Glucose Deprivation and Immunostimulation Induced Death in Rat Primary Astrocytes is Mediated by Their Synergistic Effect on the Decrease in Cellular ATP Level

  • In this study we investigated whether ATP loss was involved in the potentiated death of immunostimulated rat primary astrocytes in glucose-deprived condition. Rat primary astrocytes immunostimulated with LPS plus IFN-${\gamma}$ for 48 h underwent death upon glucose deprivation, which dependent on the production of peroxynitrite. Intracellular ATP level synergistically decreased by glucose deprivation in immunostimulated astrocytes but not in control cells, and the loss of ATP occurred well ahead of the LDH release. The synergistic cell death and ATP loss by immunostimulation and glucose deprivation were inhibited by iNOS inhibitor (L-NAME and L-NNA) or peroxynitrite decomposition catalyst (also a superoxide anion scavenger), Mn(III)tetrakis(N-methyl-4'-pyridyl)porphyrin (MnTMPyP). Exogenous addition of peroxynitrite generator, SIN-l timedependently induced ATP loss and cell death in the glucose-deprived astrocytes. Depletion of intracellular glutathione (GSH) and dis겨ption of mitochondrial transmembrane potential (MTP) were also observed under same conditions. Supply cellular ATP by the addition of exogenous adenosine or ATP during glucose deprivation inhibited ATP depletion, GSH depletion, MTP disruption and cell death in SIN-l treated or immunostimulated astrocytes. This study showed that perturbation in the regulation of intracellular ATP level in immunostimulated astrocytes might make them more vulnerable to energy challenging stimuli.

Influence of Alcohol Consumption on Oxidative Stress and Antioxidant Status in Cancer Patients - Case-control Study from Western Nepal

  • Aim: The present study assess the effect of consumption of alcohol on oxidative stress and antioxidant status in patients suffering from different types of cancer. Methods: This hospital based case control study conducted in the Western part of Nepal covered a total of 93 cancer patients with or without alcohol intake and smoking habits, along with 94 age, sex and habit-matched individuals serving as controls. Plasma thiobarbituric acid reacting substances (TBARS), total antioxidant activity (TAA), vitamin C, ${\alpha}$-tocopherol and erythrocyte reduced glutathione (GSH) were estimated and compared. Results: The TBARS level was found to be significantly higher ($p{\leq}0.001$) in all types of cancer patients when compared to controls, being aggravated in alcoholics with a smoking habit. No statistical significance ($p{\geq}0.05$) was observed in the level of vitamin C and ${\alpha}$-tocopherol. GSH and TAA level were significantly decreased ($p{\leq}0.001$) in all the groups except those who consumed both branded as well as homemade alcohol and non-alcoholics without smoking habit. Conclusion: Alcohol, irrespective of its commercial brand, increases oxidative stress in all types of cancer patients. This is even higher when alcohol intake is combined with a smoking habit. Decreased TAA and GSH are major risk factors for cancer development.

청혈산(淸血散)이 ob/ob mouse의 혈당, 고지혈증, Polyol Pathway 및 Reactive Oxygen Species에 미치는 영향 (Effects of Cheonghyul-san on Blood Glucose, Hyperlipidemia, Polyol Pathway and Reactive Oxygen Species in ob/ob Mice)

  • Etiological studies of diabetes and its complications showed that oxidative stress might play a major role. Therefore, many efforts have been tried to regulate free oxygen radicals for treating diabetes and its complications. Cheonghyul-san has been known to be effective for the antidiabetic, antihyperlipidemic and antiobesitic prescription, and composed of four crude herbs. In male ob/ob mouse with severe obesity, hyperinsulinemia, hypergiycemia, hyperlipidemia, the acting mechanisms of Cheonghyul-san were examined. Mice were grouped and treated for 5 weeks as follows. Both the lean (C57/BL6J black mice) and diabetic (ob/ob mice) control groups received standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. The effects of Cheonghyul-san extract on the ob/ob mice were observed by measuring the serum levels of glucose, insulin, lipid components, and the kidney levels of reactive oxygen species (ROS), MDA+HAE, GSH and also the enzyme activities involved in polyol pathway. Cheonghyul-san lowered the levels of serum glucose and insulin in a dose dependent manner. Total cholesterol, triglyceride and free fatty acid levels were decreased, while the HDL-cholesterol level was increased, in Cheonghyul-san treated groups. Renal aldose reductase and sorbitol dehydrogenase activities were increased in the ob/ob mice, whereas those were inhibited in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the generation of ROS in the kidney. Finally, MDA+HAE level was increased and the GSH level was decreased in the ob/ob mice, whereas those were improved in the Cheonghyul-san-administered groups. The results suggested that Cheonghyul-san exerted the antidiabetic and antihyperlipidemic activities by regulating the activities of polyol pathway enzymes, scavenging ROS, regulating the MDA+HAE and GSH levels in the ob/ob mice.

