Selection of Cryoprotectant for the Cryopreservation of Trochophores and Early D-shaped Larvae of Surf Clam, Spisula sachalinensis (북방대합, Spisula sachalinensis 담륜자와 초기 D상 유생의 냉동보존을 위한 동해방지제의 선택)

  • This study was performed to find out the cryoprotectants for cryopreservation of trochophores and early D-shaped larvae of surf clam, Spisula sachalinensis. Dimethyl sulfoxide (DMSO), ethylene glycol, 1,2-propanediol and methanol were used as cryoprotectant Each cryoprotectant was made to 1.0 M, 2.0 M, 3.0 M with dilution of 0.2 M fructose and 0.2 M sucrose. The trochophoers and early D-shaped larvae were immersed in each preparation for 10 minutes to reach equilibration and cryopreserved in liquid nitrogen. Survival rates of post-thawed trochophores and early D-shaped larvae in 2.0 M DMSO with 0.2 M sucrose were the highest as $91.4{\%}\;and\;78.9{\%}$, respectively.

  • The metamorphosis and the growth of larva in Limnoperna fortunei were studied from October, 1981 to December, 1983. 1) The larva of this species is observed in the three different stages in the form of D-shaped. The shell length of the larva in the early stage is $130.44{\mu}m$ to $143.60{\mu}m$ and in the middle stage $161.67{\mu}m$ to $184.11{\mu}m$. In the late stage the length size of the larva increases up to $194.55{\mu}m$ to $208.45{\mu}m$. The shell height is $103.19{\mu}m$, to $119.47{\mu}m$, $126.51{\mu}m$ to $157.63{\mu}m$ and $136.87{\mu}m$ to $176.35{\mu}m$ in the three stages respectively. Thus the growth ratio of shell length to shell height is 1:0.75. 2) The metamorphosing stage begins when the shell height becomes larger than $210{\mu}m$ and the shell length exceedes $260{\mu}m$.

Development and Growth of Larvae of Four Bivalve Species (4종 조개류 유생의 발생과 성장)

  • Exposure to air and increased temperature induced successful spawning in Mytilus edulis, M. coruscus Crassostrea gigas and Pinctata fucata martensii. Developmental durations required for an egg to attain D-shaped larva and the D-shaped larva to reach pediveliger stage were estimated in these bivalves. Size of fertilized eggs was the largest (70.3 ${\mu}m$) in M. coruscus and the smallest (45.3 ${\mu}m$) in P. fucata martensii. At 17$^{\circ}C$, M. edulis and M. coruscus attained D-shaped larval stage within 48 hours after fertilization but those of C. gigas and B. fucata martensiii within 24 and 22 hours at 21 and 26$^{\circ}C$, respectively. The development duration required for a D-shaped larva to attain pediveliger stage was the longest (27 days) in M. coruscus and ranged between 20 and 22 days for the others. The shell length of the pediveliger was the longest (274.9 ${\mu}m$) in C. gigas and smallest (190.9 ${\mu}m$) in P. fucata martensii Length and height of larval shell was highly correlated with each other in all the 4 species. The shell height of C. gigas was more than the shell length beyond the size of 100 ${\mu}m$ shell length. However, shell length of the others was always longer than shell height at the larval stage.

Egg Development and Lana Growth of the Scallop, Patinopecten yessoensis (가리비, Patinopecten yessoensis 난 발생과 유생의 성장)

  • In order to obtain the basic information for seedling production of the scallop, Patinopecten yessoensis, the egg development and larva growth were investigated at different conditions such as water temperature, salinity and phytoplankton. Eggs were demersal isolated eggs, which averaged 77.3${\pm}$2.7$\mu\textrm{m}$ in diameter after spawning. The fertilized eggs developed to D-shaped larva of shell length 117.5${\pm}$3.8$\mu\textrm{m}$ after 60 hours at 15$^{\circ}C$. The range of water temperature and salinity during egg development were 10-20$^{\circ}C$ and 28-34 ppt, respectively. The time of egg development was shorter with higher water temperature. After 10 days of spawning, D-shaped larva reached 160 $\mu\textrm{m}$ in shell length, and after 25 days became full-grown larva 250 $\mu\textrm{m}$ in shell length, in which could be observed eye spots. The relative growth formula between shell length (SL) and shell height (SH) was SH = 1.0425SL-27.731 (r$^2$= 0.9749) during the entire larva period. In regard to water temperature, growth and survival rates of larvae were good at 16$^{\circ}C$. Lower growth and survival rates were observed at 12$^{\circ}C$ and 20$^{\circ}C$ than that at 16$^{\circ}C$. When larvae were fed mixed phytoplankters, such as isochrysis galbana, Pavlova lutheri and Chaetoceros calcitrans, their growth and survival rates were the highest among groups.

Effect of Temperature, Salinity and Density on the Egg Development of the Sunray Surf Clam, Mactra chinensis (개량조개, Mactra chinensis의 난발생에 미치는 수온, 염분 및 수용밀도의 영향)

  • Water temperature, salinity and density on egg development of the sunray surf clam, Mactra chinensis were investigated for artificial seedling production. The required time from fertilization to D-shaped larvae were 33.8 hours in $18^{\circ}C$, 20.6 hours in $23^{\circ}C$, 18.2 hours in $28^{\circ}C$ and 15.0 hours in $33^{\circ}C$. The development duration was reduced with increasing temperature. The relationships between temperature and the required time from egg to each developmental stage were described as follows: 2-cell, 1/h = 0.1051WT - 1.4782; 8-cell, 1/h = 0.037WT - 0.3686; gastrula, 1/h = 0.008WT - 0.0521; trochophore, 1/h = 0.0041WT - 0.0235; D-shaped larva, 1/h = 0.0024WT - 0.0102. Biological minimum temperature for the egg development was estimated to be $8.0^{\circ}C$ in average. The possible range of temperature for the development of D-shaped larvae was $18-28^{\circ}C$ and optimum of water temperature for the development of egg was $23^{\circ}C$. The possible range of salinity for the development of D-shaped larvae was 20-35 psu and optimum of salinity for the development of egg was 30-35 psu over 25 psu at least. The density of fertilized egg was below 40 per 1 ml in rearing seawater for elevating the development rate from fertilized egg to D-shaped larva.

