(+)-Catechin, an Antioxidant Principle from the Leaves of Pinus densiflora that Acts on 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The antioxidant activity of Pinus densiflora was determined by measuring the radical scavenging effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The methanol extract of P. densiflora showed strong antioxidant activity, and thus fractionated with several solvents. The antioxidant activity potential of the individual fraction was in the order of ethyl acetate>n-butanol>water>dichloromethane> n-hexane fraction. The ethyl acetate soluble fraction exhibiting strong antioxidant activity was further purified by repeated silica gel column chromatography. Antioxidant (+)-catechin was isolated as one of the active principles from the ethyl acetate fraction, together with the inactive components, dihydrokaempferol and 1-O-benzoylglucose. The radical scavenging effect of (+)-catechin on DPPH radical exceeded to that of L-ascorbic acid which is a well known antioxidant.

Antioxidative and Macrophage Phagocytic Activities and Functional Component Analyses of Selected Korean Chestnut (Castanea crenata S. et Z.) Cultivars (국내산 밤 일부 품종의 기능성 성분분석과 항산화 및 대식세포 활성)

  • Antioxidative and macrophage phagocytic activities and contents of functional component in selected Korean chestnuts (Dantaek, Daebo, Okkwang, Seokchu, Byunggo) were evaluated. Coumarin, gallic acid and catechin in inner skin and whole kernel of selected Korean chestnuts were detected by HPLC. The predominant functional components in inner skin of chestnut were catechin, followed by gallic acid and coumarin. However, the whole kernel had only gallic acid. Thus, the antioxidant properties of gallic acid and catechin were evaluated through DPPH radical-scavenging activity and SOD like activity. Gallic acid and catechin at 6.0 mg/100 g exhibited 69.4% and 38.3% of scavenging activities on DPPH radical, respectively. DPPH radical-scavenging activity of gallic acid increased in a concentration-dependent manner. Gallic acid was found to possess higher DPPH radical-scavenging activity than equivalent amount of catechin at all concentrations, whereas catechin was found to have higher SOD like activity than gallic acid. In addition, pre-incubation of macrophage with white kernel extract from Byunggo resulted in a significant increase of phagocytic activity and yellow kernel extracts from Byunggo, Dantaek, Daebo and Okkwang, leading to an increase in phagocytic activity compared with untreated cells. Yellow kernel extracts was found to have higher phagocytic activity than white kernel extracts. Byunggo had the highest phagocytic activity. The results suggest that the Korean chestnut may provide a natural source of antioxidants and active immunity.

  • In recent year, We are concerned in anti-aging, disease-protection, long-life, many method are used in solving this problem. Recently, We heard that Luffae Fructus Retinervus(LFR) has effect of anti-aging, disease-protection, long-life. So I let made a experiment for this result. The purpose of this study is to; 1) the anti-oxidant effect of Luffae Fructus Retinervus(LFR) used for 3 methods, those are DPPH radical scavenging activity, Nitric oxide(NO) radical scavenging activity, Superoxide anion radical scavenging activity, 2) cultivation 3T3-L 1 Preadipocytes and Protein chip used for ProteoPlexTM 16-Well Murine Cytokine Array Kit. We measured level of DPPH radical scavenging activity. And we experienced that the ability of DPPH radical's elimination was increased by rising concentration of LFR. When the concentration of LFR was 5 mg/ml, the ability of DPPH radical's elimination was Maximum. We measured level of Nitric oxide(NO) radical scavenging activity. And we founded that the ability of NO radical's elimination was significant when concentration of LFR was from 1.25 mg/ml to 2.5 mg/ml. We measured level of Superoxide anion radical scavenging activity. And we founded that the ability of Superoxide anion radical's elimination was maximum when concentration of LFR was 0.3125 mg. When we inspected Antioxidative Effects with BSA, we experienced that ability of defense was increased by rising concentration of LFR. We known the immunity of LFR about 3T3-L1 Preadipocytes and gained the increase of Cytokines(IL-2, IL-4, GM-CSF) without IL-12p70, $INF-{\gamma}$, $TNF-{\alpha}$ So I guess that Luffae Fructus Retinervus(LFR) has effects of anti-aging, disease-protection, long-life, etc.

