Cytokines expressed specifically in leukocytes subsets and in activated cells, which are involved in chemotaxis and activation of leukocytes, are recently defined as chemokines. Macrophage inflammatory $protein-1{\alpha}(MIP-1{\alpha})$ and $MIP-1{\beta}$ are members of C-C chemokine subfamily which produces wide immunomodulatory, proinflammatory, and hematopoietic modulatory actions. We have studied their gene expression by using Northern blot analysis in various blood cells such as cytolytic T lymphocyte(CTL), helper T lymphocyte(HTL), macrophage, and B lymphocyte. Resting CTL line CTLL-R8 expressed $MIP-1{\alpha}$ mRNA which was downregulated by ConA stimulation. Both of resting and ConA stimulated HTL line Hut78 and Jurkat did not express $MIP-1{\alpha}$ mRNA. There was detectable $MIP-1{\alpha}$ transcript in HTL hybridoma 2B4.11 which was a little upstimulated by ConA stimulation. B cell line 230, and macrophage cell line RAW264.7 and WR19M.1 showed distinct $MIP-1{\alpha}$ message which were induced after LPS stimulation. Expression pattern of $MIP-1{\beta}$ in all cell lines or cell were almost identical to that of $MIP-1{\alpha}$. Also strategies employed to identify and characterize the biological functions was preceded by receptor cloning to trace the shorcut to the final goal of cytokine research. For the cloning of $MIP-1{\alpha}$ receptor(R), we used synthetic oligonucleotides of transmembrane(T) conserved sequences of already cloned human(h) IL-8-R, and performed reverse transcription-polymerase chain reaction(RT-PCR) amplification using murine(m) macrophage cell line mRNA. Among 5RT-PCR products, we isolated a homologous cDNA with hIL-8-R which were shown to be putative mIL-8-R cDNA.
Background and Aims: Dansamtongmek-tang (DSTMT) and Dansamsengmek-san (DSSMS) have been used for many years as therapeutic agents for the acute stage of cerebrovascular disease, hypertension and hyperlipidemia in Oriental medicine, but the effects of DSTMT and DSSMS on hyperlipidemia and safety for cell damage are not yet well-known. This study was done to investigate the effects of DSTMT and DSSMS on hyperlipidemia. Methods: In vivo test: after administering DSTMT and DSSMS to SHR and ICR occurred hyperlipidemia for 3 weeks, we analyzed body weight, cholesterol levels. TG, HDL-chol, LDL-chol, LDH in plasma, brain, liver and kidney tissue, and DNA by RT-PCR. In vitro test: after administering DSTMT and DSSMS to human hepatocellular carcinoma in hypoxia, we observed cell cohesion by light microscope, analyzed the inflow of Ca2+ by confocal laser scanning microscope and DNA by RT-PCR. Results: DSTMT significantly decreased the levels of triglyceride and increased the levels of HDL-cholesterol in SHR, and significantly decreased the levels of LDL-cholesterol and body weight and increased the levels of HDL-cholesterol in ICR. DSSMS significantly decreased body weight, total cholesterol levels, LDL-cholesterol, LDH and cardiac risk factor (CRE) in SHR and significantly decreased the levels of total cholesterol, triglyceride, LDL-cholesterol, LDH and CRF in ICR. DSTMT had an effect on protecting cells from damage by inhibiting production of p53 mRNA, and in DSSMS, by inhibiting production of p53 mRNA and p21 mRNA after hypoxia. DSTMT effectively blocked off Ca2+ at low density, but DSSMS effectively blocked off Ca2+ at high density. Both DSTMT and DSSMS had an effect on inhibiting lipid metabolism by blocking off production of apo B mRNA. Conclusions: These results suggest that DSTMT and DSSMS might be usefully applied for treatment of hyperlipidemia and suppression of brain damage.
