Modelling land surface temperature using gamma test coupled wavelet neural network

  • The climate change has made adverse effects on land surface temperature for many regions of the world. Several climatic studies focused on different downscaling techniques for climatological parameters of different regions. For statistical downscaling of any hydrological parameters, conventional Neural Network Models were used in common. However, it seems that in any modeling study, uncertainty is a vital aspect when making any predictions about the performance. In this paper, Gamma Test is performed to determine the data length selection for training to minimize the uncertainty in model development. Another measure to improve the data quality and model development are wavelet transforms. Hence, Gamma Test with Wavelet decomposed Feedforward Neural Network (GT-WNN) model is developed and tested for downscaled land surface temperature of Patna Urban, Bihar. The results of GT-WNN model are compared with GT-FFNN and conventional Feedforward Neural Network (FFNN) model. The effectiveness of the developed models is illustrated by Root Mean Square Error and Coefficient of Correlation. Results showed that GT-WNN outperformed the GT-FFNN and conventional FFNN in downscaling the land surface temperature. The land surface temperature is forecasted for a period of 2015-2044 with GT-WNN model for Patna Urban in Bihar. In addition, the significance of the probable changes in the land surface temperature is also found through Mann-Kendall (M-K) Test for Summer, Winter, Monsoon and Post Monsoon seasons. Results showed an increasing surface temperature trend for summer and winter seasons and no significant trend for monsoon and post monsoon season over the study area for the period between 2015 and 2044. Overall, the M-K test analysis for the annual data shows an increasing trend in the land surface temperature of Patna Urban.

  • Objectives : This research was performed to investigate the effect of herbal acupuncture(Ephedra sinica and Green tea) at Umnungchon(SP9) and Pungnyung(ST40) on weight gain, food intakes, food efficiency ratios, HDL-cholesterol, triglyceride, free fatty acid on rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into normal group(Sham), high fat diet group(Con), high fat diet and Ephedra sinica-herbal acupuncture in SP9 ST40 treated group(ES), high fat diet and Green tea-herbal acupuncture in SP9 ST40 treated group (GT), high fat diet and Ephedra sinica+Green tea-herbal acupuncture in SP9 ST40 treated group(ES+GT). Herbal acupuncture was bilaterally treated at the level of 19.125mg/ml per 2 days during 5 weeks. Results : Body weight, feed intake and feed efficiency ratio were decreased in Sham, ES, ES+GT, compared with control group. The level of serum HDL-cholesterol was increased in sham, tender to increase in ES, ES+GT. The change on that of serum triglyceride was decreased in sham, ES, ES+GT and free fatty acid was decrease in sham, ES+GT. Conclusions : Ephedra sinica with Green tea-herbal acupuncture in SP9 ST40 can control the Body weight, feed intake and feed efficiency ratio, HDL-cholesterol, triglyceride, free fatty acid.

Effects of Gyejigamchoyonggolmoryeo-tang on sleep latency and total duration of sleep induced by thiopental sodium in mice (계지감초용골모려탕(桂枝甘草龍骨牡蠣湯)이 치오펜탈 나트륨으로 유도된 수면의 입면 및 수면 시간에 미치는 영향)

  • Objectives : Gyejigamchoyonggolmoryeo-tang (GT, Guizhigancaolonggumulitang in Chinese) is a herbal medicine to be prescribed for insomnia caused by anxiety induced by Heart-Heat and elevated Liver-Yang. In the present study, the onset time (sleep latency) and the duration time of sleep were measured to find out the sleep inducing effects of GT. The expression of c-fos immunohistochemistry was also measured at the ventrolateral preoptic area (VLPO) and tuberomammillary nucleus (TMN) site in brain. Methods : The onset time (sleep latency) and the duration time of sleep were measured 30 minutes after thiopental sodium injection. Thereafter, brain tissue was obtained and c-fos immunohistochemistry was performed on the VLPO and TMN sites. Results : GT statistically significantly reduced the sleep latency required to enter sleep, and significantly increased sleep duration time. GT significantly increased the number of c-fos immunohistochemical staining-positive cells in the sleep-inducing center (VLPO), whereas GT significantly decreased the number of c-fos immunohistochemical staining-positive cells in the arousal center (TMN). Conclusions : It could be concluded that the GT shortened the sleep latency and increased the duration time for sleep induced by thiopental sodium. And it was confirmed that the mechanism was to stimulate the sleep-inducing center (VLPO) and suppress the arousal center (TMN) in the brain. The results of this study are considered to be useful as scientific evidence that can be used clinically for the treatment of insomnia caused by anxiety.

