Rheological Characterization of Dextran Solution (DEXTRAN 용액의 유동학적 특성)

  • Some rheological properties of subfractions for dextran in the molecular weights range from $3{\times}10^4$ to $2{\times}10^6$ was investigated at room temperature. The dependence of the viscosity on concentration, shear rate, pH & ionic strength, temperature and solvent effect was observed. From the experimental data the Mark-Houwink viscosity equation in water at $25^{\circ}C$ was determined for samples having the molecular weight ranging from $3{\times}10^4$ to $2{\times}10^6$ as$[{\eta}]=3.1{\times}10^{-3}\;Mw^{0.39}(in\;dl/g)$. The intrinsic stiffness of the dextran backbone was estimated by evaluating the 'characteristic ratio' $C_{\infty}$, which is below the 0.082. In the concentrated region, the viscosity was decreased with increasing shear rate and was exponentially decreased with raising temperature, the viscosity showed the maximum value at neutral condition. From the experimental data, it was concluded that dextran chain, linked by the ${\alpha}-1$, 6-glucosidic linkage, behaves like a flexible random coil chain in aqueous solution, dextran solutions were pseudoplastic power law fluids among the empirical models of non-Newtonian behavior. Urea was an active reagent which increases the viscosity and swells dextran while pyrididine and glycerol were inactive reagents. Also, it could be estimated that the formation of gel structure is promote to the neutral state, the molecular weight larger than $2{\times}10^5$, when electrolytic concentration is IN and Ureas is use to solvent.

Low-temperature Sintering Behavior of TiO2 Activated with CuO

  • In $TiO_2$-CuO systems, low-temperature sinterability was investigated by a conventional sintering method. Sintering temperatures were set at under $950^{\circ}C$, at which the volume diffusion is inactive. The temperatures are less than the melting point of Ag ($961^{\circ}C$), which is often used as an internal conductor in low-temperature co-fired ceramic technology. To optimize the amount of CuO dopant, various dopant contents were added. The optimum level for enhanced densification was 2 wt% CuO. Excess dopants were segregated to the grain boundaries. The segregated dopants supplied a high diffusion path, by which grain boundary diffusion improved. At lower temperatures in the solid state region, grain boundary diffusion was the principal mass transport mechanism for densification. The enhanced grain boundary diffusion, therefore, improved densification. In this regard, the results of this study prove that the sintering mechanism was the same as that of activated sintering.

The Enzymatic Properties of Actinidine from Kiwifruit

  • Activity and stability of kiwifruit actinidine was determined in various conditions of pH, salt, and temperature using N-${\alpha}$-CBZ-lysine P-nitrophenyl ester as the substrate. Actinidine activity was low below pH 6, and undetectable below pH 3. The enzyme was stable in a pH range of 6.0-8.5. At $4^{\circ}C$ the enzyme was inactive in the presence of greater than 36% vinegar and in 2 M NaCl. Actinidine at $25^{\circ}C$ was unstable in 24% vinegar but stable in up to 3 M NaCl. With regard to freeze-thaw stability, actinidine retained 85% residual activity after being frozen at $-20^{\circ}C$ for 3 days. Based on Arrenius and Lineweaver-Burk plots, actinidine became unstable at greater than $45^{\circ}C$ with only 30% residual activity remaining after 6 min. The Km, kcat, and kcat/Km values of actinidine were $56\;{\mu}M$, 67/sec, and $1.2\;{\mu}M/sec$, respectively.

  • Cold-start emissions are given great importance under the Euro-7 emission standard due to their significant impact on overall vehicle emissions. When an engine is started from a cold state, the combustion process is not yet optimized, leading to higher emissions. Hybrid vehicles, in particular, may face additional challenges, as their engine may remain inactive for extended periods, causing their catalysts to cool down and potentially become less effective in reducing emissions. In the present study, the performance of an electric heater was investigated as a means to enhance the catalyst heating during the start-up time. A simulation tool was utilized to develop a model for the gasoline exhaust aftertreatment system. The result indicates that the heater was able to increase the three-way catalyst temperature to 500℃ in 4 s using 20 kW power. In addition, the implementation of a secondary air supply resulted in reduced temperature overshoot and improved conversion efficiencies.