DPPH Radical Scavenging Effect and in vitro Lipid Peroxidation Inhibition by Portulaca oleracea (쇠비름(Portulace oleracea) 추출물의 DPPH radical 소거능과 in vitro 지질과산화 억제 효과와 그 활성성분)

  • The antioxidative activity of Portulaca oleracea was tested using in vitro experimental models. Antioxidative activities were determined by measuring DPPH radical scavenging activity and lipid peroxide using 2-thiobarbituric and (TBA). The crude extract was sequentially partitioned with n-hexane, 15% aq. MeOH, EtOAc, n-BuOH, $H_2O$. A remarkable antioxidative effect was observed in the EtOAc and n-BuOH fractions. The DPPH radical scavenging effect ($IC_{50}$=17.90 $\mu\textrm{g}$/ml) of the n-BuOH soluble fraction was comparable with that of the natural antioxidant, $\alpha$-tocopherol ($IC_{50}$=6.99 $\mu\textrm{g}$/ml) and the inhibition effect of lipid peroxidation in mouse liver homogenate was similar to that of the natural antioxidant, L-ascorbic acid at a concentration of 1.0 mg/ml to 5 mg/ml.

Compositions of Astragali Radix and Angelicae Radix by DPPH Radical Scavenging Activity (당귀(當歸)와 황기(黃芪)의 배합 변화가 DPPH 자유기 소거에 미치는 영향 연구)

  • Objectives : This study was conducted to compare antioxidant activity of ratio-related Angelicae Radix and Astragali Radix compositions by DPPH radical scavenging activity. Methods : The species of Angelicae Radix was compared as Angelica gigas, A. sinensis and A. acutiloba. Then various compositions of Angelicae Radix with Astragali Radix were prepared with different ratio. Diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity was measured after 10, 20 and 30 minutes. Each extracts were tested by 1, 5, 10, 50, 100, 500 and 1,000 ${\mu}g/ml$ concentrations. Results : The results showed that Angelica gigas, A. sinensis and A. acutiloba scavenged DPPH radical with 66.8, 61.7 and 56.7% in 1,000 ${\mu}g/ml$ concentration, respectively. According to this result, Angelicae Radix and Astragali Hadix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio were scavenged DPPH radical with 56.3, 49.9, 61.2, 55.7, 54.7, 55.0, 52.8 ,md 50.2% in 1,000 ${\mu}g/ml$ concentration, respectively. The extract of Angelicae Radix and Astragali Radix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio scavenged DPPH radical with the $IC_{50}$ being 85,21, 1016.26, 656.51, 785.67 and 956.74 ${\mu}g/ml$ respectively. Conclusion : In conclusion, the extract of Angelicae Radix and Astragali Radix compositions with 1:1 ratio showed dose-dependent DPPH radical scavenging activity.

Isolation and Identification of Antioxidative Compounds 3,4-Dihydroxybenzoic acid from Black Onion (흑양파로부터 항산화 활성 물질인 3,4-Dihydroxybenzoic acid의 분리 및 동정)

  • The antioxidant substance in black onion was identified. The assays that used 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzthiazo line-6-sulphonic acid) ($ABTS^+$) radicals showed that the ethyl acetate fraction of black onion methanol extract had a higher level of radical-scavenging activity than the other fractions. Two antioxidative compounds were purified and isolated from the ethyl acetate fraction via column chromatographies of silica gel and Sephadex LH-20 using the guided DPPH radical-scavenging assay. The isolated compounds were identified as 3,4-dihydroxybenzoic acid (1) and quercetin (2) based on mass spectrometry and nuclear magnetic resonance. The isolated compounds showed a high level of DPPH and ABTS+ radical-scavenging activity. Compound 2 had a higher level of radical-scavenging activity than 1.

