Herbicidal Activities of Dinitroaniline Compounds in Turfgrass (Dinitroaniline계(系) 제초제(除草劑)의 잔디밭에서의 작용특성(作用特性))

  • This study was conducted to determine the herbicidal activity, persistance, downward movement and effect on bentgrass of 7 dinitroaniline herbicides such as benefin [N-butyl-N-ethyl-2,6-dinitro-4-(trifluoromethyl) benzenamine], beslogine [N,N-dibutyl-2,6-dinitro-4-trifluoromethylaniline], prodiamine [2,4-dinitro-$N^3$,$N^3$-dipropyl-6-(trifluoromethyl)-1,3-benzenediamine], pendimethalin [N-(1-ethylpropyl)-3,4-dimethyl-2,6-dinitrobenzenamine], trifluralin [2,6-dinitro-N,N-dipropyl-4-(trifluoromethyl) benzenamine], ethalfluralin [N-ethyl-N-(2-methyl-2-propenyl)-2,6-dinitro-4-(trifluoromethyl) benzenamine], and oryzalin [4-(diprop-ylamino)-3,5-dinitrobenzene-sulfonamide) together with bensulide [O,O-bis(1-methylethyl) S-[2-[(phenyl-sulfonyl) amino]ethyl]phosphorodithioate] and siduron [N-(2-methylcyclohexyl)-N'-phenylurea) as the control. In addition, distribution of pendimethalin and benefin into bentgrass was also determined. Prodiamine, benefin, and pendimethalin at the 1/16 dose of the recommended rate showed very high herbicidal activity($LD_{90}$) on Digitaria sanguinalis, but ethalfluralin and bensulide showed $LD_{90}$ at the 1/4 dose of the recommended rate, showing difference in herbicidal activities among dinitroaniline herbicides. All the herbicides except for pendimethalin had the lower herbicidal activity in sandy soil than that of clay-loam soil. Benefin, beslogine, prodiamine, oryzalin, bensulide and siduron persisted in the soil for about 50 days, but pendimethalin persisted in the soil for about 35 days, and trifluralin and ethalfluralin for about 25 days. Ethalfluralin, oryzalin and bensulide were the most mobile(downward movement) of the 9 herbicides studied. Less mobility was observed in the turfgrass condition than that of the bare soil. Beslogine bensulide prodiamine and benefin had no injury effect on bentgrass(Agrastis atolonifera L., penncross creeping bent grass). However, herbicides like oryzalin, trifluralin and pendimethalin reduced the dry weight of bentgrass by 12%, 30% and 40%, respectively. No significant difference in distribution of pendimethalin and benefin into inner part of leaves, surface and wax layer of bentgrass was observed, and thus it seems that different phytotoxic effect between pendimethalin and benefin may be attributed to different metabolism and mode of action.

  • A new chiral stationary phase (CSP) for the liquid chromatographic resolution of enantiomers was prepared from (S)-naproxen and 3,5-dinitroaniline. The 6-alkoxy-2-naphthyl group of the CSP was presumed to act as a ${\pi}$-basic interaction site for resolving ${\pi}$-acidic racemates while the 3,5-dinitroanilide group of the CSP was presumed to play a role as a ${\pi}$-acidic interaction site for resolving ${\pi}$-basic racemates. From the chromatographic resolution trends of N-alkylamide derivatives of ${\alpha}$-arylalkylamines on the CSP prepared, the chiral recognition mode involving the intercalation of the amide alkyl chain of the less retained enantiomers between the connecting tethers of the CSP was proposed.

Effect of Oryzalin on the Gravitropic Response and Ethylene Production in Maize Roots (옥수수 일차뿌리에서 oryzalin이 굴중성 반응과 에틸렌 생성에 미치는 효과)

  • Oryzalin is a dinitroaniline herbicide, which disrupts the arrangement of microtubules. Microtubules and microfilaments are cytoskeletal components that are thought to play a role in the sedimentation of statoliths and the formation of cell walls. Statoliths regulate the perception of gravity by columella cells in the root tip. To determine the effect of oryzalin on the gravitropic response, ethylene production in primary roots of maize was investigated. Treatment with 10-4 M oryzalin to the root tip inhibited the growth and gravitropic response of the roots. However, the treatment had no effect on the elongation zone of the roots. An application of 10-4 M oryzalin for 15 hr to the root tip caused root tip swelling. The application of 1-aminocycopropane-1-carboxylic acid (ACC), a precursor of ethylene, to the root tip also inhibited the gravitropic response. To understand the role of oryzalin in the regulation of the growth and gravitropic response of roots, ethylene production in the primary roots of maize was measured following treatment with oryzalin. Oryzalin stimulated ethylene production via the activation of ACC oxidase (ACO) and ACC synthase (ACS), and it increased the expression of ACO and ACS genes. Indole-3-acetic acid (IAA) played a key role in the asymmetric elongation rates observed during gravitropism. The results suggest that oryzalin alters the gravitropic response of maize roots through modification of the arrangement of microtubules. This might reduce the distribution of IAA in the upper and lower sides of the elongation zone and increase ethylene production, thereby inhibiting growth and gravitropic responses.