Enhanced Sialylation of Recombinant Erythropoietin in CHO Cells by Human Glycosyltransferase Expression

  • Sialylation, the attachment of sialic acid residues to a protein, can affect the biological activity and in vivo circulatory half-life of glycoproteins. Human ${\alpha}2$,3-sialyltransferase (${\alpha}2$,3-ST) and ${\beta}1$,4-galactosyltransferase (${\beta}1$,4-GT) are responsible for terminal sialylation and galactosylation, respectively. Enhanced sialylation of human erythropoietin (EPO) by the expression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT was achieved using recombinant Chinese hamster ovary (CHO) cells (EC1). The sialic acid content and sialylation of N-glycans were evaluated by HPLC. When ${\alpha}2$,3-ST was expressed in CHO cells (EC1-ST2), the sialic acid content (moles of sialic acid/mole of EPO) increased from 6.7 to 7.5. In addition, the amount of trisialylated glycans increased from 17.3% to 26.1 %. When ${\alpha}2$,3-ST and ${\beta}1$,4-GT were coexpressed in CHO cells (EC1-GTST15), the degree of sialylation was greater than that in EC1-ST2 cells. In the case of EC1-GTST15 cells, the sialic acid content increased to 8.2 and the proportion of trisialylated glycans was markedly increased from 17.3% to 35.5%. Interestingly, the amount of asialoglycans decreased only in the case of GTST15 cells (21.4% to 14.2%). These results show that coexpression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT is more effective than the expression of ${\alpha}2$,3-ST alone. Coexpression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT did not affect CHO cell growth and metabolism or EPO production. Thus, coexpression of ${\alpha}2$,3-ST and ${\beta}1$,4-GT may be beneficial for producing therapeutic glycoproteins with enhanced sialylation in CHO cells.

Effects of Skin Application of Gelidium amansii Extract on Deep Second-degree Burns in Mice (석화채(石花菜) 추출물 도포가 마우스의 심재성 2도 화상에 미치는 영향)

  • Objective: The purpose of this study was to investigate the effects of skin application with Gelidium amansii extract on skin with deep second degree burns in mice. Methods: BALB/c mice were divided into four groups: normal (NOR) group; burn-elicited mice (CON) group, Silmazine-treated mice after burn elicitation (ST) group, and Gelidium amansii-extract treated mice after burn elicitation (GT) group. To examine the skin recovery effect after burn, changes of burn area, angiogenesis and histologic structure were analyzed. To measure effect of edema regulation, matrix metalloproteinase-9 (MMP-9) was analyzed. To estimate the skin regenerative & stable effect, 5-bromo-2'-deoxyuridine (BrdU) and substance P were analyzed. Results: 2 weeks later, 1. The size of burn area decreased in the GT and ST groups more than the CON group. 2. Alleviation of angiogenesis appeared in the GT and ST groups more than in the CON group. 3. Blood clot, epithelial cell hyperplasia, and inflammatory cell infiltration declined in the GT and ST groups more than in the CON group. 4. MMP-9, BrdU, and substance P positive reaction decreased in the GT and ST groups more than in the CON group 5. In the comparative study, the GT group was superior to the ST group. Conclusion: The skin application of Gelidium amansii extract could lessen skin damage by the medium of regulation MMP-9 activation. This skin stabilization was induced in mice with deep second degree burns.

The effects of Gamisipjeontang-gamibang on injured tissue after burn elicitation (가미십전탕(加味十全湯) 가미방(加味方)이 화상(火傷)의 조직 손상에 미치는 영향)

  • Objective : The purpose of this study is to examine the effects of Gamisipjeontang-gamibang(GT) on an experimental bum elicitation. Method : The absorbance of the photo cell mixed with GT at the Abs 560 nm was measured after irradiation for 1 min. In order to know the antioxidant effect on skin cell of mice after burn elicitation, superoxide dismutase(SOD) activity was measured. Also, in order to know effects on the skin regeneration in the burned mice, we counted the eosinophil in blood from animals via cardiac puncture and observed the histological structure in the epidermal basal layer and the dermal section on the 3rd, 7th and 14th day after burn elicitation. We also studied changes in angiogenesis in the capillary surrounding the basal layer and dermal papilla. The changes of HSP70 distribution and changes of p53 positive reaction were observed to investigate the changes of the stress in the skin as well. Result : The results indicated that GT has a significant impact on the antioxidant effect on skin cells of mice after bum elicitation by increasing SOD activity in vitro test. GT seems to decrease MIP-2 which induces neutrophil infiltration and promotes the angiogenesis dose-dependently. Furthermore, GT decreased HSP70, the expression of which was induced by elevated temperatures, and p53 which induce a apoptosis after stress. Conclusion : GT can be applied to burned skin through its antioxidant effect and skin regeneration property.

Multiplex PCR Detection of the GT73, MS8xRF3, and T45 Varieties of GM Canola

  • A multiplex polymerase chain reaction (PCR) method was developed to simultaneously detect three varieties of genetically modified (GM) canola. The construct-specific primers were used to distinguish the following three varieties of GM canola; GT73, MS8xRF3, and T45, using multiplex PCR. The FatA (fatty acyl-ACP thioesterase) gene was used as an endogenous canola reference gene in the PCR detection. The primer pair Canendo-FIR containing a 105 bp amplicon was used to amplify the FatA gene and no amplified product was observed in any of the 15 different plants used as templates. The GT73-KHUF1/R1 primer recognized the 3'-flanking region of GT73, resulting in an amplicon of 125 bp. The Barstar-F1/MS8xRF3-R primer recognized the junction region of bars tar and the NOS terminator introduced into MS8xRF3, resulting in a 162 bp amplicon, and the T45-F2/R2 primer recognized the junction region of PAT and the 35S terminator introduced into T45, resulting in an amplicon of 186 bp. This multiplex PCR allowed for the detection of construct-specific targets in a genomic DNA mixture of up to 1% GM canola containing GT73, MS8xRF3, and T45.

