Objectives: This study was performed to investigate the anti-obesity effects of Gambi-bang 4 (GBB4) on obesity-induced mice. Methods: C57BL/6 mice were divided into four groups (normal, high fat diet with control, high fat diet with Reductil, high fat diet with GBB4 extract) and fed for 8 weeks. We observed body weight change, the weight change of the adipocytes in body, total cholesterol, LDL-cholesterol, HDL-cholesterol and triglycerides, serum leptin level, expression of ${\beta}3AR$ and leptin genes in 3T3-L1 adipocytes and adipose tissue, and histological changes of adipose tissue and liver cells. Results: 1. Compared with the control group, the GBB4 group was significantly lower in body weight, weight of adipocytes, and amount of glucose. 2. The GBB4 group was significantly lower in the amount of total cholesterol, LDL-cholesterol and triglycerides, and HDL-cholesterol compared with the control. 3. Compared with the control group, the GBB4 group was significantly lower in the amount of serum leptin. 4. The GBB4 group was significantly higher in the revelation of ${\beta}3AR$ and leptin in 3T3-L1 adipocytes and primary adipose cells compared with the control. 5. Compared with the control group, the GBB4 group was smaller in the size of adipocytes in adipose tissue and the adipose vacuoles in liver tissue were decreased. Conclusions: These results suggested that GBB4 has inhibitory effects on obesity. GBB4 might be applied in treatment of obesity, so further studies analyzing its effects are needed.
This study was conducted to examine the effect of different types of milking instrument and season on milk composition (fat, protein, non-fat milk solids, milk urea nitrogen and somatic cell count) on Holstein-Friesian lactating cows. Raw milk samples were collected from 20 automatic milking system (AMS) installed dairy farms and 85 conventional milking system (CMS) installed dairy farms. Milk fat, protein, NFMS and MUN contents did not changed between AMS and CMS and different seasons. On the other hand, the average SCC in AMS was $239{\times}10^3cells/mL$ which was lower than in CMS ($373{\times}10^3cells/mL$, (p<0.05). In conclusion, present results indicated that SCC in milk might be influenced by AMS. However, this area needed more study to confirm the reason of reducing SCC content from AMS.
Recent studies have shown that Rosa canina L. and tiliroside, the principal constituent of its seeds, exhibit anti-obesity and anti-diabetic activities via enhancement of fatty acid oxidation in the liver and skeletal muscle. However, the effects of rosehip, the fruit of this plant, extract (RHE), or tiliroside on lipid accumulation in adipocytes have not been analyzed. We investigated the effects of RHE and tiliroside on lipid accumulation and protein expression of key transcription factors in both in vitro and in vivo models. RHE and tiliroside inhibited lipid accumulation in a dose-dependent manner in 3T3-L1 cells. We also analyzed the inhibitory effect of RHE on white adipose tissue (WAT) in high-fat diet (HFD)-induced obesity mice model. Male C57BL/6J mice were fed HFD or HFD supplemented with 1% RHE (HFDRH) for 8 weeks. The HFDRH-fed group gained less body weight and had less visceral fat than the HFD-fed group. Liver weight was significantly lower in the HFDRH-fed group and total hepatic lipid and triglyceride (TG) content was also reduced. A significant reduction in the expression of peroxisome proliferator-activated receptor gamma (PPAR${\gamma}$) was observed in epididymal fat in the HFDRH-fed group, in comparison with controls, through Western blotting. These results suggest that downregulation of PPAR${\gamma}$ expression is involved, at least in part, in the suppressive effect of RHE on lipid accumulation in WAT.
