A recent report showed that analysis of CD154 expression in the presence of the secretion inhibitor Brefeldin A (Bref A) could be used to assess the entire repertoire of antigen-specific $CD4^+\;T$ helper cells. However, the capacity of intracellular CD154 expression to identify antigen-specific $CD8^+\;T$ cells has yet to be investigated. In this study, we compared the ability of intracellular CD154 expression to assess antigen-specific $CD8^+\;T$ cells with that of accepted standard assays, namely intracellular cytokine IFN-${\gamma}$ staining (ICS) and MHC class I tetramer staining. The detection of intracellular CD154 molecules in the presence of Bref A reflected the kinetic trend of antigen-specific $CD8^+\;T$ cell number, but unfortunately showed less sensitivity than ICS and tetramer staining. However, ICS levels peaked and saturated 8 h after antigenic stimulation in the presence of Bref A and then declined, whereas intracellular CD154 expression peaked by 8 h and maintained the saturated level up to 24 h post-stimulation. Moreover, intracellular CD154 expression in antigen-specific $CD8^+\;T$ cells developed in the absence of $CD4^+\;T$ cells changed little, whereas the number of IFN-${\gamma}$-producing $CD8^+\;T$ cells decreased abruptly. These results suggest that intracellular CD154 could aid the assessment of antigen-specific $CD8^+\;T$ cells, but does not have as much ability to identify heterogeneous $CD4^+\;T$ helper cells. Therefore, the combined analytical techniques of ICS and tetramer staining together with intracellular CD154 assays may be able to provide useful information on the accurate phenotype and functionality of antigen-specific $CD8^+\;T$ cells.
This study was undertaken to investigate the effect of indomethacin on the distribution of Langerhans cells and T-lymphocytes related with immune response of 4-Nitroquinoline-1-oxide induced carcinogenesis at the palate and tongue of albino rat. 54 Sprague-Dawley strain 10 weeks old albino rats, about 150gm weighted, divided into a normal group of 6 rats without treatment, a control group of 12 rats given indomethacin, a carcinogenesis group of 18 whose palatal mucosa were appiled with 4-Nitroquinoline-1-oxide three times a week, and experimental group of 18 rats were treated with indomethacin and whose palatal mucosa were applied 4-Nitroquinoline-1-oxide. All these 54 rats were subjected to be observed as being ATPase stained specimens, specimens for the observation of light and electron microscope, and T-lymphocyte stained specimens. The obtained results were summarized as follows; 1. In carcinogenesis group, proliferation of epithelial layer and rete peg were observed early period of the experiment and showed parakeratosis, individual cell keratinization, acanthosis, and lymphocyte infiltration from 13th week of the experiment on lightmicroscopically, while experimental group showed less reaction than that of carcinagenesis group. 2. The number of Langerhans cells in normal group rarely changed until 21st week of the experiment, while the Langerhans cells increased markedly from 3rd week of the experiment in control group. 3. The number of Langerhans cells were decreased markedly and persistantly until 21st week of the experiment both in carcinogenesis and experimental groups. 4. Appearance of the T-helper cells and T-suppressor cells were minimal and irregullar in number both in normal and control groups. Thus it is assumed that administration of indomethacin and distribution of Langerhans cells showed close relation. 5. In carcinogenesis and experimental groups, the number of the T-helper cells was apparently inereased than that of the T-suppressor cells, but increasing pattern in experimental group was less than in carcinogenesis group. These cells increased most in the 21st week, decreased from the 23rd week and the appearance of these cells were irregular in general throughout the experiment.

When it uses the elementary student safe helper system which we propose before the accident occurs, it will grasp the state information of the student and it will be able to prevent an accident in advance. We will try to more efficient safe guard to elementary school student by real-time monitoring skill soon.
