  • Parasitylenchus orthotomici sp. n. is described as a nematode parasite of Orthotomicus an¬gulatus in Korea. In a sample of 170 beetles randomly collected from Pinus densiflora S. et. Z, 51. 8% were parasitized by nematodes. The number of nematodes per bark beetle was ranged from 1 to 97. The parasitisms of F, generation were ranged from 43.2% to 52%. There was no positive tendency in nematode parasitism between sun-side and shade-side.

  • The genetic analysis was caried out on the boiling-off ratio of cocoon layer, and the amounts of sericin and fibroin in the F1 generation from the diallel crosses of silkworm. The result obtained were as follows : 1. Boiling-off ratio and the amounts of sericin adn fibroin in male and female were over dominqnce and partial dominance, respectively. 2. The additive effect of gene was higher then the dominance effect in the amounts of sericin and fibroin in male and female, and in boiling off ration in female. 3. The direction of dominance in the amounts of sericin and fibroin in male and female was positive and that in boiling off ratio negative. 4. The effect of three characters on the combining ability was significant and their general combining ability was higher than the specific combining ability.

Fabrication of Porous Electrodes for Zinc-Ion Supercapacitors with Improved Energy Storage Performance (아연-이온 전기화학 커패시터의 에너지 저장 성능향상을 위한 다공성 전극 제조)

  • Zn-ion supercapacitors (ZICs) show high energy densities with long cycling life for use in electronic devices. Porous Zn electrodes as anodes for ZICs are fabricated by chemical etching process using optimized conditions. The structures, morphologies, chemical bonding states, porous structure, and electrochemical behavior are examined. The optimized porous Zn electrode shows a root mean square of roughness of 173 nm and high surface area of $153{\mu}m^2$. As a result, ZIC using the optimized porous Zn electrode presents excellent electrochemical performance with high specific capacitance of $399F\;g^{-1}$ at current density of $0.5A\;g^{-1}$, high-rate performance ($79F\;g^{-1}$ at a current density of $10.0A\;g^{-1}$), and outstanding cycling stability (99 % after 1,500 cycles). The development of energy storage performance using synergistic effects of high roughness and high surface area is due to increased electroactive sites by surface functionalization of Zn electrode. Thus, our strategy will lead to a rational design and contribute to next-generation supercapacitors in the near future.

MORE RELATIONS BETWEEN λ-LABELING AND HAMILTONIAN PATHS WITH EMPHASIS ON LINE GRAPH OF BIPARTITE MULTIGRAPHS

  • This paper deals with the λ-labeling and L(2, 1)-coloring of simple graphs. A λ-labeling of a graph G is any labeling of the vertices of G with different labels such that any two adjacent vertices receive labels which differ at least two. Also an L(2, 1)-coloring of G is any labeling of the vertices of G such that any two adjacent vertices receive labels which differ at least two and any two vertices with distance two receive distinct labels. Assume that a partial λ-labeling f is given in a graph G. A general question is whether f can be extended to a λ-labeling of G. We show that the extension is feasible if and only if a Hamiltonian path consistent with some distance constraints exists in the complement of G. Then we consider line graph of bipartite multigraphs and determine the minimum number of labels in L(2, 1)-coloring and λ-labeling of these graphs. In fact we obtain easily computable formulas for the path covering number and the maximum path of the complement of these graphs. We obtain a polynomial time algorithm which generates all Hamiltonian paths in the related graphs. A special case is the Cartesian product graph Kn☐Kn and the generation of λ-squares.

Linolenic Acid in Association with Malate or Fumarate Increased CLA Production and Reduced Methane Generation by Rumen Microbes

