The change on characterizations of fruiting body according to the growing humidity in Flammulina velutipes (팽이버섯 생육습도에 따른 균주별 자실체의 특성 변화)

  • We investigated the morphological traits, hardness and color of fruting bodies on eight strains according to changes of the humidities in mushroom house in Flammulina velutipes. The morphological traits like length and thickness of stipe, diameter and thickness of pilei didn't show any relations to humidity. Variation of these traits were rather showed by strains. ASI 4065 and ASI 4031 produced the longest stipes and ASI 4103 did the shortest ones. Fruiting body of ASI 4103 showed the most thickness of stipe, diameter and thickness of pilei. The pilei of ASI 4065, ASI 4021 and the stipe of ASI 4153 was the thinest. Brightness (L) values of white strains were increased at the low humidity except ASI 4021. But chroma (a, b) value showed little variation according to the humidity. L value of brown strains usually increased at the low humidity and a chroma (a, b) value showed little variation according to the humidity. But the chroma (a) value of pilei increased and the chroma (b) value of pilei decreased at the hight humidity. Most strains were brighter at pilei than at stipes. Hardness were not any relations to the humidity. In general, hardness of stipes were higher than that of pilei and their degrees were different according to strains. Hardness of ASI 4021 was the highest and ASI 4166 was the lowest. White strains were higher than brown strains at the hardness. As a result, hardness was rather affected by strains than by humidity.

Screening of Chemicals on Bacterial Brown Blotch Caused by Pseudomonas tolaasii on Flammulina velutipes (팽이버섯 세균성갈색무늬병(Pseudomonas tolaasii) 방제약제 선발)

  • This study was carried out in an attempt to select chemicals such as zeolite granules, chitosan, wood-vinegar, $Ca(OCl)_2$, NaOCl, against bacterial brown blotch caused by Pseudomonas tolaasii on Flammulina velutipes in laboratory and mushroom unit. Results obtained using these chemicals were summarized. Inhibitory effect on $Ca(OCl)_2$ and wood-vinegar with 0.5% concentration was shown to caual pathogen, P. tolaasii, by slight damage on the mycelial growth of F. velutipes. Those materials were recognized as promising one for control of bacterial brown bloth on F. velutipes. Disease incidence in control was the highest value as 43.8%; whereas chemical treatment was estimated as 14.6% with 0.5% of $Ca(OCl)_2$. Disease incidence was inhibited about 20% in chemical treatment with 0.5% of wood-vinegar, 1.0% of $Ca(OCl)_2$, and 26.1% with 1.0% of wood-vinegar. For the inhibition effects of chemical concentrations, it was effective at the low concentration which was 0.5% rather than that of 1.0%. Quality of mushrooms was significantly improved, and yields was also increased by 30% in the chemical treatment. In case of $Ca(OCl)_2$ treatment at 1.0% concentration, the yields was increased by 35.6% showed the highest value among tested chemicals. However, the overall effects including disease incidence, quality and yields suggests that $Ca(OCl)_2$ is relatively more effective than wood-vinegar, and the optimal concentration controlling the disease was $0.5{\sim}1.0%$ with $Ca(OCl)_2$ and 0.5% with wood-vinegar.

Effects of spent mushroom (Flammulina velutipes) substrates on in vitro ruminal fermentation characteristics and digestibility of whole crop sorghum silage (팽이버섯 수확후배지 첨가가 수수 사일리지의 in vitro 반추위 발효특성 및 소화율에 미치는 영향)

