  • 1. Objectives The purpose of this study is to find out effects of Choeongsimyeonja-tang and Taeumjowi-tang on the against decline of physical function as aging. 2. Change that is Wistar rat's hematological value by aging according to 10, 30, 50 week WBC, RBC, Hgb, monocyces, eosinophil ere. increase, and HCT, PLT ere. showed tendency that decrease accorcling to old-week, and observed improvement that is hematological value than control group form of Cheongsimyeonja-tang & Taeumjowi-tang.5. Change that is Wistar rat's biochemical value by aging was measured highest in 50 week because ALT, AST, BUN, CRN, T-bili., T-chol., TG, TP, ALB, A/G. P ete. increase according to 10, 30, 50 week, and observed improvement that is biochemical value than control group form of Cheongsimyeonja-tang & Taeumjowi-tang. Is considered by being effect that Cheongsimyeonja-tang & Taeumjowi-tang living body function decline by aging by this result.

Effects of Kuseonwangdogo on the Proliferation of Preadipocyte 3T3-L1 Cells, the Anti-Complementary and the Cytotoxic Effects (구선왕도고가 전지방세포(前脂肪細胞) 3T3-L1의 증식(增殖), 항보체활성(抗補體活性) 및 세포독성(細胞毒性)에 미치는 영향)

  • To investigate the anti-complementary and cytotoxic effects of oriental prescription, Kuseonwangdogo, on the proliferation of preadipocyte 3T3- L1 cells, we examined biological effects of Kuseonwangdogo. The results obtained were as follows. 1. After 14 days, the body weight of rats treated with Kuseonwangdogo decreased more than that in the control group (p<0.05). However, the weights of liver, spleen and kidney were unchanged. In serum biochemical test, we examined the level of glucose (GLU) and glutamic pyruvic transaminase (GPT). The levels of GOT and CHOL in serum were decreased remarkably by the administration of Kuseonwangdogo (p<0.05). The haematological examination of the tested group showed significant increment of white blood cells (WBC), hemoglobin concentration (HGB), mean corpuscular hemoglobin (MCH) and monocyte (MO). 2. The effect of Kuseonwangdogo on the proliferation of 3T3-L1 cells was tested by the sulforhodamin B(SRB) assay. The high concentration ($100{\mu}l\;and\;200{\mu}l$) of extracts inhibited the proliferation of 3T3- L1 cells. The p-value was <0.01, respectively. 3. The extract of Kuseonwangdogo showed a potent anti -complementary activity. It was suggested that the active principle may be a kind of polysaccharide molecule. 4. The cytotoxic effects of Kuseonwang dogo and its composing herbs in human liver cells (WRL68) and monkey kidney cells (Vero) were examined by the SRB and 3- (4,5- Dimethylthiazol-2-yl) -2,5 diphenyl-2H- tetrazolium bromide (MTT) assay. Cytotoxic effects were not observed.

Anti-inflammatory Effect of Haingkyunghonghwatang in cultured RAW264.7 cells and murine models of inflammation (RAW264.7세포주와 염증생쥐모델에서 항염증(抗炎症) 작용(作用)에 대한 행경홍화탕(行經紅花湯)의 효과(效果))

  • Purpose : The Purpose of this research was to investigate the effects of Haingkyunghonghwatang (HKHHT) on anti-inflammatory effects. Methods : As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators were determined in mouse lung fibroblast cells(mLFC) and RAW264.7 cells. Also, changes in pathological features by drug treatment were investigated in the in vivo edema-induced rats by carrageenin/arachidonic acid or in the colitis-induced mice by DSS treatment. Results : The cytotoxicity of HKHHT on mLFC and RAW264.7 cells wasn't observed at 100, 50, 10, and $1{\mu}g/ml$ of The treatments. $IL-1{\beta}$, IL-6 and NOS-II mRNA expression of RAW264.7 cells was inhibited by The treatments in a dose-dependent manner. HKHHT treatment of RAW264.7cells(HtRc) inhibited $TNF-{\alpha}$ and COX-2 mRNA expression. HtRc significantly inhibited IL-6 and NO production. HtRc inhibited ROS production. HKHHT inhibited rat's paw edema induced by carrageenin or arachidonate treatment in all concentrations examined. The body weight and colon length of colitis-induced mice were recovered to a normal level by DSS treatment. Clinical disease levels were significantly improved compared to the control animals. HKHHT treatment of colitis-induced mice(HtCm) significantly increased hematological values such as WBC and RBC counts, Hgb and HCT levels, but decreased PLT values. HtCm decreased IL-6 and $TNF-{\alpha}$ production significantly HtCm significantly increased CD3+(T) cell counts. In contrast, HKHHT treatment decreased CD19+ B cell counts and CD3+/CD69+ significantly, and also decreased B/T ratio (%) though not significant. Conclusion : These results indicated that HKHHT could be used for treating diverse female diseases caused by the inflammation.