환경 Hormone에 대한 삼백초의 Glutathione 및 항산화 활성 효과 (Effects of Saururus chinensis Baill on Glutathione and Antioxidative Activity Against TCDD-treated Rats)

  • 삼백초가 TCDD 투여로 유발된 rat의 간손상에 미치는 효과를 알아보기 위해 NTT, TTA, STT군의 3군으로 각각 10마리씩 나누어 TCDD를 투여하고 4주 동안 물질을 투여하였다. 4주 후 희생시켜서 생체 내 간 조직 활성 효과 및 항산화 효과를 관찰한 결과 1. AST 활성도는 STT군이 TTA군과 비교해서 70.05%,의 억제 효과를 보였다. 반면에, ALT활성도는 STT군이 TTA군에 비해 24.00% 감소하였으며 64.80%의 억제 효과를 보였다. 2. SOD는 TTA군이 NTT군보다 137.78% 감소하였으며 , STT군은 TTA군에 비해 81.94%로 상승되었다. Catalase는 TTA군은 NTT군 보다 199.13% 감소하였으며, STT군의 catalase 활성도는 TTA 군과 비교하여 52.48% 증가하였고 55.45%의 회복율을 보였다. 3. TTA군의 GSH함량은 NTT군 보다 291.29% 감소하고 STT군은 TTA군보다 비교하여 74.20% 증가하였으며, 98.72%로 회복하였다. TTA군은 GSSG함량은 NTT군 보다 46.80% 증가하였고 STT군의 간조직중의 GSSG 함량은 TTA군과 비교하여 61.08% 감소하였으며, 81.01%로 억제되었다.

Effect of Macromolecules in Maturation Medium on Oocyte Maturation and Embryonic Development after Parthenogenesis and Nuclear Transfer in Pigs

  • The objective of this study was to examine the effect of macromolecule in a maturation medium on nuclear maturation, intracellular glutathione (GSH) level of oocytes, and embryonic development after parthenogenetic activation (PA) and somatic cell nuclear transfer (SCNT) in pigs. Immature pig oocytes were cultured in maturation medium that was supplemented with each polyvinyl alcohol (PVA), pig follicular fluid (pFF) or newborn calf serum (NBCS) during the first 22 h and the second 22 h. Oocyte maturation was not influenced by the source of macromolecules during in vitro maturation (IVM). Embryo cleavage and cell number in blastocyst after PA was altered by the source of macromolecule but no difference was observed in blastocyst formation among treatments. Oocytes matured in PVA-PVA medium showed lower rates of oocyte-cell fusion (70.4% vs. 77${\sim}$82%) and embryo cleavage (75% vs. 86${\sim}$90%) after SCNT than those matured in other media but blastocyst formation was not altered (13${\sim}$27%) by different macromolecules. pFF added to IVM medium significantly increased the intracellular GSH level of oocytes compared to PVA and NBCS, particularly when pFF was supplemented during the first 22 h of IVM. Our results demonstrate that source of macromolecule in IVM medium influences developmental competence of oocytes after PA and SCNT, and that pFF supplementation during the early period (first 22 h) of IVM increases intracellular GSH level of oocytes.

Boswellic Acid Improves Cognitive Function in a Rat Model Through Its Antioxidant Activity - Neuroprotective effect of Boswellic acid -

  • Objectives: Boswellic acid (BA), a compound isolated from the gum-resin of Boswellia carterii, is a pentacyclic terpenoid that is active against many inflammatory diseases, including cancer, arthritis, chronic colitis, ulcerative colitis, Crohn's disease, and memory impairment, but the mechanism is poorly understood. This study investigated the effects of boswellic acid on spatial learning and memory impairment induced by trimethyltin (TMT) in Wistar rats. Methods: Forty male Wistar rats were randomly divided into 5 groups: Normal group, TMT-administrated rats (8.0 mg/kg, Intraperitoneally, i.p.) and TMT + BA (40, 80 and 160 mg/kg, i.p.)-administrated rats. BA was used daily for 21 days. To evaluate the cognitive improving of BA, we performed the Morris water maze test. Moreover, to investigate the neuroprotective effect of BA, we determined the acetylcholinesterase (AchE) activity, the malondialdehyde (MDA) level as a marker of lipid peroxidation, and the glutathione (GSH) content in the cerebral cortex. Results: Treatment with TMT impaired learning and memory, and treatment with BA at a dose of 160 mg/kg produced a significant improvement in learning and memory abilities in the water maze tasks. Consistent with behavioral data, the activity of AChE was significantly increased in the TMT-injected rats compared to the control group (P < 0.01) whereas all groups treated with BA presented a more significant inhibitory effect against AChE than the TMT-injected animals. In addition, TMT reduced the GSH content and increased the MDA level in the cerebral cortex as compared to the control group) P < 0.01). On the other hand, treatment with BA at 160 mg/kg slightly increased the GSH content and reduced the MDA level in comparison to the TMT-administered group (P < 0.01). Conclusion: The above results suggest that the effect of BA in improving the cognitive function may be mediated through its antioxidant activity.