  • An in vitro study was conducted to investigate the effect of malate or fumarate on fermentation characteristics, and production of conjugated linoleic acid (CLA) and methane ($CH_4$) by rumen microbes when incubated with linolenic acid (${\alpha}-C_{18:3}$). Sixty milligrams of ${\alpha}-C_{18:3}$ alone (LNA), or ${\alpha}-C_{18:3}$ with 24 mM malic acid (M-LNA) or ${\alpha}-C_{18:3}$ with 24 mM fumaric acid (F-LNA) were added to the 150 ml culture solution consisting of 75 ml strained rumen fluid and 75ml McDougall's artificial saliva. Culture solution for incubation was also made without malate, fumarate and ${\alpha}-C_{18:3}$ (Control). Two grams of feed consisting of 70% concentrate and 30% ground alfalfa (DM basis) were also added to the culture solution of each treatment. In vitro incubation was made anaerobically in a shaking incubator up to 12 h at $39^{\circ}C$. Supplementation of malate (M-LNA) or fumarate (F-LNA) increased pH at 6 h (p<0.01) and 12 h (p<0.001) incubation times compared to control and linolenic acid (LNA) treatments. Both malate and fumarate did not influence the ammonia-N concentration. Concentration of total VFA in culture solution was higher for M-LNA and F-LNA supplementation than for control and LNA treatments from 6 h (p<0.040) to 12 h (p<0.027) incubation times, but was not different between malate and fumarate for all incubation times. Molar proportion of $C_3$ was increased by F-LNA and M-LNA supplementation from 6 h (p<0.0001) to 12 h (p<0.004) incubation times compared to control and LNA treatments. No differences in $C_{3}$ proportion, however, were observed between M-LNA and F-LNA treatments. Accumulated total gas production for 12h incubation was increased (p<0.0002) by M-LNA or F-LNA compared to control or LNA treatment. Accumulated $CH_4$ production for 12 h incubation, however, was greatly reduced (p<0.0002) by supplementing malate or fumarate compared to the control, and its production from M-LNA or F-LNA treatment was smaller than that from LNA treatment. Methane production from LNA, M-LNA or F-LNA treatment was steadily lower (p<0.01 - p<0.001) from 3 h incubation time than that from the control, and was also lower for M-LNA or F-LNA treatment at incubation times of 6 h (p<0.01) and 9 h (p<0.001) than for LNA treatment. Methane production from LNA, however, was reduced (p<0.01 - p<0.001) from 3 h to 9 h incubation times compared to the control. Both malate and fumarate increased concentration of trans11-$C_{18:1}$ from 3 h to 12 h incubation (p<0.01), cis9,trans11-CLA up to 6 h incubation (p<0.01 - p<0.01), trans10,cis12-CLA at 3 h (p<0.05) and 12 h (p<0.01), and total CLA for all incubation times (p<0.05) compared to corresponding values for the ${\alpha}-C_{18:3}$ supplemented treatment (LNA). In conclusion, malate and fumarate rechanneled the metabolic $H_2 pathway to production of propionate and CLA, and depressed the process of biohydrogenation and methane generation. Linolenic acid alone would also be one of the optimistic alternatives to suppress the $CH_4$ generation.

Relationships between Dapsone Metabolic Activity and Polymorphism of Arylamine N-acetyltransferase 2 in the F2 Hybrid Rats (잡종 2세대(Fischer 계: Wistar-Kyoto 계) 흰쥐에서 Arylamine N-acetyltransferase 2의 다형성과 Dapsone의 대사능과의 연관성에 대한 연구)

  • The arylamine N-acetyltransferases (NATs) are a family of enzymes that N-acetylate mylhydrazines and arylamines through transfer of an acetyl group from acetyl coenzyme A. This activity was found to vary among individuals as a Mendalian trait and the basis of the genetic differences in human NAT activity is one of the best of the genetic studied examples of pharmacogenetic variation. The classical N-acetylation polymorphism is regulated at the NAT2 locus, which segregates individuals into rapid, intermediate, and slow acetylator phenotypes. In this study, the relationship between NAT2 activity phenotype using HPLC:UV assay for the determination of dapsone and monoacetyldapsone in plasma and NAT2 genotype by PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) was investigated in the F2 hybrid (Fischer 344 vs Wistar-Kyoto) rats. Three Common mutant alleles at the NAT2 gene locus have been identified in the F2 generation progeny of Fischer 344 rats as raid acetylator and Wistar-Kyoto rats as slow acetylator segregated into three modes (low, intermediates, and high) with simple Mendelian inheritance. The metabolic activity of NAT2 of the intermediate and rapid acetylators is significant1y greater than slow acetylator, but the metabolic activity of rapid acetylator is not significantly different from Intermediate type. Therefore, we could observe that complete trimodal NAT2 genotypic alleles and incomplete trimodal NAT2 metabolic phenotypic distribution in tile F2 hybrid rats. These observations suggest that the relationships between NAT2 genotype and metabolic phenotype exists and F2 hybrid (Fischer 344: Wistar-Kyoto) animal models about NAT2 polymorphism might be applied in the toxicity and pharmacogenetic studies of arylamine drugs and carcinogens.

The Inheritance of Anatomical Components in Red Pepper (고추 일과중(一果重)의 구성요소(構成要素)에 대(對)한 유전(遺傳))

  • Six genetic populations, viz, $P_1$, $P_2$, $F_1$, $F_2$, $B_1$, and $B_2$ of three crosses involving five varieties ($342{\times}$Kimjanggochu, Masan${\times}$Hungarian was, and Masan${\times}$Jeju) were used for estimating heterosis, heterobeltiosis, inbreeding depression, genetic components, heritability and correlations for fresh and dry anatomical components of red pepper. The results obtained were summarized as follows. Heterosis was positive in most characters of three crosses and heterobeltiosis was negative except in cross $342{\times}$Kimjanggochu. Inbreeding depression was positive, showing decrease of weight in $F_2$ generation. Duplicate and complimentary type of epistasis were concerned with the expression of characters, and broad and narrow sense heritabilities of fruit weight and pericarp weight were fairly high and stem weight, placenta weight and seed weight were low. Correlations among the anatomical components were positively significant in fresh and dry weights.