BACKGROUND/OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) is a leading cause of chronic liver disease and is closely associated with metabolic syndrome. In the present study, we observed the effect of ethanol extract of Allium fistulosum (EAF) on NAFLD and have suggested the possibility of using EAF as a natural product for application in the development of a treatment for NAFLD. MATERIALS/METHODS: The preventive effect on hepatic lipid accumulation was estimated by using an oleic acid (OA)-induced NAFLD model in vitro and a Western diet (high-fat high-sucrose; WD)-induced obese mouse model. Animals were divided into three groups (n = 7): normal diet group (ND), WD group, and WD plus 1% EAF group. RESULTS: EAF reduced OA-stimulated lipid accumulation in HepG2 cells in the absence of cellular cytotoxicity and significantly blocked transcriptional activation of sterol regulatory element-binding protein 1 and fatty acid synthase genes. Subsequently, we investigated these effects in vivo in mice fed either ND or WD in the presence or absence of EAF supplementation. In comparison to the ND controls, the WD-fed mice exhibited increases in body weight, liver weight, epididymal fat weight, and accumulation of fat in hepatocytes, and these effects were significantly attenuated by EAF supplementation. CONCLUSIONS: Allium fistulosum attenuates the development of NAFLD, and EAF elicits anti-lipogenic activity in liver. Therefore, EAF represents a promising candidate for use in the development of novel therapeutic drugs or drug combinations for the prevention and treatment of NAFLD.
중간엽줄기세포는 성체줄기세포의 한 종류로, 자기재생산능력(self-renwal)과 다분화능(multipotency)을 가지고 있고, 다양한 자양인자(trophic factors)들을 분비한다. 뿐만 아니라, 중간엽줄기세포는 골수, 지방, 탯줄과 같은 조직에서 쉽게 얻을 수 있기 때문에 줄기세포치료에 좋은 도구로 이용되고 있다. 하지만, 줄기세포치료의 효율성을 높이기 위해 추출한 세포의 개체 수를 늘리는 과정에서 중간엽줄기세포는 점차적인 노화를 겪게 되고, 이는 줄기세포 자체의 기능적인 감소를 야기한다. 인체 내에서, 노화된 줄기세포는 조직 내의 항상성 유지에 부정적인 영향 을 미치게 되고, 이러한 상태가 지속되면 대표적인 노인성 질환인 퇴행성 질환의 원인이 된다. 최근 연구들에 의하면 중간엽줄기세포가 노화를 겪을 때, 노화 관련된 DNA 메틸화 패턴의 변화와 히스톤의 변형이 일어남을 확인하였다. 또한, 중간엽줄기세포의 노화에 있어서 DNA 메틸화효소(DNA methyltransferase) 억제제와 히스톤 아세틸화효소(histone deacetylase) 억제제가 부분적으로 노화를 개선하는 효과를 관찰한 연구사례들이 있다. 본 총설에서는, 노화에 따른 후생유전학적인 변화에 의해, 조절되는 노화 관련 유전자들과 중간엽줄기세포의 노화에 대한 연구사례들을 분석하여 서술하고자 한다.

BACKGROUD/OBJECTIVES: Allomyrina dichotoma larva (ADL), one of the many edible insects recognized as future food resources, has a range of pharmacological activities. In a previous study, an ADL extract (ADLE) reduced the hepatic insulin resistance of high-fat diet (HFD)-induced diabetic mice. On the other hand, the associated molecular mechanisms underlying pancreatic beta-cell dysfunction remain unclear. This study examined the effects of ADLE on palmitate-induced lipotoxicity in a beta cell line of a rat origin, INS-1 cells. MATERIALS/METHODS: ADLE was administered to high-fat diet treated mice. The expression of apoptosis-related molecules was measured by Western blotting, and reactive oxidative stress generation and nitric oxide production were measured by DCH-DA fluorescence and a Griess assay, respectively. RESULTS: The administration of ADLE to HFD-induced diabetic mice reduced the hyperplasia, 4-hydroxynonenal levels, and the number of apoptotic cells while improving the insulin levels compared to the HFD group. Treatment of INS-1 cells with palmitate reduced insulin secretion, which was attenuated by the ADLE treatment. Furthermore, the ADLE treatment prevented palmitate-induced cell death in INS-1 cells and isolated islets by reducing the apoptotic signaling molecules, including cleaved caspase-3 and PARP, and the Bax/Bcl2 ratio. ADLE also reduced the levels of reactive oxygen species generation, lipid accumulation, and nitrite production in palmitate-treated INS-1 cells while increasing the ATP levels. This effect corresponded to the decreased expression of inducible nitric oxide synthase (iNOS) mRNA and protein. CONCLUSIONS: ADLE helps prevent lipotoxic beta-cell death in INS-1 cells and HFD-diabetic mice, suggesting that ADLE can be used to prevent or treat beta-cell damage in glucose intolerance during the development of diabetes.
