The purpose of this study was to investigate the difference in perceptions between gardens and park functions as recognized by two groups, Group A and Group B, in order to confirm the distinction between concepts and functions and then establish the importance of individual functions. The AHP was used to analyze the importance of each group's perceptions by dividing them into garden and park, Group A and non-Group A, respectively. In Group A, the importance of garden functions were considered in descending order of importance to be cultural function, ecological function, and social function. In the general group, ecological function, cultural function, and social function also appeared, but in a different order of importance. As for the park functions, Group A recognized the importance of functions in a similar order of importance to the gardens: cultural function, ecological function, and social function. Group B thought that social function, ecological function, and cultural function have the same significance. At the major classification level, Group A and Group B emphasized the social function of the parks. Group A recognized the importance of the garden's cultural function as the most important, whereas the general group emphasized the importance of the garden's ecological function. As for the mid-class level, Group A recognized the aesthetic beauty, health, ecological environment protection, and water circulation as important functions of the garden. For Group B, the ecological environment protection, aesthetic beauty, water cycle, and health were important. The concepts and functions of gardens and parks are still largely mixed but are gradually becoming differentiated. As a follow-up study, it is important to systematically manage the functions of gardens by establishing design, construction, and monitoring DB techniques for the garden type and examine the hierarchy of various other gardens.
A mixed solution of $^{54}Mn$, $^{60}Co$, $^{85}Sr$ and $^{137}Cs$ was applied to the soil of culture boxes in a greenhouse 2 days before transplanting red pepper and at 3 different times during its growth for investigating transfer factors ($m^2/kg-dry$) for its green and red fruits. Transfer factors varied with radionuclide, application time and harvest time by factors of about $20{\sim}100$. They decreased mostly in the order of $^{85}Sr>^{54}Min>^{60}Co>^{137}Cs$ while $^{54}Mn$ and $^{60}Co$ was higher than $^{85}Sr$ when time lapse between application and harvest was short. Transfer factors of $^{85}Sr$ and $^{137}Cs$ at the last application were lower than those at the previous one by factors of $3{\sim}20$ depending on harvest time. Variations in $^{54}Mn$ and $^{60}Co$ transfer factors with application time after transplanting were comparatively low. Transfer factors of $^{54}Mn$, $^{60}Co$ and $^{85}Sr$ mixed with topsoil before transplanting were up to $3{\sim}9$ times higher than those for the application onto soil surface 2 days after transplanting while there was no difference in $^{137}Cs$. The present results can be referred to in estimating root-uptake concentrations of the radionuclides in red pepper fruit and taking proper measures for its harvest and consumption at the event of an accidental release during the growing season of red pepper.

The effect of the weight ratio of carbon source to nitrogen source on the growth and the composition change of fatty acids of Thraustochytrium aureum ATCC 34304 was investigated. The cell concentration of 5 days' culture increased and then decreased according to the increase of C/N, The ratioes for the maximum biomass were unique respectively and distributed between 1 and 4 with the initial sugar concentration in the range of 5 g/L to 25 g/L. The biomass yield, $Y_{x/s}$ decreased along with the increase of C/N, but maintained a constant value 0.35 between 10 and 20. The composition of myristic acid $(C_{14:0})$, one component of the lipid synthesized by T. aureum, was not affected with the change of C/N, but the composition of palmitic acid $(C_{16:0})$ was around $20\%$ below 4 of C/N and decreased to $15\%$ according to C/N about 4. The compositions of oleic acid $(C_{18:1})$ and linoleic acid $(C_{18:2})$ increased from 0 to $20\%\;and\;7\%$ respectively. The composition of $\gamma-linoleic$ acid $(C_{18:3})$, however, reduced from $5\%\;to\;2\%$. EPA $(C_{20:5})$ and DPA $(C_{22:5})$ showed a tendency of reduction in the weight composition according to the increase of C/N, but DHA $(C_{22:6})$ had a trend maintaining an approximately constant value, around $40\%$, irrespective of the change of C/N.
