Efficient Biotinylation of Nitrocellulose Membrane for Immuno-Filtration Capture Assay

  • We investigated biotinylation of nitrocellulose membrane for immuno-filtration capture assay. In order to enhance the efficiency of biotinylation, nitrocellulose membranes were pretreated with several chemicals for the purpose of suitable protein absorption through surface modification. As a signal generating enzyme, urease was used and the concentration of avidin was optimized for the efficient binding kinetics between urease-biotin in liquid phase and biotinylated membrane in solid phase. For effective biotinylation, bovine serum albumin-biotin complexes could be immobilized at a concentration of $370\;{\mu}g$/stick ($4.4\;cm^2$). Among tested chemicals, polylysine (0.25%) showed a significant effect in biotinylation. Polylysine is thought to enhance surface area by extending unbound residues into solution. Time of treatment over 30 min and higher molecular weight of polylysines (58,100 dalton) showed positive effect on the enhancement of biotinylation. The result from this study may be useful for developing a new biosensor and other biofunctional membranes for examining molecular recognition.

  • The main objective of this study is to examine the mediating effect of 'e-trust' on the relationship between the antecedents related to e-marketplace and the intention of using e-marketplace. Data was collected from a survey of 169 exporting firms in Gwangju, Korea. The results of the study are as follows ; First, 'e-trust' is produced a mediating effect when technology and government factors affect the intention of using e-marketplace. However, "e-trust" does not play a mediating role in the relationship between company factor and the intention of using e-marketplace. Second, technology factor, government factor and company factor have a positive impacts on the intention of using e-marketplace. Accordingly, in order to enhance the intention of using e-marketplace, small & medium exporting firms in Korea should try more to prepare for factors related to technology and company. In addition to these, government should give full support to be prepared for regulations and systems related to e-marketplace. If e-trust were properly built, the enhancement of using e-marketplace would be taken place more smoothly. So most of exporting firms can participate in e-marketplace more actively.

Effect of Time of Initiating Dietary Fat Supplementation on Performance and Reproduction of Early Lactation Dairy Cows

  • Forty-two Holstein cows (21 multiparous) were assigned by calving date and parity to three dietary sequences to evaluate the effect of time of initiating fat supplementation to diets on lactation and reproductive performance. The dietary sequences were: 1) control, no supplemental fat from 1 to 98 days in milk (DIM) ; 2) control diet from 1 to 28 DIM then 3% supplemental fat (calcium salts of long-chain fatty acids) from 29 to 98 DIM; or 3) 3% supplemental fat from 1 to 98 DIM. Feeding supplemental fat did not enhance mean milk and 4% fat corrected milk (FCM) yields, but efficiency of FCM production was higher for cows fed supplemental fat. Milk fat percentage was unchanged whereas milk protein percentage was depressed with fat supplementation. Feeding supplemental fat reduced DMI and energy balance but there were no differences among treatments on time to resumption of ovarian cyclicity or conception rate to first service. Concentrations of progesterone during the first two ovulatory cycles tended to be greater in the fat-supplemented groups. Feeding supplemental fat starting at either parturition or 29 DIM increased efficiency of FCM production, but did not greatly enhance reproductive performance.

  • Aviation/tourism industry has a unique characteristic of high inter-dependance between customer and service-provider. It is the mental condition of service-provider that could influence on the attitude of the service-provider. Thus, it is important to manage the mental condition of the service employees to enhance a company's financial performance. This paper tries to analyze the combined model of both policy acceptance and service profit chain(SPC) models. First, service policy acceptance model tells how the service policy acceptance, which consists of policy compliance, trust, participation and policy failure, would influence on SPC model. According to empirical research, it was found that both the employees' policy compliance and the policy trust have a positive significant impact on their participation on service policy. In the policy acceptance model, the employee's voluntary participation based on their trust and compliance of the policy was proven to have a positive effect on increasing job satisfaction and customer orientation. Regarding SPC model, their participation in the policy had the strongest impact among variables on customer orientation. Such results implies the employees' participation on the policy could become the starting point to enhance the customer value.

Assemblage and Driving Characteristics of a Ultrasonic Fabric Washing Machine (초음파 직물수세기의 구성 및 구동 특성)

  • A new, high-efficiency ultrasonic fabric washing machine was developed to be an energy-efficient washing machine and to enhance fabric washing quality in washing processes of the dyeing and finishing process in the textile industry. This system is composed of ultrasonic wave generator, air blowing nozzle, torque motor for fabric tension control, and enclosed washing bath, multi-tube type exchanger, noiseless heater, air cylinder, expander roller, mangle upper and lower rollers, bend bar, dancer, shower spray nozzle, and solenoid valve, and so on. These elements are synergised for fabric washing. One of the very important principles is the low tension fabric running system. For an efficient washing effect, a counter flow system is also adopted. The new system also adopts the dancer and torque motor to control fabric tension and prevent fabric creasing. Shower spray nozzle, counter flow and overflow apparatus, and air-blowing apparatus are adopted to enhance the fabric washing effect. In this study, peach yoryu, exter, and moss crepe fabrics were washed by the general and ultrasonic washing systems under different conditions respectively. The washing efficiency was affected by the fabric running speed and characteristics of fabrics. Size content after washing increased with increasing the fabric running speed. The values in the general washing system were higher than those of the ultrasonic washing system. The changes of conductivity in the ultrasonic and the cooling bath were affected by the running time under the ultrasonic generating. The values of conductivity decreased as the experimental time passed.

