A positive feedback loop of heparanase/syndecan1/nerve growth factor regulates cancer pain progression

  • Background: The purpose of this research was to assess the role of heparanase (HPSE)/syndecan1 (SDC1)/nerve growth factor (NGF) on cancer pain from melanoma. Methods: The influence of HPSE on the biological function of melanoma cells and cancer pain in a mouse model was evaluated. Immunohistochemical staining was used to analyze HPSE and SDC1. HPSE, NGF, and SDC1 were detected using western blot. Inflammatory factors were detected using ELISA assay. Results: HPSE promoted melanoma cell viability, proliferation, migration, invasion, and tumor growth, as well as cancer pain, while SST0001 treatment reversed the promoting effect of HPSE. HPSE up-regulated NGF, and NGF feedback promoted HPSE. High expression of NGF reversed the inhibitory effect of HPSE down-regulation on melanoma cell phenotype deterioration, including cell viability, proliferation, migration, and invasion. SST0001 down-regulated SDC1 expression. SDC1 reversed the inhibitory effect of SST0001 on cancer pain. Conclusions: The results showed that HPSE promoted melanoma development and cancer pain by interacting with NGF/SDC1. It provides new insights to better understand the role of HPSE in melanoma and also provides a new direction for cancer pain treatment.

The Association Between Childhood Interpersonal Trauma and Psychiatric Symptom Complexity, and the Mediating Impact of Dissociation (아동기 대인관계 외상, 정신 증상의 복잡성 및 해리의 매개 효과)

  • Objective : Any traumatic event can be a risk factor, for subsequent mental disorder. However, childhood trauma, especially in interpersonal nature, is associated with later development of complex symptom patterns. This study examined the role of dissociation as a mediator between childhood trauma and symptom complexity. Methods : A pooled data of 369 psychiatric outpatients at a university-affiliated hospital was analyzed for descriptive statistics, group differences, and bivariate correlation analysis to verify a structural model. The questionnaires included the Symptom Checklist-90-Revised, the Trauma History Screen, the Dissociative Experiences Scale-Taxon, the Beck Depression Inventory, the Beck Anxiety Inventory, and the Abbreviated PTSD checklist. Results : When other trauma variables were controlled, childhood interpersonal trauma had significant correlation with symptom complexity (r=0.155, p=0.003). Among the paths analyzed, that of childhood interpersonal trauma and dissociation showed the greatest impact on symptom complexity (b=9.34, t=5.75, p<0.001). Based on the significance of the indirect impact, the results suggest a complete mediation impact of dissociation on symptom complexity. Conclusion : This study validated that childhood interpersonal trauma impacts symptom complexity, through the sequential mediating impact of dissociation. Thus, clinicians should understand childhood interpersonal trauma, dissociation, and symptom patterns in a complex and interacting mode, and develop effective pertinent treatment strategies.

Dronedarone Attenuates Ang II-Induced Myocardial Hypertrophy Through Regulating SIRT1/FOXO3/PKIA Axis

  • Background and Objectives: Long-term pathological myocardial hypertrophy (MH) seriously affects the normal function of the heart. Dronedarone was reported to attenuate left ventricular hypertrophy of mice. However, the molecular regulatory mechanism of dronedarone in MH is unclear. Methods: Angiotensin II (Ang II) was used to induce cell hypertrophy of H9C2 cells. Transverse aortic constriction (TAC) surgery was performed to establish a rat model of MH. Cell size was evaluated using crystal violet staining and rhodamine phalloidin staining. Reverse transcription quantitative polymerase chain reaction and western blot were performed to detect the mRNA and protein expressions of genes. JASPAR and luciferase activity were conducted to predict and validate interaction between forkhead box O3 (FOXO3) and protein kinase inhibitor alpha (PKIA) promoter. Results: Ang II treatment induced cell hypertrophy and inhibited sirtuin 1 (SIRT1) expression, which were reversed by dronedarone. SIRT1 overexpression or PKIA overexpression enhanced dronedarone-mediated suppression of cell hypertrophy in Ang II-induced H9C2 cells. Mechanistically, SIRT1 elevated FOXO3 expression through SIRT1- mediated deacetylation of FOXO3 and FOXO3 upregulated PKIA expression through interacting with PKIA promoter. Moreover, SIRT1 silencing compromised dronedarone-mediated suppression of cell hypertrophy, while PKIA upregulation abolished the influences of SIRT1 silencing. More importantly, dronedarone improved TAC surgery-induced MH and impairment of cardiac function of rats via affecting SIRT1/FOXO3/PKIA axis. Conclusions: Dronedarone alleviated MH through mediating SIRT1/FOXO3/PKIA axis, which provide more evidences for dronedarone against MH.

