RNA interference (RNAi)-mediated transcriptional knock-down of Crassostrea gigas big defensin 1 and 2 genes (Cg-BigDef1 and Cg-BigDef2) was investigated. The cDNA sequences of Cg-BigDef1 and Cg-BigDef2 were identical, excluding an additional fragment of 20 nucleotides in Cg-BigDef1; thus, a long double-stranded RNA (dsRNA) targeting the mRNA of Cg-BigDef2 effectively downregulated both Cg-BigDef2 and Cg-BigDef1. In addition, long dsRNA targeting green fluorescent protein (GFP) did not affect transcription of the two big defensin genes. These results suggest that the transcriptional downregulation of Cg-BigDef1 and Cg-BigDef2 was mediated by sequence-specific RNA interference (RNAi). Despite injection of long dsRNA targeting Cg-BigDef2 into only the adductor muscle, knock-down of Cg-BigDef1 and Cg-BigDef2 was observed in the adductor muscle, hemocytes, mantle, and gills, suggestive of systemic spread of RNAi in C. gigas. Furthermore, the inhibitory effect of dsRNA persisted until 72 h post-injection, indicative of a long-lasting RNAi-mediated knock-down of target genes.

The aim of this study was to evaluate the effect of the adenovirus-mediated double suicide gene (CD/TK) for selective killing of gastric cancer cells. Gastric cancer cells SCG7901 and normal gastric epithelial cell lines were infected by adenoviruses Ad-survivin/GFP and Ad-survivin/CD/TK. GFP expression and CD-TK were detected by fluorescence microscopy and reverse transcriptase polymerase chain reaction (RT-PCR), respectively. After treatment of the infected cells with the pro-drugs ganciclovir (GCV) and/or 5-FC, the cell growth status was evaluated by methyl thiazolyl tetrazolium assay. Cell cycle changes were detected using flow cytometry. In nude mice bearing human gastric cancer, the recombinant adenovirus vector was injected directly into the tumor followed by an intraperitoneal injection of GCV and/or 5-FC. The subsequent tumor growth was then observed. The GFP gene driven by survivin could be expressed within the gastric cancer line SCG7901, but not in normal gastric epithelial cells. RT-PCR demonstrated the presence of the CD/TK gene product in the infected SCG7901 cells, but not in the infected normal gastric epithelial cells. The infected gastric cancer SCG7901, but not the gastric cells, was highly sensitive to the pro-drugs. The CD/TK fusion gene system showed significantly greater efficiency than either of the single suicide genes in killing the target cells (P<0.01). Treatment of the infected cells with the pro-drugs resulted in increased cell percentage in G0-Gl phase and decreased percentage in S phase. In nude mice bearing SCG7901 cells, treatment with the double suicide gene system significantly inhibited tumor growth, showing much stronger effects than either of the single suicide genes (P<0.01). The adenovirus-mediated CD/TK double suicide gene driven by survivin promoter combined with GCV an 5-FC treatment could be an effective therapy against experimental gastric cancer with much greater efficacy than the single suicide gene CD/TK combined with GCV or 5-FC.

This study aimed to explore the effects of perceived family stress, supportive resources, intentional rumination, and coping behaviors on posttraumatic growth of cancer survivors based on the Double ABCX Family Stress Model. Data were collected from 200 cancer survivors currently living with their spouses and analyzed by path analysis using AMOS program. The results of this study were as follows. First, perceived family stress of cancer survivors had direct effects on supportive resources, intentional rumination, and coping behaviors, and had indirect effects on posttraumatic growth mediated by supportive resources, intentional rumination, and coping behaviors. Second, supportive resources, intentional rumination, and coping behaviors had direct effects on posttraumatic growth. Third, supportive resources and intentional rumination had direct effects on coping behaviors and had an indirect effect on posttraumatic growth mediated by coping behaviors. The current study supported the applicability of the Double ABCX Family Stress Model to the posttraumatic growth process of cancer survivors.

This study aimed to examine the effect of parentification on the smartphone dependency of upper elementary school children in double-income families and to verify whether ambivalence over emotional expression and loneliness sequentially mediates the relationship between parentification and smartphone dependency. The participants were 311 upper-elementary school students (4th to 6th graders; 126 boys, 40.5%) in doubleincome households residing in Seoul, Gyeong-gi, and Incheon. The data were collected through an online self-report questionnaire completed by the participants and were analyzed using SPSS 26.0 and Mplus 8.7 software. The results can be summarized as follows. Firstly, the direct effect of parentification on the smartphone dependency of the children from double-income families was statistically insignificant. Secondly, ambivalence over emotional expression mediated the effect of parentification on smartphone dependency, while loneliness did not. Lastly, parentification influenced smartphone dependency through the sequential mediating channel of ambivalence over emotional expression and loneliness. In conclusion, these findings indicate that interventions for smartphone-overdependent children from double-income families should place emphasis on children's psychological difficulties attributed to parentification. Specifically, this study highlights the importance of alleviating the levels of ambivalence over emotional expression and loneliness to address the issue of children's smartphone dependency in double-income families, suggesting possible involvement and support at both household and societal levels.

