동물성(動物性)과 식물성(植物性) 단백질(蛋白質) 섭취(攝取)가 혈청(血淸) 지질(脂質) 및 뇨중(尿中) Methylhistidine에 미치는 영향(影響) (Effect of Plant and Animal Proteins on Serum Lipids and Urinary Methylhistidine in Human)

  • 본(本) 연구자(硏究者)는 21세(歲)부터 25세(歲)까지의 건강(健康)한 여대생(女大生) 8명(名)을 대상(對象)으로 식이중(食餌中)의 단백질(蛋白質) 급원(給源)((동물성)(動物性)과 식물성(植物性) 단백질(蛋白質))을 달리했을 때 혈청중(血淸中)의 cholesterol 농도(濃度)와 triglyceride 농도(濃度) 및 24시간(時間) 뇨중(尿中)의 creatinine 농도(濃度)와 3-methylhistidine의 농도(濃度)에 미치는 영향(影響)을 살펴보았다. 조사대상자(調査對象者)들은 control diet와 meat diet와 textured soy protein (TSP) diet를 각(各) 3일씩 섭취(攝取)하였으며, 각(各) 식이(食餌)의 3일째 되는 날에는 24시간(時間) 뇨(尿)를 채취(採取)하였고, 4일째 되는 날 아침 식사(食事) 전에 채혈(採血)하여 분석(分析)한 결과(結果)는 다음과 같다. 1) 혈청(血淸) cholesterol 농도(濃度)는 meat diet 시에 control diet보다 현저(顯著)하게 증가(增加)하였으며 (P<0.05), TSP diet시에는 control diet 보다 현저(顯著)하게 감소(減少)하였고 (P<0.01) meat diet와 비교(比較)시에도 현저(顯著)하게 감소(減少)되었다(P<0.01). 2) 혈청(血淸) triglyceride 농도(濃度)는 meat diet 시에 control diet 보다 감소(減少) 되었으나, 유의적(有意的)인 차이(差異)는 없었고 meat diet와 비교(比較)시에는 현저(顯著)하게 감소(減少)되었다(P<0.01). 3) 뇨중(尿中)의 creatinine 농도(濃度)는 meat diet 시에 control diet보다 약간 증가(增加)했으나 유의적(有意的)인 차이(差異)는 없었다. TSP diet 시에는 control diet보다 약간 감소(減少)하였으나 유의적(有意的)인 차이(差異)는 없었고 meat diet와 비교(比較)시에는 현저(顯著)하게 감소(減少)되었다(P<0.05). 4) 뇨중(尿中)의 3-methylhistidine 농도(濃度)는 meat diet 시에 control diet보다 약간 증가(增加)했으나, 유의적(有意的)인 차이(差異)는 없었다. TSP diet 시에는 control diet보다 약간 감소(減少)하였으나 유의적(有意的)인 차이(差異)는 없었고 meat diet와 비교(比較)시에는 현저(顯著)하게 감소(減少)되었다 (P<0.05). 5) 3-methylhistidine가 creatinine의 비(比)는 control diet 보다 meat diet와 TSP diet 시에 약간 증가(增加)하였으나, 유의적(有意的)인 차이(差異)는 없었다.

단백질 급원의 변화와 열량제한 및 식이회복이 흰쥐의 성장발달에 미치는 영향 (Effects Protein Quality, Energy Restriction and Subsequent Rehabilitation on Growth in Young Rats)

  • Effects of dietary protein qualify, energy restriction, and subsequent rehabilitation were studied in the weanling rats. Rats were devided into the six experimental diet groups. Rats in AC (casein- containing diet ), AS (soyprotein-containing diet), and AG(gluten-containing diet) group consumed their diets ad libitum, and rats in PC, PS, PG groups were pair-fed in 60% of the of amounts eaten by corresponding rats in the AC, AS, AG groups, respectively. After 3 weeks of nutritional restriction, all rats were switched to the AC diet for 14 days. At the end of restriction period, body weight, weight gain, food intake, and FER were higher in casein- fed rats than other groups of rats. The differences were, however, disappeared with rehabilitation diet, except body weights which were not fully recovered until the end of experiment. The weights of liver, gastrocnemius muscle, kidney, small intestine, spleen, and lung were the highest in the casein group, and the lowest in gluten group at the end of restriction period. The recovery with rehabilitation diet were differ in different organs. The muscle and liver seemed to be the most affected organs by dietary protein quality and energy restriction. There were no differences in brain weights among the experimental groups during the restriction period, however, after 2 weeks of rehabilitation, rats in AC group showed lower brain weight compared to AC and AS groups, and the brain weight of PC group was lower than AC group. Protein contents in liver and muscle showed the same trends as their weights. Gluten-fed rats showed low serum protein concentration, but recovered fully with rehabilitation diet for 3 days.