TPEG-based User Created Message Service Model Design for Terrestrial DMB (지상파DMB TPEG 기반 사용자 주도형 메시지 서비스 모델 설계)

  • The terrestrial-digital multimedia broadcasting(T-DMB) is now provided for the public service and the number of receivers reaches 15 million at the present of August 2008. The service of T-DMB itself has been succeeded in the presence. It is however remained as a supplementary role of stationed TV. It is necessary to find an additional business model as a new media to acquire surplus escaping from the deficit of the present T-DMB service. In the present paper, data service of T-DMB is proposed as candidate of such solution. Especially, TTI(Traffic and Travel Information) service is proposed as a killer application using TPEG(Transport Protocol Experts Group). At present, TPEG has not been so popular in the sense of various user satisfaction. Thus the variety of service including bi-directional service using DMB2.0 as in the sphere of Web2.0 is necessary for the service providers. In the paper, the restriction of TPEG-POI(Point of Interest) application service using the conventional mobile communication infrastructure is indicated. To overcome such restriction, TPEG-UCM(User Created Message) application service model using individual bi-directional media which is initiated and interacting by the users is proposed. In the experiment, the application program shows the usability of the proposed method so that the implementation results show its operability.

  • The service that has been recognized as both a non-productive activity and auxiliary activity of manufacturing have become the driving force of the customers' demand with the 'service' itself. The service base is expanding and evolving rapidly. It is important to look at changes in service concepts to understand service systems. Because the service system itself has a cyclic nature based on the concept of service, it can help in the study and the "how" of service by looking at changing the system according to the evolution of the service concept. The ability to organize and utilize relationships is considered to be an important factor for managers in the service economy era. However, the attention of corporate is focused on their internal capabilities and they are familiar with external resources (knowledge and competence of customers). In this case study for each type of service, we analyzed the activities of interacting service providers-consumers in service relationship, and constructed a new service system model emphasizing intangible value and long-term outcome. This study is worth re-examining the role of customers in today's service economy era and actively utilizing a new service model for business performance.

Suggestion of the Scientific Argumentation PCK Developmental Model for Preservice Earth Science Teachers through an Instructional Design Program Using Argumentation Structures (논증구조 수업설계 프로그램을 통한 예비 지구과학 교사의 과학논증 PCK 발달 모델 제안)

  • In this study, after applying the argument structure class design program for 20 preservice earth science teachers, we conducted individual in-depth interviews, analyzed the data, and derived a scientific argumentation PCK development model. The scientific argumentation PCK development model consists of three dimensions: Scientific argumentation PCK, PCK ecosystem, and reflective practice. Scientific argumentation PCK is demonstrated in the process of designing or executing classes using argumentation structures as an instructional reasoning tool. PCK ecosystem, consisting of the existing conventional PCK components, is a dimension surrounding the scientific argumentation PCK, and these two dimensions develop by interacting with each other. Reflective practice regulates each dimension and develops it in various ways by mediating the two dimensions of the scientific argumentation PCK and the PCK ecosystem. The conclusions drawn based on the results are as follows: First, preservice science teachers can demonstrate scientific argumentation PCK in the process of design and implementation of lessons using argumentation structures as a pedagogical reasoning tool. Second, it is necessary to develop the PCK for pedagogical reasoning tools such as scientific argumentation PCK in advance for the development of science teachers' PCK, since the scientific argumentation PCK can develop various components of the PCK ecosystem. Finally, it is necessary to use scientific argumentation PCK to support the preservice teacher's reflective practice, seeing that the scientific argumentation PCK promotes the development of PCK ecosystem components by inducing reflective practice.

