  • This study was carried out to elucidate phylogenetic relationship of a yellow lump, Phellinus linteus by comparing the nuclear ribosomal intergenic spacer (IGS) I region with that of other genera of basidiomycetes retrieved from Genbank. IGS I region of Phellinus linteus was 730 bp long and sequence homology was conserved in the 5' region, in particular $1{\sim}280\;bp$, and decreased in the direction toward the 3' end. ITS region was widely studied in phylogenies related to basidiomycetes, but IGS region was not well understood yet. Our study indicated that IGS region can be a good tool in phylogenetic study of basidiomycetes.

  • Variation within the intergenic spacer(IGS) of the ribosomal DNA gene for twenty-two strains of E. oxysporum and its formae speciales was examined by PCR, couped with RELP analysis. The length of the amplified IGS region was about 2.6 kb in all strains except F.oxysporum f. sp. cucumer-inum from Korea and F. oxysporum f. sp. niveum. Those two strains were 2.5 kb long. Restriction digestion of IGS-RELP regions by Eco RI, NruI, HincII, SAlI, SmaI, BalIi, HindIII, XhoI and KpnI gave rise to nine IGS hapoltypes among all strains. Cluster analysis based on the presence of absence of comigrating restriction reagments show the two groups based on 44% genetic similarity. These results demonstrated that analysis of IGS showed some difference within and between F. oxysporum formae speciales.

  • The intergenic spacer (IGS) region of the ribosomal DNA of species in Fusarium section Liseola was analyzed by amplification and subsequent digestion with several restriction enzymes. The length of the amplified IGS region was about 2.6 Kb in all strains except F.moniliforme 12 Which was about 2.9 Kb. The enzymes, EcoRI, HincII, SalI, HindIII, PstI and SmaI, digested the IGS region and nine haplotypes were identified among 11 strains. In the dendrogram based on PCR-RFLP of IGS region combined the results of section Liseola in this study and section Elegans in previous study, variation in the IGS appears to offer considerable potential to resolve intraspecific relationship as well as interspecies or intersection.

Characterization of Carp (Cyprinus carpio L.) Immunoglobulin Structure

  • Serum immunoglobulins (Igs) from Israeli carp were purified using affinity chromatography. Fish were immunized with purified mouse IgG, and the specific fish antibodies were purified from the immune serum on a mouse IgG-immobilized agarose gel. Rabbit anti-Israeli carp Igs (R $\alpha$ I. carp Igs) antibodies were produced following hyperimmunization with mouse IgG specific carp antibodies. SDS-PAGE analysis under reducing condition showed that Israeli carp Igs were composed of two $\mu$-like heavy chains with about 82 and 50 kd, respectively, and one light chain with about 25 kd. On immunoblotting analysis, however, R $\alpha$ I. carp Igs failed to react with the light chain. When both protein A and protein G-purified normal carp Ig were compared with mouse IgG-specific Israeli carp Ig, no significant structural differences among them were observed. To investigate if there is any homology between other fish Ig molecules, cross-reactivity of R $\alpha$ I. carp Igs against Ig molecules from 6 different fish sera and mouse control serum was checked on immunoblotting analysis. As a result, R $\alpha$ I. carp Igs responded to Israeli carp, common carp, and tilapia Ig molecules. In flow cytometry study, however, R $\alpha$ I. carp Igs appeared to recognize 42.0%, 35.8% and <5% of Israeli carp, common carp and tilapia $Ig^+$ head kidney cells, respectively. The result suggests the heterogeneity between receptor Igs on B-like lymphocytes and soluble Igs in serum. It is crucial to obtain pure fish Igs to produce reagent antibodies as tools for the study on their specific immune responses.

