Emerging Technology - Research on Ginsenoside Characteristics of Gyeonggi Ginseng (신기술 신소재 - 경기인삼 진세노사이드 특성 연구)

  • 경기인삼 품질 특성을 확립하기 위한 기초자료로 진세노사이드 특성을 조사하였다. 경기도내 4대 인삼조합(개성, 김포 파주, 경기동부, 안성) 인삼을 인삼농가에서 직접 채취하거나 채굴현장에서 수집하여 진세노사이드 함량 및 조성을 분석하여 그 특성을 분석해 본 결과는 다음과 같다. 1. 경기인삼 4, 5, 6년근의 총 진세노사이드 함량은 3.92 mg/g, 4.34 mg/g 및 4.94 mg/g으로 각각 나타나 재배 년수가 증가할수록 함량이 높아짐을 알 수 있었다. PD/PT 비율은 1.12, 1.34 및 1.40으로 년근 수가 증가할수록 PD계열 진세노사이드 함량이 증가하였다. 2. 경기인삼 6년근의 크기별 진세노사이드 함량을 측정한 결과 큰 인삼(大)의 총 진세노사이드 함량은 5.19 mg/g, 작은 인삼은 4.69 mg/g으로 같은 년근 수에서는 인삼의 크기에 따라 진세노사이드함량 차이는 크게 나타나지 않았다. 3. 6년근 부위별 총 진세노사이드 함량은 주근이 3.70 mg/g이며 세근은 6.37 mg/g으로써 주근보다 세근에서 1.72배 함량이 높은 것으로 나타났다. 진세노사이드 각각 함량별로 비교해 볼 때 PD계인 진세노사이드 $Rb_2$, $Rb_3$, Rc, Rd와 PT계인 Re, $Rg_2$가 주근보다 세근에서 높은 함량을 나타내어, PD/PT 비율이 1.08과 2.06으로 세근은 주근보다 PD계열 함량이 2배 이상 높은 것으로 나타났다.

Optimization of Extraction of Marker Compounds from Red Ginsengs by Accelerated Solvent Extraction Using Response Surface Methodology (반응표면분석법을 사용한 가속 용매 추출에 의한 홍삼 지표성분의 추출 최적화)

  • A new method based on accelerated solvent extraction (ASE) combined with response surface methodology (RSM) has been developed for optimization of the extraction of ginsenoside [Rb1, Rg1, and Rg3(20S)], total phenolics, and benzopyrene in red ginseng. The RSM method, based on a five level and two variable central composite design, was employed to obtain the optimal combination of extraction conditions. In brief, ginsenosides Rb1, Rg1, and Rg3(20S) and total phenolics with undetectable benzopyrene were optimally extracted with 50% ethanol as an extraction solvent, extraction temperature of $158^{\circ}C$, extraction time of 20 min, extraction pressure of 2,500 psi, flush volume of 60%, and one extraction cycle. The contents of ginsenosides and total phenolics in red ginseng extracted by ASE under optimum conditions were significantly higher than those extracted by sonication and reflux extraction.

Comparisons of Antidiabetic Activities between White Ginseng Ethanol Extract and IH-901 in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨 흰쥐에서 백삼 에탄올엑스와 IH-901의 항당뇨 작용 비교)

  • Antidiabetic activities of white ginseng 50% ethanol extract (WGE) and IH901, an intestinal metabolite of ginsenoside R $b_1$, were compared in multiple low dose streptozotocin (STZ)-induced diabetic rats. WGE or IH901 were coadministered with STZ on Day 1 at dose of 100 and 300 mg or 10 and 30 mg, respectively, and continually administered for 16 days. STZ dissolved in citrate boner was injected intraperitoneally at dose of 20 mg/kg for 5 consecutive days. During the experiment, plasma glucose level and body weight were measured every 4$^{th}$ day. Amount of food and water intake were evaluated once a week and compared between groups. WGE and IH901 both significantly reduced the plasma glucose levels on Day 16 as compared with those of the diabetic control group. In the meantime, amount of food and water intake in WGE-and IH901-treated groups were significantly improved in a dose dependent fashion as compared with those of the diabetic control group. Taken together WGE and IH901 showed the comparable antidiabetic activities at the corresponding doses used in this experiment.