Effects of freeze-dried cranberry powder on serum lipids and inflammatory markers in lipopolysaccharide treated rats fed an atherogenic diet

  • This study investigated the effects of freeze-dried cranberry powder on anti-inflammation and lipid profiles of lipopolysaccharide (LPS)-treated rats fed an atherogenic diet for 6 weeks. Forty Sprague-Dawley male rats (6-weeks-old) were equally divided into the following five groups: 1) normal diet group+saline (NC); 2) atherogenic diet+saline (HFC); 3) atherogenic diet+LPS (HL); 4) atherogenic diet with 5% cranberry power+LPS (C5); 5) atherogenic diet with 10% cranberry power+LPS (C10). LPS (0.5 mg/kg) was injected into the abdominal cavities of rats 18 hours prior to sacrifice. At the end of the experimental period, we measured serum lipid profiles as well as levels of serum C-reactive protein (CRP), nitric oxide (NO), and pro-inflammatory cytokines such as tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-1${\beta}$, IL-6, and IL-10 as an anti-inflammatory cytokine. The mean serum high density lipoprotein (HDL)-cholesterol level in C5 rats was significantly higher than that in NC and HL rats (P<0.05). The mean serum levels of CRP and IL-1${\beta}$ were significantly lower (P<0.05) in the cranberry powder groups compared to those in HL rats. Additionally, mean serum IL-6 levels tended to be lower in the cranberry groups than that in the HL group, whereas serum IL-10 and NO showed 29% and 88% higher mean values in the C5 group and 49% and 24% higher in the C10 group than those in the HL group, respectively. These results suggest that freeze-dried cranberry powder may have beneficial effects on cardiovascular diseases by modifying serum lipids and the early inflammatory response.

비유에 대한 래복자 약침(藥鍼)이 고지방 식이로 유발된 비만(肥滿) 백서(白鼠)에 미치는 영향 (Effects of Herbal Acupuncture(Raphanus Sativus) at $BL_{20}$ on the Hyperlipemia Rats Induced by High Fat Diet)

  • Objectives : This research was performed to investigate the effect of herbal acupuncture(Raphanus sativus) at Bisu($BL_{20}$) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL to total cholesterol ratio of rats fed high fat diet for 5weeks. Methods : Experimental groups were divided into high fat diet group(Control), high fat diet and saline in $BL_{20}$ treated group(Saline), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-1, 0.058mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-2, 0.029mg/ml/g), high fat diet and Raphanus sativus-herbal acupuncture in $BL_{20}$ treated group(RHA-3, 0.011mg/ml/g), Herbal acupuncture was bilaterally treated at 0.2cc per 4days, total 14 times in 56days. Results : Body weight and food efficiency were decreased in saline, RHA-1, RHA-2. Feed intake was increased in saline, RHA-1, RHA- 2, RHA- 3. The level of serum total cholesterol was decreased in saline, RHA-1. The level of serum HDL-cholesterol, LDL-cholesterol were not significant. The level of serum triglyceride, Total Bilirubin were decreased in saline, RHA-1, RHA-2, RHA-3.The level of serum AST was decreased in RHA-1. But the level of serum ALT was increased in RHA-1. The level of atherogenic index was decreased in saline, RHA-1. The level of HTR was increased in saline, RHA-1. Conclusions : Raphanus sativus-herbal acupuncture in Bisu($BL_{20}$) can control body weight, feed intake and feed efficency ratio, total cholesterol, total bilirubin, and prevention cardiovascular risk.

