소나무 수피를 유기 설폰산 촉매존재 하에서 페놀액화하고 얻어진 액화물에 대한 특성을 검토하였다. 수피의 페놀액화시 유기 설폰산은 염산보다 뛰어난 촉매임을 확인 할 수 있었으며, 본 촉매의 사용에 의하여 소나무 수피의 완전한 액화가 가능하였다. 이들 유기 설폰산을 촉매로하여 제조된 수피 액화물의 경우 액화용매로 사용된 페놀이 염산을 촉매로 한 경우에 비하여 2-3배 많이 결합해 있었으며, 유리 전이점(Tg)이 낮았다 (PTSA: 85℃, MSA: 169℃, HCI: 181℃). 또한 이들 액화물은 전수산기, 페놀성 수산기 및 지방족 수산기가 많이 존재하여 반응중 액화 용매인 페놀의 도입과 2차적인 축합억제를 예상할 수 있었다. 액화물의 IR 스펙트럼 및 중성당류 분석 결과로부터 수피 액화물 중의 탄수화물은 대부분 분해되었다는 것을 알 수 있었다. 액화물 및 그 잔사의 EDS 분석결과 유기 설폰산 촉매는 염산 촉매에 비하여 반응용기의 심각한 부식을 초래하지 않는다는 것을 알 수 있었다.
Rice(Oryza stiva L.) is a major cereal food providing nourishment to over half of the world's populations and was considered only as a source of energy. However, The recent many researches have been made to suggest that rice may relate to prevention chronic disease and health-promoting properties such as postprandial glucose response, hypocholesterolemic effect and blood pressure-lowering effect. There has been numerous observation supporting that rice has hypocholesterolemic effect. Rice, rice bran, rice bran oil and unsaponifiable matter of rice bran oil reduced plasma cholesterol in rat, hamster as well as human. Components of rice showing hypocholesterolemic effect include dietary fiber(hemicellulose, phytic acid). protein, ${\gamma}$-oryzanol, $\beta$-sitosterol, and tocotrienols. Crapo et al has been studied that the effect of various of starchy foods on the postprandial blood glucose and insulin responses in healthy and diabetic humans. The results showed that rice had lower blood glucose and insulin responses compared to potato, bread and dextrose. The different physical forms in the same starch also produce the different postprandial glucose and insulin responses. In recent years, several studies have shown that some components of rice have potent antioxidant activity against Fe$^{2+}$ -ascorbate induced lipid peroxidation in rat liver microsomal membranes. Cell culture and animal studies have shown that some components of rice have inhibitory effect on the growth and proliferation of several types of human cancer cell. It was also reported that the methanol extract of brown rice has antimutagenic activity against various mutagens. In addition, the pepsine hydrolysate from rice protein is reported to inhibit angiotensin converting enzyme activity. GABA (${\gamma}$ - aminobutyric acid) and GABA enriched rice germ is also effective for lowering blood pressure and triglyceride levels.s.
A composite microbial system (XDC-2) was used to pretreat and hydrolyze corn stalk to enhance anaerobic digestion. The results of pretreatment indicated that sCOD concentrations of hydrolysate were highest (8,233 mg/l) at the fifth day. XDC-2 efficiently degraded the corn stalk by nearly 45%, decreasing the cellulose content by 22.7% and the hemicellulose content by 74.1%. Total levels of volatile products peaked on the fifth day. The six major compounds present were ethanol (0.29 g/l), acetic acid (0.55 g/l), 1,2-ethanediol (0.49 g/l), propionic acid (0.15 g/l), butyric acid (0.22 g/l), and glycerine (2.48 g/l). The results of anaerobic digestion showed that corn stalks treated by XDC-2 produced 68.3% more total biogas and 87.9% more total methane than untreated controls. The technical digestion time for the treated corn stalks was 35.7% shorter than without treatment. The composite microbial system pretreatment could be a cost-effective and environmentally friendly microbial method for efficient biological conversion of corn stalk into bioenergy.
