In silico Discovery of Genes Expressed in Liver, Kidney, Spleen and Small Intestine of Pigs

  • An in silico approach was developed to survey the genes expressed in four internal organs of pig: liver, kidney, spleen and small intestine. The major procedures of the approach included: (1) BLAST searching against GenBank "est_others" database using human cDNA sequences as queries to screen the porcine orthologous expressed sequence tags (ESTs), (2) classifying the porcine ESTs records by resources according to certain criteria and (3) analyzing data for ESTs specifically expressed in each organ. In order to do so, four Java programs were developed. Based on the ESTs available in the GenBank database, it was found that there were at least 2,100 genes expressed in these four organs, including 128 in the liver, 81 in the kidney, 780 in the spleen, and 1,423 in the small intestine respectively (a few genes co-expressed in these tissues). Gene expression patterns, such as co-expressed genes, preferentially expressed genes and basic active genes were also compared and characterized among these organs. This study provides a comprehensive model on how to use the bioinformatics approach and Genbank databases to facilitate the discovery of new genes in livestock species.

  • Department of the imperial guard is of the soldiers organization which is formed in the royal power or the absolute near by a king. Thus, department of the imperial guard absolutely is showed it is a soldiers organization for guard a king. We summary the character and the function as followed when we subdivide. 1. The main mission was the guard of the royal palace. As we know from several historical materials, the defense of the royal palace was an fundamental mission. Therefore, department of the imperial guard would take charge of the defense of the royal palace. 2. They would carry out throne of king and the royal family or a battle. As unification, they would carry out guarding for political stability and a suppress probation of internal war in regard to the security and the frequent rebellion in central and locality. 3. They carried out guarding an honored going of a king or the royal family. We know the fact that there was an honored going of the royal family or many attendants on record. When a king or the royal family go, guarding of department of the imperial guard had not only protect them but also show off the authority of the royal family. As a result, when we analysis the substance from studying the guard system of Shilla, though our guard system is imitated the foreign guard system like U.S.A, transcending space-time, the guard system can be found a lot of something common at the guard principle and a rule. Today, in looking about a political system of any countries on the world, a king and a ruler of the country represent their country international, have responsibility of protect their nation and a people internal. Therefore, the guard for a ruler absolutely must carry out at a safety situation. Moreover the guard organ, at the same time is connected directly to a welfare of the nation, so we will have to devote a constant effort.

  • In the Literatual Study on the Dysphonia, the results were as follows. 1. The causes of dysphonia are exogenous pathogenic factors,(specially cold evil)internal damage and meridian in The Yellow Emperor's Canon of Interal Medicine, since then endogenous pathogenic factors are lung-asthenia and deficiency of lung-yin etc. The main causes are disease caused by exogenous evils, general body weakness, emotional stimulation and excess of high voice rescently. 2. The pathogenesis of dysphonia originated from two factors; The first internal damages are consumption of body fluid with the formation of dryness evil resulting from the insufficienty of lung-yin and lung-collaterals damaged by heat-evil caused by deficiency of lung and kidney-yin. The second disease caused by exogenous evils is sluggishness of lung-energy caused by exogenous pathogenic factors. 3. The main relative organ are heart, lung and kidney etc. 4. The prescriptions of wind-cold symptoms are Samyoutang(三拗湯) and Hangsosan(杏蘇散), in the prescriptions of phlegm-heat symptom is Chenginyongphetang(淸咽寧肺湯), in the prescriptions of depressive syndrome due to disorder of vital energy are Sogangkitang(小降氣湯) and Shihochenggantang(柴胡淸肝湯加減), in the prescriptions of consumption of body fluid with the formation of dryness evil resulting from the insufficiency of lung-yin symptoms are Sanghangtang(桑杏湯) and Chenginguphetang(淸咽救肺湯, in the prescriptions of deficiency of lung and kidney-yin are Baekhabgokumtang(百合固金湯) and Maekmigiwhangtang(麥味地黃湯). 5. The treatment of acupunctures are used by LI-4(合谷), H-7(湧泉), Liv-3(太衝), K-3(太谿), Sp-6(三陰交), H-5(通里), GV-15(아門), CV-23(廉泉), S-40(農隆), K-6(照海), L-7(列缺), S-36(足三里) etc.