  • Administrating PalMoolGoonJaTang and SipIMiGwanJungTang to 40-weeks-(old Wistar rat for 10 weeks so, I researched into weight change, weight change of internal organ, and hemtological and serological changews. Then I got these conclusions. 1. Observed gain in weight than control group form of SipIMiGwanJungTang to aged Wistar rat. 2. Is thought to promote activation of living body action gaining intestine weight along with gain in weight. 3. Displayed decrease of MDA's content of serum than control group form of SipIMiGwan JungTang to aged Wistar rat. 4. Change that is Wistar rat's hematological value by aging according to 10, 30, 50 week WBC, RBC, Hgb, monocytes, eosinophil etc. increase, and HCT, PLT etc. showed tendency that decrease according to old-week, and observed improvement that is hematological value than control group form of SipIMiGwanJungTang 5.   • The water extract of Gamidaebo-Tang depressed the lethal toxicity of cis-platin$(45{\mu}M/kg,\;s.c.)$, and mitomycin C (6mg/kg, s.c.). Significant depression of renal toxicity(indicated by an increased blood urea nitrogen value and creatinine value) of cis-platin $(35{\mu}M/kg)\;s.c.)$, was observed in mice treated with Gamidaebo-Tang. In rats treated with mitomycin C and Gamidaebo-Tang, the increased LDH activity and reduced total protein and albumin contents by mitomycin C were significantly depressed. The number of WBC was significantly increased in rats treated with mitomycin C but in the rats treated with mitomycin C and Gamidaebo-Tang, the number of WBC was increased and the hematocrit value and the number of RBC and Hgb were significantly increased in the dose-dependent manner. Therefore, bone marrow toxicity of mitomycin C in the rats treated with Gamidaebo-Tang was depressed. In the end, alleviative effects of the side actions of cis-platin and mitomycin C were acknowledged by combined usage of Gamidaebo-Tang and these anti-neoplastic drugs.

The Effects of Kimchi on Hematological and Immunological Parameters in vivo and in vitro (In vivo와 in vitro에서 김치가 혈액성상과 면역세포배양에 미치는 영향)

  • This study was designed to know the effect of kimchi on the hematological and immunological parameters in vivo and in vitro, respectively. To study the effects of kimchi on the hematological parameters, rats(S.D., male) were divided into 4 groups and fed diets containing of 3%, 5% and 10% kimchi or kimchi free diet(control) for 6 weeks. The results of CBC(complete blood cell) tests obtained from the bloods of rates were as follows ; In 10% kimchi group, the level of WBC(white blood cells), RBC(red blood cells), Hgb(hemoglobin), Hct(hematocrit) were increased significantly than those of control group(p<0.05). MCV(mean corpuscular volume), one of the red cell indices, was also increased significantly in the animals fed 10% kimchi(p<0.05). RDW(Red cell distritution width) and PCT(plateletcrit) was lowest in 10% kimchi group(p<0.05). To examine the effects of kimchi on immune cell growth in vitro, three types of mouse immune cells-spleen cells, bone marrow cells, thymus cells-were cultured with extracts of salted Chinese cabbage, fresh kimchi and fermented kimchi(for 1 week) for 12 or 20 days. Control was supplemented with PBS(phosphate buffer saline) excluding kimchi extract. The results of spleen cell, bone marrow cell, and thymus cell cultures showed similar tendency: control medium accelerated death of cells, extracts of salted Chinese cabbage reduced the rate of cell death, and extracts of fresh kimchi and fermented kimchi promoted cell growth. From these results, it could be suggested that kimchi possibly has an effect on the hematopoietic ability and increases immune cell development and growth in vivo.