Aflatoxins are produced by Aspergillus flavus, parasiticus that grows in improperly stored cereals. Aflatoxin $B_1\;(AFB_1)$ is a potent hepatocarcinogen in a variety of experimental animals including human beings. In spite of a high attention to the hepatocarcinogenecity of aflatoxins, the relative toxicity of other types $(AFB_2,\;AFG_1\;and\;AFG_2)$ of the toxins is not fully clarified. Sprague-Dawley male rats were orally administered with $AFB_1,\;AFB_2,\;AFG_1\;and\;AFG_2$ at the dose of 250, 1250, and $2500\;{\mu}g/kg$ body weight. Animals were then killed at 12, 24 or 48 hrs following aflatoxin adminstration. Subsequently the relative weight of liver was measured and histopathological examination on the liver was performed. Level of 8-OxodG and expression of ras gene in the liver was determined. The relative liver weights at high doses of $AFB_1\;and\;AFG_1$ was significantly low. The treatment of $AFB_1$ at the high dose of $2500\;{\mu}g/kg$ showed vacuolar degeneration and centrilobular hepatic necrosis with inflammatory cells. The pathological changes by $AFB_2\;AFG_1,\;and\;AFG_2$ were not clearly found. The formation of 8-OxodG by $AFB_1$ increased in a dose-dependent manner up to 24 hrs after a single treatment of $AFB_1$ thereafter decreased to the level of the control. The treatments of $AFB_2\;AFG_1,\;and\;AFG_2$ showed an inconsistent pattern in the formation of 8-OxodG in the liver of rats with increasing time. The expression of ras oncogene in the liver by $AFB_1$ at the dose of $1250\;{\mu}g/kg$ was increased twice compared to the control. The treatments of $AFB_2\;AFG_1,\;and\;AFG_2$ at all doses decreased the expression of ras in the liver. These results in the present study indicate that $AFB_1$ among aflatoxins with low comparable levels is the most toxic as determined by early biomarkers such as 8-OxodG formation and ras expression. However, the levels of 8-OxodG and ras as biomarkers were not useful to predict the relative hepatocarcinogenicity of aflatoxins to $AFB_1$ in the present model. Further studies are required to look for other biomarkers to predict carcinogenic potency of aflatoxins.
p53 유전자는 스트레스, DNA 손상, 저산소증 및 종양 발생에 대한 세포 반응의 전사 조절에서 중요한 역할을 담당한다. 최근에 발견된 다양한 종류의 p53의 생리 활성을 생각한다면 p53이 암 조절에 관여한다는 것은 놀랄만한 일이 아니다. 인간 암의 약 50%에는 p53 유전자의 돌연변이 또는 p53을 활성화시키는 기전의 결함을 통해 p53 단백질 기능의 불활성화가 나타난다. p53 기능의 이러한 장애는 p53 의존 반응으로부터 회피를 허용함으로써 종양의 진화에 결정적인 역할을 하게 된다. 최근의 많은 연구들은 p53의 돌연변이를 대폭 감소시키거나 p53의 종양 억제 기능을 복원하기 위하여 선택적인 저분자 화합물을 동정함으로써 p53의 돌연변이를 직접 표적하는 것에 초점을 두고 있다. 이들 저분자는 좋은 약물과 유사한 특성을 유지하면서 다양한 상호작용을 효과적으로 조절해야 한다. 이 중, p53의 음성조절인자 핵심인 MDM2의 발견은 p53과 MDM2 간의 상호작용을 차단하는 새로운 저분자 억제제의 설계를 제공하였다. 저분자 화합물 중 일부는 개념 증명 연구에서 임상 시험으로 옮겨졌으며 향후 맞춤형 항암제가 추가될 전망이다. 본 리뷰에서는 야생형 p53과 돌연변이 p53의 구조적 및 기능적 중요성과 p53을 직접 표적하는 치료제 개발, p53과 MDM2 간의 상호작용을 억제하는 화합물에 대하여 검토하였다.
Streptococcus pneumoniae는 pneumolysin과 같은 병원성 인자를 가지고 있으며, 지역사회에서 전파되는 심각한 병원성균에 포함된다. Pneumolysin (PLY)은 콜레스테롤 의존적으로 세포막에 구멍을 형성하는 세포독소로서 백신의 주요한 표적 항원이다. PLY의 연구를 위하여 Streptococcus pneumoniae D39 균주에서 추출한 genomic DNA를 주형으로 하여 PCR을 시행하였다. 합성된 PLY 유전자 DNA를 pQE-30 vector에 삽입하고, E. coli M15에 형질전환 시킨 후 LB 배지에 IPTG를 첨가하여 PLY 단백질을 생산하였다. 재조합 단백질은 $Ni^{2+}$-agarose column을 사용하여 정제하였다. 또한, EGFP를 PLY C-말단에 부착한 융합단백질도 동일한 방법으로 클로닝하여 재조합 단백질을 생산하였다. 500 ng/ml 농도의 재조합 PLY는 1.0% 적혈구 현탁액을 100% 용혈시켰으며, 240 ng/ml 농도는 50% 용혈을 나타내었다. 그러나 재조합 PLY-EGFP는 용혈 활성이 전혀 나타나지 않았으나 형광현미경으로 관찰하였을 때 적혈구 막에 결합되어 있었다. 즉, EGFP의 PLY C-말단 부착은 PLY의 세포막 결합능은 유지시켰으나 용혈기능은 방해하였다. PLY C-말단은 용혈기능에 아주 중요한 영역이며, 세포막 결합은 PLY의 다른 영역이 보다 중요하게 작용할 것으로 추측된다. 따라서, 육안으로 관찰이 가능한 결합능은 가졌으나 용혈 기능이 결여된 PLY-EGFP를 대조군으로 활용함으로써 두 재조합 단백질은 폐렴 유발에 있어서 PLY 작용 연구에 사용될 수 있을 것으로 기대된다.

