Yacon의 뿌리와 잎의 추출물이 streptozotocin으로 유발된 흰쥐의 항당뇨 효과 (Antidiabetic Effect of Yacon(Smallanthus sonchifolius) of Root and Leaf in Streptozotocin-Induced Diabetic Rats)

  • This study was carried to investigate the antidiabetic effect of ethanol extract of Smallanthus sonchifolius(S.S) in Streptozotocin(STZ)- induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 43mg/kg dissolved in citrate buffer. The ethanol extract of S,S in leaves and root were orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in S.S treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased,and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in S.S treated group compared to the those of STZ-control group, but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was not significantly increased, These results indicated that ethanol extract of S.S would have antidiabetic effect in STZ-induced diabetic rats.

  • This study was done to investigate the antidiabetic and antioxidant effects of Lycii fructus in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Lycii fructus was orally administrated once a day for 7 days. The contents of serum glucose, total cholesterol were significantly decreaed (p<0.05) in Lycii fructus treated group compared to the those of STZ-control group, also content of Triglyceride (TG), Altherogenic index (AI) were decreased, but not statistically significant. But high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were increased in Lycii fructus treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6PDH), glutathione-s-transferase(GST), superoxide dismutase(SOD) were significantly increased(p<0.05), glucose-6-phos-phatase(G-6pase)was significantly decreased(p<0.05) and The glutathione(GSH), glucokinase(GK) were increased. But not statistically significant In Lycii fructus treated group compared to the those of STZ-control group These results indicated that ethanol extract of Lycii fructus would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

Spermatogenic index and hormonal profile in the rats received chromatographic fractions of ethanol extract of Crotalaria juncea L. seeds

  • The ethanol extract of the Crotalaria juncea seeds, which showed promising antispermatogenic and antiandrogenic activities in albino mice, was taken up further for the isolation of the active fractions present in it. Two fractions that were obtained from thin layer chromatography were subjected for testing to know their antispermatogenic and antiandrogenic activities. After preliminary trials the fraction I showed maximum antifertility activity at the dose level of 200 mg/kg body weight when administered orally to the rats for 50 days. The fraction I was found to affect spermatogenesis as well as the endocrine functions of the testis as indicated by gravimetric, histopathological and biochemical changes. Further this fraction has caused degenerative changes in the seminiferous tubules and Leydig cells of the testis. The accessory reproductive organs like epididymis, seminal vesicles, vas deferens, prostrate, Cowper's gland and Levator Ani muscle showed significant malfunction. Cauda epididymal sperm count and sperm motility were reduced significantly. The treatment has also resulted in increase in the cholesterol level and alkaline phosphatase activity, and decrease in protein, glycogen, sialic acid contents and acid phosphatase activity in testis. It is noteworthy that RIA studies have shown significant reduction in serum FSH, LH and testosterone. Scanning electron microscopic observations revealed abnormalities in sperm structure.

Antispermatogenic and antiandrogenic activities of various extracts of Melia azedarach Linn. seeds in albino rats

  • Petroleum ether, chloroform and ethanol extracts of the seeds of Melia azedarach Linn. administered orally to male rats at the dose level of 25 mg/100 g body weight for 48 days showed antispermatogenic activity, as the number of spermatocytes, spermatids and spermatozoa was decreased. The total cholesterol content was increased while protein and glycogen contents were decreased. The acid phosphatase content was also decreased while that of alkaline phosphatase increased. At the same time the weight of caput and cauda epididymis, prostate gland, seminal vesicle and Levator Ani muscle was decreased indicating its antiandrogenic property. Of the three extracts, the petroleum ether extract was more potent in its antispermatogenic and antiandrogenic activities and did not produce any sings of toxicity upto a dose of 25 mg/100 g body weight oral administration. After subjecting it to preliminary phytochemical screening the petroleum ether extract showed positive tests for steroids and saponins.

  • This study was done to investigate the carbohydrate metabolism related enzyme activities and antioxidative effects of Nelumbo nucifera(N.N) Root in streptozotocin (STZ)-induced diabetic rats. The contents of serum glucose, triglyceride (TG) and Total cholesterol were significantly decreaed in N.n treated group compared to the those of STZ-control group. The activity of glucose-6-pase(G-6-Pase) was significantly decreased in N.N treated group. Also the activity of glucokinase(Gk) was significantly increaed in N.N treated group. The content of hepatic glycogen was significantly increaed in N.N treated group, in addition, content of malondialdehyde(MDA) was significanly decreased in N.N treated group. Also, content of glutathione(GSH) was significanly increased in N.N treated froup. whereas, activity of catalase(CAT) was significantly decreaed in N.N treated group compared to the those of STZ-control group. activity of glutathione peroxidase(GSH-Px) was inecreaed. In conclusion, these results indicated that ethanol extract of N.N would have carbohydrate metabolism antioxidative effects in STZ-induced diabetic rats.

