Activation of JNKs is essential for BMP9-induced osteogenic differentiation of mesenchymal stem cells

  • Although BMP9 is highly capable of promoting osteogenic differentiation of mesenchymal stem cell (MSCs), the molecular mechanism involved remains to be fully elucidated. Here, we explore the possible involvement and detail role of JNKs (c-Jun N-terminal kinases) in BMP9-induced osteogenic differentiation of MSCs. It was found that BMP9 stimulated the activation of JNKs in MSCs. BMP9-induced osteogenic differentiation of MSCs was dramatically inhibited by JNKs inhibitor SP600125. Moreover, BMP9-activated Smads signaling was decreased by SP600125 treatment in MSCs. The effects of inhibitor are reproduced with adenoviruses expressing siRNA targeted JNKs. Taken together, our results revealed that JNKs was activated in BMP9-induced osteogenic differentiation of MSCs. What is most noteworthy, however, is that inhibition of JNKs activity resulted in reduction of BMP9-induced osteogenic differentiation of MSCs, implying that activation of JNKs is essential for BMP9 osteoinductive activity.

Terahertz Characteristics of Hydroxygraphene Based on Microfluidic Technology

  • Hydroxygraphene as a kind of functionalized graphene has important applications in composite, photoelectric and biological materials. In the present study, THz and microfluidic technologies were implemented to study the THz transmission characteristics of hydroxygraphene with different concentrations and residence times in magnetic and electric fields. The results show that the THz transmission intensity decreases with the increase in sample concentration and duration of an applied electric field, while it increases by staying longer in the magnetic field. The phenomenon is analyzed and explained in terms of hydrogen bond, conductivity and scattering characteristics. The results establish a foundation for future research on the THz absorption characteristics of liquid graphene based on microfluidic technology in different external environments. It also provides technical support for the application and development of graphene in THz devices.

Gene Expression Profiles of HeLa Cells Impacted by Hepatitis C Virus Non-structural Protein NS4B

  • By a cDNA array representing 2308 signal transduction related genes, we studied the expression profiles of HeLa cells stably transfected by Hepatitis C virus nonstructural protein 4B (HCV-NS4B). The alterations of the expression of four genes were confirmed by real-time quantitative RT-PCR; and the aldo-keto reductase family 1, member C1 (AKR1C1) enzyme activity was detected in HCV-NS4B transiently transfected HeLa cells and Huh-7, a human hepatoma cell line. Of the 2,308 genes we examined, 34 were up-regulated and 56 were down-regulated. These 90 genes involved oncogenes, tumor suppressors, cell receptors, complements, adhesions, transcription and translation, cytoskeletion and cellular stress. The expression profiling suggested that multiple regulatory pathways were affected by HCV-NS4B directly or indirectly. And since these genes are related to carcinogenesis, host defense system and cell homeostatic mechanism, we can conclude that HCV-NS4B could play some important roles in the pathogenesis mechanism of HCV.

  • We present an overview on the observation and research for the China seas using both field experiments and multi-sensor satellite data at ORSI/OUC, covering two topics: (1) Spatial and temporal distribution of internal waves in the China Seas and retrieval of internal wave parameters; (2) Retrieval, validation, and cross-comparison of multi-sensor ocean color data as well as ocean optics in situ experiments in the East China Sea. We also present an incoherent Doppler wind lidar, developed by ORSI, and its observation for marine-atmospheric boundary layer.