Purpose : The purpose of this study was to investigate the double mediating effect of ego-resilience and impulsivity in the effects of smartphone addiction on depression among office workers. Research design, data and methodology : The subjects of this study were office workers working in four cities: Seoul, Daejeon, Chungnam, and Chungbuk Province, and the total number of respondents was 286. Descriptive statistics, difference test (t-test, ANOVA), correlation analysis, and double-mediated effect analysis were used for statistics. Results : The results of this study are as follows. First, as a result of the difference analysis, there was no difference in depression according to the general characteristics of the subjects of this study, such as gender, age, marital status, and salary. Second, as a result of correlation analysis, smartphone addiction showed a negative correlation with ego-resilience, and a positive correlation with impulsivity and depression. Also, ego-resilience showed negative correlation with impulsivity and depression. Impulsivity showed a positive correlation with depression. Third, as a result of dual mediating effect analysis, it was found that ego-resilience and impulsivity played double-mediated role between smartphone addiction and depression. Conclusions : Based on the results of this study, theoretical measures centered on ego-resilience were discussed and presented in order to positively induce the decrease in impulsivity and depression due to smartphone addiction among office workers.

This study aimed to provide basic data for the promotion of flourishing by analyzing whether welfare and hope mediated and double-mediated in the relationship between boss incivility and flourishing with 400 childcare teachers. The subjects of the survey were collected through offline questionnaires. The data were analysed using SPSS PC+ Win ver. 25.0 and PROCESS macro. Frequency, reliability, correlation, and double mediation effect analysis were performed. As a result of the study, first, experience of incivility from bosses showed a negative correlation with welfare, hope, and flourishing. welfare, hope and flourishing showed a positive correlation with each other. Second, welfare and hope played a double mediating role in the relationship between boss and co-worker incivility and flourishing. Welfare and hope played a role in partially offsetting the negative (-) effect of incivility on flourishing. The conclusion of this study confirmed that the promotion of flourishing can be promoted by using the dual mediating role of welfare and hope.

Despite the rapid reorganization of the center of consumption online, the fashion industry is still strengthening brand marketing using offline stores. This study investigates the psychological mechanisms of shopping flow by three mediators that influence the e-word of mouth(e-WOM) as a marketing performance variable in recent marketing. Data collection was conducted online for 241 women in their 20s and 30s. The significance of multi-mediated pathways was verified using Process 3.5 Model 6. The results for multiple mediation paths are as follows. First, the direct effect of shopping flow on brand advocacy was not significant. Second, analysis of the significance of the indirect effect via simple mediation found that the path mediated by shopping value in the path from the shopping flow to brand advocacy was significant. Third, analysis of the indirect effect by double mediation revealed all three double mediating effects of shopping flow on brand advocacy were significant(emotional response and shopping value, emotional response and store attachment, shopping value and store attachment). Fourth, analysis of the significance of the indirect effect by sequential multiple mediation established the indirect effect leading to emotional response, shopping value, and store attachment was significant. These results indicate that the operation of an experiential fashion store is not just a means for sales, but a communication tool that improves and promotes the brand advocacy by providing brand experience in a store.

This study is a descriptive survey research for identifing double mediating effect of ego-integrity and cultural competency and the moderated mediation effect of Wisdom on the relationship between basic pychological needs and successful aging. The participants were 211 elderly people aged at least 60 from June 2020 to March 2021. Data were collection by self-report questionnaire. Data was analyzed by Hayes' number 6 and 91 models using the SPSS/25.0 program and SPSS PROCESS MACRO. According to the study, the ego-integration and cultural competence were effected to double mediated in successful aging, and the ego-integration and cultural competence were effected to successful aging through moderated mediation by wisdom. Therefore, it is needed to develop welfare policy and strategy for the elderly to enhance ego-integration and cultural competence by effort to satisfy basic pychological needs and wisdom.

Photoheterotrophic evolution of molecular hydrogen by Rhodobacter sphaeroides is mediated by nitrogenase that is regulated transcriptionally and post-translationally by ammonium ion. Two PII-like proteins, GlnB and GlnK, play key roles in mediating inhibition and repression of nitrogenase in the presence of ammonium ion. glnB and glnK of R. sphaeroides were interrupted to abolish the ammonium ion effect controlling nitrogenase. Ammonium ion effect was still observed in mutant having an interruption in either glnB or glnK. However, the nitrogenase activity of glnB-glnK double mutant is not affected by ammonium ion. $H_2$ evolution was improved by increasing gene dosages of nitrogenase-coding genes, nifHDK in trans in glnB-glnK double mutant.

This study aims to determine whether learning motivation and grit mediate in the relationship between emotional intelligence and learning engagement among Chinese high school students. Data were collected through a survey targeting 304 high school students purposively sampled from a high school in China. The collected data was analyzed using SPSS PC+ Win ver. 25.0 and SPSS PROCESS macro ver. 4.2. The statistical methods applied were frequency analysis, reliability analysis, correlation analysis, and dual mediation effect analysis. The conclusion of the study is as follows. First, emotional intelligence, learning motivation, grit, and learning engagement all showed significant positive correlations. Second, high school students' learning motivation and grit double-mediated in the relationship between emotional intelligence and learning engagement. Based on these results, this study proposed a plan to improve high school students' academic engagement by utilizing not only emotional intelligence but also learning motivation and grit.
