Experimental infection method for Edwardsiellosis was studied to imitate histopathologically natural infections in the Tilapia Oreochrimis niloticus. Prior to the bacterial challenge, the intestine of tilapia was damaged by 0.2ml of 30% hydrogen peroxide introduced through a silicon tube which was inserted 4 to 5cm into the intestine from anus. E. tarda was mixed a 10% Gum arabia and administered into the stomach by a cannula 20 hours after the hydrogen peroxide treatment. Bacterial doses used were from $4.2{\times}10^6$ to $6.4{\times}10^\;CFU$/fish. Fish 72 hours after the challenged showed swollen and erosious lesion and focal necrosis with bacterial-eaden inflammatory cell in the Liver, Spleen, Kidney. Fish 96 and 120 hours after bacterial challenge were moribund showing swollen Liver, Spleen, Kidney and focal accumulation of macrophages and production of granulomas in the infected lesions.

Bacillus SW1-1 is a probiotic isolated from shrimp intestines. We investigated the effects of Bacillus SW1-1 coated diets on the growth, feed utilization, innate immunity, hematological parameters and resistance to Edwardsiella tarda in olive flounder (Paralichthys olivaceus). A commercial diet was used as the control (AP0) and two other diets were prepared by coating 0.25% (AP25) or 0.50% (AP50) probiotic powder which contains 1.0 × 107 CFU/g Bacillus SW1-1. Four replicate groups of olive flounder (153 ± 2 g) were fed one of the diets for 12 weeks. Growth performance and feed utilization of the fish were not significantly affected by the dietary Bacillus SW1-1. After the challenge with E. tarda, AP50 group showed significantly higher survival than AP0 and AP25 groups. Innate immunity and anti-oxidant capacity of the fish were not significantly affected after the feeding trial. However, after the E. tarda challenge, the innate immune parameters (immunoglobulin, lysozyme and anti-protease) were significantly improved in fish fed AP25 and AP50 diets compared to those in fish fed AP0 diet. After the challenge test, significantly lower glucose level was observed in AP50 group compared to AP0 group. These results indicate that dietary supplementation of Bacillus SW1-1 could increase the disease resistance of olive flounder against E. tarda infection. The optimum coating levels of Bacillus SW1-1 needs to be further elucidated.
The present study investigated the effects of dietary Edwardsiella tarda (E. tarda) specific bacteriophage (phage) and Bacillus subtilis (B. subtilis) mixture on innate immune responses and antibacterial activity of Nile tilapia, Oreochromis niloticus. In a dietary experiment, tilapia were fed the control diet (C), a phage-only supplemented diet (P), a B. subtilis only supplemented diet (B), or a B. subtilis and phage mixed diet (B+P). A respiratory burst and significant increase in lysozyme activity (P<0.05) were noted in the B+P group, as compared to other groups after 4 days of feeding. The B group showed a significant (P<0.05) increase in respiratory burst and lysozyme activity versus the C and P groups, whereas no significant increases (P<0.05) were observed in the P and C groups. $ACH_{50}$ was significantly up-regulated in the B+P group versus other groups after 8 days of feeding (P<0.05). In vivo antibacterial activity was significantly enhanced in the B+P fed group, as compared to other groups (P<0.05) after 7 days of E. tarda challenge. A significant (P<0.05) increase in antibacterial activity was seen in the B group, as compared to C or P groups after 14 days of feeding. These results suggest that a B. subtilis and phage mixture could be utilized as an alternative to antibiotics in the control of fish diseases caused by E. tarda.
We compared the pathogenicity and antigenicity of two different Edwardsiella tarda (E. tarda) strains KFE and Edk-2 isolated in Korea and Japan respectively. In the pathogenicity analysis with challenge test against loach, E. tarda KFE isolate showed stronger pathogenicity compared to that of E. tarda Edk-2 isolate. The differences were also confirmed by the comparison of OMP (outer membrane protein) in SDSPAGE which showed three major bands, 41kDa, 37kDa and 30kDa, for E. tarda KFE isolates and two major bands, 41kDa and 30kDa, for E. tarda Edk-2 isolates. On the base of these results, we tried to determine the differences of antigenicities of these two isolates in loach which is one of the important species in freshwater aquaculture in Korea. Numbers of specific antibody secreting cells (SASC), appeared to be higher in loach immunized with FKC of E. tarda Edk-2 than loach immunized with FKC of E. tarda KFE. ELISPOT-assay for the comparison of antigenicity showed relatively high percentage of cross-reaction and implied the presence of some common epitopes in the antigens of these two E. tarda isolates.
In this study, effect of mating group, mating dam and mating sire was analyzed using olive flounder Paralichthys olivaceus challenged with Edwardsiella tarda. A challenge test was conducted using 1,708 olive flounder fingerlings obtained through $4{\times}4$ factorial cross from parents selected based on resistant degree. Regarding the means of resistant index, survival rate and days of survival by each mating group, the group(RR) produced by mating among resistant parents were 17.98, 13.77%, and 18.36 days, respectively; the group(SS) produced by mating among susceptible parents were 12.46, 2.71%, and 12.40 days, respectively. This study gives an indication that challenge test results may be successfully used as selection criteria for disease resistance to E. tarda in olive flounder, P. olivaceus.