Strength Development Characteristics of Clay Stabilized with Electric Furnace Steel Slag (전기로 제강슬래그로 안정화된 연약점토의 강도 발현 특성)

  • This study aimed to investigate the changes in chemical components that occur when weak clay is mixed with steel slag modified with calcium oxide, and to understand the expression characteristics of compressive strength according to hydrophilicity and curing time. XRF testing, SEM imaging, vane shear strength and uniaxial compressive strength testing were conducted. Calcium (Ca) released from the steel slag increases the Ca content in clay by increasing the number of crystal particles and forming a coating layer known as calcium silicate hydrate (CaO-SiO2-H2O) through chemical reactions with SiO2 and Al2O3 components. The weak clay stabilized with steel slag is classified into an initial inactive zone where strength relatively does not increase and an activation zone where strength increases over curing time. The vane shear strength of the initial inactive area was found to be 4.4 to 18.4 kN/m2 in the state of the weight mixing ratio Rss 30% (steel slag 30% + clay 70%). In the case of the active area, the maximum uniaxial compressive strength increased to 431.8 kN/m2 after 480 hours of curing time, which increased due to the apparent adhesion strength of clay through pozzolanic reaction. Therefore, considering the strength expression characteristics of stabilized mixed clay based on the mixing ratio (Rss) during the recycling of steel slag can enhance its practicality in civil engineering sites.

  • Fertilized egg, by successive cell divisions, differentiates into different tissues and organs with various structures and functions. Different cells and tissues contain different proteins, products of selective gene expression. Not all the genes in any genomes are equally active, temporal and spatial gene expression being the general rule. Present paper attempts to review the tanscriptional mechanisms or the initiations of transcription from several angles. In some of the organisms the genes in the process of transcription or the genes in the inactive state can be seen under the light microscope. Some bands of Drosophila polytene chromosomes may exhibit a swollen or puff appearance under certain conditions. A puff, unfolded or decondensed form of chromomere, represents sets of intense transcriptional activity or RNA synthesis. The heterochromatic X chromosome whose genes remain inactive in the female mammals can be visualized as a dark staining structure called Barr body, Configuration of chromatin differs between transcribed and nontranscribed chromatin. Modification to the chromatin facilitates RNA synthesis. The movement of large polymerase molecule along the DNA would probably be facilitated if some modifications of the chromatin configuration is effected. Methylation of cytosines in CG sequences is associated with inactive genes. Methylation can play a role in determination of mammalian cells during embryogenesis. Demethylation is necessary for the gene to be expressed during development A histone modification that is also known to be correlated with transcriptional capacity of chromatin is acetylation of the lysine residues of the core histones. Chromatin containing a high level of histone acetylation is very sensitive to DNase 1. For the transcription to occur TBP must first bind to the TATA box. Another TF, TF IIB, then binds to the promoter-TBP complex, facilitating the access of RNA polymerase to the transcription initiation site. As recently as eight years ago researchers assumed that histones were irrelevant to the regulation of gene expression. Histones combine with the DNA to form nucleosome of the chromatin. Histones are vital participant in gene regulation. Histone and basal factors compete for access to TATA box. When DNA is exposed to basal factors before histones are introduced, the basal factors assemble on TATA boxes preventing the access of histones, allowing transcription to occur, for transcription to begin, activator protein at the upstream activation sequence or enhancer must interact with the tail of histone H4 at TATA box and cause the histone role particle to dissociate from the TATA box leading to partial breakup of the histone core particle and allowing the basal factors to bind to the TATA box. New concept of genomic flux in contrast to the old concept of static genome has been developed based on the powerful new molecular techniques. Genomic changes such as repetitive DNAs and transposable elements, it is assumed but not yet proved, may affect some of the developmental patterns that characterize particular cells, tissues, organs, and organisms. In the last decade or so remarkable achievement have been made in the researches of the structures and functions of TFs and the specific target sequences located in promoters or enhancers where these TFs bind. TFs have independent domains that bind DNA and that activate transcription. DNA binding domain of TFs serves to bring the protein into the right location. There are many types of DNA binding domains. Common types of motifs can be found that are responsible for binding to DNA. The motifs are usually quite short and comprise only a small part of the protein structure. Steroid receptors have domains for hormone binding, DNA binding, and activating transcription. The zinc finger motif comprises a DNA binding domain. Leucine zipper consist of a stretch of amino acids with a leucine residue in every seventh position Two proteins form a dimer because they interact by means of leucine zippers on similar α-helical domain. This positions their DNA binding basic domains for interaction with the two halves of a DNA sequence with dyad symmetry of TGACTCA, ACTGAGT.

Developing Equipment to Detect the Deterioration Status of 6.6kV Power Cables in Operation at Power Station (발전소에서 운전 중인 활성 6.6kV 전력 케이블의 고장상태를 파악하는 장치의 개발)

  • The technology to predict and prevent an accident of the cable system in power station is required. The techniques of inactivated diagnosis, partial discharge and $tan{\delta}$, have been adopted to diagnoze the operating characteristics of cables, but it is not so easy to find out problems in cables in an inactive state before the cable accident happens. In this paper, we did a research on the 6.6kV high-power cables, installed at Korean Western Power Station Co., Ltd. in order to diagonize the cables, playing a major role at the station. We have developed an equipment to measure an insulation resistance based on the temperature and current of the cable. By installing the system in a power station, we could find abnormal status for evaluation of the lifetime. In the short term, by analyzing the data, we apply the research result to the diagnosis and evaluation of the 6.6kV power cables. In the long run, however, we plan to reduce the cost of the installation and operation of cable systems at power stations.

