Two-stage Enzymatic Conversion of Lactose to Galactooligosaccharides by Two-type ${\beta}-galactosidases$ (두 종류 ${\beta}-galactosidases$ 의 이단 반응을 이용한 갈락토올리고당의 제조)

  • In an attempt to increase the conversion of lactose to galactoligosaccharides, two types of ${\beta}-galactosidases$ originated from Thermus caldophilus and Bacillus sp. A4442 reacted with 60% (w/w) lactose consecutively. Concentration of galactooligosaccharides reached up to 60% at the 85% conversion of the initial lactose maintaining transgalactosylation ratio ca. 90%.

The Effect of Acid Hydrolysis and Enzymatic Saccharification in Bioethanol Production Process Using Fruit Peels (과일껍질을 이용한 바이오에탄올 생산 공정에서 산 가수분해 및 효소당화의 영향)

  • The acid hydrolysis and enzymatic saccharification were carried out for the production of cellulosic ethanol. The possibility of bio-energy production from tangerine peel and apple and watermelon rind was evaluated by determining the optimum production condition. The optimum conditions for the production of cellulosic ethanol from fruit peel were as follows: the sulfuric acid concentration and reaction time of acid hydrolysis for the ethanol production from an apple rind were 20 wt% and 90 min, respectively. The concentration of sulfuric acid for tangerine peel and a watermelon rind at the hydrolysis time of 60 min were 15 wt% and 10 wt%, respectively. A viscozyme was proven as the best conversion for the ethanol production when using enzymatic saccharification from fruit peels. The optimum enzymatic saccharification time for tangerine peel and apple and watermelon rind were 60, 180, and 120 min, respectively.

Immobilization of a Mediator onto Carbon Cloth Electrode and Employment of the Modified Electrode to an Electroenzymatic Bioreactor

  • 5,5'-Dithiobis(2-nitrobenzoic acid) (DTNB) was selected as an electron transfer mediator and was covalently immobilized onto high porosity carbon cloth to employ as a working electrode in an electrochemical $NAD^+$-regeneration process, which was coupled to an enzymatic reaction. The voltammetric behavior of DTNB attached to carbon cloth resembled that of DTNB in buffered aqueous solution, and the electrocatalytic anodic current grew continuously upon addition of NADH at different concentrations, indicating that DTNB is immobilized to carbon cloth effectively and the immobilized DTNB is active as a soluble one. The bioelectrocatalytic $NAD^+$ regeneration was coupled to the conversion of L-glutamate into ${\alpha}$-ketoglutarate by L-glutamate dehydrogenase within the same microreactor. The conversion at 3 mM monosodium glutamate was very rapid, up to 12 h, to result in 90%, and then slow up to 24 h, showing 94%, followed by slight decrease. Low conversion was shown when substrate concentration exceeding 4 mM was tested, suggesting that L-glutamate dehydrogenase is inhibited by ${\alpha}$-ketoglutarate. However, our electrochemical $NAD^+$ regeneration procedure looks advantageous over the enzymatic procedure using NADH oxidase, from the viewpoint of reaction time to completion.

Evaluation of Pitch Pine for Bioethanol Production by Organosolv Pretreatment (Organosolv 전처리를 통한 리기다소나무의 바이오에탄올 생산 적용성 평가)

  • In this study, the feasibility of utilizing wood chips from pitch pine (Pinus rigida) was evaluated for bioethanol production by an organosolv pretreatment and enzymatic saccharification. When wood chips from pitch wood were pretreated with 75% (v/v) ethanol and 1.7% sulfuric acid as a catalyst at H-factor 2000, average pulp yield was 43.3%, which pretreated wood fibers showed higher glucan (55.8%) and lower lignin (12.2%) contents than untreated control (43.9% glucan and 27.8% lignin). After enzymatic saccharification, the organosolv pulps with 56.2% delignification rate reached above 97% conversion rate of cellulose to glucose. These results indicated that increasing the delignification rate causes micro pores on the surface of organosolv pulps resulting in improved the accessibility of enzyme onto the substrate. Moreover, it was in agreement with the SEM examination of wood fibers.

  • The effective saccharification of cellulosic biomass to glucose is the most critical step for the conversion of renwable biomass to alternative liquid fuel. The enzymatic hydrolysis of biomass can be significantly enhanced provide the attrition milling media is added during hydrolysis. The enhancing mechanism of hydrolysis reaction in an agitated bead system was investigated. An attrition-reactor (bioattritor) which installed specially designed torque measuring apparatus was developed, and the potimal saccharification conditions of bioattritor were determined. The relationship between the power consumption required for agitation of attrition-milling media and enhanced extent of hydrolysis of biomass was compared to evaluatic economic feasibility of the process.