TCDD를 투여한 rat의 간손상에 대한 어성초의 효과 (Effects of Houttuynia Cordata thunb on the liver damage of TCDD-treated rats)

  • TCDD를 투여한지 일주일 후부터 어성초를 200mg/kg 복강 내로 4주간 격일로 투여하였다. TTA군과 NTT군은 saline을 투여하였다. 혈청에서 GOT, GPT의 활성도를 측정하였고, 간조직중에서 MDA, GSH, GSSG, GPx, SOD, Catalase의 level을 측정하였다. GOT 활성도는 HTT군이 TTA군과 비교해서 49.00%의 억제 효과를 보였다. 반면 GPT활성도는 TTA(29.70)군에 비해 68.36%의 억제효과를 보였다. MDA는 TTA군이 NTT군 보다 높게 나왔으며, HTT군은 NTT군에 비교하여 30.47%의 억제율을 보였다. HTT군의 GSH는 TTA군에 비해 1.91배 증가하였고, GSSG는 TTA군에 비해 46.72% 감소하였다. SOD는 TTA군이 NTT군보다 낮게 나타났고, HTT는 TTA군에 비해 46%의 증가를 보였다. Catalase는 TTA군(1.12mU/mg protein)의 활성은 NTT(3.44mU/mg protein)군 보다 32.56% 감소하였다. HTT군의 간의 Catalase 활성도는 TTA 군과 비교하여 50.00% 증가 하였다. 이러한 결과에서 보듯이 TCDD로 인해 생성된 free radical은 항산화 효소에 의해서 감소되었으며, 그리고 어성초가 항산화 효소 대신에 노화를 저해하는 작용을 하는 것으로 사료 된다.

Reduced glutathione 및 인삼추출액(人蔘抽出液)이 X-선전신조사(線全身照射)를 입은 마우스 간조직(肝組織) 및 혈중(血中) NP-SH 및 NP-SS에 미치는 영향(影響) (Effects of Reduced Glutathione and Ginseng Extract on Non-Protein Sulfhydryl, and Non-Protein Disulfide of Mouse Liver and Blood Following Whole Body X-Irradiation)

  • In an attempt to better understand the radioprotective effect of reduced glutathione(GSH), and to observe a possible radioprotective effect of Ginseng extract, whole body X-irradiation of 1,200 r was administered to the mouse either independently or immediately following the injection of GSH or Ginseng extract to the mouse intraperitoneally. The non-protein sulfhydryl (NP-SH) and non-protein disulfide (NP-SS) levels of the liver, and NP-SH level of NP-SH of the blood of the mouse were measured at 30, 60 and 120 minutes, and results were compared with the normal. The results thus obtained are summarized as follows; 1) The normal values of NP-SH and NP-SS of the mouse liver were $5.90{\pm}0.46\;{\mu}\;mol/gm\;wet\;wt.,\;and\;3.02{\pm}0.42\;{\mu}\;mol/ml$ wet wt., respectively, and the normal value of NP-SH of NP-SH of the mouse blood was $3.98{\pm}1.29\;{\mu}\;mol/ml$ 2) The injection of both GSH and Ginseng extract produced the highest values of NP-SH in the liver at 30 minutes, but a gradual decrease to the normal was observed thereafter. When X-irradiation alone was applied, the liver NP-SH value was lower than the normal at 60 minutes post-irradiation and thereafter. When Ginseng extract was injected immediately prior to X-irradiation, the liver NP-SH was lower than the normal throughout the experiment with the lowest value at 60 minutes. However, the combination of GSH and X-irradiation produced higher than the normal values throughout the entire experiment. 3) The liver NP-SS value was most significantly elevated at 30 minutes after the injection of GSH, hut the recovery to the normal was observed thereafter. The injection of Ginseng extract produced slightly higher liver NP-SS values at 30 and 60 minutes, but the value at 120 minutes was similar to the normal. The single application of X-irradiation resulted in the lower then normal liver NP-SS values throughout the entire experiment. When GSH was injected price to X-irradiation, the liver NP-SS values were higher than the normal at 30 and 60 minutes followed b the recovery to the normal at 120 minutes. The combination of Ginseng extract and X-irradiation showed generally lower liver NP-SS values throughout the experiment. 4) The blood NP-SH showed the higher than the normal values in all the experimental groups except when GSH was injected prior to X-irradiation alone produced e significantly elevated blood NP-SS value at 30 minutes post-irradiation.