레토르트 열처리 조건에 따른 굴자숙수 농축물의 식품성분 특성 비교 (Comparison of Food Component of Oyster Drip Concentrates Steamed under Different Retort Pressures)

  • This study was conducted to investigate the food components of cooking drips from shell oysters steamed under various retort pressures. Among the drips from shell oyster steamed under different retort pressures (1.0, 2.0, 2.5, 3.0, 3.5, 4.0 and 4.5 kg/$cm^2$) the oyster drip obtained at the 4.5 kg/$cm^2$ of retort pressure had the highest degree of brix, yield, crude protein, glycogen and trichloroacetic acid soluble-N contents, while its salinity was the lowest. In the results of food safety test, the presence of E. coli (18 MPN/100 g>) and viable cell (30 CFU/g>) in the oyster drip was in acceptable level as a food-stuff. However, the sensory evaluation such as color, flavor and taste, total amino acid and free amino acid contents of cooking drip from shell oyster steamed at 4.5 kg/$cm^2$ were inferior to those of oyster wash water. Differences in the major amino acids of total amino acid and free amino acid between oyster cooking drip and oyster wash water were also found. The results suggested that the effective use methods of oyster cooking drip should be investigated.

삼채 보충식이가 당뇨흰쥐 간의 항산화효소 함량에 미치는 영향 (Effects of Dietary Supplementation with Allium hookeri Root on Hepatic Enzyme Contents in Streptozotocin-induced Diabetic Rats)

  • 본 연구는 streptozotocin(STZ)으로 당뇨를 유발시킨 Sprague-Dawley계 수컷 흰쥐에게 4주간 삼채(Allium hookeri; AH) 뿌리를 식이에 보충한 정상대조군 normal-control(N-control)과 STZ-control, STZ-AH 5%와 STZ-AH 10% 첨가군을 당뇨실험군으로 하여 삼채의 잠재적인 항산화 식재료로써의 활용성을 확인하였다. 해당식이를 4주간 공급한 후, 간과 근육의 글리코겐, 간의 단백질과 MDA, 간의 세포질에서 XOD, GST, SOD, CAT, GR 및 GPX의 항산화효소 함량을 분석하여 다음과 같은 결과를 얻었다. 간의 글리코겐 함량은 N-control에 비해 STZ-control에서 유의적으로 낮았고, 당뇨실험군에서는 STZ-AH 5%에서 STZ-control보다 유의적으로 높은 함량 차이를 보였다. 근육의 글리코겐 함량은 N-control에 비해 STZ-control에서 유의적으로 낮았고, STZ-control에 비해 STZ-AH 5%와 STZ-AH 10%에서 유의적으로 높은 차이었다. 간의 단백질 함량은 N-control과 STZ-control에서는 유의적인 차이를 보이지 않았으나, STZ-control보다 STZ-AH 첨가군 모두에서 유의적으로 높았다. 간의 MDA 함량은 N-control보다 STZ-control에서 유의적으로 높은 함량이었고, STZ-control보다 STZ-AH 첨가군 모두에서 낮은 함량이었다. 간의 세포질에서 측정한 XOD 함량은 N-control과 STZ-control은 유의적인 차이를 보였고, 당뇨실험군에서는 STZ-control보다 STZ-AH 5%와 STZ-AH 10% 첨가군 모두에서 유의적으로 낮은 함량 차이를 보였다. GST 함량은 N-control에 비해 STZ-control에서는 유의적으로 낮았고, STZ-control 보다 STZ-AH 5%와 STZ-AH 10% 첨가군 모두에서 유의적으로 높은 함량이었다. SOD는 N-control과 STZ-control에서 유의적인 함량 차이를 보였으나, 당뇨실험군 간에는 STZ-control과 유의성을 보이지 않았다. CAT는 N-control과 STZ-control에서 유의적인 차이는 아니었으나, STZ-control과 비교하여 STZ-AH 10%에서 유의적으로 높은 함량 차이를 보였다. GR과 GPX 함량에서는 유의적인 차이를 보이지 않았다. 이상의 연구결과, 삼채뿌리 분말을 식이로 첨가하였을 때 당뇨시 간과 근육의 글리코겐과 간의 단백질 함량이 높아졌고, MDA 함량은 낮아졌다. 효소 함량은 XOD에서 낮아졌고, CAT와 GST에서 높아지는 경향이었다. 삼채는 STZ에 의한 포도당의 과부하에 의해 증가된 과산화물의 생성억제와 조직 내 세포의 산화적 스트레스 감소작용으로 생체내 항산화 방어력을 증가시킬 수 있으며, 당뇨 예방 및 치료를 위한 식사요법에 이용 가능한 식품으로 사료된다.