  • Ginseng (Panax ginseng Meyer) has been used as traditional herbal drug in Asian countries. Ginsenosides are major components having pharmacological and biological efficacy like anti-inflammatory, anti-diabetic and anti-tumor effects. To control the quality of the components in diverse ginseng products, we developed a new quantitative method using LC-MS/MS for 13 ginsenosides; Rb1, Rb2, Rc, Rd, Re, Rf, 20(S)-Rh1, 20(S)-Rh2, Rg1, 20(S)-Rg3, F1, F2, and compound K. This method was successfully validated for linearity, precision, and accuracy. This quantification method applied in four representative ginseng products; fresh ginseng powder, white ginseng powder, red ginseng extract powder, and red ginseng extract. Here the amounts of the 13 ginsenosides in the various type of ginseng samples could be analyzed simultaneously and expected to be suitable for quality control of ginseng products.

Enhancement of Ginsenoside Rg1 and Rg5 Contents in an Extract of Wood-cultivated Ginseng by Lactobacillus plantarum (Lactobacillus plantarum을 이용한 산양삼 추출물의 진세노사이드 Rg1 및 Rg5의 함량 증대)

  • Twelve lactic acid bacteria harboring ${\alpha}$-rhamnosidase (EC 3.2.1.40) activity were isolated from traditional Korean foods. The 6 strains (Weissella confuse [n = 1], Lactobacillus pentosus [n = 1], and Lactobacillus plantarum [n = 4]) with the highest rhamnosidase activity were selected for bioconversion of an extract of wood-cultivated ginseng. The L. plantarum MBE/L2990 strain increased ginsenoside content (0.58 mg for Rg1 and 0.24 mg for Rg5) and showed higher bioconversion activity than the control strain L. plantarum KCTC21004 (56% and 42% increase for Rg1 and Rg5, respectively). L. plantarum MBE/L2990 was deposited at the Korean Collection for Type Cultures as Lactobacillus plantarum KCTC18529P.

Composition of Saponin and Free Sugar of Some White Ginsengs with Processing Conditions (가공방법에 따른 몇가지 백삼 제품의 사포닌 및 유리당 조성 변화)

  • To analyse components of fresh ginseng and some white ginsengs with different processing conditions, approximate composition, extraction yield, total saponin content and composition and free sugar composition of fresh ginseng, white ginseng, Taeguksam A and Taeguksam B were examined. Yield of hot water extraction was two times higher than that of 80% methanol extraction. Hot water extraction yields of fresh ginseng, white ginseng, Taeguksam A and Taeguksam B were 56.4, 39.9, 42.9 and 46.6%, respectively, while the 80% methanol extraction yields ranged from 15.8% to 21.9%. Total saponin contents of the above were 2.40, 1.73, 1.45 and 1.79%, respectively, with hot water extraction and were 2.15, 2.99, 2.81 and 2.35%, respectively, with 80% methanol extraction. Ginsenoside compositions of the above varied with processing conditions and extraction solvents. Hot water and 80% methanol extracts of fresh and white ginseng composed of fructose, glucose, sucrose and maltose. Rhamnose was detected only in the extract of Taeguksam A and B.

Studies on the Activation Mechanism of c-src Protein Tyrosine Kinase by Ginsenoside-Rgl

  • We have studied an activation mechanism of $pp60^{c-src}$ protein tyroslne kinase (PTK) by ginsenoside-$Rg_1$ (G-$Rg_1$ ) in NIH(pMcsrc/foc)B c-src overexpressor cells. It was previously reported that G--$Rg_1$ stimulated the activation of c-src kinase at 20 pM with a 18 hr-incubation, increasing the activity by 2-4-fold over that of untreated control, and this effect was blocked by treatments of in- hibitors of either protein synthesis (cycloheximide) or RNA synthesis (actinomycin D) (Hong, H.Y. et at. Arch. Pharm. Res. 16, 114 (1993)). However, an amount of c-src protein itself in wild-type cells was not changed by G-$Rg_1$. When the cells mutated at one or two tyrosine residue(s) (Y416/527) that are important sites to regulate the kinase activity were treated with G-$Rg_1$, increases both in the activity of c-src kinase and in the expression of the protein were not observed. In addition, removal of extracellular calcium ion by EGTA or inhibition of PKC by H-7 canceled the G-$Rg_1$-induced activation of the kinase. Although the activation was little affected by G-$Rg_1$ with a calcium ionophore A23187, it was synergistically stimulated by treatment of G-Rgl and PMA, a PKC activator. Taken together, these results suggest that the activation of c-src kinase by G-$Rg_1$ is caused by an increase in the specific activity of the kinase, but not in amount of it, and is involved with both collular calcium ion and PKC. Further the increase in the specific activity of c-src kinase may result from altered phosphorylation at tyro-416 and -527.