The degradation mode of lignocellulose by anaerobic ruminal cellulolytic bacterium Ruminococcus albus F-40 was investigated. Birchwood holocellulose and filter paper were incubated as the sole carbohydrate sources with using the Hungate techniques. After 2 or 4 days of incubation, samples were employed for chemical and electron microscopic evaluations. The degradation rate of cellulosic substrates and the adhesion rate of bacteria to the substrates increased proportionally with the decrease of relative crystallinity of cellulose, indicating the preferential breakdown of amorphous cellulose, by this bacterium. X-ray diffraction analyses and polarized light microscopy showed, however, that crystalline cellulose was also degraded by R. albus. FT-IR spectra indicated that not only cellulose but hemicellulose was also degraded by this bacterium. Electron microscopic investigations showed the protuberant structures on the surface of R. albus. These structures were much more significant when bacterial cells were grown in the media containing insoluble substrates, such as cellulose, indicating clearly that bacterial protuberant structures were induced by the substrates. Protuberant structures extended from the bacterial cells adhered tightly to the substrates and numerous vesicles covered the surface of cellulosic substrates affected. Cellulosome-like structures were distributed on the cellulose matrix. Electron microscopic works showed that diverse surface organells of R. albus were involved in the degradation of cellulosic materials. SEM examinations showed the breakdown of cellulose by R. albus was proceeded by severeal routes : short fiber formation, defibrillation and destrafication of cellulose microfibril.

This study aimed to increase the sugar and ethanol yield from the mengkuang plant biomass through biological and liquid hot water (LHW) pretreatment and their combination. The results showed that biological pretreatments with 5% inoculum of the fungus Trametes versicolor resulted in the highest alpha cellulose content incubated for 30 days, and 10% inoculum resulted in the lowest lignin content. LHW pretreatment decreased the hemicellulose content of pulps from 10.17% to 9.99%. T. versicolor altered the structure of the mengkuang pulp by degrading the lignin and lignocellulose matrix. The resulting delignification and cellulose degradation facilitate the hydrolysis of cellulose into sugars. The alpha cellulose content after biological-LHW pretreatment was higher (78.99%) compared to LHW-biological pretreatment (76.85%). Scanning electron microscopy analysis showed that biological-LHW combinated treatment degrades the cell wall structures. The ethanol yield for biological-LHW pretreated sample was observed 43.86% (13.11 g/L ethanol by weight of the substrate, which is much higher than that of LHW-biological pretreatment (34.02%; 9.097 g/L). The highest reducing sugar content about 45.10% was observed with a resulting ethanol content of 15.5 g/L at LHW pretreatment temperature of $180^{\circ}C$ for 30 min.
Leaching ($60^{\circ}C$, 5 min) and wet torrefaction ($200^{\circ}C$, 5 min) of empty fruit bunch (EFB) were carried out to improve the fuel properties; each liquid fraction was reused for leaching and wet torrefaction, respectively. In the leaching process, potassium was effectively removed because the leaching solution contained 707.5 ppm potassium. Inorganic compounds were accumulated in the leaching solution by increasing the reuse cycle of leaching solution. The major component of the leached biomass did not differ significantly from the raw material (p-value < 0.05). Inorganic compounds in the biomass were more effectively removed by sequential leaching and wet torrefaction (61.1%) than by only the leaching process (50.1%) at the beginning of the liquid fraction reuse. In the sequential leaching and wet torrefaction, the main hydrolysate component was xylose (2.36~4.17 g/L). This implied that hemicellulose was degraded during wet torrefaction. As in the leaching process, potassium was effectively removed and the concentration was accumulated by increasing the reuse cycle of wet torrefaction hydrolysates. There was no significant change in the chemical composition of wet torrefied biomass, which implied that fuel properties of biomass were constantly maintained by the reuse (four times) of the liquid fraction generated from leaching and wet torrefaction.

Torch ginger (Etlingera elatior Jack) is a potential source of lignocellulose material for various derivative products. This study aims to determine the chemical components, ratio of syringyl to guaiacyl units (S/G) in lignin, and crystallinity of the biomass of torch ginger. The effects of soda pulping on the chemical characteristics of torch ginger pulp were also studied. Pulping of the chips was conducted with active alkali of 15%, 20%, and 25% and a Liquor-to-Wood (L/W) ratio of 4:1, 5:1, and 6:1. The impregnation and pulping times at maximum temperature (170℃) were 120 and 90 min, respectively. To assess the effect of treatments on the properties of pulping, a two-factorial experimental design was applied. Results showed that the content of α-cellulose and hemicellulose in the torch ginger was 48.48% and 31.50%, respectively, with an S/G ratio of 0.70 in lignin. Soda pulping changed the crystalline structure of the biomass from triclinic to monoclinic. Active alkali, L/W ratio, and interactions considerably influenced the observed responses. The degree of delignification increased with an increase in the loading of active alkali, which lead to a decrease in the kappa number of the pulp. An active alkali content of 25% and an L/W ratio of 6:1 resulted in the highest delignification selectivity with a kappa number of 2.78 and a yield of 24%. Given its cellulose content and ease of pulping, torch ginger can be a potential raw material for derivative products that require delignification as pretreatment. However, the increase in cellulose crystallinity should be considered when converting torch ginger to bioethanol.