Effect on lymphocyte subpopulations of Quil A-ISCOMs with recombinant Aujeszkay's disease virus(ADV) gp50, gIII and α-ADV protein (돼지 오제스키바이러스 재조합단백질 gp50, gIII와 α-ADV을 이용한 Quil A-ISCOMs 접종시 백혈구아군 분포율에 관한 연구)

  • An effective candidate subunit vaccine was prepared by using the immunostimulating complexs(ISCOMs) with Quil A and recombinant protein(gp50, gIII and inactive $\alpha$-ADV) Aujeszky's disease virus(ADV). The weaned pigs were twice immunized with a ADV-ISCOMs, and followed by intramuscular challenge with $1{\times}10^4$ $TCID_{50}$ ADV(strain Yangsan). The unvaccinated pigs were also challenged with same dose of ADV. At 5 days after challenge, the control pigs have developed ADV clinical signs. Whereas, the vaccinated pigs protected them from ADV-induced acute symptoms and death. Also, to identify the lymphocyte subpopulation in peripheral blood with pigs from ADV-ISCOMs vaccinated and control group, lymphocyte reacted with a panel of monoclonal antibodies which are specific to swine leukocyte surface antigens and assayed by the flow cytometry. MHC class I, CD2, CD8, N cells, CD11a, and CD45 antigen positive cells were decreased after inoculating virulent ADV Yangsan strain in control group. The data indicated that ISCOMs technique was useful in ADV subunit vaccine preparation and demonstrated the importance of gp50, gIII as a component of ADV vaccine.

Sintering and Electrical Properties of Ni-doped ZnO-Bi2O3-Sb2O3 (Ni를 첨가한 ZnO-Bi2O3-Sb2O3계의 소결과 전기적 특성)

  • The present study aims at the examination of the effects of 1 mol% NiO addition on the reaction, microstructure development, resultant electrical properties, and especially the bulk trap and interface state levels of $ZnO-Bi_2O_3-Sb_2O_3$ (Sb/Bi=0.5, 1.0, and 2.0) systems (ZBS). The samples were prepared by conventional ceramic process, and characterized by density, XRD, SEM, I-V, impedance and modulus spectroscopy (IS & MS) measurement. The sintering and electrical properties of Ni-doped ZBS (ZBSN) systems were controlled by Sb/Bi ratio. Pyrochlore ($Zn_2Bi_3Sb_3O_{14}$) was decomposed more than $100^{\circ}C$ lowered in ZBS (Sb/Bi=1.0) by Ni doping. The reproduction of pyrochlore was suppressed by the addition of Ni in ZBS. Between two polymorphs of $Zn_7Sb_2O_{12}$ spinel ($\alpha$ and $\beta$), microstructure of ZBSN (Sb/Bi=0.5) composed of a-spinel was more homogeneous than $Sb/Bi{\geq}1.0$ composed of $\beta$-spinel phase. In ZBSN, the varistor characteristics were not improved drastically (non-linear coefficient $\alpha\;=\;6{\sim}11$) and independent on microstructure according to Sb/Bi ratio. Doping of Ni to ZBS seemed to form ${V_0}^{\cdot}$ (0.33 eV) as dominant bulk defect. From IS & MS, especially the grain boundaries of Sb/Bi=0.5 systems were divided into two types, i.e. sensitive to oxygen and thus electrically active one and electrically inactive intergranular one with temperature.

Design and Implementation of Virtual and Invisible Private Disk (VIPDISK) having Secure Storage Device (보안 저장장치를 구비한 가상의 인비저블한 보안 디스크 (VIPDISK) 설계 및 구현)

  • This paper proposes a virtual and invisible private disk (VIPDISK) technology equipped with the secure storage devices. As a software based security technology, it can create hidden partitions on any data storage device which can not be identified by the windows OS, so the program running on it, does not have any evidence of the existence of the hidden storage space. Under inactive state, it maintains an unexposed secure partition which can only be activated with a matching combination of a unique digital key and a user password to open the decryption tool. In addition, VIPDISK can store data to secure storage device with real-time encryption, it is worry-free even in the case of lost or theft. Simulation results show that VIPDISK provides a much higher level of security compared to other existing schemes.