Ojeoksan's Toxicological Effects on Pregnant Rats (오적산이 실험동물의 모체에 미치는 생식독성학적 연구)

  • Purpose : The experiments was undertaken to evaluate the effects of herbal medicine, Ojeoksan, in pregnant rats Methods : Female Sprague-Dawley rats were orally administered with the Ojeoksan at dose of 5mg/kg/day for 20 days. Pregnant rats were sacrificed at 20th day of gestation, and observed internal and reproductive organs. Approximately live fetuses in the 20th day of gestation were randomly selected and fixed in 95% ethanol. Results : Maternal body weight of Ojeoksan treated group has a tendency to increase compared to that of control group. There were no significant difference in internal and reproductive organs. There were no significant changes between two groups in blood chemistry and hematological values. There were no significant changes in number of corpus luteum, implantation, live fetuses and sex ratio. But Ojeoksan administered group showed higher delivery rate, early resorption rate than the control group. Also Ojeoksan administered group showed higher implantation rate, late resorption rate than the control group. Conclusion : From these results, it can be concluded that Ojeoksan showed no toxicity effects on maternal body weight and number of live fetuses. There were no significant changes in organ weight, hematological data, reproductive organs. We need more precise study to investigate the mechanism of early or late resoption by the herbal medicines such as Ojeoksan.

Different Pathology between General and palms-and-soles hyperhidrosis in Korean Medicine and Medicine (자한(自汗)과 수족한(手足汗)에 대한 한의학 및 의학적 고찰)

  • Objectives: We noticed that hyperhidrosis can be differentiated by whether it is topical or systemic in both Korean medicine(KM) and Modern medicine(MM). Comparing between topical and systemic sweating, we will figure out similarity between KM and MM about stimuli on sweat. Methods: All research is done by finding information on text-book, article, books. Results: Hyperhidrosis is differentiated by whether it is topical or systemic in both Korean medicine(KM) and Modern medicine(MM). First, systemic sweating(SS) is affected by body temperature. In KM, Heat and Cold(plus yang deficiency) can make human sweat systemically. In MM, heat is also mentioned as stimulus. Second, topical sweating(TS) can occur on emotionally-stressed situation especially on palms-and-soles. In KM, this phenomenon is explained by heart spirit(心神) and disease transmitted by pericardium meridian(手厥陰心包經 是動病). In MM, anatomically hyperhidrosis on palms-and-soles is generated by adrenergic sympathetic nerve which is involved with stress. Third, sweating on palms-and-soles also can be generated by internal organ. In KM, hyperhidrosis on palms-and-soles is explained as illness on stomach meridian(足陽明胃經). The 70% of parasympathetic nerve is vagus nerve which is located at internal organs-usually gastrointestinal tract. In that point, stomach and parasympathetic nerve seem to be involved in hyperhidrosis on palms-and-soles. Conclusion: Hyperhidrosis is differentiated similarly by whether it is topical or systemic in both Korean medicine and Modern medicine. Conserving each perspective of KM and MM, one perspective can be useful to other by supplementing other's weak point.

Study of Single-dose Toxicity of Aconitum Kusnezoffii Reichb. Pharmacopuncture in Rats

  • Objective: This study was performed to analyze the single-dose toxicity of Aconitum kusnezoffii Reichb. pharmacopuncture (AKRP). Methods: All experiments were conducted at the Korea Testing & Research Institute (KTRI), an institute authorized to perform non-clinical studies, under the regulations of Good Laboratory Practice (GLP). Twenty (20) Sprague-Dawley rats were chosen for the pilot study. The animals were divided into four groups of five animals per group: group 1 (G1) being the control group with each animal receiving an injection of 0.3 ml of saline and groups 2, 3, and 4 (G2, G3, and G4) being the experimental groups with each animal receiving an injection of 0.1, 0.2 or 0.3 ml of AKRP, respectively. This study was conducted with the approval of the Institutional Animal Ethics Committee. Results: No deaths occurred in any of the 4 groups, and the $LD_{50}$ of AKRP administered via IV was higher than 1.77 ml/kg. Some changes in the weights of the male rates were observed between the control group and the experimental groups, but no significant differences were noted in the weights of the female rats. To check for abnormalities in organs and tissues, we stained representative sections of each specified organ with Hematoxylin & Eosin for light microscopic examination. The results showed no significant differences in any of the organs or tissues. Conclusions: The above findings suggest that Aconitum kusnezoffii Reichb. pharmacopuncture is a relatively safe treatment. Further studies on the subject should be conducted to yield more concrete evidence.

