Colony Count with Mixed Culture of Enteric Bacteria by in vitro Quantitative Method (장내세균의 시간차 혼합배양이 보여주는 균수측정의 비교)

  • This study was attempted to see more clear relationships among the enterobacteria, especially between the intestinal normal flora and pathogenic bacteria. It has been known that some intestinal normal flora produce the bactrial metabolites that are harmful to other enteric bacteria. One of the metabolites is known as colicin, the protein fraction, which possesses certain degree of inhibitory effect against other bacterial growth fraction, whih possesses certain degree of inhibitory effect against other bacterial growth. As a preliminary study for a colicin purification, the antagonistic effect of E, coli to groups of Salmonella and Shigella has been studied by means of in vitro quantitative culture method. 1. E.coli showed definite inhibitory effects aganist both Salmonella and Shigella groups in the mixture of two organisms. 2. The inhibitory effects of E.coli in the E.coli-Salmonella and the E.coli-Shigella mixture occurred from 4 hours incubation following the inoculation. 3. Even the complete inhibition of pathogenic enteric bacterial growth was noticed in the E.coli-Salmonella mixture at overnight incubation. 4. Among the diluted mixtures, 1:100, 1:1,000, and 1:10,000, survival rate of pathogenic enteric bacteria in the mixtures with E.coli showed least affected at the 1:1,000 dilution. 5. It was found that the antagonistic effect aganist groups of Salmonella-shigella was depending upon the groups of the genera.

Synthesis of 2-Thio-4-aminopyrimidine Derivatives as Anti-cancer Agent

  • The screening of the anti-cancer activity of the chemical library provided 2-thio-4-aminopyrimidine as the initial hit. The confirmation of structure and biological effect of hit was performed by synthesis and biological evaluation. The optimization of hit was performed by derivatization of substituents while keeping the core structure. The evaluation of growth inhibitory activity was carried out using SRB assay against 6 human cancer cell lines and human fibroblast. The growth inhibitory activity of compounds showed substituent dependency and more than 5 compounds showed complete growth inhibition of cancer cell lines at 10 ${\mu}M$ concentration. Chemical library screening followed by synthetic modification provided possibility that 2-thio-4-aminopyrimidine can be used as a new scaffold for the development of anti-cancer agent.

Study of the Mechanism for the Growth Inhibitory Effects of Conjugated Linoleic Acid on Caco-2 Colon Cancer Cells (Conjugated Linoleic Acid에 의한 대장암 세포 증식 억제 기전 연구)

  • Conjugated linoleic acid (CLA) is a group of positional and geometric isomers of linoleic acid (LA) and exhibits anticarcinogenic activity in a variety of animal models. We have previously observed that CLA inhibited the growth of Caco-2 cells, a human colon adenocarcinoma cell line. The present study was performed to determine whether the growth inhibitory effect of CLA is related to change in secretion of IGF- II and/or IGF-binding proteins (IGFBPs) that have been shown to regulate Caco-2 cell proliferation by an autocrine mechanism. Cells were incubated in serum-free medium with various concentrations of CLA or linoleic acid (LA). Immunoblot analysis of 24-hours, serum-free, conditioned medium using a monoclonal anti-IGF-IIantibody revealed that Caco-2 cells secreted both mature 6,500 Mr and higher Mr forms of pro IGF-II. The levels of pro IGF-II and mature IGF-IIwere decreased by 43 $\pm$ 2% and 53 $\pm$ 6%, respectively by treatment with 50 $\mu$ M CLA. LA slightly increased pro IGF- II levels. Results from Northern blot analysis showed that CLA decreased IGF-II mRNA levels at 50 $\mu$ M concentration suggesting that CLA regulation of IGF-II protein expression occurs partly at the transcriptional level. Ligand blot analysis of conditioned media using 1251-IGF-II revealed that CLA slightly decreased IGFBP-2 levels and increased IGFBP-4 levels. We confirmed our previous results that CLA inhibited cell growth in a dose-dependent manner but LA slightly increased cell growth. Exogenous IGF-II mitigated the growth inhibitory effect of CLA. These results indicate that the growth inhibitory effect of CLA may be at least in part mediated by decreasing IGF-II and IGFBP-2 secretion and increasing IGFBP-4 secretion in Caco-2 cells.