Preparation of Diacylglycerol from Lard by Enzymatic Glycerolysis and Its Compositional Characteristics

  • The aim of this study was to prepare diacylglycerol (DAG) by enzymatic glycerolysis of lard. The effects of reaction parameters such as lipase type, reaction temperature, enzyme amount, substrate molar ratio (lard/glycerol), reaction time, and magnetic stirring speed were investigated. Lipozyme RMIM was found to be a more active biocatalyst than Novozym 435, and the optimal reaction conditions were 14:100 (W/W) of enzyme to lard substrate ratio, 1:1 of lard to glycerol molar ratio, and 500 rpm magnetic stirring speed. The reaction mixture was first incubated at $65^{\circ}C$ for 2 h and then transferred to $45^{\circ}C$ for 8 h. At the optimum reaction conditions, the conversion rate of triacylglycerol (TAG) and the content of DAG in the reaction mixture reached 76.26% and 61.76%, respectively, and the DAG content in purified glycerolized lard was 82.03% by molecular distillation. The distribution of fatty acids and Fourier transform infrared spectra in glycerolized lard samples were similar to those in lard samples. The results revealed that enzymatic glycerolysis and molecular distillation can be used to prepare more highly purified DAG from lard.

  • Since 1950 there has been a dramatic progress in rationalization of the production of sweet potato and potato starch in Japan. This enabled dextrose industry by enzymatic process to develop rapidly due to the success of enzymatic liquefaction and saccharification. Isomerization of glucose to fructose has been studied, and the immobilization of isomerases prompted its products on industrial scale in 1970. Another advance is the development of effective methods of producing high purity maltose. A malto-hexaose forming amylase was discovered in 1971 and attempts are being made for its pharmaceutical utilization. Saccharification of cellulose by cellulase has been studied. Conversion of starch to other polysaccharides is another example for the numerous Japanese activities.

  • Recently the production of ethanol from lignocecllulosics has received much attention due to immense potential for conversion of renewable biometerials into biofuels and chemicals. Fomitopsis palustris causes a typycal brown-rot and is unusual in that it rapidly depolymerize the cellulose in wood without removing the surrounding lignin that normally prevents microbial attack. This study demonstrated that the brown rot basidiomycete F. palustris was able to degrade crystalline cellulose. This fungus could also produce the three major cellulases (BGL, EXG and EG) when the cells were grown on 2.0% Avicel. The fungus was able to degrade both the crystalline and amorphous forms of cellulose from woody biomasses. Moreover, we found that this fungus has the processive EG like CBH which are able to degrade the crystalline region of cellulose. To establish the cellulase system in relation with degradation of woody biomass, we performed that purification, characterization and molecular cloning of a BGL, EGs and GLA from F. palustris grown on Avicel.

  • Glycosyl acrylate and methacrylate were synthesized by lipase-catalyzed esterification of vinyl acrylate and vinyl methacrylate with ${\beta}$-methyl glucoside in t-butanol as a reaction medium. At the optimum conditions of initial concentration of 150g/l ${\beta}$-methyl glucoside, molar ratio of 1 : 3, 5%(w/v) lipase(Novozym 435) and $50^{\circ}C$, we attained up to 100% conversion for enzymatic glycosylation of vinyl acrylate and vinyl methacrylate by supersaturated solvent process. The polymerizable glycosyl acrylates and methacrylate are expected to have biomedical application as hydrophilic monomers and hydration modifiers to be used for biocdmpatible hydrogel.

Simultanceous Saccharification and Fermentation of Cellulose for Lactic Acid Production

  • Lactic acid production from ${\alpha}$-cellulose by simultaneous saccharification and fermentation (SSF) was studied. The cellulose was converted in a batch SSF using cellulase enzyme Cytolase CL to produce glucose sugar and Lactobacillus delbrueckii to ferment the glucose to lactic acid. The effects of temperature, PH, yeast extract loading, and lactic acid inhibition were studied to determine the optimum conditions for the batch processing. Cellulose was converted efficiently to lactic acid, and enzymatic hydrolysis was the rate controlling step in the SSF. The highest conversion rate was obtained at 46$^{\circ}C$ and pH 5.0. The observed yield of lactic acid from ${\alpha}$-cellulose was 0.90 at 72 hours. The optimum pH of the SSF was coincident with that of enzymatic hydrolysis. The optimum temperature of the SSF was chosen as the highest temperature the microoraganism could withstand. The optimum yeast extract loading was found to be 2.5g/L. Lactic acid was observed to be inhibitory to the microorganisms' activity.