Enhancement of Pendimethalin Degradation Activity in Bacillus sp. MS202 using Gamma Radiation

  • To induce the enhanced mutants of dinitroaniline herbicide pendimethalin degrading bacterium, Bacillus sp. MS202 was irradiated with gamma radiation at the dose of $LD_{99}$ (3.35 kGy). Three enhanced mutants (MS202m7, MS202m14, MS202m18) were isolated from the candidates by the generation - isolation method. Clear zone formation and the GC analysis confirmed that the degrading activity of each enhanced mutant (MS202m7, MS202m14, MS202m18), the formation of pendimethalin metabolite, increased by $11\%,\;45\%,\;and\;32\%$ than a wild type, respectively. It suggested that these mutants induced by gamma radiation could be useful for the application of pesticide degradation.

Partial Reduction of Dinitroaniline Herbicide Pendimethalin by Bacillus sp. MS202 (Bacillus sp. MS202에 의한 Dinitroaniline계 제초제인 Pendimethalin의 부분환원)

  • The persistence of pendimethalin in soil and ground water has an injurious effect on ecosystem. Pendimethalin-degrading bacterium was isolated from Masan, Gyeongnam province and temporarily identified as Bacillus sp. MS202 by the analysis of API CHB50, kit, FAME, and 16S rDNA sequence. from the analysis of pnedimethalin metabolite using TLC, GC, and GC-MS, we found that the degradation of pendimethalin by Bacillus sp. MS202 did not result in the dealkylated form, but the formation of the reduced compound, 6-amino-2-nitro-N(1-ethylpropyl)-3,4-xylidine or 2- amino-6-nitro-N(1-ethylpropyl)-3,4-xylidine.

Syntheses, X-ray Structures and Second Harmonic Generation Efficiencies of MAP (Methyl (2,4-dinitrophenyl)-aminopropanoate) Analogues

  • An attempt to improve the second harmonic generation (SHG) efficiency of MAP (methyl (2,4-dinitrophenyl)aminopropanoate) by modifying the substituents on the amino group of MAP is described. Several MAP analogues have been prepared using optically active amino acids alanine, phenylalanine and serine, and their SHG efficiencies measured. None of the MAP analogues exhibited SHG efficiencies as high as that of MAP. X-ray crystal structures of three MAP analogues have been determined. In the crystal structures of two of them, which were the derivatives of phenylalanine, two crystallographically-independent molecules existing in the asymmetric unit are aligned almost antiparallel. These structures are consistent with the very low SHG efficiencies of these compounds. On the other hand, the crystal structure of a serine derivative reveals substantial alignment of the dinitroaniline chromophore along the polar axis. However, the angle of 86.2° between the molecular charge tranfer axis and the polar axis of the crystal is still far away from the optimum value of 54.74° for the phase-matchable SHG. The structure is consistent with the SHG efficiency of this compound which is much higher than those of the phenylalanine derivatives but still lower than that of MAP. This study demonstrates the importance of the orientation of molecules in the crystal lattice in determining secod-order nonlinear optical properties of crystalline materials.

Simultaneous Pesticide Analysis Method for Bifenox, Ethalfluralin, Metolachlor, Oxyfluorfen, Pretilachlor, Thenylchlor and Trifluralin Residues in Agricultural Commodities Using GC-ECD/MS (GC-ECD/MS를 이용한 농산물 중 Bifenox, Ethalfluralin, Metolachlor, Oxyfluorfen, Pretilachlor, Thenylchlor 및 Trifluralin의 동시 분석)

  • BACKGROUND: This experiment was conducted to establish a simultaneous analysis method for 7 kinds of herbicides in 3 different classes having similar physicochemical property as diphenyl ether(bifenox and oxyfluorfen), dinitroaniline (ethalfluralin and trifluralin), and chloroacetamide (metolachlor, pretilachlor, and thenylchlor) in crops using GC-ECD/MS. METHODS AND RESULTS: All the 7 pesticide residues were extracted with acetone from representative samples of five raw products which comprised apple, green pepper, Kimchi cabbage, hulled rice and soybean. The extract was diluted with saline water and directly partitioned into n-hexane/dichloromethane(80/20, v/v) to remove polar co-extractives in the aqueous phase. For the hulled rice and soybean samples, n-hexane/acetonitrile partition was additionally employed to remove non-polar lipids. The extract was finally purified by optimized Florisil column chromatography. The analytes were separated and quantitated by GLC with ECD using a DB-1 capillary column. Accuracy and precision of the proposed method was validated by the recovery experiment on every crop samples fortified with bifenox, ethalfluralin, metolachlor, oxyfluorfen, pretilachlor, thenylchlor, and trifluralin at 3 concentration levels per crop in each triplication. CONCLUSION: Mean recoveries of the 7 pesticide residues ranged from 75.7 to 114.8% in five representative agricultural commodities. The coefficients of variation were all less than 10%, irrespective of sample types and fortification levels. Limit of quantitation (LOQ) of the analytes were 0.004 (etahlfluralin and trifluralin), 0.008 (metolachlor and pretilachlor), 0.006 (thenylchlor), 0.002 (oxyfluorfen), and 0.02 (bifenox) mg/kg as verified by the recovery experiment. A confirmatory technique using GC/MS with selected-ion monitoring was also provided to clearly identify the suspected residues. Therefore, this analytical method was reproducible and sensitive enough to determine the residues of bifenox, ethalfluralin, metolachlor, oxyfluorfen, pretilachlor, thenylchlor, and trifluralin in agricultural commodities.

