Lactulose는 lactose의 이성질체로 galactose와 fructose의 ${\beta}$-1,4-glycosidic 결합으로 구성된 비소화성 기능성 이당으로 소장에서 분해되지 않고 대장에 도달하여 장내 유산균에 의해 이용되어 대장의 pH를 저하시켜 유해균의 증식을 억제 시키고 장내 균총을 유익한 방향으로 개선하는 효과를 가지고 있다. 또한 수용성 식이섬유로 작용하여 변비와 간성뇌질환등의 치료에 이용되고 있고 lactose에 비해 감미도 및 용해도가 우수하여 다양한 식품산업으로 유용하게 사용될 수 있는 높은 잠재적 활용가치를 보유하고 있는 기능성 당류이기도 하다. Lactulose를 생산하기 위하여 화학적 방법과 효소적 방법이 보고 되어있는데, lactose를 강알칼리 조건에서 이성화시키는 화학적 방법에 의한 lactulose의 생산은 고온, 고압의 반응 조건 및 중화 과정에서 사용되는 강산으로 인해 생성물의 과분해 및 부산물 생성에 의한 복잡한 정제과정이 요구되며 환경오염에 대한 심각한 문제를 내포하고 있다는 단점이 있다. 이러한 화학적 방법과는 달리 ${\beta}$-galactosidase 또는 cellobiose 2-epimerase와 같은 효소를 이용한 lactulose의 생산은 반응의 정밀성, 특이성, 반응공정의 안전성 및 친환경적 생산방법이라는 다양한 장점을 가지고 있다. 하지만, 효소적 생산 방법 중에서 ${\beta}$-galactosidase를 이용한 lactulose의 생산은 기질로서 유당뿐 아니라 과당을 함께 사용해야 하며 높은 기질농도에서만 반응이 이루어 지기 때문에 경제적으로 비효율적이라는 단점이 지적되고 있기도 하다. Lactulose의 효소적 생산방법에 있어서 이러한 단점을 극복하기 위하여 lactose 단일 기질로부터 높은 수율의 lactulose를 생산할 수 있는 cellobiose 2-epimerase를 이용한 lactulose생산 방법에 대한 연구가 활발하게 진행되고 있지만 향후 효소 반응 공정 및 효소특성개량에 대한 지속적인 연구를 통하여 lactulose 산업적 생산을 위한 다양한 연구가 필요한 실정이다.
Gingival fibroblasts are embedded in an extracellular matrix. The matrixs have influence on the development, polarity, and behavior of nearby cells. The major component of periodontal extracellular matrix is a glycosaminoglycan. The glycosaminoglycan are large carbohydrates that are composed of repeating disaccharide units and exist in three main form: dermatan sulfate, chondrotitin sulfate, heparan sulfate. The purpose of present study is to examine the biologic effects of glycosaminoglycan on human gingival fibroblast. Human gingival fibroblasts were supplemented with each glycosaminoglycan, and cellular attachment and proliferation was determined by MTT assay. Dermatan sulfate and chondroitin sulfate did not stimulate the attachment and proliferation of human gingival fibroblasts, but heparan sulfate increased the proliferation and attachment in a time- and dose dependent manner. These results indicated that heparan sulfate seems to have a high potential for gingival regeneration and root surface attachment.

Di-D-fructose-2,6':6,2'-dianhydride (DFA-IV) is a disaccharide consisting of two fructose residues that are prepared from levan by levan fructotransferase. Levan is a homopolysaccharide composed of D-fructofuranosyl residues joined by $\beta$-(2,6) and $\beta$-(2,1) linkages. We compared the immunomodulatory effects of levan with DFA-IV. Tumoricidal activity, phagocytosis and nitric oxide (NO) production were examined in levan- and DFA-IV-treated RAW264.7 cells. The NO production, tumoricidal and phagocytic activities were significantly increased in both treated cells. The results indicate that levan has significantly greater effects on tumoricidal activity than DFA-IV at low concentrations (1 ${\mu}g/mL$) and its effect on NO production shows a similar pattern. These results suggest that tumoricidal activity induced by both samples is mediated by NO production.
Lactan gum produced by Rahnella aquatilis is a high viscous, anionic polysaccharide and has shear thinning behaviour. Lactan gum yield and cencentration was greater on disaccharide such as lactose and sucrose than on monosaccharides such as glucose and galactose. When initial carbon source concentration was 45g/l of sucrose of lactose, the microorgnisms produced 28 g/l and 27 g/l of lactan, respectively with a yield more than 60%. $\beta$-Galactosidase, hydrolyzing lactose into galactose and glucose, was induced by lactose or galactose. When initial corbon source was 45 g/l of mixed carbon I (glucose:galactose=1:1), lactan gum concentaration was higher than that from 45 g/l of monosaccharide (glucose pf galactose) but was similar to the result from 45 g/l of lactose. Therefore, lactose hydrolysis reaction by $\beta$-galactosidase does not seem to be a rate determining step in lactan gum biosynthesis. When initial carbon source was 45 g/l of mixed carbon II (glucose:fructose=1:1). total carbon source consumption rate was slower than that from sucrose, but glucose consumption rate was faster than that from fructose.