Prostaglandin $E_2$ ($PGE_2$) promotes tumor-persistent inflammation, frequently resulting in cancer. Curcumin is a diphenolic turmeric that inhibits carcinogenesis and induces apoptosis. $PGE_2$ inhibits curcumin-induced apoptosis; however, the underlying inhibitory mechanisms in colon cancer cells remain unknown. The aim of the present study is to investigate the survival role of $PGE_2$ and whether addition of exogenous $PGE_2$ affects curcumininduced cell death. HCT-15 cells were treated with curcumin and $PGE_2$, and protein expression levels were investigated via Western blot. Reactive oxygen species (ROS) generation, lipid peroxidation, and intracellular glutathione (GSH) levels were confirmed using specific dyes. The nuclear factor-kappa B ($NF-{\kappa}B$) DNA-binding was measured by electrophoretic mobility shift assay (EMSA). $PGE_2$ inhibited curcumin-induced apoptosis by suppressing oxidative stress and degradation of PARP and lamin B. However, exposure of cells to the EP2 receptor antagonist, AH6809, and the PKA inhibitor, H89, before treatment with $PGE_2$ or curcumin abolished the protective effect of $PGE_2$ and enhanced curcumin-induced cell death. $PGE_2$ activates PKA, which is required for cAMP-mediated transcriptional activation of CREB. $PGE_2$ also activated the Ras/Raf/Erk pathway, and pretreatment with PD98059 abolished the protective effect of $PGE_2$. Furthermore, curcumin treatment greatly reduced phosphorylation of CREB, followed by a concomitant reduction of $NF-{\kappa}B$ (p50 and p65) subunit activation. $PGE_2$ markedly activated nuclear translocation of $NF-{\kappa}B$. EMSA confirmed the DNA-binding activities of $NF-{\kappa}B$ subunits. These results suggest that inhibition of curcumin-induced apoptosis by $PGE_2$ through activation of PKA, Ras, and $NF-{\kappa}B$ signaling pathways may provide a molecular basis for the reversal of curcumin-induced colon carcinoma cell death.
포자형성균은 사람과 동물용의 프로바이오틱 제제로서 사용되어져 왔다. 포자형성균의 낮은 pH에 대한 안정성과 위내 생육저해 환경에 생존능은 프로바이오틱 제제로서 매력적이다. 본 연구는, 한국 전통 대두 발효 식품의 종균인 BCNU 9028균주를 메주로부터 분리하였다. 생리학적, 생화학적 특성과 16S 리보좀 DNA 염기서열 분석 결과 BCNU 9028균주는 $Bacillus$에 속하는 것을 확인하였다. $Bacillus$ sp. BCNU 9028은 pH 2.5에서 92%의 생존률을 보였으며, 0.3% 담즙산에서도 저항성을 나타냈다. 그리고, 식품 병원성균과의 응집정도와 자가결합능에 의해 $Bacillus$ sp. BCNU 9028는 $Listeria$$monocytogenes$, $S.$$aureus$ 및 $E.$$coli$와 같은 식품 병원성균의 생물막형성과 부착을 저해할 수 있는 것을 확인하였다. BCNU 9028의 소수성 특성(63.3%)은 장관내 부착능이 우수할 것으로 나타났다. 특히, $Bacillus$ sp. BCNU 9028 균주는 그람양성 및 그람음성 병원성균 모두에 항균력을 갖는 것을 확인하였다. 이상의 결과로 $Bacillus$ sp. BCNU 9028균주가 프로바이오틱스로서 이용가능성이 있음을 제시하였다.
