Detection of transgene in early developmental stage by GFP monitoring enhances the efficiency of genetic transformation of pepper

  • In order to establish a reliable and highly efficient method for genetic transformation of pepper, a monitoring system featuring GFP (green fluorescent protein) as a report marker was applied to Agrobacteriummediated transformation. A callus-induced transformation (CIT) system was used to transform the GFP gene. GFP expression was observed in all tissues of $T_0$, $T_1$ and $T_2$ peppers, constituting the first instance in which the whole pepper plant has exhibited GFP fluorescence. A total of 38 T0 peppers were obtained from 4,200 explants. The transformation rate ranged from 0.47 to 1.83% depending on the genotype, which was higher than that obtained by CIT without the GFP monitoring system. This technique could enhance selection power by monitoring GFP expression at the early stage of callus in vitro. The detection of GFP expression in the callus led to successful identification of the shoot that contained the transgene. Thus, this technique saved lots of time and money for conducting the genetic transformation process of pepper. In addition, a co-transformation technique was applied to the target transgene, CaCS (encoding capsaicinoid synthetase of Capsicum) along with GFP. Paprika varieties were transformed by the CaCS::GFP construct, and GFP expression in callus tissues of paprika was monitored to select the right transformant.

Soil Environment and Soil-borne Plant Pathogen Causing Root Rot Disease of Ginseng (인삼 뿌리썩음병 발병에 미치는 토양전염성병원균과 토양환경요인)

  • Disease is the major problem in ginseng cultivation from seed stratification, soil preparation prior to planting, right through to drying of the roots. There are many soil-borne disease pathogen in rhizosphere soil environment, furthermore occurrence of diseases by a diverse group of fungi and related organisms are closely related to various soil condition. Observable symptoms for soil-borne diseases include wilting, leaf death and leaf fall, death of branches and limbs and in severe cases death of the whole plant. The fungus Cylindrocarpon destructans is the cause of root rot characterized by a decay of the true root system in many ginseng production areas in Korea. Some pathogens are generally confined to the juvenile roots whilst others are capable of attacking older parts of the root system. However, the relation between the soil environmental characteristics and ginseng root rot by soil-borne disease pathogen is not clearly identified in ginseng field. In this paper, we reviewed soil-borne plant pathogen causing root rot disease of ginseng with respect to soil environment.

  • More recently, the ecocide like an environmental shocks of acid rain, oil spills and now the ozone layer have driven home the fact that we're all responsible for looking after our natural environment and threaten the our right to live. At such a situation enterprises has been to develop techniques of enhancing environment friendly design. However it is difficult to producing environment friendly product - especially at electronic product like a high accumulated thing and produced with a small amount - while they know the importance of environment. Because of a direct and actual disadvantage of enterprise and customer. Moreover it is rare and negative for physical point that an adaptation case of recycled material or natural one for designing. Therefore it is important to positive consideration and action of design process for the nature. In this paper the focus of interest is find out the active and real environmental friendly design practice scheme as a designer and understanding of basic of the environmental friendly design for a coexistence of the enterprise, customer, and environment.

Numerical Simulation of 3-Dimensional Fluid Flow and Dust Concentrations in a Steel Foundry (제강 작업장내 삼차원 유동장 및 먼지농도의 수치 모사)

  • The steel foundries with electric arc furnaces handling metal scraps have recently gained an attention as a potential source of dusts. The present study focuses on the analysis of dust emissions and removals during furnace charging and melting processes by commercial CFD software named FLUENT. A body fitted grid system consisting of 880,000 meshes was first generated by Gambit for the electric arc furnace with the capacity of 60 ton/cycle and then FLUENT was invoked to solve the corresponding NavierStokers equation for the momentum, temperature and dust concentration. The entire processes from metal charging to metal melting were simulated to investigate the unsteady behaviors of fluid flows and dust concentrations. The model simulation results showed that as the top of the electric arc furnace opened for metal charging, hot plumes bursted out from the furnace rose strongly by buoyance and escaped mostly through the main hood. Therefore, the capacity of main hoods determined the vent efficiency in the metal charging process. As the furnace was closed after the metal charging and the metal melting processes was followed, the hot flow stream stretching from the furnace to the main hood was dissipated fast and the flow from the inlet of the bottom of the left hand side to the main and monitoring hoods constituted the main stream. And there was only a slow flow in the right hand side of the furnace. Therefore, the dust concentrations were calculated higher in the left hand side of the furnace, which was consistent with observations.

  • Smart Pole Measurement System was constructed with not only the core sensors of a GNSS receiver, a TRS sensor and a soil moisture sensor but supplementary installation of power supply and radio communication for monitoring steep slope sites. Also a data processing software for displacement extraction and visualization was developed. Smart Pole Measurement sensor is composed of a GNSS antenna at the top of the pole, a TRS sensor and a gyro sensor vertical below right of the antenna and a soil moisture sensor at the bottom of the pole. The sensor combination extracts not only ground combination in real time but transltion, slide, settlement and soil moisture content. This measuring/monitoring system which cosists of data receiving part, data collection/transfer part and data processing part was built to exercise their functions and then test measuring/monitoring was conducted by introducing artificial displacement and the results were analyzed to evaluate field applicability.