곤충의 난황형성에는 각종호르몬들이 먹이 등과 함께 매우 중요하게 작용하고 있으며, 이러한 호르몬들 중에서는 신경분비호르몬, 유약호르몬 및 엑디스테로이드 등이 주작용을 한다. 본 총설에서는 곤충호르몬을 포함한 곤충성장조절제가 곤충의 난황형성에 작용하여 어떤 영향을 주는지에 대하여 살펴보고 종합적 해충관리 인자로서 이용 가능성을 조사하였다. 곤충의 난황형성은 일반적으로 뇌에서 분비하는 신경분비호르몬의 작용과 난소에서 분비되는 엑디스테로이드 및 알라타체에서 분비되는 유약호르몬이 지방체를 자극하여 난황소전구물질을 합성하게 하며, 혈림프로 분비된 난황소 전구물질은 난모세포로 흡수되어 난발육이 이루어진다. 곤충호르몬을 포함한 곤충성장조절제는 이러한 난황형성과정 중에 각종 호르몬분비 기관(알라타체와 난소 등) 및 여러 가지 생리기관(지방체, 중장상피 그리고 침샘 등)에 작용하여 용해, 파괴 및 세포독성을 나타내 정상적인 생식작용을 하지 못하도록 한다. 따라서 곤충의 종류에 따라서는 포유동물에 독성이 낮은 곤충호르몬을 포함한 곤충성장조절제를 종합적 해충관리의 일환으로 사용함으로서 효과적인 해충방제가 가능할 것으로 보인다.
본 연구는 소포체스트레스(endoplasmic reticulum stress)에 의해 유발되는 지방간(hepatic steatosis)에 대한 오미자추출물(Schisandra chinensis)의 주요성분인 gomisin A의 지방간 보호 효능에 대하여 연구하였다. 이를 위해 HepG2 세포에 소포체스트레스 유도물질인 tunicamycin 또는 palmitate을 처리하여 세포에서의 지방간 모델을 만들어 실험을 진행 하였으며, 소포체스트레스 표지자(marker)인 GRP78, CHOP, XBP-1의 발현을 측정하였다. Tunicamycin 처리한 세포에서는 GRP78, CHOP, XBP-1의 발현이 증가되었으나, gomisin A를 처리 한 세포에서는 이들의 발현 증가가 억제됨을 확인하였다. 이는 palmitate를 처리한 HepG2 세포에서도 palmitate에 의해 증가하는 소포체스트레스 표지자들이 gomisin A을 처리한 세포에서 발현이 감소함을 확인하였다. 이러한 결과에 의해, gomisin A는 소포체스트레스를 억제함을 알 수 있었다. 다음으로 gomisin A가 in vivo에서 소포체스트레스 및 지방간에 대한 보호효과가 있는지 확인하기 위해, tunicamycin과 고지방(high fat diet)으로 식이 한 쥐에서 소포체스트레스와 지방간의 보호효능에 대해 실험을 진행하였다. Tunicamycin과 고지방식이을 한 쥐의 간에서 중성지방이 증가하였으나, gomisin A를 처리한 쥐의 간에서 중성지방의 수준이 유의적으로 감소함을 확인하였다. 소포체스트레스 표지자들 역시 tunicamycin이나 고지방식이을 한 쥐에서 증가되나 gomisin A를 처리한 쥐에서 감소됨을 확인하였다. Gomisin A의 염증 반응에서의 조절기능을 확인하기 위하여 $TNF-{\alpha}$, IL-6 그리고 MCP1과 같은 염증관련 유전자들의 발현을 분석한 결과, tunicamycin이나 고지방식이을 한 쥐에서 염증유전자들의 발현이 증가하였으나 gomisin A를 처리한 쥐에서는 유의적으로 감소하였다. 종합적으로 본 연구 결과에 의하면, gomsin A는 소포체스트레스를 억제하여 지방간의 생성을 저해함을 알 수 있었다.