Triazine Herbicide Treatment and Survival Rate of Pearl Millet Seedlings (Triazine 제초제(除草劑) 처리(處理)와 진주(眞珠)조의 생존율(生存率))

  • Pearl millet (Pennisetum americanum (L.) Leeke) has been found as a promising new forage crop of higher quality and yield in Korea since 1985. The objective of the study was to select preemergence herbicides for weed control in growing pearl millet in order to reduce production cost and with increased yields. Propazine (Milogard 4L) was the best in controlling weeds without its phytotoxicity at the standard application rate, and with the highest survival rate of seedlings of peal millet "Suwon 1" hybrid. Sprayings with Atrazine and Simazine caused lower survival rate of peal millet and with their light phytotoxicity as compared with manual weeding and Propazine spraying. Alachlor killed pearl millet seedlings almost completely. Pendimethalin also was not good for weed control of peal millet due to its severe phytotoxicity and with very low survival rate of pearl millet seedlings.

Development of Tetraploid Watermelon Using Chromosome Doubling Reagent Treatments (염색체 배수화제를 이용한 4배체 수박품종 개발)

  • To produce high quality watermelon, three tetraploid watermelon breeding lines (‘SA03-1’, ‘SA06-1’ and ‘SB01-1’) were developed by treatment with different chromosome doubling reagents. To identify the optimal tetraploid inductive conditions, the three watermelon breeding lines were selected by counting the number of doubled chloroplasts in guard cells. Tetraploid induction rates differed depending on the genotypes and treatment with doubling reagents. However, the highest induction rate occurred with 1.0% colchicine (82.2%). These putative tetraploid lines were re-confirmed for ploidy using flow cytometric analysis and chromosome counting. The internode length of the tetraploid breeding lines was different when the leaf size was larger in all three tetraploid lines compared to their diploids. The fruit weight of the tetraploid fruits for ‘SA03-1’ and ‘SB01-1’ was lower than for their diploid, and the rind thickness and total sugar content (°Brix) of tetraploid SB01-1 were significantly different from those of its diploid. Tetraploid lines were sterile, yielded a lower number of seeds per fruit for ‘SA03-1’ (21), ‘SA06-1’ (62), and ‘SB01-1’ (34.7), and the seeds were larger and thicker than those of their diploids. These tetraploid breeding results will be useful for breeding new seedless watermelon cultivars.

The Effect of Oryzalin on Growth and Gravitropism in Arabidopsis Roots (Oryzalin이 애기장대 뿌리 생장과 굴중성 반응에 미치는 작용)

  • Oryzalin is a dinitroaniline herbicide that has been known to disrupt microtubules. Microtubules and microfilaments are components of cytoskeletons that are implicated in plant cell growth, which requires the synthesis of cellulose when cell walls elongate. In addition, microtubules are also involved in the sedimentation of statoliths, which regulate the perception of gravity in the columella cells of root tips. In this study, we investigated the effect of oryzalin on the growth and gravitropic response of Arabidopsis roots. The role of ethylene in oryzalin's effect was also examined using these roots. Treatment of oryzalin at a concentration of 10-4 M completely inhibited the roots' growth and gravitropic response. At a concentration of 10-6 M oryzalin, root growth was inhibited by 47% at 8 hr when compared to control. Gravitropic response was inhibited by about 38% compared to control in roots treated with 10-6 M oryzalin for 4 hr. To understand the role of oryzalin in the regulation of root growth and gravitropic response, we measured ethylene production in root segments treated with oryzalin. It was found that the addition of oryzalin stimulated ethylene production through the activation of ACC oxidase and ACC synthase genes, which are key components in the synthesis of ethylene. From these findings, it can be inferred that oryzalin inhibits the growth and gravitropic response of Arabidopsis roots by stimulating ethylene production. The increased ethylene alters the arrangement of the microtubules, which eventually interferes with the growth of the cell wall.