Mapping QTLs for drought tolerance using an introgression line population from a cross between Ilpumbyeo and Moroberekan in rice (벼 일품벼/모로베레칸 조합의 이입계통을 이용한 내건성 유전자 탐지)

  • This study was conducted to map quantitative trait loci (QTLs) related to drought stress tolerance. An introgression line population derived from a cross, "Ilpum" / "Moroberekan" was used in this study. $F_1$ plants were backcrossed three times to Ilpum to produce $BC_3F_1$ plants. These plants were advanced by selfing for four generation and a total of 117 $BC_3F_5$ introgression lines were developed. These lines were evaluated for percent seed set and spikelets per panicle under the control (field) and drought condition. To identify QTLs related to drought tolerance, 134 SSR markers showing polymorph isms between the parents were genotyped for the 117 $BC_3F_5$ lines. A total of 6 QTLs associated with drought stress were detected on chromosomes 1, 3, 4, 7 and 10. These include two QTLs for phenotypic acceptability, two QTLs for percent seed set ($R^2$ = 19.0 - 20.9%), and two QTLs for spikelets per panicle ($R^2$ = 22.3 - 23.10%). The Moroberekan alleles at three loci contributed the positive effect for drought tolerance. The SSR markers linked to drought stress tolerance can not only facilitate the selection of valuable genes from Moroberekan, but also allow identification of lines with drought tolerance.

The Secretion Optimization of Oligopeptide with His-Pro Repeats in Bacillus subtilis and Its Anti-Diabetic Effects (고초균에서 His-Pro 반복서열을 갖는 Oligopeptide의 분비 최적화 및 항당뇨 효과)

  • To verify anti-diabetic effect of oligopeptide with His-Pro repeats (mHP peptide), the oligopeptide was first secreted and optimized using the secretion vector, pRBAS with alkaline protease gene promoter and the signal sequence in Bacillus subtilis and directly the anti-diabetic effect of the mHP peptide was investigated in insulinoma cell, RINm5F cell line. The oligopeptide gene was obtained by annealing oligonucleotides with repeated His-Pro sequence and finally was constructed as 18 dipeptides (108 bp and 4.0 kDa) coding gene, named oligopeptide with His-Pro repeats (mHP peptide) to make cyclo(His-Pro) known to be anti-diabetic effects. The region encoding the oligopeptide gene was subcloned into the pRBAS secretion vector (E.coli-Bacillus shuttle vector) after PCR amplification using the designed primers including initiation and termination codons and His tag, named pRBAS-mHP (6.56 kb). To optimize secretion of the oligopeptide, various culture conditions were investigated in Bacillus subtilis LKS. As a result, the secreted oligopeptide was maximally measured (approximately $59.6{\mu}g/mL$) in 3 L batch culture and the highest secretion was achieved at $30^{\circ}C$, PY medium, and carbon sources (particularly barley and glycerol). In the RINm5F cells treated with 2 mM STZ, the oligopeptide treatment (0.1 mg/mL) restored the cell viability (10%) and reduced the nitric oxide (NO) generation (35%) and DNA fragmentation (90%). And also, insulin secretion level was increased to 17% higher than in STZ-treated RINm5F cells. These results suggest that the oligopeptide with His-Pro repeats could be a candidate material for anti-diabetic agent against STZ-induced diabetes.

Productivity of $F_1$ Hybrid Maize and of $F_2$ Material Derived from by Multiplication (옥수수 잡종이세대의 건물생산과 수량구성요소)

  • The objective of this study was to find out the productivity of F_2 material derived from F_1 hybrid Maize (Zea mays L.) by multiplication and their response to plant population. Suweon 19 and Junju-Ok were grown at three population densities. Populations were ca. 8.3, 10.4 and 13.9 thousand plants per 10a. The results are summarized as follows; 1. The number of barren plants differed significantly for the two generations but other yield components comparisons were not significant at P(0.05). 2. Ear and TDN yield were higher from the $F_1$ hybird of Suweon 19 than from the F_2 material, while yields of Jinju-Ok and stover yield of both hybrids were not affected by generation. 3. Plant and ear height were not affected by population density but barren plants were increased at high population. Length and weight of a ear, and ear (including cobs)-to-stover ratio decreased with increased population. 4. Increasing population density resulted in significant decrease in ear and TDN yield, while stover yield not decreased.