  • Objective : This study was undertaken to determine the inhibitory effect of Whallak-tang against Nitric Oxide, the scavenging effect of Whallak-tang on DPPH radical. Methods : Both nitric concentration 1, 3, 6 and 24 hours after treatment with Vit. C and Whallak-tang at 0.1, 1, 10 mg/ml and scavenging effect 10, 20, 30 and 60 minute after treatment with Vit. C and Whallak-tang at 0.1, 1, 10 mg/ml were estimated. Results : The inhibitory effect of Whallak-tang against Nitric Oxide was dose-dependent. The scavenging effect of Whallak-tang on DPPH radical was dose-dependent. Conclusion : These results would provide important basic data on the possibility of the clinical treatment of Whallak-tang in musculoskeletal disease.

  • The antioxidant activity of Desmodium oldhami (Leguminosae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of D. oldhami showed the strong antioxidant activity. Five compounds, kaempferol-3-O-rhamnopyranoside (afzeline) (1), quercetin-3-O-[$\alpha$-L-rhamnopyranosyl($1{\rightarrow}6$)-$\beta$-Dglucopyranoside] (rutin) (2), kaempferol-3-O-glucopyranoside (astragalin) (3), genistein-7-O-$\beta$-D-glucopyranoside (genistin) (4), kaempferol-3-O-rutinoside (5) were isolated from the active ethyl acetate soluble fraction of D. oldhami through repeated silica gel and Sephadex LH-20 column chromatography. Among them, compound 2 showed the most significant antioxidative effect on DPPH free radical scavenging test. Compounds 1-5 are reported for the first time from this plant.

Antioxidant Activity of Partially Purified Extracts Isolated from Bacillus polyfermenticus SCD Culture

  • The antioxidant activity of Bacillus polyfermenticus SCD was studied by partially purified culture extracts using various methods: ammonium sulfate precipitation, adsorption to Diaion HP-20 columns using polar solvents, and extraction using non-polar solvents. The 1,1-diphenyl-2-picyrylhydrazyl (DPPH) radical scavenging activity of these partially purified fractions was then investigated. The precipitate isolated using 75%-saturated ammonium sulfate was shown to contain about 77.2% DPPH radical scavenging activity. Using the Diaion HP-20 resin adsorption method, the fraction obtained using 60% ethanol and 60% methanol possessed 76.7 and 89.5% DPPH radical scavenging activity, respectively. Fractions obtained by extracting with the non-polar solvents 80 mg/mL chloroform, 80 mg/mL n-hexane, 80 mg/mL ethyl acetate, and 80 mg/mL butanol contained 68.4, 75.0, 70.7, and 87.5% DPPH radical scavenging activity, respectively. Further study is needed to characterize the antioxidant substance(s) released by B. polyfermenticus SCD cultures.

Inhibitory Effects of Methanol Extract, Phenolic Acids and Flavonoids from the Leaves of Eucalyptus darylmpleana against 1,1-Diphenyl-2-picrylhydrazyl Radical

  • The inhibitory effects of the leaves of Eucalyptus darylmpleana (Myrtaceae) on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical was examined. The scavenging effect of the ethyl acetate fraction of Eucalyptus darylmpleana leaves on DPPH radical was stronger than the other fractions, and further purified by silica gel and Sephadex LH-20 column chromatography. 3,4-Dihydroxybenzoic acid, gallic acid, quercetin, quercetin $3-O-{\alpha}-_L-rhamnoside$, quercetin $3-O-{\beta}-_D-glucoside$ and quercetin 3-O-rutinoside were isolated and elucidated by spectroscopic data. Among these components, gallic acid and quercetin $3-O-{\alpha}-_L-rhamnoside$ exhibited potent scavenging activities on DPPH radical with $IC_{50}$ values of 6.02 and $5.54\;{\mu}M$, respectively.
