Comparison of Dry Medium Culture Plates for Mesophilic Aerobic Bacteria in Milk, Ice Cream, Ham, and Codfish Fillet Products

  • This study was performed to compare the performance of Sanita-Kun dry medium culture plate with those of traditional culture medium and Petrifilm dry medium culture plate for the enumeration of the mesophilic aerobic bacteria in milk, ice cream, ham, and codfish fillet. Mesophilic aerobic bacteria were comparatively evaluated in milk, ice cream, ham, and codfish fillet using Sanita-Kun aerobic count (SAC), Petrifilm aerobic count (PAC), and traditional plate count agar (PCA) media. According to the results, all methods showed high correlations of 0.989~1.000 and no significant differences were observed for enumerating the mesophilic aerobic bacteria in the tested food products. SAC method was easier to perform and count colonies efficiently as compared to the PCA and PAC methods. Therefore, we concluded that the SAC method offers an acceptable alternative to the PCA and PAC methods for counting the mesophilic aerobic bacteria in milk, ice cream, ham, and codfish fillet products.

A Study on Antibacterial Activity of Natural Material Treated Cotton Fabric (천연물 가공 면포의 항균성 연구)

  • Water-insoluble chitosan with molecular weight of 2,000,000, 500,000, 80,000, and 40,000 and more than 90% of degree of deacetylation were used to test antibacterial activity of chitosan against a pathogenic bacteria, methicillin resistant Staphylococcus aureus(MRSA), which is being issued in the world. As experimental method, Agar plate Smear Method and Agar plate Contact Method were used. The moleculur weight of chitosan didn't exert significant influences on its antibacterial activity against MRSA but chitosan having molecular weight 40,000, 80,000 and 150,000 showed the excellent antibacterial activity. The antibacterial efHciency was excellent in applying it after chitosan was dissolved in acetic acid solution, while the antibacterial efficiency was not expressed nearly in case of applying after chitosan was dissolved in neutral water. Therefore, it is considered that chitosan can show the antibacterial efficiency only if a positive ion status of -NH₃/sup +/ is maintained. MIC of chitosan/acetic acid solution and cotton fabrics finished with chitosan/acetic acid solution showed in concentration of 0.05%.

Lateral performance of CRCS connections with tube plate

  • This paper presents experimental and analytical studies to evaluate the cyclic behaviour of Circular Reinforced Concrete column Steel beam (CRCS) connections. Two 3/4-scale CRCS specimens are tested under quasi-static reversed cyclic loading. Specimens were strengthened with a tube plate (TP) and a steel doubler plate (SDP). Furthermore; nine interior beam-through type RCS connections are simulated using nonlinear three-dimensional finite element method using ABAQUS software and are verified with experimental results. The results revealed that using the TP improves the performance of the panel zone by providing better confinement to the concrete. Utilizing the TP at the panel zone may absorb and distribute stress in this region. Results demonstrate that TP can be used instead of SDP. Test records indicate that specimens with TP, with and without SDP maintained their maximum strength up to 4% drift angle, satisfying the recommendation given by AISC341-2016 for composite special moment-resisting frames.

Effectiveness of Nanocrystalline Silver(Acticoat®) Dressing at Wound Infected by Multidrug Resistant Bacteria (다제내성(多劑耐性)을 지닌 균주에 감염된 상처에서 Nanocrystalline Silver(Acticoat®) 드레싱의 효용)

  • Purpose: The emergency of multi-drug resistant stains of bacteria represents a challenge in the field of plastic surgery. Especially, MRSA(methycillin-resistant Staphylococcus aureus) and Pseudomonas aeruginosa have strong pathogenicity as well as multi-drug resistance so that they have become a lot more problematic strains. This study has been planned to reduce the bacterial burden by applying $Acticoat^{(R)}$(Smith & Nephew Healthcare, Hull, England)dressing into the chronic wounds infected by multi-drug resistant strains and to facilitate their healing. Methods: Nanocrystalline silver dressings($Acticoat^{(R)}$) were applied to chronic wound infected by MRSA or Pseudomonas aeruginosa. Multi-drug resistant bacteria were smeared over a slide glass using sterilized cotton swabs and gram stains were performed directly before and after applying $Acticoat^{(R)}$ dressings at 1, 24, 48 and 72 hours. The gram-stained slides were observed using an optical microscope magnified 1000 times(${\times}1000$). The bacterial counts of the control group(0 hour) were compared to those of the experimental groups(1, 24, 48, and 72 hour). Paired T-test was used to assess a statistical significance. MRSA was cultured in two BAPs(blood agar plate) and two MacConkey plates with streak plate method. None were interventions on one culture plate, while on the other culture plate, $Acticoat^{(R)}$ was placed in a square shape and cultured for 72 hours at $37^{\circ}C$, then plates were examined. Pseudomonas aeruginosa was cultured in the same manner as MRSA. Results: There are the large amount of declination of bacterial counts with statistical significance after $Acticoat^{(R)}$ dressing. The bacteria grew in culture plate without specific intervention, but no bacteria grew in culture plate with applying of $Acticoat^{(R)}$ dressing. Conclusion: We believe that $Acticoat^{(R)}$ dressing could be used as an effective method of treating chronic wounds which are infected by multi-drug resistant organisms.

