Degradation and Detoxification of Disperse Dye Scarlet RR by Galactomyces geotrichum MTCC 1360

  • Galactomyces geotrichum MTCC 1360 degraded the Scarlet RR(100 mg/l) dye within 18 h, under shaking conditions(150 rpm) in malt yeast medium. The optimum pH and the temperature for decolorization were pH 12 and $50^{\circ}C$, respectively. Enzymatic studies revealed an induction of the enzymes, including flavin reductase during the initial stage and lignin peroxidase after complete decolorization of the dye. Decolorization of the dye was induced by the addition of $CaCO_3$ to the medium. EDTA had an inhibitory effect on the dye decolorization along with the laccase activity. The metabolites formed after complete decolorization were analyzed by UV-VIS, HPLC, and FTIR. The GC/MS identification of 3 H quinazolin-4-one, 2-ethylamino-acetamide, 1-chloro-4-nitro-benzene, N-(4-chloro-phenyl)-hydroxylamine, and 4-chloro-pheny-lamine as the final metabolites corroborated with the degradation of Scarlet RR. The phytotoxicity study revealed the nontoxic nature of the final metabolites. A possible degradation pathway is suggested to understand the mechanism used by G. geotrichum and thereby aiding development of technologies for the application of this organism to the cleaning-up of aquatic and terrestrial environments.

Anti-tumor Initiating Potential of Andrographolide in 7,12-dimethylbenz[a]anthracene Induced Hamster Buccal Pouch Carcinogenesis

  • The aim of the study was to investigate the chemopreventive potential of andrographolide in 7,12-dimethylbenz(a) anthracene (DMBA)-induced hamster buccal pouch carcinogenesis. Oral tumors developed in the buccal pouch of golden Syrian hamsters at a 100% incidence on painting with 0.5% DMBA in liquid paraffin three times a week for 14 weeks. Marked abnormalities in the status of detoxification enzymes, lipid perxodiation and antioxidants were noticed in hamsters treated with DMBA alone. Oral administration of andrographolide at a dose of 50 mg/kg bw to hamsters treated with DMBA not only completely prevented the tumor formation but also restored the status of the above mentioned biomarkers. The present study thus demonstrates the chemopreventive potential of andrographolide in DMBA-induced hamster buccal pouch carcinogenesis, which is probably due to its antioxidant potential as well as modulating effect on xenobiotic metabolising enzymes during DMBA-induced oral carcinogenesis.

Up-regulation of Aldo-keto Reductase 1C3 Expression in Sulforaphane-treated MCF-7 Breast Cancer Cells

  • The chemopreventive activity of sulforaphane (SFN) occurs through its inhibition of carcinogen-activating enzymes and its induction of detoxification enzymes. However, the exact mechanisms by which SFN exerts its anti-carcinogenic effects are not fully understood. Therefore, the mechanisms underlying the cytoprotective effects of SFN were examined in MCF-7 breast cancer cells. Exposure of cells to SFN (10 ${\mu}M$) induced a transcriptional change in the AKR1C3 gene, which is one of aldo-keto reductases (AKRs) family that is associated with detoxification and antioxidant response. Further analysis revealed that SFN elicited a dose- and time-dependent increase in the expression of both the NRF2 and AKR1C3 proteins. Moreover, this up-regulation of AKR1C3 was inhibited by pretreatment with antioxidant, N-acetyl-L-cysteine (NAC), which suggests that the up-regulation of AKR1C3 expression induced by SFN involves reactive oxygen species (ROS) signaling. Furthermore, pretreatment of cells with LY294002, a pharmacologic inhibitor of phosphatidylinositol 3-kinase (PI3K), suppressed the SFN-augmented Nrf2 activation and AKR1C3 expression; however, inhibition of PKC or MEK1/2 signaling with $G\ddot{o}6976$ or PD98059, respectively, did not alter SFN-induced AKR1C3 expression. Collectively, these data suggest that SFN can modulate the expression of the AKR1C3 in MCF-7 cells by activation of PI3K via the generation of ROS.

cDNA Microarray Analysis of Transcriptional Response to Hyperin in Human Gastric Cancer Cells