  • The in vitro experiment was conducted to ensure the supplemental level of spent Flammulina velutipes mushroom substrates (SMS) as an energy source in manufacturing of whole crop sorghum silage. Sorghum harvested at heading stage was ensiled with spent mushroom substrates of 20% (S-20), 40% (S-40) and 60% (S-60) as fresh matter basis for 6 week. The experiment was conducted by 3, 6, 9, 12, 24, 48 hrs of incubation time with 3 replications. The silages were evaluated fermentation characteristics and dry matter digestibility (DMD) in vitro. The pH of in vitro solution was inclined to decrease with elapsing the incubation time, and that of the S-20 was significantly (P<0.05) lower than the other treatment at 48 hr of incubation. Gas production was greater (P<0.05) in the S-20 than the other treatments at 6 and 12 hrs of incubation. The microbial growth in vitro was inclined to decrease following 24 hr of incubation, and thereafter sustained the similar levels. In vitro dry matter digestibility (IVDMD) was lowered by increasing the supplemental level of spent mushroom substrate, and was a low level in the S-60 throughout whole incubation time. Although the IVDMD for S-40 was steadily increased from 9 hr of incubation and reached to similar level with the S-20 at 48 hour of incubation, however SMS for whole crop sorghum silage fermentation might as well add about 20 to 30% in fresh matter basis when considering DMD.

Effects of supplementation of spent mushroom (Flammulina velutipes) substrates on the fermentative quality of rye silage (팽이버섯 수확후배지 첨가가 호밀 사일리지의 발효 품질에 미치는 영향)

  • This study was conducted to know the optimal supplementary level of spent mushroom substrates (Flammulina velutipes) as an energy source and fermentation period in manufacturing of rye silage. Whole crop rye was harvested at full bloom stage and ensiled with spent mushroom substrates of 20%(S-20), 40%(S-40) and 60%(S-60) as fresh matter basis. Each silage was prepared in plastic buckets included with vinyl bag by three replications and fermented for 3, 6 and 9 weeks, respectively. Moisture contents of whole crop rye at full bloom stage and spent mushroom substrates were 62.3% and 54.3%, respectively, and those of silages was ranged from 58% to 64%. Ether extracts content of silages was significantly (P<0.05) increased in 6 weeks of fermentation period. The pH of silages ranged from 4.46 to 5.05, and fluctuated in the changes by fermentation period beside of decreased with elapsing the period in the S-60. The organic acid content of silages was higher in the order of lactic acid, butyric acid, acetic acid and propionic acid. Lactic acid content was higher when fermented for 6 weeks compared to the other fermentation period. Flieg's score for estimation of silage quality ranged from 63 to 80, and was relatively high level in the S-20 and the S-40 fermented for 6 weeks, and was high level in S-20, and was relatively low level in the silages fermented for 3 week. Hence, we suggest that supplementation of 20% spent mushroom substrates in fresh matter basis and fermentation for 6 weeks are resonable for a good quality of rye silage.

Fruiting body development and genetic analysis of somatic hybrids by protoplast fusion in edible fungi (식용버섯의 원형질체 융합체의 자실체 발생 및 유전분석)

  • Somatic hybrids of inter-compatible and inter-incompatible strains were obtained by protoplast fusion. The fusion products between compatible strains, Pleurotus ostreatus and P. florida, formed heterokaryons, while fusants between incompatible strains such as P. cornucopiae + P. florida, P. ostreatus + Ganoderma applanatum, P. florida + Ganoderma lucidum, and P. ostreatus + Flammulina velutipes formed synkaryons that retained genes from both parents. The heterokaryons showed the same level of basidioma development. In contrast, the synkaryons showed unique characteristics including clamp connection formation at mitosis, either partner basidioma development, and abnormal segregation and recombination compared with inter-compatible strains. Synkaryons can be classified into homokaryoyic and heterokaryotic type. A comparison of somatic hybrids with compatible and incompatible strains was made using random amplified polymorphic DNA (RAPD) analysis. The heterokaryons between compatible species showed the same level of variability and contained both parental RAPD bands. In contrast, most of the synkaryons between incompatible species showed similarity to those of either parental bands and non-parental RAPD bands. Synkaryons can be classified into microgenome insertion type and macrogenome insertion type. A tetrapolar mating system was found among monospore isolates in somatic hybrids and wild type P. ostreatus. Homokaryons from each somatic hybrid combination were paired with tester homokaryons of the initial wild type of P. ostreatus. The changed mating types were identified in progenies. The pattern of mating type switching in somatic hybrids depends on compatibility of fusion partner. There are several factors related to the mechanism of clamp connection formation and fruiting body development of synkaryons. Of these,the major factor may be associated with self-fertility and mating type switching such as homokaryotic fruiting of wild type P. ostreatus. This review will discuss these aspects.

