정수처리시설에서 막공정 도입시 침전공정생략 가능성에 관한 연구 (A Study on Possibility of Sedimentation Basin Omission After Installed Membrane System in Drinking Water Treatment)

  • The objective of this study was to evaluate the possibility of sedimentation basin omission when installed hybrid membrane filtration process in the field plant with the capacity of $500m^3/day$ for 11 months in the "G", water purification plant in Seoul. In order to evaluate the possibility of the sedimentation basin omission, we measured the change of DOC by coagulant dosage. Dosage of PAC(power activated carbon) 4mg/L and coagulant($AI_2O_3$ 10%) 1.67mg/L were compatible to meet the water quality. Also according to the experiment without settlement process, optimization G values were determined to be 300/s, 64/s, and 32/s at the mixing tank, the first flocculator and the second flocculator, respectively. The test was performed under the conditions PAC-coagulation-no settlement-MF. As a result, a dosage of 4.0mg/L as PAC and 0.86 to 1.22mg/L as $Al_2O_3$(10%) in the condition of flux of 62.5LMH were determined to keep TMP value less than $1.0kg_f/cm^2$.

  • Ankyrins are a ubiquitously expressed family of intracellular adaptor proteins involved in targeting diverse proteins to specialized membrane domains in both the plasma membrane and the endoplasmic reticulum. We described here that the C-terminal domain of ankyrin-B interact specifically with Z-line portion of titin in yeast two-hybrid analysis, in vitro pull-down assays and localization experiments in COS-7 cells. In this study we provide the first experimental evidence that Z-line portion of titin is necessary for the localization of ankyrin-B and ankyrin-B links between the sarcolemma and the myofibril in costameres.

연속식 전기탈이온 장치의 원리와 기술 동향 (Principles and Current Technologies of Continuous Electrodeionization)

  • 전기탈이온 시스템은 전기투석과 이온교환수지법의 혼합 공정으로서 외부전기장 하에서의 이온교환매개체를 통해 이온이 제거되어 고순수를 제조할 수 있도록 고안된 장치이다. 전기탈이온 장치는 이온의 제거 메커니즘과 이온교환 매개체의 전기적 재생 메커니즘으로 설명되며 이를 이용하여 희석조의 성능을 최적화하는 것이 시스템의 효과적인 사용을 위해서 필요하다. 또한 전기탈이온 장치의 다양한 분야에서의 적용을 위해서는 스택구성을 다양하게 변경하여 적용가능성을 판단하는 것이 필요하다. 이를 효과적으로 달성하기 위해서는 기존에 제시되었던 다양한 시스템 특성분석법과 이동현상 관련 수식들을 이해하여 전기탈이온 스택구성에 적용하는 것이 무엇보다 중요하다. 본 총설에서는 전기탈이온 장치의 최적화를 위해 필요한 기초 이론들과 방법들에 대해서 살펴보고 현재의 연구동향에 대해서 고찰하여 분리공정으로서의 이해를 높이고자 하였다.

Preparation and Characterization of $TiO_2$Filled Sulfonated Poly(ether ether ketone) Nanocomposite Membranes for Direct Methanol Fuel Cells

  • A series of inorganic-organic hybrid membranes were prepared with a systematic variation of titanium dioxide nanoparticle content. Their water uptake, methanol permeability and proton conductivity as a function of temperature were investigated. The results obtained show that the inorganic oxide network decreases the proton conductivity and water swelling. It is also found that increase in inorganic oxide content leads to decrease of methanol permeability. In terms of the morphology, membranes are homogeneous and exhibit good adhesion between inorganic domains and the polymer matrix. The properties of the composite membranes are compared with the standard nafion membrane.

Possible Implication for an Indirect Interaction between Basic Fibroblast Growth Factor and (Na,K)ATPase

  • The (Na,K)ATPase is responsible for generating the ionic gradients and membrane potentials by the exchange of intracellular $Na^+$ for $K^+$. It has been recentl y shown that (Na,K)ATPase is involved in the exocytic pathway of basic fibroblast growth factor (bFGF), although it is not known that bFGF is secreted to the outside of cell through direct interaction with (Na,K) ATPase. To understand the role for (Na,K)ATPase in the secretary pathway of bFGF, we have sought to identify the cytoplasmic domains of the alpha1 isoform of (Na,K)ATPase interacting with bFGF by yeast two-hybrid system. We have also investigated the interaction between the alpha2 isoform of (Na,K)ATPase and bFGF to find out whether the interaction is isoform-specific. We found that none of the cytoplasmic domains of (Na,K)ATPase isoforms interacted with bFGF. The result suggests that the interaction between bFGF and (Na,K)ATPase might be indirect, thus requiring other proteins which are involved in the formation of protein complexes for the interaction, although we cannot exclude the possibility that the interaction requires the element of the whole alpha subunit structure that was not present in the isolated alpha subunit cytoplasmic domains.