Effects of Dietary Inclusion of Astaxanthin on Growth, Muscle Pigmentation and Antioxidant Capacity of Juvenile Rainbow Trout (Oncorhynchus mykiss)

  • This study was designed to investigate the effects of dietary astaxanthin levels on growth performance, feed utilization, muscle pigmentation, and antioxidant capacity in juvenile rainbow trout. Four experimental diets were formulated to contain 0, 50, 75, and 100 mg/kg astaxanthin (designed as AX0, AX50, AX75, and AX100). Each diet was fed to triplicate groups of fish (18.5 g/fish) for 10 weeks. Growth performance and muscle composition of fish were not affected by dietary astaxanthin levels. Total carotenoid concentration in the muscle of fish fed the AX50 diet was higher than that of fish fed the AX0 diet, but no significant differences were observed between these fish and those fed the AX75 and AX100 diets. Muscle astaxanthin content increased with increased astaxanthin in the diet. Deposition of astaxanthin in the flesh resulted in a decrease in lightness and an increase in redness and yellowness. The fillets from trout fed the AX75 diet had significantly lower lightness than trout fed the AX50 and AX100 diets. Fish fed the AX50 and AX75 diets showed significantly lower catalase activity than those fed the control diet. Total antioxidant status increased significantly in all astaxanthin supplemented groups when compared to the control group. Superoxide dismutase activity was significantly decreased in fish fed the AX50 diet compared to fish fed the AX0 diet. These findings suggest that while fillet pigmentation increased with increasing dietary astaxanthin concentration, indices of fish antioxidant capacity may not be affected in a dose dependent manner.

Functions of Monocyte Chemotactic Protein-3 in Transgenic Mice Fed a High-Fat, High-Cholesterol Diet

  • Monocyte chemotactic protein-3 (MCP-3), a chemokine that is in a superfamily of structurally related small chemotactic cytokines involved in leukocyte trafficking, is regarded as a key factor in atherogenesis. In this study, we examined the changes in atherogenic parameters including hepatic lipid accumulation and oxidative balance in MCP- 3-overexpressing transgenic mice (MCP-3 mice) under atherogenic conditions. To induce an extreme atherogenic condition, mice were fed a high-fat, high-cholesterol (HFHC) diet for 12 weeks. The body weight and food intake were not changed by MCP-3 overexpression in the aorta. On a HFHC diet, the MCP-3 mice had higher plasma levels of total cholesterol and a higher atherogenic index compared with wild-type mice, although there were no differences in the plasma HDL-cholesterol and triglyceride levels. Furthermore, an increase in lipid accumulation was observed in the aortas as well as the livers of the HFHC diet-fed MCP-3 mice compared with wild-type mice. The activities of antioxidant enzymes increased in the livers of the HFHC diet-fed MCP-3 mice, whereas supplementation with antioxidants, naringin and hesperidin, reversed the activities of the hepatic antioxidant enzymes in HFHC diet-fed MCP-3 mice, indicating that there might be more oxidative damage to the tissues in the HFHC diet-fed MCP-3 mice leading to progression towards atherosclerosis and hepatic steatosis. Microarray analyses of the aorta revealed atherosclerosis-, PPARs-, lipoprotein receptor, and apolipoprotein-related genes that were affected by the HFHC diet in MCP-3 mice. These findings suggest that aortic MCP-3 overexpression may contribute to the development of atherosclerosis and hepatic steatosis under atherogenic conditions.

구기자 분말 첨가 식이가 1% 고콜레스테롤 급여 흰쥐의 지질 대사에 미치는 영향 (Effect of Lycii Fructus Powder on Lipid Metabolism in 1% Cholesterol Fed Rats)