본 연구에서는 발아 검은 쌀로부터 분리한 올리고펩타이드($Oligosproutin^{(R)}$)의 피부 탄력 개선 효과에 대하여 연구하였다. 올리고펩타이드가 각질형성세포의 유전자 발현에 미치는 영향을 알아보고자 DNA microarray를 수행하였다. 각질형성세포에 올리고펩타이드를 처리하여 유전자의 발현이 2배 이상 증가하는 유전자는 745개이고, 2배 이상 감소하는 유전자는 1011개로 나타났다. 이 중 hyaluronan synthase 2 (HAS2)의 유전자 발현이 2배 이상 증가함을 확인하였다. 올리고펩타이드를 처리한 각질형성세포에서 세포활성 증가, HAS2 유전자 발현 증가, 세포내 항산화 효과를 확인하였다. 올리고펩타이드를 함유한 O/W 에멀젼을 이용한 임상실험에서 피부의 탄력 개선 효과를 확인하였다. 본 연구를 통하여 발아 검은 쌀로부터 분리한 올리고펩타이드는 피부 탄력 개선을 주는 항노화 화장품 개발에 응용될 수 있을 것이라 기대된다.
본 연구실에서는 이전의 실험에서 suppression subtractive hybridization(SSH)을 통하여 생쥐의 생후 1일자 난소와 5일자 난소에서 차이 나게 발현하는 유전자들의 목록을 얻었고 그 중에서 MT transposon-like element, clone MTi7(MTi7)이 성장하는 난포에서 더 높게 발현한다는 것을 알아냈다(Park et al., 2002). In situ hybridization과 RNA interference를 이용한 연구결과, MTi7은 난자에서 특이 적으로 발현하는 유전자로 특히 난자성숙에 관여하는 것으로 관찰되었다(Park et at., 2003). 그러나 현재까지 MTi7의 염기서열은 생쥐에서만 알려져 있다. 따라서 본 연구는 두 부분으로 나누어서 첫째, MTi7이 다른 포유류에도 존재하는지 알아보기 위해 소,돼지, 흰쥐 그리고 사람 등 각기 다른 네 종의 난소 cDNA를 사용하여 새로운 MTi7을 분리하고자 하였으며, 둘째, 생쥐의 MTi7이 transposon의 특징을 갖고 있어 다른 유전자에 삽입되어 있는지를 알아보기 위해 inverse PCR을 시행하여 MTi7주변의 유전자가 있는지를 조사하였다. 네 종의 난소 cDNA를 사용하여 생쥐의 MTi7과 매우 유사한 염기서열을 갖고 있음을 알았다(87%∼98%). Inverse PCR 결과, 생쥐의 MTi7은 beta-carotene 15, 15'-monooxygenase(Bcdo) 유전자 혹은 serine protease inhibitor, Kunitz type I(Spint 1) 유전자에 삽입되어 있음을 알 수 있었다. 본 연구의 결과로 여러 포유류의 MTi7 sequence를 알아내고, 또한 생쥐의 MTi7이 삽입되어 있는 유전자를 알아냄으로써 머지 않은 장래에 난자형성 및 난포형성과정에 있어서 MTi7의 역할을 밝혀 낼 수 있을 것으로 기대된다.
유아 분변, 우분, 염소 분변, 개 분변, 돼지 분변, vaginal tract 채취물, 채소류, 부패한 과일류, 김치, 젓갈, 발효 소세지, 원유, 치즈, 발효유, 청국장, 메주, 막걸리 등 다양한 시료로부터 3,000주의 미생물을 분리하였다. 분리미생물은 L. monocytogenes, E. coli O157:H7, S. enterica serovar Enteritidis에 대해 항균활성을 측정하여 박테리오신 생산 미생물 26주를 최종 선발하였다. 16S rDNA gene sequencing 분석을 통하여 동정한 결과, Enterococcus faecalis(7주), E. faecium(8주), E. hirae(5주), Lactobacillus acidophilus(1주), L. amylovorus(1주), L. curvatus(2주), L. plantarum(1주), Pediococcus acidilactici(1주)와 같이 8종의 미생물이 박테리오신을 생산하는 것으로 확인되었다. 미생물이 생산한 박테리오신은 대부분이 단백질 또는 펩타이드성 물질이어서 인간 또는 동물의 소화효소에 의해 분해되므로 식품에서 안전한 천연보존제로 사용할 수 있을 것으로 기대된다.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