Hydrate Characteristics of Cement Mixtures with Expansion Additive According to Age and Improvement Effect on Initial Strength (팽창재를 사용한 시멘트 혼합물의 재령별 수화물의 특성과 초기강도 개선 효과)

  • CSA, a cement mineral compound that is mainly composed of $3CaO{\cdot}3Al_2O_3{\cdot}CaSO_4$, generates ettringite as a hydration product after a reaction with glass (lime), gypsum and water to speed up the hardening process and enhance the strength and degree of expansion. When used as a cement admixture, there is increased production of ettringite, which can improve the initial strength in the first three days and ameliorate the reduction in the initial strength caused by the use of fly ash in particular. In this study, a hydrate analysis was performed using XRD and SEM after substitution with fly ash (30%) and CSA (8%) with the goal of observing the effect of CSA on the initial strength of a cement mixture containing fly ash. The results of the analysis showed that an addition of CSA promoted the production of ettringite and improved the initial strength, resulting in the generation of hydrates, which can effectively enhance the long-term strength of these materials.

  • The present study examined the ex vivo effect of Job's tear on immune function. Seven to eight week old mice(Balb/c) were fed a chow diet ad libitum two different concentrations (50 and 500 mg/kg BW) of water extract of Job's tear were orally administ every other day for two weeks. The results indicated that macrophage activation had occurred in the mice receiving 50 mg/kg B. W. of Job's tear water extract. Overall, using a mouse model, this study demonstrated that Job's tear extract may enhance immune function by regulating the $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ and IL-10 cytokine production capacity of activated macrophages in mice. This study may suggest that supplementation of Job's tear water extracts may enhance the immune function by regulating the enhancing the cytokine production by activated macrophage ex vivo.

  • Recently, interest in technology for eco-friendly energy harvesting has been increasing. Polyvinylidene fluoride (PVDF) is one of the most fascinating materials that has been used in energy harvesting technology as well as micro-filters by utilizing an electrostatic effect. To enhance the performance of the electrostatic effect-based nanogenerator, most studies have focused on enlarging the contact surface area of the pair of materials with different triboelectric series. For this reason, one-dimensional nanofibers have been widely used recently. In order to realize practical energy-harvesting applications, PVDF nanofibers are modified by enlarging their contact surface area, modulating the microstructure of the surface, and maximizing the fraction of the ν-phase by incorporating additives or forming composites with inorganic nanoparticles. Among them, nanocomposite structures incorporating various nanoparticles have been widely investigated to increase the β-phase through strong hydrogen bonding or ion-dipole interactions with -CF2/CH2- of PVDF as well as to enhance the mechanical strength. In this study, we report the recent advances in the nanocomposite structure of PVDF nanofibers and inorganic nanopowders.

Utilizing vacuum bagging process to enhance bond strength between FRP sheets and concrete

  • This paper investigates the effect of utilizing vacuum bagging process to enhance the bond behavior between fiber reinforced polymer (FRP) composites and concrete substrate. Sixty specimens were prepared and tested using double-shear bond test. The effect of various parameters such as vacuum, fiber type, and FRP sheet length and width on the bond strength were investigated. The experimental results revealed that utilizing vacuum leads to improve the bond behavior between FRP composites and concrete. Both the ultimate bond forces and the maximum displacements were enhanced when applying the vacuum which leads to reduction in the amount of FRP materials needed to achieve the required bond strength compared with the un-vacuumed specimens. The efficiency of the enhancement in bond behavior due to vacuum highly depends on the fiber type; using carbon fiber showed higher enhancement in the bond strength compared to the glass fiber when vacuum was applied. On the contrary, specimens with glass fiber showed higher enhancement in the maximum slippage compared to specimens with carbon fibers. Utilizing vacuum does not affect the debonding failure modes but lead to increase in the amount of attached concrete on the surface of the debonded FRP sheet.

The Effect of Promotion and Job Training on Job Satisfaction of Employees: An Empirical Study of the SME Sector in Bangladesh

  • SME sector's success also depends on its employees' job satisfaction as satisfied employees are likely to be more productive at the workplace and positively enhance SME business performance. Small and medium firms are the heart of the economy, and employees are the main and valuable asset for the SME firms. If SME business managers can increase employee satisfaction, then SMEs' performance will also increase in the future. Hence, the current study aims to determine the job satisfaction of SME employees by analyzing the impact of job training (JT) and promotion (PRO) opportunities on employee job satisfaction. Purposive sampling is applied in the study, and 202 SME employees have participated as sample respondents. The final sample size is n = 202. SPSS 26.0 version is used to analyze the hypotheses. The study findings show that both job training (JT) and promotion (PRO) have a positive effect on SME employee job satisfaction. It does indicate that SME managers need to provide necessary training programs and timely promotion to their current working employees to keep them satisfied with their job. Promotion and effective job training will certainly enhance employees' job satisfaction. The study has also offered a few strategic implications for SME business managers.