Characteristic Effects of Dangnyo-hwan for Diabetes Control Studied Using LC-MS/MS and ICP (LC-MS/MS 및 ICP를 이용한 당뇨환의 화학적 특성 분석)

  • Objectives: The primary aim of this study was to identify bioactive compounds in Dangnyo-hwan, a Korean herbal medicine, through instrumental analysis using LC-MS/MS and ICP, and investigate its potential use in diabetes treatment. Methods: The extract of Dangnyo-hwan has 12 medicinal herbs; these were compared with 18 marker substances selected from literature survey. Results: LC-MS/MS analysis could detect 9 of the 18 bioactive compounds: citruline, catalpol, berberine, ginsenoside Rb1, ginsenoside Rg1, oleanolic acid, β-sitosterol, mangiferin, and schizandrin. While harmful heavy metals such as As, Pb, Cd, Hg, Ni, and Cu were not present in high concentrations, Zn concentration was 4.2 mg in 100 g Dangnyo-hwan. Conclusions: Instrumental analysis such as LC-MS/MS and ICP was successfully used to identify bioactive compounds in Dangnyo-hwan. Detection of 9 bioactive substances and Zn from the herb medicine is a valuable finding, and suggests that Dangnyo-hwan is a candidate medicine for diabetes. Further investigations like in vitro assay, percent GPR 119 activity, and percent human DGAT-1 inhibition are underway.

처치온도에 따른 초미분화 산양삼의 홍삼화 연구

  • Objectives : The aim of this study was to find optimal conditions for producing red ginseng from cultivated wild ginseng using the Turbo Mill. Methods : Characteristics of powdered cultivated wild ginseng based on various temperature settings of the Turbo Mill were observed, and changes in the content was measured by HPLC for various ginsenosides. Results : 1. The diameter of cultivated wild ginseng powder ground by the Turbo Mill was around 10${\mu}m$. 2. As the temperature rose, pressure, Specific Mechanical Energy(SME), and density decreased, whileas Water Solubility Index(WSI) increased. 3. As the temperature rose, super fine powder showed tendency to turn into dark brown. 4. Measuring content changes by HPLC, there was no detection of ginsenoside Rg3 and ginsenosideRg1, Rb1, and Rh2 concentrations decreased with increase in temperature. Conclusions : Super fine powder of cultivated wild ginseng produced by the Turbo Mill promotes easy absorption of effective ingredients by breaking the cell walls. Using this mechanism to produce red ginseng from cultivated wild ginseng, it yielded less than satisfactory results under the current experiment setup. Further researches are needed to verify more suitable condition for the production of red ginseng.

Inhibitory Activity of Korean Ginseng on Lipolytic Action of Toxohormone-L from Cancerous Ascites Fluid (고려인삼이 암독소의 지방분해작용에 미치는 저해활성)

  • A substance that inhibit the lipolytic action of Toxohormone-L was a crude acidic polysaccharide isolated from Korean red and white ginseng. The total inhibitory activities (units) of PGI and PG4 fraction in white ginseng and those of PG,, PG4 and PG43 fraction in red ginseng were higher than other fractions in vitro test. Each water extract of ginseng was effective against the lipolysis induced by the Toxohormone-L at the concentration over 10~100$\mu$l/ml. The total inhibitory activities (units) were highest at the concentration of 100ug/ml and 1,000ug/ml in the 4-year and 5-year old white ginseng root respectively, while, it was higher in the 6-year old ginseng than other ages regardless of the reaction concentration In the red ginseng. The inhibitory effect of ginsenos1de - Rb2 on the lipolysis by Toxohormone-L was higher than other ginsenosides at the concentration of 100 $\mu$g to 500 $\mu$g/ml of reaction mixture, and total inhibitory activities (units) of ginsenoside -Rb2 were also higher than other treatments.