Present Food Legislation compels dairy industry to carry out analyses in order to guarantee the food safety and quality of products. Furthermore, in many cases industry pays milk according to bacteriological or/and nutritional quality. In order to do these analyses, several expensive instruments are needed (Milkoscan, Fossomatic, Bactoscan). NIRS technology Provides a unique instrument to deal with all analytical requirements. It offers as main advantages its speed and, specially, its versatility, since not only allows determine all the parameters required in milk analysis, but also allows analyse other dairy products, like cheese or whey. The objective of this study is to develop NIRS calibration equations to predict several quality parameters in goat milk, cheese and whey. Three sets of 123 milk samples, 190 cheese samples and 109 whey samples, have been analysed in a FOSS NIR Systems 6500 I spectrophotometer equipped with a spinning module. Milk and whey were analysed by folded transmission, using circular cells with gold surface and pathlength of 0.1 m, while intact cheese was analysed by reflectance using standard circular cells. NIRS calibrations were obtained for the prediction of chemical composition in goat milk, for fat (r$^2$=0.92; SECV=0.20%), total solids (r$^2$=0.95: SECV=0.22%), protein (r$^2$=0.94; SECV=0.07%), casein (r$^2$=0.93; SECV=0.07%) and lactose (r$^2$=0.89; SECV=0.05%). Moreover, equations have been performed to determine somatic cells (r$^2$=0.81; SECV=276.89%) and total bacteria (r$^2$=0.58; SECV=499.32%) counts in goat milk. In the case of cheese, calibrations were obtained for the prediction of fat (r$^2$=0.92; SECV=0.57), total solids (r$^2$=0.80; SECV=0.92%) and protein (r$^2$=0.70; SECV=0.63%). In whey, fat (r$^2$=0.66; SECV=0.08%), total solids (r$^2$=0.67; SECV=0.19%) and protein (r$^2$=0.76; SECV=0.07%) NIRS equations were obtained. These results proved the viability of NIRS technology to predict chemical and microbiological parameters and somatic cells count in goat milk, as well as chemical composition of goat cheese and whey.
Hypertension caused by high-fat and high-salt diets is is a well-known significant risk factor for cardiovascular and cerebrovascular diseases. In this study, to confirm the relationship between hypertension and immune cells, angiotensin (Ang) II was administered to Dahl salt-sensitive (SS) rats and Dahl salt-resistant (SR) rats. Then the expression of immune cells and the proinflammatory cytokines were compared between the SS and SR rats. It was observed that after administration of Ang II (50ng/kg/min) for three weeks, blood pressure was increased in the SS rats, but there was no significant change in the SR rats. In addition, the expression of T helper (Th) cells and Th 17 cells in the spleen and the expression of Th cell Rorγt and regulatory T regulatory (Treg) cells in the peripheral blood mononuclear cells did not show a significant difference between the two experimental groups even after the administration of Ang II.IL-1β expression was significantly increased in the kidney tissue of the SS rats, while there was no significant difference in the IL-6 expression in all the experimental groups. The results of this study suggest that Ang II induces hypertension by stimulating IL-1β secretion from renal macrophage in SS rats.