This study was carried out to investigate into the effect of light-emitting diode (LED) for the light quality as a light source on the broccoli seed germination and the physiological activity of vegetable sprouts. We have also germinated seeds of the broccoli and applied LED as a light quality such as blue, green, red, white, yellow and red + blue color lights to their sprouts for 14 hours and kept dark for 10 hours at the temperature of $25^{\circ}C$ (day)/$18^{\circ}C$ (night). Broccoli sprouts were extracted by methanol and their physiological activities were examined. All broccoli seeds were germinated at 3 days after seeding regardless of the light color. Total sprout fresh weight were mostly became highest by 0.389g (10 plants) at 8 days after seeding when their sprouts were grown under blue color light. Total phenol compound contents in broccoli sprouts were extremely increased by $83.0\;mg{\cdot}L^{-1}$ under the white light, and total flavonoid contents were most much more by $72.6\;mg{\cdot}L^{-1}$ under the blue light. DPPH radical scavenging activity at $2,000\;mg{\cdot}L^{-1}$ were most highest by 93.5% in broccoli sprouts grown under the white light. Nitrite radical scavenging activity at the concentration of $500\;mg{\cdot}L^{-1}$ in sprout extracts were the most increased by 66.9% under the yellow light, and tyrosinase inhibition activity at $2,000\;mg{\cdot}L^{-1}$ in sprout extracts were by 14.5% under red light.
Background: Epigallocatechin-3-gallate (EGCG) is the major catechin in green tea, and has shown antiproliferative, antiangiogenic, antimetastatic and cell cycle pertubation activity in various tumor models. Hypoxia can be induced because angiogenesis is insufficient for highly proliferating cancer. Hypoxia-inducible factor-1$\alpha$ (HIF-1$\alpha$) and its downstream target, vascular endothelial growth factor (VEGF), are important for angiogenesis, tumor growth and metastasis. The aim of this study was to determine how hypoxia could cause changes in the cellular phenomena and microenvironment in a non-small cell culture system and to examine the effects of EGCG on a HIF-1$\alpha$ and VEGF in A549 cell line. Methods: A549 cells, a non-small cell lung cancer cell line, were cultured with DMEM and 10% fetal bovine serum. A decrease in oxygen tension was induced using a hypoxia microchamber and a $CO_2-N_2$ gas mixture. Gas analysis and a MTT assay were performed. The A549 cells were treated with EGCG (0, 12.5, 25, 50 ${\mu}mol/L$), and then examined by real-time-PCR analysis of HIF-1$\alpha$, VEGF, and $\beta$-actin mRNA. Results: Hypoxia reduced the proliferation of A549 cells from normoxic conditions. EGCG inhibited HIF-1$\alpha$ transcription in A549 cells in a dose-dependent manner. Compared to HIF-1$\alpha$, VEGF was not inhibited by EGCG. Conclusion: HIF-1$\alpha$ can be inhibited by EGCG. This suggests that targeting HIF-1$\alpha$ with a EGCG treatment may have therapeutic potential in non-small cell lung cancers.
Purpose: Several studies have proven that EGCG, the primary green tea catechin, and glucosamine-6-phosphate (PGlc) reduce triglyceride contents in 3T3-L1 adipocytes. The objective of this study is to evaluate the combination effect of EGCG and PGlc on decline of accumulated fat in differentiated 3T3-L1 adipocytes. Methods: EGCG and PGlc were administered for 6 day for differentiation of 3T3-L1 adipocytes. Cell viability was measured using the CCK assay kit. In addition, TG accumulation in culture 3T3-L1 adipocytes was investigated by Oil Red O staining. We examined the expres-sion level of several genes and proteins associated with adipogenesis and lipolysis using real-time RT-PCR and Western blot analysis. A flow cytometer Calibar was used to assess the effect of EGCG and PGluco on cell-cycle progression of differentiating 3T3-L1 cells. Results: Intracelluar lipid accumulation was significantly decreased by combination treatment with EGCG $60{\mu}M$ and PGlc $200{\mu}g/m$ compared with control and EGCG treatment alone. In addition, use of combination treatment resulted in directly decreased expression of $PPAR{\gamma}$, $C/EBP{\alpha}$, and SREBP1. In addition, it inhibited adipocyte differentiation and adipogenesis through downstream regulation of adipogenic target genes such as FAS, ACSL1, and LPL, and the inhibitory action of EGCG and PGlc was found to inhibit the mitotic clonal expansion (MCE) process as evidenced by impaired cell cycle entry into S phase and the S to G2/M phase transition of confluent cells and levels of cell cycle regulating proteins such as cyclin A and CDK2. Conclusion: Combination treatment of EGCG and PGlc inhibited adipocyte differentiation through decreased expression of genes related to adipogenesis and adipogenic and cell cycle arrest in early stage of adipocyte differentiation.