We studied the T lymphocyte and its subpopulation percentage change in 40 patients immediately after the radiation therapy. Study population consisted of 12 patients treated at the site of head and neck region,14 patients treated at the site of thoracic region, and 14 patients treated at the site of pelvic region. Twenty two patients received radiotherapy as radical modality, and remaining 18 patients received radiotherapy as postoperative modality. Immediately after radiotherapy, total T lymphocyte (T1) percentage was decreased from $56.4\%$ to $55.2\%$, helper T cell (T4) percentage was decreased from $36.4\%$ to $34.1\%$, but suppressor T lymphocyte (T8) percentage was increased from $23.5\%$ to $25.4\%$. As a result, T4/T8 ratio was decreased from 1.57 to 1.39. This study suggested that immediate change after radiotherapy of the T lymphocyte and its subpopulation percentage was not related to the treatment volume and the degree of helper T cell decrement was not pronounced by the radiation dose increment. Long-term follow-up study En larger scale is needed to determine long term changing pattern in T lymphocyte subpopulation and its relationship to the prognosis of patients.
Purpose: Lymphocyte subset recovery is an important factor that determines the success of hematopoietic stem cell transplantation (HSCT). Temporal differences in the recovery of lymphocyte subsets and the factors influencing this recovery are important variables that affect a patient's posttransplant immune reconstitution, and therefore require investigation. Methods: The time taken to achieve lymphocyte subset recovery and the factors influencing this recovery were investigated in 59 children who had undergone HSCT at the Department of Pediatrics, The Catholic University of Korea Seoul St. Mary's Hospital, and who had an uneventful follow-up period of at least 1 year. Analyses were carried out at 3 and 12 months post-transplant. An additional study was performed 1 month post-transplant to evaluate natural killer (NK) cell recovery. The impact of pre- and post-transplant variables, including diagnosis of Epstein-Barr virus (EBV) DNAemia posttransplant, on lymphocyte recovery was evaluated. Results: The lymphocyte subsets recovered in the following order: NK cells, cytotoxic T cells, B cells, and helper T cells. At 1 month post-transplant, acute graft-versus-host disease was found to contribute significantly to the delay of $CD16^+/56^+$ cell recovery. Younger patients showed delayed recovery of both $CD3^+/CD8^+$ and $CD19^+$ cells. EBV DNAemia had a deleterious impact on the recovery of both $CD3^+$ and $CD3^+/CD4^+$ lymphocytes at 1 year post-transplant. Conclusion: In our pediatric allogeneic HSCT cohort, helper T cells were the last subset to recover. Younger age and EBV DNAemia had a negative impact on the post-transplant recovery of T cells and B cells.
Objective : This study was undertaken to investigate effects of Opuntia ficus-indica (OP)extract on an asthma murine model. Methods : The total number of cells and eosinophils in BALF and the infiltration of inflammatory cells into lung tissues were determined by hematoxylin and eosin staining. Results : The number of OVA-induced total cells and eosinophils, a phenomenon of asthma, were decreased by treatment of the animals with OP extract (200 mg/kg) (respectively, p < 0.001 and p<0.01). Furthermore, we showed that OP extract reduced the increased immune cell infiltration induced by ovalbumin (p < 0.01). The levels of interleukin (IL)-4 in BAL fluid were measured by enzyme-linked immunosorbent assay, because the eosinophilia is associated with a T helper (Th) 2 response including IL-4. The level of OVA-induced IL-4 was decreased by OP extract in BAL fluid (p < 0.05). We investigated whether OP extract reduced nitrite (NO) production on lipopolysaccharide (LPS)-stimulated RAW 264.7 sells, because asthma is an inflammatory disease. The level of LPS-induced NO production was decreased by OP extract (50, 100 and 200 mg/ml) on RAW 264.7 cells (respectively, p < 0.05, p<0.01 and p<0.05). Conclusions : Our results indicate that OP extract has an inhibitory effect on lung eosinophilia of asthma and suggest that OP extract is a therapeutic candidate in the treatment of inflammatory disease, including asthma.