Analysis of growth environment of Flammulina velutipes using the smart farm cultivation technology (병재배 팽이버섯의 스마트팜 재배를 통한 생육환경 분석)

  • In this study, smart farm technology was used by farmers cultivating 'CHIKUMASSHU T-011' in order to develop an optimal growth model for the precision cultivation of bottle-grown winter mushroom and the results of the same are mentioned herein. Farmers participating in the experiment used 60 ㎡ of bed area with 4 rows and 13 columns of shelf shape, 20 horsepower refrigerator, 100T of sandwich panel for insulation, 6 ultrasonic humidifiers, 12 kW of heating, and 20,000 bottles of Flammulina velutipes mushroom spores. The temperature, humidity, and carbon dioxide concentrations, which directly affect the growth of the mushroom, were collected and analyzed from the environmental sensors installed at the winter mushroom cultivation area. The initial temperature was found to be 14.5℃, which was maintained at 14℃ to 15℃ until the 10th day. In the restriction phase, the initial temperature was 4℃ and was maintained between 2℃ and 3℃ until the 15th day, while during the growth phase, it was maintained between 7.5℃ to 9.5℃. Analysis of the humidity data revealed initial humidity to be 100%, which varied between 88% to 98% during primordia formation period. The humidity remained between 77% to 96% until the 15th day, in the restriction phase and between 75% to 83% during the growth phase. The initial carbon dioxide concentration was 3,500 ppm and varied between 3,500 ppm to 6,000 ppm during primordia formation period and was maintained at 6,000 ppm until the 15th day. During the growth phase, the carbon dioxide concentration was found to be over 6,000 ppm. Fruiting body characteristics of 'CHIKUMASSHU T-011' cultivated in the farmhouse were as follows: Pileus diameter of 7.5 mm and thickness of 4.1 mm, stipe thickness of 3.3 mm, and length of 154.2 mm. The number of valid fruiting bodies was 1,048 unit per 1,400 mL bottle, and the individual weight was 0.71 g per unit. The yield of fruiting bodies was 402.8 g per 1,400 mL bottle.

Effects of Dietary Flammulina velutipes Mycelium on Physico-chemical Properties and Nutritional Components of Chicken Meat (팽이버섯 균사체의 급여가 닭고기의 이화학적 특성과 영양성분에 미치는 영향)

  • The purpose of this study was to evaluate the effects of dietary Flammulina velutipes mycelium (FVM) on physico-chemical properties and nutritional components of chicken meat. Ninety-six broiler chicks (HanHyup No. 3, Korea) were divided into four groups: control (basal diet), T1 (supplemented with 1% FVM), T2 (3%), and T3 (5%). Broiler chicks in each group were slaughtered at 7 weeks of age, and their breast and thigh meat were collected. Lightness ($L^*$) and yellowness ($b^*$) in the T2 and T3 groups were higher than in the control group (p<0.05). The loss of breast meat during cooking was increased in the T1, T2, and T3 groups, and the water-holding capacity of the breast meat was also decreased in these three groups (p<0.05). The water-soluble protein solubility was lower in the T1, T2, and T3 groups than in the control group (p<0.05), but the salt-soluble protein solubility in these three groups was higher than that in the control group (p<0.05). The crude fat content of the breast meat was decreased and that of the thigh meat was increased as the level of FVM was increased (p<0.05). The unsaturated fatty acid content of the breast meat was higher in the T2 and T3 groups than in the control group (p<0.05). The free amino acid content was high in the T1, T2, and T3 groups compared to the control. In conclusion, these results showed that feeding with 3% and 5% FVM increased the L and b values in thigh meat, improved the salt-soluble protein solubility, and increased the content of unsaturated fatty acids and free amino acids.

