생강나무의 수피를 아세톤-물 (7:3)의 혼합용액으로 추출하여 에틸아세테이트용성과 수용성 화합물로 분리하였고 이중에서 에틸아세테이트용성 부분을 Sephadex LH-20과 TSK 40F로 충진한 칼럼크로마토그래피를 사용하여 5개의 화합물, 즉 (+)-catechin, (-)-epicatechin, quercetin 그리고 epicatechin-($4{\beta}{\rightarrow}6$-catechin 이량체와 epicatechin-($2{\beta}{\rightarrow}7$, $4{\beta}{\rightarrow}8$)-epicatechin 이량체를 단리 하였다. 단일물의 확인을 위하여 박층크로마토그래피를 실시하였으며, 화합물의 정확한 구조는 $^1H$-NMR과 $^{13}C$-NMR 스펙트럼을 이용하여 규명하였다.

Effect of fried beancurd containing powered green tea on the serum lipid metabolism in rats were investigated. HPLC analysis of green tea-fried beancurd showed that epigallocatechin, epicatechin, epicatechin gallate and epigallocatechin gallate were intact without degradation during manufacturing process. Serum HDL-cholesterol, LDL-cholesterol, triglyceride and glucose concentrations were measured in rats fed experimental diet containing green tea-fried beancurd. Serum LDL-cholesterol contents and blood glucose were significantly decreased with treatment of green tea-fried beancurd comparing to control.
Four flavonoide glycosides and (-)-Epicatechin were isolated from the aqueous extracts of dried whole part of Lysimachia clethroides Duby(Primulaceae). They were 3-0-Methyl-quercetin-7-0-[$\alpha$-L- rhamnopyranosyl (1-2) glucopyranoside], Quercetin-3-0-$\beta$-D-glucopyranoside, Kaempferol-3-0-$\beta$-D-glucopyranoside, Kaempferol-3-0-[$\alpha$-L-rhamnopyranosyl (1-6)-$\beta$-D-glucopyranoside] and (-)-Epicatechin. 3-0-[$\alpha$-L-rhamnopyranosyl (1-6)-$\beta$-D-glucopyranoside]and (-)-Epicatechin. 3-0-Methyl-quercetin-7-0-[$\alpha$-L-rhamnopyranosyl (1-2)-$\beta$-D-glucopyranoside] and (-)-Epicatechin were first isolated from this plant. Their structures were elucidated by spectral analysis [$^{1}$H-, $^{13}C-$ NMR, $^{1}$H-$^{1}$H-COSY, DEPT-analysis, HMQC(Heteronuclear multiple quantum coherence), FAB-MS].
The five main green tea catechin components such as (+)-catechin, (-)-epicatechin, (-)-epigallocatechin, (-)-epicatechin gallate, and (-)-epigallocatechin gallate were analyzed quantitatively from commercial green tea by HPLC. Amounts of catechins decreased in the following order : (+)-catechin > (-)-epigallocatechin gallate >(-)-epigallocatechin >(-)-epicatechin >(-)-epicatechin gallate. In this study, the stability of the following green tea catechins to pH in the range from 3 to 11 was studied using of ultraviolet spectroscopy : (+)-catechin, (-)-epicatechin, (-)-epicatechin gallate, and (-)-epigallocatechin gallate. This study demonstrated that green tea catechins were not stable at high pH and that the pH-, and time-dependent spectral alternatives were not reversible In conclusion, low pH is important to maintain the efficient utilization of green tea catechins.
From the aerial parts of Sorbaria sorbifolia var. stellipila, three flavonoids, (+)-catechin (1), (-)-epicatechin (2) and astragalin (3) have been isolated and characterized by physicochemical and spectral means.