  • Antioxidants either scavenge superoxide and free radicals or stimulate the detoxification mechanisms within cells, resulting in increased detoxification of free radicals formation. Hyperin, isolated from the stem of Uncaria rhynchophylla, prevented oxygen radical formation and inhibited lipid oxidation. The effective concentrations were 31.3 $\mu$M for a radical scavenging assay and 2.2 $\mu$M for a microsome assay. cDNA microarray analysis to determine which genes were modulated by hyperin found that 50 genes were upregulated and 37 genes were downregulated in SNU-668 human gastric cancer cells. Among these genes, thirteen genes that were significantly affected by hyperin were verified by RT-PCR for their effect of genetic reprogramming.

  • In order to investigate the effect of selenium (Se) on the liver damage, metallothionein synthesis and hepatic antioxidative detoxification system in cadmium(Cd) administered rats. Sprague-Dawley male rats(60\\5g) were divided into two diet groups, depending on with (CdS groups) or without (Cd groups) 0.5ppm Se supplementation and fed experimental diets ad libidum for 4 weeks. And then each group was again subdivided into five groups, depending on injection number of Cd, i.e., 0, 1, 2, 3, and 4 times of 2.5mg Cd/kg of body wt once a day. Hemoglobin concentration, hematocrit values, superoxide dismutase, glutathione peroxidase and glutathione S-transferase activite were decreased progressively with increasing number of Cd injection, but increased by the supplementation of Se. The reduced form of glutathione (GSH) contents in blood and liver and vitamin E content were decreased and oxidized form (GSSG) increased in Cd groups, but these of Se supplemented groups were not very different from controls. Cd reduced liver vitamin E content which was not restored by Se supplementation. Liver lipid peroxide values were elevated with increasing doses of Cd, but Se supplementation reduced these elevated levels. Accumulation of metallothionein in liver and kidney was increased with increasing number of Cd injection, but Se did not affect on them. Histological examination revealed that lysosomes were significantly increased and mitochondria and Golgi apparatus were enlarged by Cd, however, these changes were reduced by Se. It was concluded that Se administration promoted antioxidative detoxification and alleviated peroxidative damage in rat liver by Cd.

  • Chemoprevention refers to the use of non-toxic chemical agents to prevent the neoplastic development by inhibiting, delaying, or reversing a multi-stage carcinogenesis. The primary goal of chemoprevention research is to identify or produce effective agents and strategies for clinical trials for applications to normal or high risk human populations. A large number of compounds have been tested for their possible chemopreventive activities, and it is of interest to note that many of them are naturally occurring substances. Thus, a variety of plant and vegetable constituents, particularly those included in our daily diet, have been found to possess substantial protective properties against experimental carcinogenesis. These substances, collectively known as dietary phytochemicals, exert their chemopreventive effects by influencing specific step(s) of multi-stage carcinogenesis: some inhibit metabolic activation or enhance detoxification of carcinogens, others interfere with covalent interactions between ultimate eloctrophilic carcinogens and the target cell DNA and still others may exert anti-promoting or anti-progressing effects. Mechanism-based interventions by use of safe dietary phytochemicals may provide one of the most practical and promising cancer chemopreventive strategies.

Regulation of Ascorbate Peroxidase Gene Expression in Response to Stresses and Phytohormone in Rehmannia glutinosa

  • Ascorbate peroxidase (APX) plays a crucial role in the detoxification of hydrogen peroxide. APX activity is maintained significantly higher in the paraquat­treated leaves of the paraquat-tolerant Rehmannia glutinos. This study was conducted to understand structural and regulatory characteristics of APX gene in R. glutinosa. A putative APX cDNA clone (RgAPX1) was isolated from a leaf cDNA library using a partially sequenced expressed sequence tag clone. RgAPX1 is consisted of 1148 bp nucleotides and contains an open reading frame encoding a 250 amino acid-long polypeptide. Deduced RgAPX1 amino acid sequence shares higher sequence similarity to cytosolic APXs. RgAPX1. expression was higher in the leaf than in the flower and root. Southern blot result indicates the presence of one or two RgAPX1-related genes in R. glutinosa genome. RgAPX1 transcription was affected differentially by various stresses and phytohormone. The results indicate that RgAPXl is constitutively expressed in most tissues and its expression is modulated for the immediate and efficient detoxification of $H_2O_2$ under normal and stress conditions.