The Growth Inhibition against Gastric Cancer Cell in Germanium or Soybean Sprouts Cultured with Germanium (게르마늄 및 게르마늄 분말 용해수로 재배한 콩나물의 위암세포 성장억제 작용)

  • The growth inhibitory effect of germanium, or soybean sprouts cultured with germanium, on cancer cells was determined in the cultured gastric cancer cell line, AGS. The growth of AGS was significantly inhibited by the addition of 0.01-1% organic germanium (Ge-132) and germanium stone powder in MTT cytotoxicity assays. The juice from germanium treated soybean sprouts (GTS) inhibited the growth of AGS gastric cancer cells by 78-88% at concentrations of 2.5 or 5${\mu}\ell$. The juice from Seomoktae GTS revealed an especially higher growth inhibitory effect than that from the control soybean sprouts (germanium non-treated soybean sprouts, GNTS) in AGS. The results suggest that soybean sprouts cultured with germanium may exert an anticancer effect against gastric cancer cells.

Quinone Reductase Inductive Activity and Growth Inhibitory Effect against Hepatoma Cell of Oriental Melon Extract (참외 추출물의 Quinone Reductase 유도활성 및 간암세포 증식 억제효과)

  • This study was performed to elucidate anticancer activities of various parts, such as peel, flesh, placenta, seed, stalk and stem leaf of oriental melon. Chemopreventive and anticancer effects of oriental melon extract were evaluated by detoxifying enzyme, quinone reductase (QR) inductive activity, cytotoxicity and growth inhibitory effect against hepatoma cell. Stalk and stem leaf extracts of oriental melon showed the increment of QR inductive activity with dose-dependent manner and induced quinone reductase 3.9, 1.5-fold at $200{\mu}g/mL$ respectively compared to control. The growth inhibitory effect of oriental melon extract against mouse hepatoma cell (Hepa1c1c7) was investigated by crystal violet (CV) assay. Stalk and stem leaf of oriental melon showed potent growth inhibitory effect. Based on these result, the growth inhibitory effects of stalk, stem leaf at various concentration were examined in detail by MTT assay using human hepatoma cancer cell (HepG2). All of two parts showed growth inhibitory effects and expecially stalk exhibited inhibitory effect of 60.3% at maximum concentration. The above results suggest that stalk of oriental melon has a possibility as a source of natural cancer chemopreventive materials.

Fermented Wheat Grain Products and Kochujang Inhibit the Growth of AGS Human Gastric Adenocarcinoma Cells

  • The growth inhibitory effect of wheat grain, fermented wheat grain products, red pepper powder and kochujang was examined in AGS human gastric adenocarcinoma cells. The kochujang samples were prepared by mixing red pepper powder and each fermented wheat grain products, such as first fermented wheat grain (FFWG), second fermented wheat grain (SFWG) and final fermented wheat grain (FiFWG). The methanol extract of FiFWG showed the highest growth inhibitory effects. In order to investigate the effects of fermented wheat grain products at each stage of fermentation on anticancer activity, the kochujang prepared with red pepper powder and each of FFWG, SFWG and FiFWG were fermented until reaching pH 5.1. The kochujang adjusted to pH 5.1 by fermentation increased the growth inhibitory effect; however, the kochujang prepared with FiFWG showed the highest effect. Increased fermentation time increased the growth inhibitory effect of kochujang when prepared with FiFWG. These results suggested that the anticancer effect of the kochujang on the gastric cancer cells was affected by the fermentation periods of the wheat grains. The well-fermented end products of the wheat grains seem to be one of the major components that showed anticancer activity of kochujang.

Production of a Phytotoxic Compound, 3-Phenylpropionic Acid by a Bacterial Endophyte, Arthrobacter humicola YC6002 Isolated from the Root of Zoysia japonica

  • An endophytic bacterial strain, Arthrobacter humicola YC6002, was isolated from a surface sterilized root of Korean turf grass (Zoysia japonica) collected from Jinju, Korea. This strain showed inhibitory effect on germination and shoot growth of radish. The inhibition of germination and shoot growth of radish seeds varied depending on the age of culture and the temperature at which it was incubated. The culture filtrate of 1/10-strength Tryptic Soy Broth medium, incubated for 48 hours at $30^{\circ}C$, showed the highest inhibitory effect on radish seed germination and shoot growth (92% inhibition as compared to control). The active compound with seed germination and shoot growth inhibition was purified and identified as 3-phenylpropionic acid. The purified compound had 53% and 93% inhibitory effect on seed germination and shoot growth of radish for 500 and 1000 ppm solutions, respectively.