Purpose : We tried to assess the optimal conditions to improve low transduction efficiency and their effect on target cells. Methods : Cultured NIH 3T3 cells were incubated with retroviral vectors bearing an enhanced green fluorescent protein (eGFP) gene. We varied the ratio of viral vectors to target cells (1:1-1:8) and the number of transfections (${\times}1$, ${\times}2$), and compared transduction efficiencies. Also, the effects of polybrene on transduction efficiency and viability of target cells were assessed. Transduction of the eGFP gene was evaluated by observing NIH 3T3 cells under a fluorescence microscope and efficiencies were measured by the percentage of eGFP positive cells using FACscan. Results : As the ratio of retroviral vectors to target cells increased, transduction efficiency was greatly improved, from 7% (1:1) to 38% (1:4). However, transduction efficiency did not increase any more when the ratio increased from 1:4 to 1:8. Cells transfected twice showed higher transduction efficiencies than cells transfected once, at a ratio of 1:8. The eGFP gene transduced to NIH 3T3 cells sustained its expression during repeated passages. However, after the third passage (day 9), the percentage of eGFP positive cells began to decline. The degree of this decline in eGFP expression was lower in cells transfected twice than in cells transfected once (P<0.05). The addition of polybrene did not have any toxic effect on NIH 3T3 cells and greatly increased transduction efficiency (P=0.007). In addition to vector component, transduction efficiency was very sensitive to culture confluence. Cells cultured and transfected in 24-well plate showed higher transduction efficiency, although cells cultured in 6- well plate proliferated more (P=0.024). Conclusion : Our data could be used as a basis for retrovirus-based gene therapy. Further study will follow using human cells as target cells.

I-cell disease (mucolipidosis type II; MIM 252500) and pseudo-Hurler polydystrophy (mucolipidosis type III; MIM 252600) are disorders caused by abnormal lysosomal transport in cells. The presence of numerous inclusion bodies in the cytoplasm of fibroblasts, a lack of mucopolysacchariduria, increased lysosomal enzyme activity in serum, and decreased GlcNAc-phosphotransferase activity are hallmark. Here, we attempted to investigate phenotypical and biochemical characteristics of the knockoutmouse of GlcNAc-phosphotransferase ${\alpha}/{\beta}$ subunits; in addition, we also attempted to determine whether the lysosome enriched fraction derived from placenta can be beneficial to phenotype and biochemistry of the knockout mouse. We found that the knockout mouse failed to thrive and had low bone density, as is the case in human. In addition, skin fibroblasts from the animal had the same biochemical characteristics, including increased lysosomal enzyme activity in the culture media, in contrast to the relatively low enzyme activity within the cells. Intravenous injection of the lysosome rich fraction derived from placenta into the tail vein of the animal resulted in a gain of weight, while saline injected animals didn't.In conclusion, our study demonstrated the phenotypical and biochemical similarities of the knockout mouse to a mucolipidosis type II patient and showed the therapeutic potential of the lysosome enriched fraction. We admit that a larger scale animal study will be needed; however, the disease model and the therapeutic potential of the lysosome enriched fraction will highlight the hope for a novel treatment approach to mucopolipidosis type II, for which no therapeutic modality is available.