Effects of Using Cold Water on Mixing Sawdust Media for Flammulina velutipes (고온기 팽이버섯 병재배 배지제조시 저온수 이용 효과)

  • This study was conducted to verify the cause of suppression symptom in mycelial growth during summer and to be able to establish a countermeasure. Cultivation of Flammulina velutipes was experimented with varying elapsed time of 0, 3, 6, 9 hours after mixing the sawdust media and two kinds of water temperature (normal water, $24^{\circ}C$ and cold water, $6^{\circ}C$) for mixing sawdust media. There were trends of increased media temperature from $24^{\circ}C$ to $31^{\circ}C$ and decreased pH from 6.5 to 5.2~5.6 with varying elapsed time from mixing the media to sterilization. Bacterial density also increased with bacterial density in Medium $24^{\circ}C$ being 1.9~4.1 times higher than that in Medium 6. Growth of F. velutipes was delayed with dual culture of bacteria isolated from sawdust media. Total nitrogen content of sawdust media was lowered by elapsed time from mixing the media to sterilization. The use of normal water($24^{\circ}C$) delayed mushroom growth by 1~2 days compared with that of cold water($6^{\circ}C$). Furthermore, mycelial growth of F. velutipes in the bottle cultivation ceased 9 hours after mixing the media. Primordia formation of F. velutipes was delayed by 1~3 days by elapsed time after mixing sawdust media, while fruit-body yield decreased by 7~12% 6 hours after mixing the media. Fruit-body yield was more stabilized with the use of cold water($6^{\circ}C$) than with that of normal water($24^{\circ}C$). Results showed that it is effective to use cold water as low as $6^{\circ}C$ in mixing media for F. velutipes cultivation, especially during summer when environmental temperature is high, high pressure sterilization after bottling work can prevent bacterial propagation in the media and can stabilize media ingredient.

  • The in vitro experiment was conducted to ensure the supplemental level of spent Flammulina velutipes mushroom substrates(SMS) as an energy source in manufacturing of rye silage. Rye harvested at heading stage was ensiled with spent mushroom substrates of 0%(Control), 20%(R-20), 40%(R-40) and 60%(R-60) as fresh matter basis for 6week. The rumen fluid for preparation of in vitro solution was collected from two cannulated Holstein bulls fed a 40:60 concentrate:timothy diet. The experiment was conducted by 3, 6, 9, 12, 24, and 48 hrs of ncubation time with 3 replications. The silages were evaluated fermentation characteristics and dry matter digestibility(DMD) in vitro. The pH of in vitro solution was inclined to decrease with elapsing the incubation time, and that of the R-60 was significantly(p<0.05) lower than the other treatment at 48 hr of incubation. The microbial growth in vitro was inclined to increase with elapsing the incubation time, and that of the R-20 was significantly(p<0.05) greater than the Control at 48 hr of incubation. Gas production was greater(p<0.05) in the Control than the other treatments at 48 hr of incubation. In vitro dry matter digestibility(IVDMD) was higher with increasing the supplemental level of SMS, and was significantly(p<0.05) lower in the Control compared with other treatments throughout whole incubation time. The IVDMD for R-60 was the highest(p<0.05) among treatments at 24 hr and 48 hr of incubation. Considering of above results and the availability of SMS, SMS could be supplemented by 60% in fresh matter basis for rye silage fermentation.

Effects of Addition of Eggshell to Sawdust Substrate on the Growth and Development of Winter Mushroom, Flammulina velutipes (계란껍질 첨가배지(添加培地)가 팽이버섯의 균사생장(菌絲生長)과 자실체(子實體)에 미치는 영향(影響))

  • Tile effects of addition of eggshell to sawdust substrate for the growth of F. velutipes were investigated. Eggshell used in this study contained 20.7% C, 0.81% N, 2530 ppm $P_2O_5$ and 44.37% Ca. The addition of eggshell resulted in the increase in bulk density and decrease of moisture content of the substrate. The addition of eggshell significantly increased the yield of the mushroom fruitbody. The addition rate of 15% (v/v), by 25% and at the rates of 5% and 10%, about 20%. Although the addition of eggshell to substrate did not improve the quality of mushroom, it increased the number of effective stipes as compared to control plot; approximately 13% more than in the control plot.