Green tea leaves 12.5 g were extracted twice with 500 ml boiling water. The green tea extract (GTE) contained 4.67 mg solid. The GTE contained polyphenols sush as 54.12% (-) epicatechin gallate, 26.21% (-) epicatechin, 10.71% epicatechin gallate, 7.09% (-) epicatechin and 1.85% catechin. The GTE inhibited the copper-catalyzed oxidation of human LDL at the concentrations of 50 and $100\;{\mu}g/ml$ GTE in the presence of $5\;{\mu}M$$CuSO_{4}$. The electrophoretic mobility of the LDL oxidized in the presence of $5\;{\mu}M\;CuSO_{4}$ was higher than that of the native LDL. The GTE also inhibited LDL oxidation induced by J774, human monocyte-derived macrophages and vascular endotherial cells. The LDL modified by copper or cells was inhibited by human macrophages at a much greater rate than native LDL in the presence of GTE. The GTE was found to be a potent inhibitor of modification of LDL. GTE inhibited the uptake of cell-modified $^(125)I-labelled$ LDL by macrophages. The formation of conjugated dienes was strongly inhibited in the presence of 50 or $100\;{\mu}g/ml$ GTE.
For glucosyltransferase(GTase) inhibitors, two flavan-3-ols were isolated from Theobroma cacao beam husk. They showed positive reaction with $anisaldehyde-H_2SO_4$ solution, $FeCl_3$ to be confirmed as dimeric flavan-3-ols on TLC and were identified as procyanidin B-1 [(-)-epicatechin-$(4{\beta}{\rightarrow}8)$-catechin] and procyanidin B-3 [(+)-catechin-$(4{\beta}{\rightarrow}8)$-catechin by spectroscopic analysis. Their inhibitory effect on glucosyltranaferase activity was also investigated and procyanidin B-1 showed 50% inhibition at 0.3 mM. They inhibited on the glucosyltransferasa noncompetitively and dimeric flavan-3-ol containing (-)-epicatechin had higer inhibitory activity.
To investigate and evaluated the scavenging and antioxidative effects of various flavonoids on paraquat induced toxicity, in vivo and vitro tests of eight flavonoids (catechin, epocatechin, flavone, chrysin, apigenin, quercetin, morin and biochanin A) were carried out. The generation of reactive oxygen substances(ROS) in PMS-NADH system $H_2O_2$ induced hemolysis and lipidperoxidation to blood, NADPH dependent lipidperoxidation to liver and lung microsome by paraquat were studied.The results are summerized as follows; 1) In the concentration ranges from 3.3 to 9.8$\mu$M of catechin,epicatechin, quercetin and biochanin A removed the 50% of DPPH radical scavenging effects. 2) In the concentration ranges from 0.60 to 1.86 mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on superoxide anion which gernerated in PMA-NADH system. 3) In the concentration ranges from 0.12 to 0.49mM of catechin, epicatechin, quercetin and biochanin A showed the inhibitory and antioxidative activity on H202 which generated in PMA-NADH system. 4) In the concentration ranges from 0.6 x10$^{-5}$ to 6.3 x 10$^{-5}$mM of catechin, epicatechin, flavone, chrysin, quercetin and morin showed the inhibitory and antioxidative activity on $H_2O_2$ induced hemolysis to blood 5) All flavonoids tested exhibited inhibitory and antioxidative effects on paraquat induced liver and tung microsomal lipidperoxidation. Therefore, all flavonoids evaluated showed the useful compounds for scavenger and antioxidant on paraquat induced toxicity.
The needles of torreya(Torreya nucifera S. et Z) were collected, extracted with acetone-$H_2O$(7:3, v/v), fractionated with hexane, chloroform and ethylacetate, and freeze dried to give some dark brown powder. The ethylacetate soluble mixture was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. Most of the needle extractives were flavan and its methyl ether compounds such as (+)-catechin, (-)-epicatechin, (+)-gallocatechin, (-)-epigallocatechin, 3'-O-methyl-(+)-catechin and 3'-O-methyl-(-)-epicatechin including protocatechuic acid, one of benzoic acids. $^1H$- and $^{13}C$-NMR, NOE and CI-MS analyses were performed to elucidate the structures of the isolated compounds.
The flavonoids kaempherol and epicatechin, and a proanthocyanidin fraction were isolated from the seed of allspice (Pimenta dioica). Their structures were determined by chemical and spectral analyses. Antioxidant activities of kaempherol, epicatechin and the proanthocyanidin fraction were tested using the reaction with the stable radical diphenyl-p-picrylhydrazyl (DPPH) and exhibited IC/sub 50/ values of 7.83, 4.27 and 2.92 g/mL, respectively. These results demonstrate that allspice proanthocyanidins and flavonoids might act as effective antioxidants.