  • Objectives The purpose of this study is to research related to Taeeumjowui-Tang(Taiyintiaowei-tang) in oriental medicine. Methods "Taeeumjowui-Tang" was used as a search term. In order to see detail review, we classified all the searched study into type, method, purpose, item of evaluation, composition of herbal medication. Results We found out total twenty six studies. They were fifteen pieces of experimental studies, five clinical case studies, five case reports and one literature review according to type and method. There were fourteen thesis that experiments on efficacy and effect in the purpose of study. Only one thesis showed about stability and toxicity. There were eleven prescription of different composition and dosage. In the item of evaluation, serum biochemical analysis, cell study, body and organ weight, food consumption, body mass index, waist circumference, waist-hip circumference ratio, bioelectrical impedance analysis were used. Conclusion To improve more objective and continuous research in oriental medicine, it is needed unity about prescription and dosage, activation of clinical trial research and variation of research subject.

  • In order to study the effects of Goosunsan known clinically for their effects of treatment for cough and asthmas the study was carried out to investigate the effect of Goosunsan extract on the contractile force of the isolated guinea pig's vaviovs kinds smooth muscles and elucidate its mechanism The result were obtained as follows: 1. The isolated trachea & ileum smooth muscles of guinea pig was suspended in the organ bath with oxygenated kreb's Henselsite bicarbonate buffer solution at $37^{\circ}C$, and the developed tension by the drug was recorded with Isometric transducer (nacro F-60). The resting tension was approximately 0.5g. 2. The isolated trachea & ileum smooth muscles of guinea pig was remakably relaxed by the administration of Goosunsan. 3. The contractile response of the trachea smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 4. The contractile response of the ileum smooth muscle of the isolated guinea pig to histamine 10-4 M was remakably by Goosunsan extract. 5. The contractile response of the trachea smooth muscle of the isolated guinea pig to acetylcholine 10-4 M was remakably by Goosunsan extract. 6. The contractile response of the ileum smooth muscle of the isolated guinea pig to acetylcholine 10-4 M was remakably by Goosunsan. 7. The contractile response of the trachea smooth muscle of the isolated guinea pig to 5-hydroxytryptamine 10-4 M was remakably by Goosunsan. 8. The contractile response of the ileum smooth muscle of the isolated guinea pig to 5-hydroxytryptamine 10-4 M was remakably by Goosunsan.

Short-Term High Expression of Interferon-Alpha Modulates Progression of Type 1 Diabetes in NOD Mice

  • Type I diabetes (T1D) is an organ-specific autoimmune disease caused by the T cell-mediated destruction of the insulin-producing ${\beta}$ cells in the pancreatic islets. The onset of T1D is the consequence of a progressive destruction of islet ${\beta}$ cells mediated by an imbalance between effector $CD4^+$ T helper (Th)1 and regulatory $CD4^+$ Th2 cell function. Since interferon-alpha (IFN-${\alpha}$) has been known to modulate immune function and autoimmunity, we investigated whether administration of adenoviralmediated IFN-${\alpha}$ gene would inhibit the diabetic process in NOD mice. The development of diabetes was significantly inhibited by a single injection of adenoviral-mediated IFN-${\alpha}$ gene before 8 weeks of age. Next, we examined the hypothesis that Th2-type cytokines are associated with host protection against autoimmune diabetes, whereas Th1-type cytokines are associated with pathogenesis of T1D. The expression of IFN-${\alpha}$ induced increase of serum IL-4 and IL-6 (Th2 cytokines) levels and decrease of serum IL-12 and IFN-${\gamma}$ (Th1 cytokines) levels. Therefore, overexpression of IFN-${\alpha}$ by adenoviralmediated delivery provides modulation of pathogenic progression and protection of NOD mice from T1D.
