MicroRNA-orchestrated pathophysiologic control in gut homeostasis and inflammation

  • The intestine represents the largest and most elaborate immune system organ, in which dynamic and reciprocal interplay among numerous immune and epithelial cells, commensal microbiota, and external antigens contributes to establishing both homeostatic and pathologic conditions. The mechanisms that sustain gut homeostasis are pivotal in maintaining gut health in the harsh environment of the gut lumen. Intestinal epithelial cells are critical players in creating the mucosal platform for interplay between host immune cells and luminal stress inducers. Thus, knowledge of the epithelial interface between immune cells and the luminal environment is a prerequisite for a better understanding of gut homeostasis and pathophysiologies such as inflammation. In this review, we explore the importance of the epithelium in limiting or promoting gut inflammation (e.g., inflammatory bowel disease). We also introduce recent findings on how small RNAs such as microRNAs orchestrate pathophysiologic gene regulation.

Effect of mushroom (Schizophyllum spp.) derived β-glucan on low-fiber diet induced gut dysbiosis

  • Dietary pattern has paramount importance in shaping the gut microbiota and its associated host health. Herein this study, long term (12 weeks) impact of mushroom derived dietary fiber, ${\beta}-glucan$, is investigated for its effect on low fiber diet consumption. Inclusion of dietary fiber into the low fiber diet (LFD) increased the abundance of genera Lactobacillus and Anaerostipes, the microbes responsible for butyrate (major 'fuel source' of colonocytes) production. Mice fed LFD with ${\beta}-glucan$ showed significant increase in the length of small intestine compared to that of the LFD group without ${\beta}-glucan$. Further, dietary fiber consumption enhanced goblet cell density along with mucosal layer thickness. These results indicate promising effects of ${\beta}-glucan$ towards maintenance of healthy gut and gut microbiota.

  • Plasmacytoid dendritic cells (pDCs) are a unique subset of cells with different functional characteristics compared to classical dendritic cells. The pDCs are critical for the production of type I IFN in response to microbial and self-nucleic acids. They have an important role for host defense against viral pathogen infections. In addition, pDCs have been well studied as a critical player for breaking tolerance to self-nucleic acids that induce autoimmune disorders such as systemic lupus erythematosus. However, pDCs have an immunoregulatory role in inducing the immune tolerance by generating Tregs and various regulatory mechanisms in mucosal tissues. Here, we summarize the recent studies of pDCs that focused on the functional characteristics of gut pDCs, including interactions with other immune cells in the gut. Furthermore, the dynamic role of gut pDCs will be investigated with respect to disease status including gut infection, inflammatory bowel disease, and cancers.

The effects of plant extracts on lipid metabolism of chickens - A review

  • The fat deposition is an important factor affecting chicken meat quality, which is closely related to lipid metabolism of chickens. Therefore, it is important to regulate the lipid metabolism of chickens to improve the chicken meat quality. Plant extracts have special regulatory effects on animal's growth and health and have been widely used in chicken breeding. Some plant extracts have been reported to have functions of changing the fatty acid composition, reducing abdominal fat percentage, and enhancing the intramuscular fat content of chickens by improving the antioxidant capacity, regulating the expression of genes, enzymes, and signaling pathways related to lipid metabolism, modulating intestinal microbiota, affecting hormones level, and regulating DNA methylation. This paper reviewed the application and mechanism of plant extracts on regulating lipid metabolism of chickens to provide a reference for the further application of plant extracts in chicken breeding.

Potential role of phytochemicals in brain plasticity: Focus on polyunsaturated fatty acids

  • [Purpose] Functional foods are thought to strongly influence the structure and function of the brain. Previous studies have reported that brain-boosting diets may enhance neuroprotective functions. Certain foods are particularly rich in nutrients like phytochemicals that are known to support brain plasticity; such foods are commonly referred to as brain foods. [Methods] In this review, we briefly explore the scientific evidence supporting the neuroprotective activity of a number of phytochemicals with a focus on phenols and polyunsaturated fatty acids such as flavonoid, olive oil, and omega-3 fatty acid. [Results] The aim of this study was to systematically examine the primary issues related to phytochemicals in the brain. These include (a) the brain-gut-microbiome axis; (b) the effects of phytochemicals on gut microbiome and their potential role in brain plasticity; (c) the role of polyunsaturated fatty acids in brain health; and (d) the effects of nutrition and exercise on brain function. [Conclusion] This review provides evidence supporting the view that phytochemicals from medicinal plants play a vital role in maintaining brain plasticity by influencing the brain-gut-microbiome axis. The consumption of brain foods may have neuroprotective effects, thus protecting against neurodegenerative disorders and promoting brain health.