Phylogenetic relationships of Arthrospira strains inferred from 16S rRNA gene and cpcBA-IGS sequences

  • $Arthrospira$ $platensis$ and $Arthrospira$ $maxima$ are species of cyanobacteria used in health foods, animal feed, food additives, and fine chemicals. This study conducted a comparison of the 16S rRNA gene and $cpcBA$-intergenic spacer ($cpcBA$-IGS) sequences in $Arthrospira$ strains from culture collections around the world. A cluster analysis divided the 10 $Arthrospira$ strains into two main genotypic clusters, designated I and II, where Group I contained $A.$ $platensis$ SAG 86.79, UTEX 2340, $A.$ $maxima$ KCTC AG30054, and SAG 49.88, while Group II contained $A.$ $platensis$ PCC 9108, NIES 39, NIES 46, and SAG 257.80. However, although $A.$ $platensis$ PCC 9223 belonged to Group II-2 based on its $cpcBA$-IGS sequence, this strain also belonged to Group I based on its 16S rRNA gene sequence. Phylogenetic analyses based on the 16S rRNA gene and $cpcBA$-IGS sequences showed no division between $A.$ $platensis$ and $A.$ $maxima$, plus the 16S rRNA gene and $cpcBA$-IGS sequence clusters did not indicate any well-defined geographical distribution, instead overlapping in a rather interesting way. Therefore, the current study supports some previous conclusions based on 16S rRNA gene and $cpcBA$-IGS sequences, which found that $Arthrospira$ taxa are monophyletic. However, when compared with 16S rRNA sequences, $cpcBA$-IGS sequences may be better suited to resolve close relationships and intraspecies variability.

Genetic and Biochemical Characterization of Monokaryotic Progeny Strains of Button Mushroom (Agaricus bisporus)

  • To promote the selection of promising monokaryotic strains of button mushroom (Agaricus bisporus) during breeding, 61 progeny strains derived from basidiospores of two different lines of dikaryotic parental strains, ASI1038 and ASI1346, were analyzed by nucleotide sequencing of the intergenic spacer I (IGS I) region in their rDNA and by extracellular enzyme assays. Nineteen different sizes of IGS I, which ranged from 1,301 to 1,348 bp, were present among twenty ASI1346-derived progeny strains, while 15 different sizes of IGS I, which ranged from 700 to 1,347 bp, were present among twenty ASI1038-derived progeny strains. Phylogenetic analysis of the IGS sequences revealed that different clades were present in both the ASI10388- and ASI1346-derived progeny strains. Plating assays of seven kinds of extracellular enzymes (${\beta}$-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease) also revealed apparent variation in the ability to produce extracellular enzymes among the 40 tested progeny strains from both parental A. bisporus strains. Overall, this study demonstrates that characterization of IGS I regions and extracellular enzymes is useful for the assessment of the substrate-degrading ability and heterogenicity of A. bisporus monokaryotic strains.

Genotypic Characterization of Cherry Witches' Broom Pathogen Taphrina wiesneri Strains (벚나무 빗자루병균 Taphrina wiesneri의 유전적 특성)

  • The ascomycetous fungus Taphrina wiesneri, the pathogen of cherry witches' broom, is highly pathogenic to Prunus yedoensis, the most widely planted cherry trees in Korea as park and roadside trees. A collection of 13 strains of the pathogen in Korea and Japan was characterized by 18S rDNA gene sequence and restriction fragment length polymorphism (RFLP) analysis. In cluster analysis based on 18S rDNA gene sequence the strains were divided into 2 clusters. In RFLP analysis of the rDNA-IGS region using HhaI, the strains were separated into four patterns, B, C, D and G, of which pattern G was new.