This study was conducted to get basic information of dry matter production and agronomic characteristics for increasing grain ripening in rice cultivar with large grains. Three cultivars which have different grain size were used. Of the two large grain cultivars, SR7796-3-1-5-1-l(SR7796F$_{7}$) had long culm and SR11269-13-3-4-2(SR11269F$_{6}$) had short culm. On the other hand, Tamjinbyeo was recommended cultivar had medium size of grain and culm. LAI was highest in Tamjinbyeo, followed by the SR11269F$_{6}$ and SR7796F$_{7}$, LAI decrement was higher in large grain cultivars than that of Tamjinbyeo after 20days after heading. Total dry weight(TDW) of areal part was highest in SR11269F$_{6}$, followed by the Tamjinbyeo and SR7796F$_{7}$. Dry weight (DW) of stem (leaf sheath+culm) at harvesting stage was decreased 15%, 12% in Tamjinbyeo, SR11269F$_{6}$, respectively, but was increased 5% in SR7796F$_{7}$ compared to heading stage. In distribution rate of each organ to TDW, distribution rate of leaf blade was low in large grain cultivar, but that of stem in Tamjinbyeo, SR7796F$_{7}$ and SR11269F$_{6}$ was 36%, 38% and 42%, respectively. In canopy architecture leaf blade weight was the highest 30~40cm, 40~50cm and 70~8Ocm above the ground in SR11269F$_{6}$, Tamjinbyeo and SR7796F$_{7}$, respectively. Range of panicle distribution in the canopy was 40cm in Tamjinbyeo, and was 70cm in large-grain cultivars. The results suggest that it has to take a growing interest in developing culture method for increasing LAI during growth stage, especially, around heading, in keeping green leaf till ripening stage and also in growing unformly plant within rice hill in large grain cultivars.cultivars.ltivars.

This paper deals with the historical geography of land-use and agriculture along the Lower Nam-River floodplains. The reclamation process of the river floodplains, the cultivation methods on the reclaimed lands, and the land-use patterns and processes are investigated. The Nam River, one of the major tributaries of the Nakdong River, flows through the boundary between Ham-An and Eu-Ryong Guns. Larger floodplains are located in Ham-An Gun. The floodplains of Ham-An Gun have been surveyed intensively in this study. In South Korea, the alluvial plains, mostly located along the river valleys, have been reclaimed to provide fertile agricultural lands. Those along the upper river valleys were reclaimed before those along the lower river valleys. The flood-plains of Han-An Gun were reclaimed to be the largest agricultural lands of the Gun. The natural levees along the Lower Nam-River Valley were identified before the reclamation processes but now hardly identified. Relatively larger floodplains are located along the tributary streams of the Nam River. Often there are low-lying back swamps between the natural levees and the hills/mountains that rise above the floodplains. The back swamps, called 'natural bog lands' in this region, have been reduced in size and in number through reclamation for the purpose of agricultural and industrial land-uses. Now about ten 'natural bog lands' are found in the Ham-An floodplains, and some of them are being reclaimed for the industrial land-use. This study suggests the emergent need of conservation for the remaining 'natural bog lands' in terms of ecology. Seven agricultural fields of large size, originated from the Nam-River floodplains, are identified in this study: Kun(큰들), Chung-Am(정암들), Chang-chi(장지들), Baek-San(백산들), Ha-Ki(하기들), Gu-Hae(구혜들), and Chang-Po(장포들) fields. The Kun field was reclaimed during the Japanese control and the Gu-Hae, in the 1950s. All of those except the above two fields were reclaimed after the mid-1960s. The Nam-River Dam in Chinju, completed in 1969, contributed the reclamation processes along the Lower Nam-River floodplains. The rice acreage of the region has been reduced slowly since 1970 but the rice production of the region has been relatively stable (Table 4). Rice culture had been the most important agriculture on the reclaimed lands for decades before the greenhouse vegetable cultivation became more important in the 1980s. Among the vegetables cultivated in the greenhouse, the watermelon is the dominantly leading one. Watermelons are usually harvested two or three times in a year though it is possible to harvest four times in one year. The rotation of watermelons and rice is common in the region. It is known the physical conditions of the Nam-River floodplains in Ham-An Gun is the most suitable for watermelon cultivation in South Korea.