생약 열수추출물의 근력향상 효능 평가 (Evaluation on the Muscular Strength Activity of Medicinal Herb Hot-Water Extracts)

  • 생약 열수추출물의 근력 향상 효능을 평가하기 위하여 근력 측정 및 혈액의 피로요소를 분석한 결과 간 글리코겐 함량은 모든 식이군들이 대조군에 비하여 높은 수치를 나타내었다. 특히 맥문동, 오미자, 오가피, 함초 식이군의 경우 높은 글리코겐 함량을 나타내었다. 4주간 측정한 근력 향상효능은 오가피, 함초, 홍삼 열수추출물 섭취군의 경우 다른 열수추출물 섭취군에 비하여 높은 근력 향상 효과를 보여주었다. 무기인산은 맥문동, 오미자, 오가피, 두충, 구기자, 홍삼 열수추출물 섭취군의 경우 대조군에 비하여 낮은 수준을 나타내었다. 젖산 함량은 모든 생약재 열수추출물 식이군에서 대조군과 비교하여 낮은 함량을 나타내어 생약 열수 추출물을 이용한 근력 향상용 기능성 식품의 개발 가능성을 보여주었다.

Meat quality, post-mortem proteolytic enzymes, and myosin heavy chain isoforms of different Thai native cattle muscles

  • Objective: This study investigated the meat quality characteristics, endogenous proteolytic enzymes, collagen content, and myosin heavy chain (MyHC) isoforms of different muscles of Thai native cattle (TNC). Methods: Infraspinatus (IF), Longissimus thoracis (LT), and Supraspinatus (SS) muscles were obtained from two TNC breeds, Kho-Lan (KL, n = 7) and Kho-Isaan (KI, n = 7). The muscle and meat characteristics of TNC breeds and their relationship with MyHC expression were examined. Results: Three MyHC isoforms namely MyHC I, MyHC IIa, and MyHC IIx were detected in the muscles. The KL had higher (p<0.05) MyHC IIx than the KI. The IF muscle had higher (p<0.05) MyHC I compared to other muscles. The LT muscle had the least MyHC I. The LT had higher (p<0.05) MyHC IIx than the IF and SS muscles. The IF presented the least MyHC IIx. The KL had higher (p<0.05) lightness and moisture content and lower crude protein, redness, cooking loss, shear force, and calpastatin than the KI. The glycogen, total collagen, soluble collagen, crude protein, ash contents, and troponin T degradation product of IF and SS were lower (p<0.05) than that of LT. Ether extract in LT was lower (p<0.05) than that of IF and SS. The percentage of MyHC I, MyHC IIa, and MyHC IIx were significantly correlated with muscle and meat characteristics of TNC. Conclusion: These results suggest that the differences in the MyHC isoforms may partly account for the variation in meat quality between breeds and among muscles of TNC.

조피볼락(Sebastes schlegeli)의 성숙에 따른 간세포 활성과 간 및 근육의 단백질$\cdot$총 RNA 함량변화 (Activities of Hepatocytes and Changes of Protein and Total RNA Contents in Liver and Muscle of Sebastes schlegeli with the Gonadal Maturation)

  • 난태생 어류인 조피볼락(Sebastes schlegeli)의 성숙과 안정적인 출산을 위한 기초 정보를 얻고자, 간중량지수의 월별 변화와 간세포의 활성이 생식소의 발달과 어떤 관계에 있는지 조사하고, 아울러 생식소 발달에 따른 간 및 근육조직내의 단백질과 총 RNA 함량변화도 조사하였다. 암컷의 HSI는 9월부터 높아지기 시작하여 2월에 최대 값(2.58)에 달한 뒤, 3월(성숙기) 부터 5월(출산기) 사이에는 낮아졌다 수컷에서는 10월(성숙기)부터 높아지기 시작하여 4월(회복기)에 최대 값(3.20)에 달했다. 암컷의 난황형성기에 간세포내 글리코겐과 지방적의 양이 점차 감소되나, 총 RNA의 함량은 증가하였다. 그러나 수컷에서는 다량의 지방적들이 소모되지 않고 성숙기에도 세포질내에 잔존하고 있었다. 생식소의 발달과 함께 간 세포질에는 잘 발달된 조면소포체 가 증가하였는데, 이 들은 단백질 합성과 난황 전구물질의 형성에 중요한 역할을 하는 것으로 간주된다. 암컷의 간과 근육조직의 단백질 및 총 RNA 함량은 1월(성장기)부터 점차 증가되기 시작하여 4월 초순(성숙기)에 최대 값을 나타낸 다음, 4월 하순(임신기)에 감소되는 경향을 보였다. 수컷에서는 초기 성숙기인 10월부터 점차 증가하여 성숙 및 교미기인 12월에 최대 값을 보인 다음, 퇴화기인 1월에 다시 감소하였다.