BALB/C Mice에서 노루궁뎅이버섯 균사체 보충이 LPS로 인한 산화적 스트레스와 염증 반응에 미치는 효과 및 기전 (Effect of Hericium erinaceus Mycelia Supplementation on the Oxidative Stress and Inflammation Processes Stimulated by LPS and Their Mechanisms in BALB/C Mice)

  • 본 연구는 in vivo에서의 6주령의 BALB/C 생쥐(평균체중 $24.73\pm0.11$ g)에게 1주일 간 노루궁뎅이버섯 균사체(Hericium erinaceus mycelia)를 첨가한 식이를 급여한 후 내독소인 LPS로 인한 산화적 스트레스 및 염증반응에 미치는 영향을 조사하고자 하였다. 실험군은 5군으로 분류하였으며 정상 대조군(NC)과 양성 대조군(LC)은 chow를, HMP 10%군과 HMP 3%군은 chow에 노루궁뎅이버섯 균사체를 각각 3%와 10%를 첨가하였으며 HMP 대조군(HMP-C)은 chow에 노루궁뎅이버섯 균사체를 10% 첨가하여 pellet 형태로 조제하여 1주일 간 각각의 실험 식이를 급여하였다. LC군과 HMP 3% 및 HMP 10%군은 각각 LPS(100 mg/kg BW)를 투여하였으며 NC군과 HMP-C군은 생리식염수를 LPS와 동일한 농도로 투여하였다. 실험동물의 식이효율은 노루궁뎅이버섯 균사체를 보충한 군에서 저하되었으며 노루궁뎅이버섯 균사체의 보충군에서 체중감소 효과를 보였다. 노루궁뎅이버섯 균사체 10%를 보충한 HMP-C군과 HMP 10%군은 분변의 배설량이 가장 많았고 분변 중 총지방 양은 노루궁뎅이버섯 균사체를 섭취한 군에서 높은 수준을 보였으나 섭취량에 따른 현저한 차이는 보이지 않았고, 혈장 TG 함량은 군 간에 유의적인 차이는 보이지 않았으나 노루궁뎅이버섯 균사체 10%를 첨가한 식이를 섭취한 군에서 다소 높은 경향을 보였다. 분변 100 g 당 $\beta$-glucan의 함량은 노루궁뎅이버섯 균사체 3%를 보충한 군에서 가장 높았다. 혈장의 ALT 수준은 LPS를 투여한 후 8시간 경과 후에는 LC군에서 현저하게 증가한 반면 노루궁뎅이버섯 균사체를 첨가한 식이를 급여한 HMP 3%와 HMP 10%군에서는 유의적으로(p<0.05) 저하되어 NC군과 비슷한 수준을 보였다. 간에서의 SOD, GSH-red, GSH-px 활성은 LPS 투여 후 LC군에 비해 노루궁뎅이버섯 균사체를 섭취한 군에서 증가하였으며 GSH-red의 단백질 발현과 GSH 함량 또한 높아지는 것으로 나타났다. 이상의 결과로 미루어 볼 때 노루궁뎅이버섯 균사체의 보충은 내독소인 LPS로 유도된 산화적 스트레스에 저항하여 간 기능을 보호하는 효과가 있는 것으로 보이며 그 기전으로는 항산화 효소계 활성과 GSH-red 단백질 발현 증가, 그리고 GSH 수준 향상 등을 열거할 수 있다.

녹차가 전자파 조사 흰쥐 간조직의 Superoxide Dismutase 및 Glutathione Peroxidase 유전자 발현에 미치는 영향 (Effect of Green Tea on Gene Expression of Superoxide Dismutase and Glutathione Peroxidase in Rat Liver Exposed to Microwaves)

  • The purpose of this study was to investigate the effects of green tea on gene expression of superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) in rat liver exposed to microwave. Sprague-Dawley male rats with 200$\pm$10g body weight were assigned to normal and microwave exposed groups : microwave exposed groups ; microwave exposed groups were divided two groups : microwave(MW) group which was administrated the distilled water and green tea(GT) group which was administrated the green tea extracts. The rats were irradiated with microwave at frequence of 2.45 GHz for 15 min and then the gene expression in the damaged tissue were investigated at 0.1, 3, 4,6 and 8 days after the microwave irradition to compared with the normal group. The level of SOD gene expression in MW group was lower than the normal group within 6 days but that of GT group as higher than MW group. These results may imply that green tea stimulates SOD expression and there by protecting tissues from free radicals. The GSH-Px gene was expressed a little bit lower than the normal group but that of GT group was expressed to higher lever than MW group from 4 days after irradiation. These results suggest that the administration of green tea extract may activate antioxidative gene expressions such as SOD and GSH-Px in rat and that may help to recover liver tissues from microwave damage by removing hazardous free radicals and oxidized by products from cells.
