Potato (Solanum tuberosum L.), one of the most important food crops, is susceptible to a number of devastating fungal pathogens in addition to bacterial and other pathogens. Producing disease-resistant cultivars has been an effective and useful strategy to combat the attack of pathogens. Potato was transformed with Agrobacterium tumefaciens strain EHA101 harboring chitinase, (ChiC) isolated from Streptomyces griseus strain HUT 6037 and bialaphos resistance (bar) genes in a binary plasmid vector, pEKH1. Polymerase chain reaction (PCR) analysis revealed that the ChiC and bar genes are integrated into the genome of transgenic plants. Different insertion sites of the transgenes (one to six sites for ChiC and three to seven for bar) were indicated by Southern blot analysis of genomic DNA from the transgenic plants. Expression of the ChiC gene at the messenger RNA (mRNA) level was confirmed by Northern blot analysis and that of the bar gene by herbicide resistance assay. The results obviously confirmed that the ChiC and bar genes are successfully integrated and expressed into the genome, resulting in the production of bialaphos-resistant transgenic plants. Disease-resistance assay of the in vitro and greenhouse-grown transgenic plants demonstrated enhanced resistance against the fungal pathogen Alternaria solani (causal agent of early blight).
In this study, the efficacy of the essential oil of Mentha longifolia, Achillea arabica and Artemisia absinthium plants were evaluated against important soil-borne fungal pathogens as Verticillium dahliae, Rhizoctonia solani, and Fusarium oxysporum. Essential oils were obtained from plants by hydrodistillation method and the chemical components of essential oils were determined by analyzing by gas chromatography-mass spectrometry. The main components found as piperitone oxide (13.61%), piperitenone oxide (15.55%), pulegone (12.47%), 1-menthone (5.75%), and camphor (5.75%) in M. longifolia, á-selinene 13.38%, camphor 13.34%, L-4-terpineneol 8.40%, (-)-á-Elemene 7.01%, 1,8-cineole 4.71%, and (-)-spathulenol 3.84% in A. arabica, and á-thujone (34.64%), 1,8-cineole (19.54%), pulegone (7.86%), camphene (5.31%), sabinene (4.86%), and germacrene-d (3.67%) in A. absinthium. The antifungal activities of the oils were investigated 0.05, 0.1, 0.25, 0.5, 1.00, and 2.00 μl/ml concentrations with the contact effect method. M. longifolia oil (1.00 and 2.00 μl/ml) has displayed remarkable antifungal effect and provided 100% inhibition on mycelial growth of V. dahliae, R. solani and F. oxysporum. The results obtained from this study may contribute to the development of new alternative and safe methods against soil-borne fungal pathogens.

To examine the effects of water stress and Cenangium ferruginosum (CF) on the fungal endophytic community of needles of Pinus koraiensis (PK), fungal endophytes isolated from the needles of 5-year-old PK seedlings were compared before and after exposure to water stress conditions and artificial inoculation with CF ascospores. Artificial CF inoculation was successfully confirmed using PCR with CF-specific primers (CfF and CfR). For comparison of the degree of water deficit in water-stressed and control groups of PK seedlings infected with CF, the water saturation deficit and water potential were measured. Lower water potential estimates were found in the water-stressed seedlings than in the control group. The fungal endophytes isolated from the second-year needles of non-water-stressed seedlings before and after CF inoculation revealed that primary saprobes were approximately 30% and 71.7%, respectively, and the remaining endophytes were rot fungi or pathogens. Sixty days after CF inoculation, diverse fungal endophytes in the first-year needles were isolated from the water-stressed seedlings. However, some fungal endophytes isolated from the non-water-stressed seedlings were also identified. Fungal endophytes in the second-year needles of the water-stressed and non-water-stressed seedlings were approximately 8% and 71.7% of saprobes, respectively, and the remaining endophytes were rot fungi or pathogens. On the basis of the results, we conclude that water deficit and CF can have an effect on fungal endophytic communities in the needles of PK seedlings.
Bioconverted linolenic acid (bLNA) obtained from linolenic acid by Pseudomonas aeruginosa PR3, showed anti-fungal activity against plant pathogens such as B. cinerea, F. oxysporum, F. solani, P. capsici and C. capsici. The oil sample also showed anti-fungal activity with MIC values, ranging from >250 to >1,000 ${\mu}g/ml$. Varied concentrations of bLNA had a great potential effect on spore germination of different fungi.
Many of the fungicides and antibiotics currently available against plant pathogens are of limited use due to the emergence of resistant strains. In this study, we examined the effects of diphenyleneiodonium chloride (DPIC), an inhibitor of the superoxide producing enzyme NADPH oxidase, against fungal and bacterial plant pathogens. We found that DPIC inhibits fungal spore germination and bacterial cell proliferation. In addition, we demonstrated the potent antibacterial activity of DPIC using rice heads infected with the bacterial pathogen Burkholderia glumae which causes bacterial panicle blight (BPB). We found that treatment with DPIC reduced BPB when applied during the initial flowering stage of the rice heads. These results suggest that DPIC could serve as a new and useful antimicrobial agent in agriculture.