Molybdenum 후면전극을 통한 CIGS우선배향성의 제어 및 변환효율에 미치는 영향

  • 최근에 보고된 양질의 고효율Cu(In,Ga)Se2 (CIGS) 태양전지는 CIGS광흡수층이 강한 (220:204) 우선배향성을 갖는 것으로 알려져 있다 [1]. 이러한 CIGS우선배향성은 Se 증착압력, Na농도, 기판온도 및 Mo후면전극의 표면상태에 영향을 받는 것으로 알려져 있지만 정확한 상호관계는 아직 명확히 알려져 있지 않으며, 특히 Mo후면전극의 영향에 대해서는 체계적인 연구결과조차 극히 드문 상황이다 [2]. 본 연구에서는 CIGS 박막의 우선배향성에 대해 Mo후면전극의 미세구조가 미치는 영향 및 이에 따른 cell특성의 변화에 대해서 연구하였다. Mo후면전극의 미세구조는 2 mTorr~16 mTorr까지 증착압력을 변화시켜 제어되었고, CIGS광흡수층은 이렇게 준비된 Mo후면전극상에 3단계 동시증밥법(3-stage process)을 사용하여 형성하였다. XRD를 통한 박막의 우선배향성 평가에서, Mo 증착압력에 대한 IGS I(300)/I(006) 및 CIGS I(220:204)/I(112)의 거동은 Mo 미세구조와 밀접한 관련이 있는 잔류응력(residual stress)의 변화 거동과 상당히 일치함을 보였다. 이에 반해, 높은 압력의 Mo위에 형성된 강한 (220:204) 우선배향성의 CIGS와 bare-glass위에서 형성된 강한 (112) 우선배향성의 CIGS내 Na농도는 서로 유사하였다. 상기의 결과는 Mo미세구조 그 자체가 CIGS 박막 우선배향성의 원인이 됨을 나타낸다. Selenized Mo시편의 XRD분석 및 IGS/Mo 시편의 TEM분석결과을 통해 MoSe2의 반응성이 잔류응력과 비례하는 Mo in-gain 밀도에 의존하는 함을 알 수 있었고, 이러한 MoSe2반응성(reactivity)과 IGS우선배향성 사이에 상당히 밀접한 관련이 있으며 이에 CIGS의 우선배향성이 결정됨을 확인하였다. 마지막으로, Mo변수에 의해 제작된 cell의 특성분석으로부터 cell의 효율이 주로 VOC의 증가에 기인하여 CIGS (220:204) 우선배향성의 정도에 비례하였다.

Real-time LSTM Prediction of RTS Correction for PPP by a Low-cost Positioning Device (저가형 측위장치에 RTS 보정정보의 실시간 LSTM 예측 기능 구현을 통한 PPP)

  • The international gnss service (IGS) provides real-time service (RTS) orbit and clock correction applicable to the broadcast ephemeris of GNSS satellites. However, since the RTS correction cannot be received if the Internet connection is lost, the RTS correction should be predicted and used when a signal interruption occurs in order to perform stable precise point positioning (PPP). In this paper, PPP was performed by predicting orbit and clock correction using a long short-term memory (LSTM) algorithm in real-time during the signal loss. The prediction performance was analyzed by implementing the LSTM algorithm in RPI (raspberry pi), the processing speed of which is not high. Compared to the polynomial prediction model, LSTM showed excellent performance in long-term prediction.

Cultural characteristics and phylogenetic relationships based on RFLP analysis in Pleurotus species

  • Pleurotus has increased rapidly production and consumption because of highly nutritional value, natural healthy food and so on. The basic studies for Pleurotus need for development of mushroom industry. This study was to investigate the cultural characteristics among 15 strains of 6 species and to analyze their phylogenetic relationships. The cultural characteristics were investigated by mycelial growth activity at different media, temperature and pH. The optimum media for mycelial growth were YM and MCM in most species. The optimum temperature for mycelial growth were $25^{\circ}C$ and $30^{\circ}C$. The optimum pH for mycelial growth were widly range from pH 5.1 to 7.4. Through the RFLP (restriction fragment length polymorphism) of IGS (intergenic spacer) I region in ribosomal DNA, it was analyzed phylogeny of interspecies and intraspecies. Each species was discriminated well as isolates within each species formed clade to be distinguished other species. P. florida was highly similar to P. floridanus, and P. flabellatus was P. cornucopiae. P. fuscus var. ferulae was highly similar to P. eryngii but discriminated different species in analysis of RFLP of IGS I region and showed different characteristics in mycelial culture. RFLP of IGS I region was useful of studying phylogenetic relationships of species and population.