By-products of marine organisms including salmon, skate, flatfish, and yellow goosefish were investigated to search for new source of chondroitin sulfate (CS). Agarose gel electrophoresis with chondroitinase depolymerization showed that purified chondroitin sulfate did not contain any other glycosaminoglycans. 1H-nuclear magnetic resonance (NMR) spectra were acquired to confirm the structure and purity. The average molecular weight ranging from 22 to 64 kDa was determined by high performance size exclusion chromatography. Disaccharide compositions and purities were determined by strong anion exchange-high performance liquid chromatography (SAX-HPLC) after chondroitinase ABC depolymerization. SAX-HPLC data exhibited that the purity was from $81.7{\pm}1.3$ to $114.2{\pm}2.5%$ and the yield was from 1.3 to 12.5%. All analytical results indicate that salmon cartilage, skate cartilage, and yellow goosefish bone could be promising sources of CS to substitute shark cartilage CS in commercial neutraceuticals.
Currently, enzymatic synthesis of lactulose, a synthetic prebiotic disaccharide, is commonly performed with glycosyl hydrolases. In this work, a new type of lactulose-producing biocatalyst was developed by displaying β-galactosidase from Bacillus stearothermophilus IAM11001 (Bs-β-Gal) on the surface of Bacillus subtilis 168 spores. Localization of β-Gal on the spore surface as a fusion to CotX was verified by western blot analysis, immunofluorescence microscopy, and flow cytometry. The optimum pH and temperature for the resulting spore-displayed β-Gal was 6.0 and 75℃, respectively. Under optimal conditions, it showed maximum activity of 0.42 U/mg spores (dry weight). Moreover, the spore-displayed CotX-β-Gal was employed as a whole cell biocatalyst to produce lactulose, yielding 8.8 g/l from 200 g/l lactose and 100 g/l fructose. Reusability tests showed that the spore-displayed CotX-β-Gal retained around 30.3% of its initial activity after eight successive conversion cycles. These results suggest that the CotX-mediated spore-displayed β-Gal may provide a promising strategy for lactulose production.
This study intended to compare the hypolipidemic effects among six experimental groups fed by three different dietary carbohydrates on hyperlipodemic rats. Sixty experimental animals were divided into 6 groups, SB, ST, SP and CB, CT, CP after production of hyperlipidemia fed by SB diet on Sprague-Dawley rats for 4 weeks. Rats were fed by six experimental diets for eight weeks. Hyjperlipidemic rats showed three times higher in plasma TG level not in cholesterol content compare to control group fed by stock diet. Two different dietary carbohydrates seem to be effective in body weight gain and fat cumulation as weight of epididymal fat pad. In comparison of S and C groups, C fed groups showed lowering effect in plasma TG and total lipid contents, but among S fed groups, ST and SP group showed lower than SB in this respects. Dietary carbohydrates seem to be more effective than fat in plasma lipid contents. When we compare among three different fat groups, only T groups with different carbohydrates increased in peroxisomal lipid oxidation and decreased in lipogenic enzyme activites. As same token, sucrose fed group with three fat sources seem to increase activiteies enzyme activities. In epididymal fat pad and Heart, SP and CP effect more in LPL activites than other groups. In conclusion, we can recommed to consume polysaccharides rather than disaccharide and n-3 fatty acids such as perilla and tuna oils to alleviate hypertringlycemic condition.
It is indispensable to increase the conversion rate of a reducing sugars such as pentose and hexose derived from cellulosic wastes for a highly efficient bioethanol fermentation from food wastes. The saccharification liquid from cellulosic substrates such as vegetable food wastes contained lots of hexose like glucose and pentose like xylose. Since Saccharomyces-based yeasts could not convert xylose to bioethanol, Pichia stipitis which could directly ferment xylose to ethanol was chosen. After selecting Saccharomyces coreanus and P. stipitis, fermentation characteristics by mixture of two yeasts were investigated. As a result, it was verified the production of ethanol was enhanced by the co-fermentation, although there were somewhat differences between the fermentation characteristics by the aeration methods. Moreover, the consumption of pentose, hexose and disaccharide was obviously observed, and aeration in the process of fermentation seemed to stimulate the activity of P. stipitis.
Three cases of hepatic encephalopathy were diagnosed in scimitar-horned oryxs housed at a zoo. Administration of amino acid and ornithine-aspartate fluid therapy via an intravenous injection decreased serum levels of ammonia and liver enzymes in cases 1 and 2. Further, additional oral ingestion of non-absorbable disaccharide lactulose to eliminate intestinal nitrogenous products enabled recovery of two oryxs. However, in case 3, the serum levels of ammonia and liver enzymes increased even after treatment, and the oryx died. Necropsy revealed cecum and colon compaction due to stiff dried feces, and this condition could have an adverse effect on increased blood ammonia levels that may have caused mortality. Overconsumption of pellets may have been the primary cause of hepatic encephalopathy. Thus, a fiber rich diet with decreased amount of pellets is needed to prevent hepatic encephalopathy, since the normal diet of scimitar-horned oryxs is rich in fiber.