Dual biocontrol of both insects and plant pathogens has been reported for certain fungal entomopathogens, including Beauveria bassiana and Lecanicillum spp. In this study, we demonstrate, for the first time, the dual biocontrol potential of two fungal isolates identified by morphological and phylogenetic analyses as Isaria javanica. Both these isolates caused mortality in the greater wax moth, and hence can be considered entomopathogens. Spores of the isolates were also pathogenic to nymphs of the green peach aphid (Myzus persicae), with an $LC_{50}$ value of $10^7spores/mL$ 4 days after inoculation and an $LT_{50}$ of 4.2 days with a dose of $10^8spores/mL$. In vitro antifungal assays also demonstrated a strong inhibitory effect on the growth of two fungi that are pathogenic to peppers, Colletotrichum gloeosporioides and Phytophthora capsici. These results indicate that I. javanica isolates could be used as novel biocontrol agents for the simultaneous control of aphids and fungal diseases, such as anthracnose and Phytophthora blight, in an integrated pest management framework for red pepper.

A small cationic peptide (JH8944) was tested for activity against a number of pathogens of agricultural crops. JH8944 inhibited conidium growth in most of the tested plant pathogens with a dose of $50{\mu}g/ml$, although one isolate of Fusarium oxysporum was inhibited at $5{\mu}g/ml$ of JH8944. Most conidia of Fusarium graminearum were killed within 6 hours of treatment with $50{\mu}g/ml$ of JH8944. Germinating F. graminearum conidia required $238{\mu}g/ml$ of JH8944 for 90% growth inhibition. The peptide did not cause any damage to tissues surrounding maize leaf punctures when tested at a higher concentration of $250{\mu}g/ml$ even after 3 days. Liposomes consisting of phosphatidylglycerol were susceptible to leakage after treatment with 25 and $50{\mu}g/ml$ of JH8944. These experiments suggest this peptide destroys fungal membrane integrity and could be utilized for control of crop fungal pathogens.
Anti-fungal materials producing bacteria were isolated from soil by bennett's agar and actinomycete isola-tion agar medium. The bacterla were identified as synonym of Actinomycetes. Based on the data obtained from its morphological and colony characteristics. The medium for production of anti-fungal materials was YEME (yeast extract 4 g, malt extract l0g, glucose 4 g, D.W 1ι, pH 7.0${\pm}$0.2). The culture conditions were 30$^{\circ}C$, 7 days and 200 rpm in shaking incubator. No. 13, No. 15 and No.28 strains were produced anti-fungal materials against fungal plant pathogens. Specially, The No. 28 strain showed a powerful biopesticide activity and broad spectrum effects of anti -fungal materials on Collectrichum coccodes, Botrytis cinerea, Cladosporium cucumerinum, Didymella bryoniae.
The increasing emergence of lead drugs for the resistance produced by the pathogenic strains and arrival of new diseases have initiated the need for searching novel metabolites with best anticancer and antimicrobial properties than the existing one. With this view, the investigation was conducted for the isolation, identification, and biological evaluation of potential endophytic fungi of Aegle marmelos, a medicinal tree used for more than three decades, for curing various disorders. A total of 169 endophytic fungal strains obtained from sampling and among those 67 were pigmented strains. Upon antagonistic screening, five endophytic fungal strains exhibited antagonistic potentiality by inhibiting the pathogens. These five potent strains were characterized at molecular level by sequencing the amplified internal transcribed spacer (ITS) 1 and ITS 4 regions of rDNA and they were grouped under order Pleosporales, Eurotiales, and Capnodiales. The metabolites from the respective strains were produced in fungal culturing media and extracted using polar solvents. Further, the extracts of five endophytes manifested antimicrobial activity against tested clinical pathogens and Alternaria alternata (FC39BY), Al. citrimacularis (FC8ABr), and Curvularia australiensis (FC2AP) exhibited significant antimicrobial profile against 9 of 12 tested pathogens, showing broad spectrum activity. The antioxidant levels of all the five endophytes revealed the highest activity at least concentrations, and major activity was unveiled by the members of order Pleosporales FC2AP and FC8ABr. This research explains the value of endophytic fungal extracts and its significance of antimicrobial and antioxidant properties.
Lichen-forming fungi (LEF) were isolated from 67 Hungarian lichen species from ascospores or thallus fragments. LFF were successfully isolated from 26 species with isolation rate of 38.8%. Of the total number of isolation from ascospores (27 species) and thallus fragments (40 species), 48% and 32.5% of the species were successfully isolated, respectively. Comparison of rDNA sequences of ITS regions between the isolated LFF and the original thallus confirmed that all the isolates originated from the thallus fragments were LEF. The following 14 species of LEF were newly isolated in this study; Acarospora cervina, Bacidia rubella, Cladonia pyxidata, Lasallia pustulata, Lecania hyaline, Lecanora argentata, Parmelina tiliacea, Parmotrema chinense, Physconia distorta, Protoparmeliopsis muralis, Ramalina pollinaria, Sarcogyne regularis, Umbilicaria hirsuta, Xanthoparmelia conspersa and X. stenophylla. Antifungal activity of the Hungarian LFF was evaluated against plant pathogenic fungi of Colletotrichum acutatum, C. coccodes and C. gloeosporioides, causal agent of anthracnose on hot pepper. Among the 26 isolates, 11 LFF showed more than 50% of inhibition rates of mycelial growth of at least one target pathogen. Especially, LFF of Evernia prunastri, Lecania hyalina and Lecanora argentata were remarkably effective in inhibition of mycelial growth of all the tested pathogens with antibiotic mode of action. On the other hands, five isolates of Cladonia furcata, Hypogymnia physodes, Lasallia pustulata, Ramalina fastigiata and Ramalina pollinaria exhibited fungal lytic activity against all the three pathogens. Among the tested fungal pathogens, C. coccodes seemed to be most sensitive to the LFF. The Hungarian LFF firstly isolated in this study can be served as novel bioresources to develop new biofungicides alternative to current fungicides to control hot pepper anthracnose pathogenic fungi.