The objective of this study was to investigate the effect of alkali-washing with different sodium hydroxide concentrations on the chemical compositions of steam-treated Betung bamboo strand. Strands were subjected to steam treatment at 126 ℃ for 1 h under 0.14 MPa pressure and followed by washing with 1-5% sodium hydroxide solution for 30 sec. The alteration of structural and non-structural chemical components content of bamboo strands was evaluated. Steam and washing treatments with various concentrations of sodium hydroxide solution considerably reduced the extractive content of bamboo strands, and the cell wall chemical components of the strand in the small degree. FTIR analysis showed noticeable changes in peaks related to hemicellulose and lignin. The relative crystallinity increased significantly after steam and washing treatment with sodium hydroxide up to 3% concentration. SEM Images showed smooth and clean strands surface after washing with 3% sodium hydroxide.
α-Galactosidase is a debranching enzyme widely used in the food, feed, paper, and pharmaceuticals industries and plays an important role in hemicellulose degradation. Here, T26, an aerobic bacterial strain with thermostable α-galactosidase activity, was isolated from laboratory-preserved lignocellulolytic microbial consortium TMC7, and identified as Parageobacillus thermoglucosidasius. The α-galactosidase, called T26GAL and derived from the T26 culture supernatant, exhibited a maximum enzyme activity of 0.4976 IU/ml when cultured at 60℃ and 180 rpm for 2 days. Bioinformatics analysis revealed that the α-galactosidase T26GAL belongs to the GH36 family. Subsequently, the pET-26 vector was used for the heterologous expression of the T26 α-galactosidase gene in Escherichia coli BL21 (DE3). The optimum pH for α-galactosidase T26GAL was determined to be 8.0, while the optimum temperature was 60℃. In addition, T26GAL demonstrated a remarkable thermostability with more than 93% enzyme activity, even at a high temperature of 90℃. Furthermore, Ca2+ and Mg2+ promoted the activity of T26GAL while Zn2+ and Cu2+ inhibited it. The substrate specificity studies revealed that T26GAL efficiently degraded raffinose, stachyose, and guar gum, but not locust bean gum. This study thus facilitated the discovery of an effective heat-resistant α-galactosidase with potent industrial application. Meanwhile, as part of our research on lignocellulose degradation by a microbial consortium, the present work provides an important basis for encouraging further investigation into this enzyme complex.
Fusarium root rot caused by the soil-borne fungus Fusarium solani is one of the most important fungal diseases of cassava in Thailand, resulting in high yield losses of more than 80%. This study aimed to investigate if the exogenous application of salicylic acid formulations (Zacha) can induce resistance in cassava against Fusarium root rot and observe the biochemical changes in induced cassava leaf tissues through synchrotron radiation based on Fourier-transform infrared (SR-FTIR) microspectroscopy. We demonstrated that the application of Zacha11 prototype formulations could induce resistance against Fusarium root rot in cassava. The in vitro experimental results showed that Zacha11 prototype formulations inhibited the growth of F. solani at approximately 34.83%. Furthermore, a significant reduction in the disease severity of Fusarium root rot disease at 60 days after challenge inoculation was observed in cassava plants treated with Zacha11 at a concentration of 500 ppm (9.0%). Population densities of F. solani were determined at 7 days after inoculation. Treatment of the Zacha11 at a concentration of 500 ppm resulted in reduced populations compared with the distilled water control and differences among treatment means at each assay date. Moreover, the SR-FTIR spectral changes of Zacha11-treated epidermal tissues of leaves had higher integral areas of lipids, lignins, and pectins (1,770-1,700/cm), amide I (1,700-1,600/cm), amide II (1,600-1,500/cm), hemicellulose, lignin (1,300-1,200/cm), and cellulose (1,155/cm). Therefore, alteration in defensive carbohydrates, lipids, and proteins contributed to generate barriers against Fusarium invasion in cassava roots, leading to lower the root rot disease severity.