Effects of Various Stimulants on Spawning Induction and Early Development at Different Water Temperatures in the Noble Scallop (흔한가리비, Chlamys nobilis의 자극방법별 산란유발 효과와 난 발생에 미치는 수온의 영향)

  • We examined the effects of stimulants including sunlight and UV-irradiation on the spawning induction and early development of the noble scallop, Chlamys nobilis. The sunlight stimulation resulted in nuch faster spawning induction (100% success within 40 minutes) compared to UV-irradiation (100% success within 70 minutes). Early development of the scallop larva took place between 15$^{\circ}C$ to 3$0^{\circ}C$. The time to reach the early D-shaped stage was 63.5, 31.5, 18.5 and 17.0 hours at 15, 20, 25 and 3$0^{\circ}C$, respectively. The correlations between the water temperature-(WT) regimes and the time (t) required for each developmental stage are as follows. 2 cell stage: 1/t=0.0606WT-0.6194 ($r^2$=0.9791) 8 cell stage: 1/t=0.0304WT-0.3453 ($r^2$=0.9941) Morula: 1/t=0.0100WT-0.1049 ($r^2$=0.9663) Trochophore: 1/t=0.0058WT-0.0618 ($r^2$=0.9848) D-shaped larva: 1/t=0.0030WT-0.0282 ($r^2$=0.9731) These correlations indicated that the biological minimum temperature of the species is around 10.44$^{\circ}C$. The highest survival rate up to D-shaped larva at different water temperature was observed at $25^{\circ}C$.

Survival Rates of Frozen-thawed Surf Clam, Spisula sachalinensis Larvae in Five Developmental Stages (북방대합, Spisula sachalinesisr 유생의 발생단계별 냉동-해동후 생존율)

  • This study was peformed to find out the optimum larval stage among trochophore, early Dshaped larva, late D-shaped larva, early umbo and late umbo stages for cryopreservation of surf clam, Spisula sachalinensis larvae. Dimethyl sulfoxide (DMSO)and ethylene glycol were used as cryoprotectant, The larvae were immersed to cryoprotectants for 10 minutes and thereafter, cryopreserved in liquid nitrogen. Survival rates of trochophores frozen-thawed in 2.0 M dimethyl sulfoxide and 2.0 M ethylene glycol were the highest as 97.4% and 85.0%, respectively and post-thaw survival rates were decreased with the larval growth.

Study on Spawning Induction and Larvae Breeding of the Hard Clam, Meretrix petechiails (Lamarck) (말백합, Meretrix petechiails (Lamarck) 의 산란유발 및 유생사육에 관한 연구)

  • For industrialization of the hard clams, Meretrix petechiails (Lamarck), spawning was induced per spawning induction technique in the artificial maturation group administered of parent maturation control and the natural maturation group of which parents were transported for artificial spawning per time period. Then, fertilization rates, hatching rates and D-shaped larva development rates were investigated. In addition, growth and survival rates of larvae were investigated per larva breeding technique. The results of spawning induction by exposure in the artificial maturation group indicated that response rates were relatively higher at 23% and 32% respectively at the 4th hour and the 8th hour of exposure. In terms of water temperature increase, responses began only when the temperature reached $28^{\circ}C$ or higher. In the experiment group administered with both exposure and water temperature increase techniques, response rate was found to be 45% or higher at the 4th hour of exposure and the temperature of $28^{\circ}C$. At the temperatures of 29, 30 and $31^{\circ}C$, significant differences were not observed. Therefore, it was indicated that the response rates of parent hard clams were higher toward water temperature increase than exposure time. As for spawning induction per time period of the transported parent group, response rate and D-shaped larva development rate were the highest at 67.6% and 96% respectively on August 6, 2009. In terms of water temperatures during larva breeding experiment, growth was faster as water temperature was higher. In addition, growth and survival rates were relatively higher at the salinity of 25. In terms of stocking density, growth and survival rates were relatively higher at 5 inds./mL.

  • The investigated amounts according to microalgae for stable supply of artificial seed of the hard clam, Meretrix petechialis and also observed the effect according to the repower of the microalgae after the specified period starvation. The stage of specimen used in the test was the D-shaped larva. The microalgae was Chaetoceros calcitrans, Isochrysis galbana and Nannochloris oculata. When the mixture of Chaetoceros calcitrans, Isochrysis galbana and Nannochloris oculata or alone C. calcitrans as food was supplied with 3,000-5,000 cells/ind., it turned to be the most effective. When the food was provided after starvation for some period, the shell length of D-shaped larva was grown to over $192.5{\mu}m$ at fourth day from the initial feeding. The survival rate tended to be lower, the longer the starvation period.

  • To know on the distribution and ecological characteristics of genus Corbicula field survey made from August 1997 to July 1998 in the North Kyungsang Province. Three species, i.e. Corbicula japonica, C. leand and C. papyracea, were inhabited in the North Kyungsang Provice. C. japonica was dioecious and oviparous species and C. leana and C. papyracea were monoecious and viviparous species.