항암제 유발 말초신경병증환자와 유기산검사 마커와의 상관성 연구 (Correlation Analysis of Organic Acid Comprehensive Profile Markers with Chemotherapy Induced Peripheral Neuropathy in Cancer Patients)

  • Objectives: The purpose of this study is to evaluate the urinary organic acid comprehensive profile for chemotherapy induced peripheral neuropathy (CIPN). Methods: Participants are 66 patients with CIPN who had symptom (Visual analog scale ${\geq}30mm$, Eastern Cooperative Oncology Group ${\leq}2$). Participants were tested with organic acid comprehensive profile markers. Results: Positive Correlation was observed in the neurotransmitter metabolism markers, N-methyl-D-aspartate (NMDA) modulators markers, detoxification markers, energy production markers, amino acid metabolism markers, and intestinal dysbiosis markers. Especially, all the neurotransmitter metabolism markers were showed positive rate of 44%. In addition, neuro-endo-immune was associated with energy metabolism (mitochondrial dysfunction) in CIPN of cancer patient. especially detoxification, intestinal bacterial hyperplasia, vitamin deficiency (folate, complex B group, vitamin C). Conclusions: Significant urinary organic acid comprehensive profile results were obtained in cancer patients who induced peripheral neuropathy by chemotherapy.

  • Studies on detoxification of Paralytic Shellfish Poison (PSP)-infested oyster, Crassostrea gigas were carried out using available processing resources. Changes of paralytic shellfish toxin components and specific toxicity during canning process were also investigated with high performance liquid chromatography (HPLC). Toxic oysters collected at Hachong in Koje Bay were used for experimental samples. The toxicity of oysters with range of 185-778 ug/100g was reduced below the quarantine limit of 80 ug/100g or not detected level by the mouse bioassay after canning process. The mole $\%$ of toxin components in the shucked oyster was in the order of 25.1 mole $\%$ of gonyautoxin 1, 19.2 mole $\%$ of gonyautoxin 3, 17.2 mole $\%$ of gonyautoxin 4 and 14.6 mole $\%$ of gonyautoxin 2. This sample had tracing amounts of Cl, C2, saxitoxin and neosaxitoxin. In the case of specific toxicity, the major toxins were consisted of gonyautoxin 1-4. The sum of gonyautoxin 1, 2, 3 and 4 was 80% of total toxicity of oyster. Saxitoxin and decarbamoylsaxitoxin were the more thermostable than any other toxin components.

양식산과 방류산 조피볼락(Sebastes schlegeli)의 간장 중 해독 및 항산화 효소계의 비교 (Comparison of Xenobiotic Metabolism and Antioxidative Status in Cultivated and Stocked Rockfish Sebastes schlegeli)

  • This study compared the composition and texture of muscle and levels of detoxification and antioxidant enzymes in the livers of stocked and cultivated rockfish Sebastes schlegeli released after the primary culture stage in Tongyoung, South Korea. The crude lipid content of muscle was significantly higher ($P$<0.05) in cultivated rockfish than stocked rockfish, while the texture did not differ significantly ($P$>0.05). The condition factor (CF) and hepatosomatic index (HSI) did not differ significantly and the growth of stocked and cultivated rockfish was similar. The levels of the detoxification enzymes cytochrome P450 (CYP) and Ethoxyresorufin-O-deethylase (EROD) were significantly lower in the livers of stocked rockfish, perhaps because of their reduced exposure to xenobiotic compounds. In addition, stocked rockfish had a significantly ($P$<0.05) lower CAT and higher GR than cultivated rockfish, but similar levels of tGPx, SOD, GSH, and GSSG. The present study shows that the growth rates of stocked and cultivated rockfish are similar and that stocked rockfish are exposed to fewer xenobiotic compounds and less oxidative stress.