Integrated Algorithm for Identification of Long Range Artillery Type and Impact Point Prediction With IMM Filter (IMM 필터를 이용한 장사정포의 탄종 분리 및 탄착점 예측 통합 알고리즘)

  • In this paper, we present an algorithm that identifies artillery type and rapidly predicts the impact point based on the IMM filter. The ballistic trajectory equation is used as a system model, and three models with different ballistic coefficient values are used. Acceleration was divided into three components of gravity, air resistance, and lift. And lift acceleration was added as a new state variable. The kinematic condition that the velocity vector and lift acceleration are perpendicular was used as a pseudo-measurement value. The impact point was predicted based on the state variable estimated through the IMM filter and the ballistic coefficient of the model with the highest mode probability. Instead of the commonly used Runge-Kutta numerical integration for impact point prediction, a semi-analytic method was used to predict impact point with a small amount of calculation. Finally, a state variable initialization method using the least-square method was proposed. An integrated algorithm including artillery type identification, impact point prediction and initialization was presented, and the validity of the proposed method was verified through simulation.

Exploring Science Communicators' Competencies for Public Understanding of Science (PUS): Focusing on National and International Science Communicators' Curriculums (과학대중화를 위한 과학관 해설사 역량 탐색 : 국내·외 과학관 해설사 교육과정을 중심으로)

  • Science museums are platforms of PUS (public understanding of science). The purpose of this study was to explore the science communicators' competencies critical in interacting with visitors from the comparison of their training and professional development programs nationally and internationally. The managers running science communicators' programs and communicators themselves from six different national science museums and those from five different international ones participated in the structured survey. The data from surveys were compared among respondents to draw common and specific descriptions for communicators' competencies. In addition, the experienced exemplary science communicator, Ms. Park, participated in this study and the data from her observation field notes, her own developed science communicators' manual, and interviews were used to support the result. The contextual model of learning in science museums (Falk, 2006) was used to illustrate science communicators' expertise. National managers and science communicators showed difference in their perception about science communicators' roles, difficulties, and improvement for their competencies. Internationally, the managers and science communicators showed similar perception about communicators' competencies in terms of the contextual model of learning. It is highly suggested that practice-based science communicators' training and professional development programs must be offered on the basis of interaction between experienced and novice communicators as mentors and mentees systems.

The Old Future of Christian Education : Education for Shalom - Thoughts on UNESCO 2050 - (기독교교육의 오래된 미래 : 샬롬을 위한 교육 - UNESCO 교육의 미래 2050에 대한 소고 -)

  • Purpose of study: The purpose of this study is to propose an education for biblical Shalom for the future of education in relation to UNESCO 2050. Research content and method: The education for Shalom is about experiencing Shalom in fellowship with God. Moreover, it expands that shalom into relationships with self, neighbors, the earth, and technology, and then helps achieving balance between Shalom and those mentioned above. In order to provide education for Shalom, this study presented five relational dimensions of experiencing Shalom. First, the joy of serving God and neighbors in a proper personal relationship with God is most important. Second, it is the joy of building a right community and living in it through harmonious relationships with neighbors. Third, it is the joy of living in a harmonious relationship with nature. Fourth, it is the joy of being respected for human rights that are dignified as the image of God and living while enjoying rights. Fifth, it is the joy of enjoying fair use and benefits from technological innovation without being alienated, excluded and treated unfairly, or receiving disadvantages. Based on that, a model of education for Shalom has been developed. Conclusions and Suggestions: The educational model for Shalom forms view of values, knowledge, and human nature through the Bible. It consists of learning strategies to maintain a balance between the form of knowledge and the five relational dimensions. This model has a structure that carries out education for Shalom while interacting with each other.

A Study on the Risk Control Measures of Ship's Collision (선박충돌사고 위험성 제어방안에 관한 연구)

  • The prevention of marine accidents has been a major topic in marine society for long time and various safety policies and Countermeasures have been developed and applied to prevent those accidents. In spite of these efforts, however significant marine accidents have taken place intermittently. Ship is being operated under a highly dynamic environments and many factors are related with ship's collision and those factors are interacting. So, the analysis on ship's collision causes are very important to prepare countermeasures which will ensure the safe navigation. This study analysed the ship's collision data over the past 10 years(1991-2000), which is compiled by Korea Marine Accidents Inquiry Agency. The analysis confirmed that ‘ship's collision’ is occurred most frequently and the cause is closely related with human factor. The main purpose of this study is to propose risk control countermeasures of ship's collision. For this, the structure of human factor is analysed by the questionnaire methodology. Marine experts were surveyed based on major elements that were extracted from the human factor affecting to ship's collision. FSM has been widely adopted in modeling a dynamic system which is composed of human factors. Then, the structure analysis on the causes of ship's collision using FSM are performed. This structure model could be used in understanding and verifying the procedure of real ship's collision. Furthermore it could be used as the model to prevent ship's collision and reduce marine accidents.