Effect of β-glucan on Epithelial Inflammation Induced by Atopic Dermatitis through Endocannabinoid System Activity (β-glucan의 Endocannabinoid system 활성을 통한 아토피피부염 유발 상피 염증 억제 효과)

  • Objectives This study was conducted to confirm the inhibitory effect of β-glucan on epithelial inflammation induced by atopic dermatitis through Endocannabinoid system (ECS) activity. Methods Six-week-old NC/Nga mice were divided into a control group (Ctrl), atopic dermatitis elicitation group (ADE), and a β-glucan-treated group (β-glucan treatment after atopy dermatitis elicitation, β-GT). After 3 weeks, CB1, CB2, and GPR55 were observed to confirm the regulation of ECS activity, and filaggrin in the stratum corneum and Kallikrein-related peptidase (KLK) 7 in the stratum corneum and protease activated receptor (PAR)-2 were observed to confirm the inhibition of the inflammation, Phosphorylated extracellular signal-related kinase (p-ERK), Phosphorylated mammalian target of rapamycin (p-mTOR), and E-Cadherin were observed to confirm microenvironmental regulation. Results β-GT was significantly increased in CB1, CB2, and GPR55 positive reactions compared to that of the ADE. In positive reaction of the filaggrin in the stratum corneum, β-GT was significantly increased than that of the ADE. For KLK7 positive and PAR2 positive, β-GT was significantly reduced compared to the ADE. The p-ERK-positive and p-mTOR-positive reactions were significantly reduced in β-GT than in ADE. E-cadherin positive reaction was significantly increased in β-GT than in ADE (All p < 0.01). Conclusions It was confirmed that β-glucan has the effect of inhibiting the epithelium induced by atopic dermatitis through the ECS activity.

The Quantitative Analysis on the Criterion Elements for Collision Avoidance Action in Collision Avoidance maneuver and Its Application (피항조선시의 피항개시기준요소의 양적파악 및 그 이용에 관한 연구)

  • The Steering and Sailing Rules of International Regulation for Preventing Collisions at Sea now in use direct actions to avoid collision when two power-driven vessels are meeting on reciprocal or nearly reciprocal courses so as to involve risk of collision. But these rules do not refer to the minimum relative distances and safety relative distances between two vessels when they should take such actions.In this paper the ship's collision avoiding actions being analyzed from a viewpoint of ship motions, the mathematical formulas to calculate such relative distances necessary for taking actions to avoid collision were worked out. The values of maneuvering indices being figured out through experiments of 20 actual ships of small, medium, large and mammoth size and applied to calculating formulas, the minimum relative distances and safety relative distances were calculated. The main results were as follows. 1. It was confirmed that the criterion elements for collision avoiding actions in head-on situation of two vessels shall be the minimum relative distances and safety relative distances between them. 2. On the assumption that two vessels same in size and condition were approaching each other in head-on situation, the minimum relative distance of small vessel(GT : 160~650tons) was found to be about 4.7 times her own length, and those of medium (GT:2,300~4,500tons),large(GT:15,000~62,000tons) and mommoth (GT:91,000~194,000tons) vessels were found to be about 5.2 times, about 5.2 times and about 6.1 times their own lengths respectively. 3. On the assumption that two vessels same in size and condition were approaching each other in head-on situation, the safe relative distance of small vessel (GT : 160~650tons) was found to be about 6.8 times her own length, and those of medium (GT : 2,300~4,500tons), large (GT: 15,000~62,000tons) and mammoth (GT : 91,000~194,000tons) vessels were found to be about 9.0 times, about 6.3 times, and about 8.0 times their own lengths respectively. 4. It is considered to be helpful for the safety of ship handling that the sufficient safe relative distances for every vessels shall be more than about 12~14 times which are 2 times minimum relative distance, their own length on above assumption.

Quality characteristics and antioxidant activity of rice sponge cake with added green tea powder (녹차 분말 함유 쌀스펀지 케이크의 품질특성 및 산화방지 활성)

  • We investigated the quality and antioxidant activities of sponge cake prepared containing 1-5% of green tea powder (GT). The water content in the cake increased with an increase in the amount of GT added. Although the hardness and springiness of the rice sponge cake increased, the adhesiveness decreased with increasing the level of GT. The cohesiveness, chewiness, and resilience of the rice sponge cake with GT were similar to those of the control. Chromaticity determination revealed that lightness, redness, and yellowness of the crust decreased with increasing GT content. Total polyphenol and flavonoid contents increased proportionally with increasing GT level. Antioxidant activity, measured by DPPH and ABTS radical scavenging as well as reducing power activities, was significantly higher in the rice sponge cake with GT than in the control. Sensory evaluation determined that addition of 1-3% GT to the rice sponge cake maintained the color, taste, texture, flavor, and overall acceptance similar to that of the control.