The Effect of Serum and Tramsferrin on the Growth of Malassezia pachydermatis in vitro (In vitro에서의 Malassezia pachydermatis의 성장에 대한 혈청과 Transferrin의 억제효과)

  • The inhibitory effect of pooled canine serum on the growth of ten strains of Malassezia pachydermatis in vitro was investigated. Studies were also carried out to observe the effect of different concentrations of unsaturated bovine transferrin on Malassezia growth in vitro. Ten strains of Malassezia pachygedrmatis in normal canine serum (11.1%, 44.4%) were found to significantly inhibit the growth (p<0.0005) not only in a dose dependent but also in a time dependent manner. The same strains of yeast treated with 1, 2, 4 and 8 times the normal value of serum transferrin (3.0 mg/ml, 6.0 mg/ml, 12.0 mg/ml, 24.0 mg/fl), were shown to have significantly lower OD readings (p<0.05) when compared to yeast treated in lower concentrations of transferrin (1.5 mg/ml). The optical density (OD) of the ten strains of yeast were significantly lower (p<0.005) when treated with various concentrations of transferrin than with the saline control except at 72 hours post incubation. These results indicate that serum has inhibitory effects on Malassezia pachydermatis growth in vitro, and transfferin is one of the components that contribute towards this inhibitory role. The inhibitions are dose and time dependent.

파두가대황(巴豆加大黃)이 항종양작용(抗腫瘍作用)과 자연살해세포(自然殺害細胞)의 활성(活性)에 미치는 영향에 대한 실험적 연구

  • This experiment was designed to study the antitumor effects and Activity of Natural Killer Cell of semen Tiglii plus Rhizoma Rhei. The cytotoxic and antitumor effects were evaluated on human cell lines(A549, Caki-1, LL2, Sarcoma 180, NIH/3T3) after exposure to prebrewed Semen Tiglii plus Rhizoma Rhei water extract 0.1, 0.2, 0.4, 0.8, 1.6mg/ml using in MTT assay, LDH, colony forming efficency and SRB assay which were regarded as a valuable method for cytotoxic and antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows. 1. From the result of MTT assay, the cytotoxicity of ST(生巴豆霜), ST+RR(生巴豆霜加大黃) were concentration-dependently increased in both group of the ST and ST+RR, the cytotoxicity of ST+RR(生巴豆霜加大黃) was similar to that of ST(生巴豆霜). 2. From the result of LDH, the cytotoxicity of ST, ST +RR were concentrati -on-dependently increased in both group of the ST and ST+RR, the cytotoxicity of ST+RR was similar to that of ST. 3. The antitumor effect on A549 tumor cell from the result of colony forming efficiency showed the inhibitory effect on the growth in both group of the ST and ST+RR, the inhibitory effect on growth was low slightly in the ST+RR. 4. From the result of SRB assay, the antitumor effect on caki-1 tumor cell of ST, ST+RR showed the inhibitory effect on the growth in both group of the ST and ST+RR, the antitumor effect of ST+RR was similar to that of ST. 5. Median survival time and increased life span were increased slightly in both group of the ST and ST+RR. 6. The inhibitory effect on the growth of Sarcoma 180 and Lewis lung carcinoma tumor cell were increased slightly in both group of the ST and ST+RR. 7. The activity of NK cell was increased in the ST+RR.

Growth Inhibitory Patterns by Adenoviral p16 Transduction in HCC Cell Lines with Different pRB Status

  • To evaluate the diagnostic significance of p16 overexpression in human hepatocellular carcinoma (HCC), we analyzed p16 status and growth inhibitory patterns by p16 overexpression in HCC cell lines having different pRE status. SKHep1 and SNU449 cells show homozygous deletion of p16. The p16 gene in SNU398 cell is inactivated at posttranscription level. Adenovira1-p16 (Ad-p16) infection inhibits the cell growth in Hep3B, SNU398, and SNU449. Failure of growth inhibition in SKHepl results from the low transduction efficiency of adenovirus. The p16-mediated growth inhibition shows G 1 phase arrest in pRE-positive SNU449 but not in pRE-negative Hep3B. These results suggest that therapeutic efficacy of p16 gene might be considered on the transduction efficiency and the toxicity of adenoviral vector. Beside, growth inhibitory effect of p16 could be exerted through either pRE-dependent or -independent pathway.
