.5 mg/kg of TMT intraperitoneal injection, which causes dementia, was used for 14 days (1 per day). In the positive control group (Tacrine group) 10 mg/kg of Tacrine, the dementia treatment, was medicated in an oral inoculation. After 30 mintues, 1 mg/kg of TMT intraperitoneal injection, which causes dementia, was used for 14 days (1 per day). The present author observed the passive avoidance performance test, and memory ability test (Y maze test), the values of MDA, acetlycholinesterase (AchE) activity in the brain and antioxidant enzyme in serum. MH extracts significantly improved memory of AD model rats in the Y-maze test, and also significantly improved memory of AD model rats in the passive avoidance test. MH extracts significantly reduced AChE activity, and significantly increased the SOD level, but not catalase and MDA. From the results above, MH extracts is thought to be effective in the improvement of antioxidant enzymes and memory ability.

Effects of Ikgukwhan and Ikgukbowhawhan on the Production of Collagen and the Regeneration of Liver Cells Damaged by Bile Duct Ligation and Dimethylnitrosamine (익국환과 익국보화환의 실험적(實驗的) 간경변(肝硬變)에 대(對)한 효과(效果))

  • This study was to investigate the protective and effects of Ikgukwhan and Ikgukbowhawhan on the liver cirrhosis or fibrosis induced by prolonged bile duct ligation; a new experimental model for cirrhosis and the intraperitoneal injection of dimethylnitrosamine in the rat. The development of fibrosis or cirrhosis and its inhibition by the two prescriptions were examined by the chemical analysis of AST, ALT, and hydroxyproline. The results obtained were as follows. 1. The increase of serum asparate aminotransferase induced by bile ductligation was inhibited by the administration of Ikgukwhan and Ikgukbowhawn extract. 2. The increase of serum alanine aminotransferase induced by bile duct ligation was inhibited by the administration of Ikgukwhan and Ikgukbowhawhan extract. 3. The increase level of serum AST and ALT induced by the intraperitoneal injection of dimethylnitrosamine was inhibited by the administration of Ikgukwhan and Ikgukbowhawhan extract. 4. The increase level of hydroxyproline volume in the damaged liver tissues in the rat was decreased by the oral administration of Ikgukwhan and Ikgukbowhawhan extract. But there were no significant differences in the inhibition rate between the two experimental groups.

The Effects of Aflatoxin $B_1$Co-administrated with Antioxidant Vitamins on Lipid Contents and Fatty Acids Composition of Liver in Mice (항산화 비타민과 Aflatoxin$B_1$의 혼합 투여가 마우스간의 지질 함량 및 지방산 조성에 미치는 영향)

  • Lipid peroxidation is one of the main manifestations of oxidative damage and has been found play an important role in the toxicity and carcinogenesis of many carcinogens. This study was carried out to investigate the effects of aflatoxin B$_1$co-administrated with antioxidant vitamins on lipid contents and fatty acids components of liver in mice. For this work, vita-min C and vitamin E, the major antioxidants, were administrated with 10 mg/kg and 63.8 mg/kg respectively, through intraperitoneal(i.p) injection to male ICR mice, and 0.4 mg/kg of the AFB$_1$injected by i.p. 1hr later. The results were as follows: two fold amounts of free cholesterol, triglyceride, and total cholesterol in serum and liver of mice treated with only AFB$_1$were observed, when compared to those of mice co-administrated with antioxidant vitamins. However, the levels of phospholipids in serum and liver of mice treated with only AFB$_1$were decreased. Concerning to fatty acids composition of liver from AFB$_1$-treated mice, P/S ratio was shown more low level in cholesteryl ester, triglyceride, total cholesterol and phospholipid than those of mice co-administrated with antioxidant vitamins. In these data which provide with a reliable evidence on their antioxidantal effects to aflatoxicosis.

Studies on Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Fruiting Body of Oudemansiella radicata (민긴뿌리버섯(Oudemansiella radicata)의 자실체로부터 추출한 조다당류의 항암 및 면역 활성 효과에 관한 연구)

  • Oudemansiella radicata, edible and medicinal mushroom belonging to Agaricales of Basidiomycota, has been known to exhibit outstanding curative effects on the fungal infection and hypertension caused by high blood pressure. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr, NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity test showed that Fr. NaCl was not cytotoxic against NIH3T3 and Sarcoma 180 at the concentration of $10{\sim}1,000\;{\mu}g/ml$. Intraperitoneal injection with Fr. NaCl exhibited antitumor activity with life prolongation effect of $42.9{\sim}66.7%$ in mice inoculated with Sarcoma 180. Fr. NaCl improved the immunopotentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.4{\sim}3$ folds compared with controlled and LPS groups, respectively. Intraperitoneal injection with Fr. MeOH increased the numbers of peritoneal exudate cells and circulating leukocytes by 3.5 and 2.5 folds, respectively. Therefore, the antitumor effect exhibited on mouse Sarcoma 180 cells was likely due to immune-modulating activity of crude polysaccharides extracted from fruiting body of O. radicata.

  • Tremella fuciformis, one of edible and medicinal mushroom belonging to Tremellaceae of Basidiomycota, has been known to have a curative effect on sarcoma 180 of mice and lowering high blood pressure of human beings. Neutral salt soluble [0.9% NaCl (Fr. NaCl)], hot water soluble (Fr. HW) and methanol soluble (Fr. MeOH) substances were extracted from Tremella fuciformis. In vitro cytotoxicity tests, Fr. HW and Fr. NaCl were not cytotoxic against cancer cell lines such as NIH3T3, Sarcoma 180, and HT-29 at the concentration of $2000{\mu}g/ml$, while Fr. MeOH was cytotoxic to NIH3T3 and Sarcoma 180. Intraperitoneal injection with Fr. NaCl showed antitumor effect with life prolongation of 53% in mice inoculated with Sarcoma 180. Fr. NaCl improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by $3.0{\sim}8.1$ folds, respectively. Intraperitoneal injection with Fr. NaCl increased the numbers of peritoneal exudated cells and circulating leukocytes by 7.4 folds and 1.6 folds, respectively, than in the control group. The antitumor effect of T. fuciformis against Sarcoma 180 of mice was likely due to immunopotentiation activity.