Effect of deep-sea mineral water on growth performance, water intake, blood characteristics and serum immunoglobulins in the growing-finishing pigs

  • Brine mineral water (BMW) is groundwater near the deep sea, and the mineral component of the BMW is different from the water of the deep sea because the components of BMW are derived from the unique geographical features surrounding it. Recently, BMW has attracted attention due to the unique health-related minerals it possesses; however, the influence of BMW on physiological function has not yet been determined in domestic animals. Therefore, this experiment investigated the influence of BMW on the growth performance, water intake, blood properties, and immunoglobulin (Ig) levels of serum in growing-finishing pigs. A total of 64 pig barrows (Landrace × Yorkshire × Duroc) with an average initial weight of 40.56 ± 0.17 kg were used in the experiment, and 0%, 2%, 3%, and 5% samples of BMW diluted with freshwater were provided to experimental animals during the 56 days. We found that the gain/feed ratio in the 3% BMW group was significantly higher than that in the 5% BMW group of growing-finishing pigs (p < 0.05). The water intake was significantly increased in the 5% BMW group compared with the other groups (p < 0.05) of growing-finishing pigs. Additionally, the concentrations of red blood cells (RBCs), hemoglobin (HGB), and hematocrit (HCT) were significantly higher in the 3% BMW group than in the control group. The level of high-density lipoprotein cholesterol was higher in the 3% BMW group than in the 5% BMW group (p < 0.05). Furthermore, IgG and IgM levels in the serum were significantly higher in the 2% and 3% BMW groups than in the control group (p < 0.05). These results suggest that a dilution of 3% BMW in the intake water could improve the levels of RBCs and serum Igs in growing-finishing pigs.

Evaluation of Worker's Health and Occupational Exposure to Perchloroethylene in Laundries (Perchlotoethylene을 사용하는 세탁소 작업자들의 직업성 폭로와 건강에 관한 연구)

  • In this study we examined several subjective symptoms in an exposed group and an unexposed group to perchloroethylene. The exposed group consists to 30 workers employed in 23 dry-cleaning establishments located in Chung Cheung Do, whereas the unexposed group consists of 42 officers employed in a certain university. This study was conducted from October, 1993 to March, 1994. Also we investigated personal exposure levels and area concentration of PCE, and performed hematological and biochemical examination in blood and urine samples from the exposed group. The results are : 1. The exposed group highly complains several subjective symptoms, particularly in neuropsychiatry, eye, respiratory system and digestive system, compared with the unexposed group. But complaint rates in the exposed group are not related with PCE exposure levels. 2. PCE exposure levels for all dry-cleaning establishment varied widely, from 0.18 to 37.58ppm. The mean exposure level for centers of chain was 16.85ppm, and for local laundries was 8.83ppm, while for self-service establishment it was 3.07ppm. Eighty three percent of the workers were exposed less than the half-level(25.0ppm) of the Korea 50.0ppm standard for eight-hour PCE exposure. Seventeen percent of them exceeded the half-level of the Korea TLV. 3. Statistical correlation exists between the work load(number of operation cycles of equipment and the mass of processed textiles) and the personal exposure levels to be observed. 4. In hematological examination the values of WBC, RBC, Hgb, Hct, MCV, MCH, MCHC and PLT in the exposed group were within normal ranges. In biochemical examination(GOT, GPT, ALP, U-A and LDH) most of the items were within normal ranges. From this surveys conducted on 23 dry-cleaning establishments, the complaint rates of the exposed group to PCE in several subjective symptoms were higher than those of the unexposed group, and PCE exposure levels in all dry-cleaning establishment were below the Koera standard 50.0ppm. In hematological and biochemical examination the significant abnormalities by occupational exposure to PCE were not observed. However, the workers employed in dry-cleaning establishments are always faced with risk, becauses they are also exposed to several organic solvents used to remove stains. Therefore, various measures to improve occupational environment in dry-cleaning establishment should be considered.