Effects of dietary Scutellaria baicalensis extract (SBE) on growth, feed utilization and challenge test of olive flounder (Paralichthys olivaceus) were determined. Six hundred thirty fish averaging 5.0 g were distributed into 18, 180-L tanks. Six experimental diets were prepared in triplicate: SBE-0, SBE-0.5, SBE-1, SBE-2, SBE-3 and SBE-5 diets containing SBE at the concentrations of 0, 0.5, 1, 2, 3 and 5%, respectively. Fish were hand-fed to apparent satiation twice a day for 8 wks. At the end of 8-wk feeding trial, ten fish from each tank were infected by Edwardsiella tarda for challenge test. Survival and weight gain of fish were not affected by dietary concentrations of SBE. However, specific growth rate of fish fed the SBE-2 diet was higher than that of fish fed the SBE-0, SBE-1, SBE-3 and SBE-5 diets. Neither feed efficiency nor serum chemical composition of fish was affected by dietary concentrations of SBE. The cumulative mortality of fish fed the SBE-0 diet was 100% at 96 h after E. tarda infection, but 77 to 87% for fish fed the other diets. Dietary inclusion of 2% SBE appears to be recommendable to improve specific growth rate of fish and SBE had the potential to mitigate mortality of fish at E. tarda infection.
The immunogenicity of Edwardsiella tarda was surveyed under two different culture conditions. In SDS-PAGE patterns of the outer membrane proteins (OMPs) extracts of E. tarda, grown under Trypic soy broth (TSB) and TSB supplemented iron chelate 2,2‘-dipyridyl iron-restricted condition, were examined. The results showed that the iron-regulated outer membrane protein (IROMPs) with molecular masses of 68 and 73 kDa were expressed by bacteria grown in iron-chelate TSB.The pathogenicity was examined by intraperitoneal injection with live E. tarda grown under TSB, iron-chelate TSB and iron-supplemented TSB. The result of pathogenicity test showed significantly high mortality in the group of live E. tarda grown under iron-chelate TSB.The effect of formalin killed cell (FKC) of TSB cultured bacteria and 2,2'-dipyridyl FKC (DP-FKC) of cultured bacteria on the iron-chelate TSB on the development of protective immunity in olive flounder was studied. The level of immune response was evaluated with immunized fish at 1, 2, 3 and 4 weeks after immunization. The numbers of specific antibody secreting cells (SASCs) showed significantly increased level at 2 week after immunization in each group. The agglutination titre of immunized fish was significantly high level at 3 weeks after immunization.The level of protection in olive flounder at 1, 2, 3 and 4 weeks after vaccination was examined by intraperitoneal challenge test with live E. tarda.
Edwardsiella tarda, the causative organism of Edwardsiellosis, was isolated from the water and the eel, Anguilla japonica of the recirculating eel culture ponds in 1988. E. tarda was always isolated from the water of eel culture ponds in the number of log 2.30~log 4.78 CFU/$m{\ell}$ without distinguishable seasonal characteristics. The counts of isolated E. tarda were changed with the health condition of the eel, i. e., a number of E. tarda were isolated from all checked organs, intestine contents, liver, spleen and kidney of the diseased eel, however, it was hardly isolated from spleen and kidney of the healthy eel. The isolates were classified according to o-agglutinating test into three serotypes (I, II, III). There were no distinct differences in the composition of the serotypes between intestine contents of eel and pond water samples(I, 52.4~53.3% ; II, 20.0~23.8% ; III, 4.8~6.7%). Serotype I was ascendant in the spleen(66.7%) and the kidney(73.7%) samples. By experimental challenge, most of serotype I proved highly virulent to eel in comparison with the other serotypes tested.
A formalized bacterin(FKC) of Edwardsiella tarda, conjugated with/without Freund's complete adjuvant(FCA), Freund's incomplete adjuvant(FIA) and potassium aluminium sulfate(PAS), was administered by intraperitoneal injection to tilapia respectively. The tilapia were reared under indoor recirculating filter system. There were agglutinin titer increment in the tilapia which were administered not only FKC bacterin but also adjuvanted vaccines. But the producing time of the highest level of antibody in the immunized tilapia was different among vaccine preparation, i. e. FKC+FCA group and PAS+FKC group are between the 2nd and the 4th week, and FKC alone group is the 2nd week. In the challenge experiment with $2.5{\times}10^7\;CFU/ml$ or $2.5{\times}10^8\;CFU/ml$ of E. tarda T1123 to the tilapia immunized with vaccines, RPS above sixty was recorded both challenge dose in the tilapia which were 3 weeks after immunization with FKC+FCA, PAS+FKC and FKC alone, and the former dose 8 weeks after immunization with FCA+FKC and PAS+FKC. There were some resistance enhancement against E. tarda in the tilapia which were injected with adjuvant alone than that of control. As the results, the FCA or PAS adjuvanted vaccine is effective to sustain the defensible period against edwardsiellosis.
To study the supplementaing effects of kugija, Lycium chinense, in commercial diet on the immune response of nile tilapia, Oreochromis niloticus, nonspecific immune responses were investigated. The activities of complement and lysozyme were higher in sera of the fish fed diet with kugija than control diet. The effects of kugija on vaccination of fish to Edwardsiella tarda were compared in three different vaccination methods after feeding kugija for ten weeks. Intraperitoneal injection and bath administration with formalin killed cells (FKC) and bath with extracellular products (ECP) after injection of FKC showed differences in immune responses of vaccinated fish. Bath administration with ECP after injection of FKC was more stimulated than any other methods in each of nonspecific and specific responses such as the activities of complement and lysozyme, antibody production and rosette forming cells. Moreover, cumulative mortality was significantly lower in the fish vaccinated with combination FKC and ECP after injection challenge with live E. tarda.