A low computational cost method for vibration analysis of rectangular plates subjected to moving sprung masses

  • A low computational cost semi-analytical method is developed, based on eigenfunction expansion, to study the vibration of rectangular plates subjected to a series of moving sprung masses, representing a bridge deck under multiple vehicle or train moving loads. The dynamic effects of the suspension system are taken into account by using flexible connections between the moving masses and the base structure. The accuracy of the proposed method in predicting the dynamic response of a rectangular plate subjected to a series of moving sprung masses is demonstrated compared to the conventional rigid moving mass models. It is shown that the proposed method can considerably improve the computational efficiency of the conventional methods by eliminating a large number of time-varying components in the coupled Ordinary Differential Equations (ODEs) matrices. The dynamic behaviour of the system is then investigated by performing a comprehensive parametric study on the Dynamic Amplification Factor (DAF) of the moving loads using different design parameters. The results indicate that ignoring the flexibility of the suspension system in both moving force and moving mass models may lead to substantially underestimated DAF predictions and therefore unsafe design solutions. This highlights the significance of taking into account the stiffness of the suspension system for accurate estimation of the plate maximum dynamic response in practical applications.

A Study on the Microbial Quality Control of Chicken Meat Salad by Adding Green Tea Extracts in Foodservice Operations (급식소에서 생산되는 닭고기 샐러드의 녹차추출물 첨가에 따른 미생물적 품질 평가)

  • This study was aimed to determine microbiological quality by adding green tea extracts to chicken meat salad. For this study, Chicken meat salad were prepared with two production method. (method 1: addition of green tea extracts to boiling phase, method 2: addition of green tea extracts to salad dressing) Microbiological effects of green tea extracts were assessed during production process by measuring process time, temperature, pH and Aw and determining total plate counts and coliforms. Effects of green tea extracts on total plate counts and coliforms were observed during holding at 3, 10, $25{\pm}1^{\circ}C$ for 12 hours. Green tea extracts improved the microbiological quality and showed antibacterial properties when they are added to chicken meat salad.

Nonlinear vibration of hybrid composite plates on elastic foundations

  • In this paper, nonlinear partial differential equations of motion for a hybrid composite plate subjected to initial stresses on elastic foundations are established to investigate its nonlinear vibration behavior. Pasternak foundation and Winkler foundations are used to represent the plate-foundation interaction. The initial stress is taken to be a combination of pure bending stress plus an extensional stress in the example problems. The governing equations of motion are reduced to the time-dependent ordinary differential equations by the Galerkin's method. Then, the Runge-Kutta method is used to evaluate the nonlinear vibration frequency and frequency ratio of hybrid composite plates. The nonlinear vibration behavior is affected by foundation stiffness, initial stress, vibration amplitude and the thickness ratio of layer. The effects of various parameters on the nonlinear vibration of hybrid laminated plate are investigated and discussed.

  • In this work, an improved semi-analytical technique is adopted to track the dynamic response of thin rectangular plates excited by sequential traveling masses. This technique exploits a so-called indirect definition of inertial interaction between the moving masses and the plate and leads to a reduction, in the equations of motion, of the number of time-varying coefficients linked to the changing position of the masses. By employing this optimized method, the resonance of the plate can be obtained according to a parametric study of relevant maximum dynamic amplification factor. For the case of evenly spaced, equal masses travelling along a straight line, the resonance velocity of the masses themselves is also approximately predicted via a fast methodology based on the fundamental frequency of the system only.

  • To investigate the antibiotic constituents of Korean basidiomycetes the carpophores of the wood-rotting basidiomycetes were collected from several locations of Korea, and from them 17 mycelial strains were isolated on potato-dextrose-agar plates supplemented with tetracycline ($20\;{\mu}g/m{\ell}$. The isolated strains were shake-cultured in glucosepeptone-yeast extract medium and then the antibacterial activities of the culture filtrates were assessed by disc-plate method. Among them, 12 strains (70.6%) were active, and basidiomycete strain LMCB-109 (Daedalea quercina) and LMCB-116 showed potent activities against all the six bacterial target organisms including Serratia marcescens.