  • Advanced and innovative breeding and management of meat-producing animals are needed to address the global food security and sustainability challenges. Beef production is an important industry for securing animal protein resources in the world and meat quality significantly contributes to the economic values and human needs. Improvement of cattle feed efficiency has become an urgent task as it can lower the environmental burden of methane gas emissions and the reduce the consumption of human edible cereal grains. Cattle depend on their symbiotic microbiome and its activity in the rumen and gut to maintain growth and health. Recent developments in high-throughput omics analysis (metagenome, metatranscriptome, metabolome, metaproteome and so on) have made it possible to comprehensively analyze microbiome, hosts and their interactions and to define their roles in affecting cattle biology. In this review, we focus on the relationships among gut microbiome and beef meat quality, feed efficiency, methane emission as well as host genetics in beef cattle, aiming to determine the current knowledge gaps for the development of the strategies to improve the sustainability of beef production.

  • Despite considerable advancements achieved using next-generation sequencing technologies in exploring microbial diversity, several species of the gut microbiome remain unknown. In this transformative era, culturomics has risen to prominence as a pivotal approach in unveiling realms of microbial diversity that were previously deemed inaccessible. Utilizing innovative strategies to optimize growth and culture medium composition, scientists have successfully cultured hard-tocultivate microbes. This progress has fostered the discovery and understanding of elusive microbial entities, highlighting their essential role in human health and disease paradigms. In this review, we emphasize the importance of culturomics research on the gut microbiome and provide new theories and insights for expanding microbial diversity via the optimization of cultivation conditions.

  • Kimchi is a traditional Korean dish made with salted fermented vegetables and contains various nutrients and functional substances with potential health benefits. The fermentation process used to make kimchi creates chemical changes in the food, developing nutrients and functional substances that are more easily absorbed and enhanced by the body. Recent studies have shown that several lactic acid bacteria strains isolated from kimchi exhibit probiotic properties and have several health benefiting properties such as such as anticancer, anti-obesity, and anti-constipation; they also promote colon health and cholesterol reduction in in vitro and in vivo experiments, as well as in epidemiological cohort studies. Kimchi contains prebiotics, non-digestible fibers that nourish beneficial gut bacteria; therefore, its intake effectively provides both probiotics and prebiotics for improved gut health and a fortified gut-derived immune system. Furthermore, fermentation of kimchi produces a variety of metabolites that enhance its functionality. These metabolites include organic acids, enzymes, vitamins, bioactive compounds, bacteriocins, exopolysaccharides, and γ-aminobutyric acid. These diverse health-promoting metabolites are not readily obtainable from single food sources, positioning kimchi as a valuable dietary option for acquiring these essential components. In this review, the health functionalities of kimchi ingredients, lactic acid bacteria strains, and health-promoting metabolites from kimchi are discussed for their properties and roles in kimchi fermentation. In conclusion, consuming kimchi can be beneficial for health. We highlight the benefits of kimchi consumption and establish a rationale for including kimchi in a balanced, healthy diet.

  • Recent gut microbiota studies have revealed the important roles of gut microbiota for our health. Increasing numbers of health functional foods have been developed every year. Development of functional food often includes ex- and in-vivo experiment to verify the beneficial effects of the functional food. To investigate effects of functional food on gut microbiota, animal models were often conducted. Beneficial effects of food can be evaluated based on how gut microbiota was shifted by food, which results in either increase in beneficial bacteria, decrease in potentially pathogenic bacteria or both. As animal experiments are generally time-consuming and laborious, we investigate how well in-vitro investigation of fecal microbiota may reflect dietary health benefits. Here, we tested 15 kinds of diets using two human subjects' fecal materials. Our results showed varying gut microbiota shifts according to diets, which suggested generally known beneficial diets (i.e. Kimchi, Chunggukjang) increased Lactobacillus and Bifidobacterium. Therefore, we suggest that in vitro fecal microbiota analysis could be used to evaluate beneficial effects of diets. Moreover, this method may be ideal to establish personalized diet.