Nebulin in Z-discs and Costameres

  • Deciphering the molecular interactions of proteins forming Z-lines is pivotal for understanding the regulation of myofibril assembly, sarcomeric organization, and mechanical properties of striated muscle. The purpose of this study is to searched for potential novel ligands of the Z-line portion of nebulin. In this study interacting proteins with intra-Z-line region of nebulin were screened using a yeast two-hybrid approach. The interaction was conformed by GST pull-down assay. We identified 269 residues within villin/gelsolin homology domain of supervillin that intreacts with the serine rich region of nebulin. The specific interactions of nebulin and supervillin were confirmed in vitro by GST pull-down experiments. Our data suggest that supervillin attaches directly to the Z-line through its interaction with the serine rich domain of nebulin in skeletal muscles. This interaction may link myofibrillar Z-discs to the membrane-associated complexes, costameres.

Silica를 함유하는 Polyurethane dispersion 투습방수 Film의 제조 (Fabrication of Silica-Containing Breathable Waterproof Polyurethane Dispersion Film)

  • Silica-polyurethane hybrid breathable films were prepared and their breathabilities were assessed. Appropriately aggregated silica was prepared through sol-gel reaction of water glass and its particle size ranged 360~500nm. The polyurethane dispersion was prepared by the reaction of isophorone diisocyanate(IPDI) as diisocyanate and polytetramethylene glycol(PTMG) and dimethylol propionic acid(DMPA) as polyol, particle size ranging 30~120nm. The reaction between isocyanate and hydroxyl group to form urethane bonding was checked by the intensity of the stretch peak of isocyanate at $2270cm^{-1}$ in the FT-IR. The silica was incorporated into polyurethane dispersion and casted into film. It was shown that the incorporated silica(1~5wt.%) increased water vapour permeability of the films by 30~100%, and decreased the hydrostatic pressure by 10~40%. From the results, it could be concluded that the appropriate hybridization of silica can increase the breathability of polyurethane dispersion film, while minimizing the loss of hydrostatic pressure.

  • In this study, we conducted an impact assessment of the amount of volatile organic solvents addition on hybrid water-proofing system of urethane waterproof coating material and modified asphalt sheet. Also, we conducted a comparative assessment of whether modified asphalt sheet is dissolved or not and oil leakage by dissolution in order to perform a comparative analysis of characteristics of the impact on modified asphalt sheet according to the volatility of volatile organic solvents included in urethane waterproof coating material. The test was carried out by adding the same amount of organic solvents into each experimental group which is subject to volatility and non-volatility of organic solvents, respectively. The results of the test showed that in both experimental groups modified asphalt sheet was dissolved when adding over 10 percent of organic solvents regardless of volatility, and oil leakage observed only in the experimental group subject to volatility.

Biological Synthesis of Genistein in Escherichia coli

  • Genistein is a type of isoflavonoid found predominantly in leguminous plants. Genistein has diverse biological activities, such as anthelmintic and antioxidant effects, as well as inhibitory effects on the growth of several cancers. In addition, genistein is well known as a phytoestrogen. In this study, we attempted to biologically synthesize genistein from either p-coumaric acid or naringenin using Escherichia coli as a biotransformation host. Four genes, Os4CL, PeCHS, RcIFS, and OsCPR, were used for genistein production. To functionally express RcIFS and OsCPR, two members of the cytochrome P450 family, in E. coli, the membrane-binding anchor domain of each gene was removed, and RcIFS and OsCPR were translationally fused to generate an RcIFS-OsCPR hybrid. Os4CL and PeCHS, or the RcIFS-OsCPR hybrid, were then transformed into E. coli BL21(DE3). Using these strains, we optimized our culture system at a laboratory scale in terms of the cell density, concentrations of substrate and isopropyl-β-D-thiogalactoside, temperature, and culture medium. Under the optimized culture conditions, genistein was produced at up to 35 mg/l and 18.6 mg/l using naringenin and p-coumaric acid, respectively.

탄소계 복합담지체에 담지된 고내구성 고분자전해질 연료전지용 백금촉매 (Highly Durable Pt catalyst Supported on the Hybrid Carbon Materials for Polymer Electrolyte Membrane Fuel Cell)

  • 본 연구에서는 산화그래핀과 카본블랙의 혼합담체를 이용하여 내구성이 향상된 백금촉매를 폴리올법으로 제조하였다. 삼전극 순환전압전류법을 이용한 전기화학성능 측정결과 적절한 비율로 조절된 혼합담지체에 백금을 담지시켰을 경우 초기 성능 감소없이 장기내구성이 향상되는 것으로 나타났다. 또한 회전원판전극을 이용하여 산소환원반응을 수행한 결과 혼합담체에 담지된 백금촉매가 카본블랙 단일담체에 담지된 백금촉매보다 우수한 고유활성값을 나타내었다.