Objectives : This trial aimed to determine if Wasong (Orostachys japonicus) extract can enhance immune system and is safe enough to be approved as a health functional food. Methods : Total 62 people, aged 45 and older, will be recruited to participate in a randomized, double-blind, placebo-controlled clinical trial. This study will compare Wasong extract and placebo. Wasong group will take 1g of Wasong extract, once a day, for 8 weeks. Placebo group will take 1g of crystalline cellulose as placebo, once a day, for 8 weeks. Outcomes will be measured at the baseline, the end of 4th week, and 8th week. Primary outcomes are the ratio of NK cells/total lymphocytes and the ratio of T-helper cells/T-suppressor cells. Secondary outcomes are total white blood cell count, the ratio of neutrophils, lymphocytes, and monocytes in total leukocytes, the ratio of total T cells, T-helper cells, T-suppressor cells, and B cells to lymphocytes, the amount of blood IgM, IgG, IgA, and cytomegalovirus (CMV) IgG, and blood metabolite target &global analysis. Results : This trial was approved by institutional review board of Pusan National University Korean Medicine Hospital (registry number: 2016006), and registered in Clinical Research information Service, one of WHO International Clinical Trials Registry Platform (registry number: PRE20161006-002). Recruitment opened in February 2017 and is supposed to be completed by August 2017. The result is expected to be published by June 2018. Conclusion : This trial will provide clinical information to determine the efficacy and safety of Wasong in enhancing immune system of middle-aged and older people.
A rise in the occurrence of allergic diseases is attributed to the dysregulated balance of type 1/type 2 immunity, where type 2 T-helper (Th2) cells predominate over type 1 T-helper (Th1) cells, leading to an abnormally increased production of IgE in response to unharmful antigens. Kimchi, a traditional Korean fermented food, is a rich source of beneficial lactic acid bacteria. In this study, we investigated the ability of Enterococcus faecium FC-K derived from kimchi to induce type I immunity in the presence of Th2 polarizing conditions in vitro and in vivo. Stimulation of mouse peritoneal macrophages with E. faecium FC-K induced the production of tumor necrosis factor alpha, interleukin (IL)-6, and IL-12. Under the in vitro Th2 conditions in which splenic T cells were activated in the presence of IL-4, E. faecium FC-K enhanced the ability of T cells to produce interferon $(IFN)-{\gamma}$. Using the ovalbumin (OVA)-induced allergy model, male BALB/c mice receiving E. faecium FC-K reduced the serum level of total IgE, but not that of OVA-specific IgE. Furthermore, the population of activated splenic B cells during OVA immunization was decreased in E. faecium FC-K-treated mice, accounting for a reduction of total IgE in the serum. Restimulating splenocytes from OVA-immunized mice with OVA ex vivo resulted in an increased production of $IFN-{\gamma}$, with no effect on IL-4, in E. faecium FC-K-treated mice. These observations provide the evidence that E. faecium FC-K can be a beneficial probiotic strain that can modulate the Th2-mediated pathologic response.
This study was to investigate the effects of interpersonal relationships on job satisfaction of home-visiting care helpers and the category of interpersonal relationships was subdivided into elderly clients, guardians, colleagues and directors considering the person-to-person nature of home-visiting care helper's job characteristics. A group of 336 care helpers who engaged in home-visiting care service providing facilities in Daejeon City was selected as a research subject, and the data were analyzed with SPSS 22.0 program using reliability, frequency, correlation and hierarchical multiple regression. The results for the final analysis revealed that the effect of socio-demographic features was not significant while, among characteristics of job performances, work priority, turnover experience, and career choice reasons turned out to have significant impacts on job satisfaction. In particular, interpersonal relationships except the one with guardians were found to be the most influential variables and the effects of those relationships on job satisfaction were significant. The influencing power was stronger in relationships with director, elderly clients, and colleagues in order, and the job satisfaction level was higher as they were in better relationships with director, elderly clients, and colleagues. The results of this research shows that it is necessary to set an atmosphere where harmonious interpersonal relationships can be formed inside and outside of facilities in order to improve job satisfaction of home-visiting care helpers.
As the Internet and mobile services increase, the use of wired/wireless web contents services increase and the demand for various contents explosively grows. To survive in competitive market, and to minimize the errors and warnings for web accessibility and standardization, and then to maximize the web usability, the periodical evaluation for web site should be performed with the events of web marketing and campaign. Through the web evaluation, the errors for technical programming language and contents offering can be found and diagnosed. In this paper, the quantitative and qualitative evaluation method for web site providing web contents are presented, and the analytic results for the 138 home pages in domestic are evaluated to validate the quantitative methodology.