  • This study examined the effects of 2% Lycii fructus powder (LFP) supplementation on lipid metabolism in rats. Male Sprague-Dawley rats were administered 1% cholesterol and 0.25% sodium cholate to induce hypercholesterolemia. Then, 40 rats were divided into four diet groups: a normal diet group (NC), high cholesterol diet group (HC), normal diet plus 2% Lycii fructus powder (NC-LFP) group, and high cholesterol diet plus 2% Lycii fructus powder (HC-LFP) group. The HC group presented higher growth rates and liver weights than NC and NC-LFP however, growth rates and liver weights in the 2% LFP administered groups gradually decreased. HC also showed increased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels and decreased atherogenic index values, HDL-cholesterol, and phospholipid levels, whereas LFP group showed decreased serum triglyceride, total cholesterol, free cholesterol, and LDL-cholesterol levels as compared to HC. There were no differences in serum triglyceride, phospholipid, LDL-cholesterol, HDL-cholesterol, and free cholesterol concentrations between the normal diet groups (NC and NC-LFP). The high cholesterol diet groups (HC and HC-LFP) had significant increases in serum alanine aminotransferase (ALT), asparate aminotransferase (AST), alkaline phosphatase (ALP), and lactate dehydrogenase (LDHase) activities. And the 2% LFP administered groups had lower hepatic concentrations of total cholesterol and triglycerides than the HC groups. Overall, the results suggest that Lycii fructus powder has hypochloesterolemic effects by reducing serum and liver cholesterol contents.

김 다당류 porphyran의 급이가 흰쥐의 혈청과 간의 효소활성 및 마우스의 면역에 미치는 영향 (Effect of Porphyran isolated from Laver, Porphyra yezoensis, on Liver Lipid Peroxidation in Hyperlipidemic Rats and on Immunological Functions in Mice)

  • 남해안에서 다량 생산되고 있는 김(Porphyra yezoensis)의 생리활성 물질은 porphyran을 분리 추출한 후, 흰쥐에서 고지혈증 및 고 콜레스테롤혈증을 유발시켜 간장의 항산화 효소활성 및 면역능을 측정하였다. 간의 효소활성 측정을 위해 Sprague-Dawley계 흰쥐를 9마리씩 5군으로 나누어 1군은 정상군, 2군은 대조군(고지혈증 유발군), 3, 4 및 5군은 porphyran 급이군으로서 대조군에 porphyran을 각각 1%, 5% 및 10%의 비율로 사료에 첨가하여 4주간 사육하였다. 면역능은 Balb/c female mouse를 이용하여 3마리씩 4군으로 대조군 및 porphyran 1%, 2% 및 5%로 조제된 시료를 실험동물에게 2일에 1회 1mL씩 복강 주사를 연속적으로 20일간 투여한 후 비교하였다. 혈청중의 alanine aminotransferase(ALT), aspartate aminotransferase(AST), alkaline phosphatase(ALP) 및 lactic dehydrogenase(LDH) 활성은 대조군에 비해 porphyran 급이군에서 낮았으며, porphyran의 급이 수준이 증가할수록 대조군에 비해 유의하게 낮은 경향을 보였다. 간장의 superoxide dismutase와 catalase활성은 정상군에 비하여 대조군이 유의하게 높았으나, 대조군에 비해 porphyran 급이군에서는 낮았으며, 특히 10% porphyran 급이군에서는 정상군 수준으로 유의하게 감소되었다. TBARS 역시 대조군에 비해 porphyran 급이군 모두 유의하게 감소하였으며, 정상군 수준이하로 감소되었다. Interleukin-1농도는 porphyran 2% 및 5%를 투여한 흰쥐의 혈청에서 정상군보다 각각 26.81%, 77.95%의 증가를 나타내었으나, Interleukin-2는 감소하였다. 본 연구 결과에서 porphyran 급이는 고지혈증을 유발한 흰쥐에서 과산화물 농도를 저하시켜 항산화 효소활성을 낮추었으며, 면역능에서 일정한 현상은 나타나지 않았다.