  • Since the chemical pesticides have been played a major role in crop protection practices during last 5 to 7 decades, social concerns on the pesticide residues in and on food commodities as well as environmental compartments have also growing with endless demands and interests. Most national regulation authorities over the world have paid a special attention on the data requirements for pesticide registration. In addition, even the registered pesticides also should follow the re-registration process, which meets today's guidelines and regulatory triggers and safety profiles. More recently, a defined interest in the international bodies has given to the global conservation program from the environmental contamination; these involves persistent organic pollutants (POPs), endocrine disrupting chemicals (EDs), biocides, etc.. In order to secure the food safety and keep our circumference sound, in-depth efforts getting information from global networks have perpetually to be given under relevant national agencies. At the same time, a nation-wide survey of the residues has also to be in operation to monitor the tendency of the toxicant in/on foods, feeds, and environmental segments. In final, the scientifically assessed results on safety should be opened to the public to provide the right-to-know for the consumers.

  • This paper is focused on the research of environmental and human factors for a design of PMVbased air conditioning system in the ship. In the results from environmental factor research, it is possible to dissatisfy thermally in the wheel house and communication room of 25 ton and engine room of 100 ton. The clothing and activity in the ship is modified using inland indoor characteristics. Thermal resistance of clothing may be more sensitive to PMV(predicted mean vote) than metabolic rate because of large deviation among maritime polices. The distribution of human factor is right long-tailed than standard normal distribution.

  • International concern over the environmental pollution is ever increasing, and diversified countermeasures must be devised in Korea also. Global trend, damages, problems and countermeasures with respect to issues mentioned in the Rio Declaration, such as prevention of ozone layer destruction, reduction of migratory atmospheric pollution between neighboring countries, and prevention of global greenhouse effect, were discussed in this report. Conclusion of the report is summarized as follows : A. Measurement, Planning and Monitoring (1) Development and implementation of a global network for measurement and monitoring from the global aspects such factors as related to acid rain(Pioneer substances, pH, sulfate, nitrate), effect of global temperature(Air temperature, $CO_2$, $CH_4$, CFC, $N_2O$) and destruction of ozone layer($CFC_S$). (2) Establishment of network system via satellite monitoring movement of regional air mass, damage on the ozone layer and ground temperature distribution. B. Elucidation of Present State (1) Improvement and development of devices for carbon circulation capable of accurately forecasting input and output of carbon. (2) Developmental research on chemical reactions of greenhouse gas in the air. (3) Improvement and development of global circulation model(GCM) C. Impact Assessment Impact assessment on ecosystem, human body, agriculture, floodgate, land use, coastal ecology, industries, etc. D. Preventive Measures and Technology Development (1) Development and consumption of new energy (2) Development of new technology for removal of pioneer substances (3) Development of substitute matter for $CFC_S$ (4) Improvement of agriculture and forestry means to prevent the destruction of ozone layer and the greenhouse effect of the globe (5) Improvement of housing to prevent the destruction of ozone layer and the greenhouse effect of the globe (6) Development of new technology for probing underground water (7) Preservation of forest (8) Biomass 5. Deducing environmentally conscious factors for apartment complex planning and weight evaluation (환경요소를 고려한 공동주택 단지계획요소 도출 및 가중치 평가)

  • Purpose: As declines in the quality of residential environments occur, such as urban heat island effect, tropical night phenomenon, and violations of right to light and privacy due to urban densification and high rise building, these problems are emerging as social issues. In order to improve these issues, design factors which consider environmental aspects must be selected when planning apartment complexes, and ways to reflect them in the planning phase must be explored. Method: In this study, the analytical hierarchy process(AHP) was used to deduce design factors that considered environmental elements during the planning of apartment complexes. Furthermore, the priority and weight for each evaluation index were assessed. The objective was to propose a guideline for planning apartment complexes by finding the best solution for each evaluation index using complex weight values. Result: Floor area ratio was selected as the most important evaluation criterion in the environmentally conscious evaluation index for apartment complex planning. The shape and placement of skylights were selected as the most important evaluation criteria in the sunshine environment for a pleasant residential environment. Ground surface cover design was selected as the most important criterion in the outdoor thermal environment index for improving the microclimate within cities and apartment complexes. Thus, the results of this study can serve as an investigation guideline that concerns policy and regulations, and as reference data that can be used in planning apartment complexes.

DNA Damage of Lymphocytes in Volunteers after 4 hours Use of Mobile Phone (일부 자원자들의 이동전화 4시간 연속 사용 후 림프구 DNA 손상 평가)

  • Objectives : There has been gradually increasing concern about the adverse health effects of electromagnetic radiation originating from cell phones which are widely used in modern life. Cell phone radiation may affect human health by increasing free radicals of human blood cells. This study has been designed to identify DNA damage of blood cells by electromagnetic radiation caused by cell phone use. Methods : This study investigated the health effect of acute exposure to commercially available cell phones on certain parameters such as an indicator of DNA damage for 14 healthy adult volunteers. Each volunteer during the experiment talked over the cell phone with the keypad facing the right side of the face for 4 hours. The single cell gel electrophoresis assay (Comet assay), which is very sensitive in detecting the presence of DNA strand-breaks and alkali-labile damage in individual cells, was used to assess peripheral blood cells (T-cells, B-cells, granulocytes) from volunteers before and after exposure to cell phone radiation. The parameters of Comet assay measured were Olive Tail Moment and Tail DNA %. Results : The Olive Tail Moment of B-cells and granulocytes and Tail DNA % of B-cells and granulocytes were increased by a statistically significant extent after 4-hour use of a cell phone compared with controls. Conclusion : It is concluded that cell phone radiation caused the DNA damage during the 4 hours of experimental condition. Nonetheless, this study suggested that cell phone use may increase DNA damage by electromagnetic radiation and other contributing factors.