옥수수 껍질 분말의 In Vitro 에서의 항산화 특성과 고지방식이로 유도된 Mice의 혈청지질 수준에 미치는 영향 (Effect of In Vitro Antioxidant Properties and Extract of Corn Husk on Serum Lipids in Mice)

  • This study was performed to verify the effects of corn husk powder on serum lipid levels of high-fat diet-induced mice from a nutritional viewpoint. Powder of corn husks has been used as a folk remedy from ancient times in Korea. In the case of corn husk measurement of TPC (p<0.05), ABTS+ radical scavenging activity of trolox (p<0.05), DPPH radical scavenging activity (p<0.05), and measured value of FRAP (p<0.0001) were higher in ethanol extract than water extract. Weight of livers was significantly higher in the control group and high-fat diet group compared to groups fed 10% and 20% corn husk powders with high-fat diet (p<0.0001). For blood HDL-cholesterol concentration, mice fed 10% corn husk powder with high-fat diet ($160.00{\pm}16.17mg/dL$) showed significantly higher levels compared with the other three groups (p<0.05). Mice fed 10% and 20% corn husk powders with high-fat diet showed high numerical values of $13.50{\pm}2.51$ and $11.25{\pm}2.87mg/dL$, respectively. For triglyceride concentration in blood, mice fed 10% ($83.50{\pm}19.30mg/dL$) and 20% ($88.50{\pm}22.25$) corn husk powder with high-fat diet showed very low levels compared to the control group and high-fat diet group. In the case of leptin, the high-fat diet group showed a significantly high level of $5.88{\pm}3.53ng/dL$. Groups fed 10% and 20% corn husks with high-fat diet showed low numerical values of $2.72{\pm}2.34ng/dL$ and $1.10{\pm}0.62ng/dL$, respectively (p<0.05). Corn husk powder increased high HDL-cholesterol concentration in blood while lowering triglyceride concentration and leptin secretion. Therefore, powder of corn husks has utility in the food industry and food processing.

고콜레스테롤 식이 섭취 흰쥐에 대한 적송잎 추출물의 항산화 효과 (Antioxidative Activity of Pine (Pinus densiflora) Needle Extracts in Rats Fed High-Cholesterol Diet)

  • 본 연구는 선행연구에서 적송잎 열수추출물의 항산화 활성이 높았으며 아임계 추출물 중에 항산화 활성이 높은 proanthocyanidin 함량이 높은 것으로 나타났으므로, 이러한 in vitro 실험의 결과를 in vivo 실험을 통하여 입증하고자 하였다. 적송잎의 열수추출물과 아임계 추출물이 콜레스테롤 식이를 투여한 흰쥐의 체내 항산화능에 미치는 영향을 검토하기 위하여 혈청 중의 AST, ALT, ALP 및 간장중의 항산화 효소인 SOD, CAT, GPx의 활성을 측정하였다. 체중 증가량은 콜레스테롤 식이를 투여한 CHOL군이 일반 대조식이군인 CON군보다 유의적으로 증가하는 결과가 나타났다. 콜레스테롤 식이 투여 시 식이섭취량은 일반식이군에 비하여 감소하였으나 식이효율은 유의적으로 증가하였다(p<0.05). 적송잎 추출물을 투여하였을 경우 체중증가량은 감소하였으나 유의차가 없었으므로 적송잎 추출물이 콜레스테롤 식이로 인한 체중증가에는 별다른 영향을 주지 않는것으로 나타났다. 혈청 중의 ALT 및 AST 활성은 콜레스테롤 식이를 투여한 CHOL군이 CON군에 비하여 활성이 증가되었으나 적송잎 추출물의 투여로 인하여 감소하는 경향을 타나내었다. 특히 열수 추출물보다 아임계 추출물에서 AST의 활성은 유의적으로 낮게 나타났으며, ALT는 유의성은 없었으나 CHOL군보다 활성이 낮게 나타났다. 간 조직 중의 항산화 효소 결과에서 SOD 유사활성은 CHOL군에서 대조군에 비해 유의적으로 감소하였으나 적송잎 아임계추출물 투여에 의해 증가하는 결과가 나타났다. GPx활성은 CON군에 비해 CHOL군에서 유의적으로 감소하였으나 적송잎 추출물 투여에 의해 CHOL군에 비해 현저하게 높은 결과로 나타나, 콜레스테롤식이 투여로 인한 효소활성 저하가 적송잎에 의해 회복됨을 알 수 있었다. 이상의 결과, 고콜레스테롤 유발 흰쥐에게 적송잎 추출물의 급여는 체내 항산화효소의 활성에 영향을 미치는 것으로 나타나 적송잎 추출물이 체내 항산화능 개선에 효과가 있을 것으로 기대된다.
