Identification of the Anopheles Mosquitoes(Diptera: Culicidae) of Southern Iran Using Analysis of Cuticular Hydrocarbons

  • Cuticular hydrocarbons(CHCs) of the epicuticle wax layer are so far used to differentiate the insects in species and subspecies levels. In this study, four species of malaria vectors(genus Anopheles) were collected from various localities in southern Iran. Twenty specimens of each species were randomly selected and one epicuticular extract was prepared of every five specimens. FID-GC profiles of the extracts did not show any qualitative difference. Using significant difference of CHC mass at retention time(RT) 39.6, the two species of An. sacharovi and An. fluviatilis could be distinguished. Similarly, the two species of An. superpictus & An. sacharovi and An. dthali & An. sacharovi were differentiated by their CHC level at RT 28.5. An. sacharovi was distinguished by integratable peaks at RTs 29.7, 30.6, 30.7, 31 and 32.6 while the other three species just indicated trace peaks at the same RTs. Similarly, An. dthali could be known by an integratable peak at RT 26.2 while An. fluviatilis and An. superpictus indicated trace peaks at the same RT. Integratable peaks and traces at RTs 27.4 and 28.5 were respectively used to differentiate An. superpictus from An. fluviatilis. Lastly, CHC trace amount of An. superpictus at RT 39.6 is another indicator to distinguish it from An. fluviatilis with an integratable peak at the same RT. In harmony with other studies worldwide we hereby report that quantitative analysis of CHCs was successfully applied to differentiate the four Anopheles species of southern Iran.

Evaluation of Aquatic Animals on the Water in a Rice Field with No-tillage Rice Cover Crop Cropping Systems (무경운 피복작물 작부체계에서 논물의 미소동물 평가)

  • The objectives of the present study evaluated aquatic animals on the water in a rice field. Field investigation was carried out in conventional tillage without rice straw or green manure crop treatment (CTFS, check plot), no-tillage without cover crops (NTNT), no-tillage amended with rape (NTRA), no-tillage amended with rye (NTRY), no-tillage amended with hairyvetch (NTHV), and no-tillage amended with Chinese milk vetch (NTCM). Total dense population of aquatic animals in HTHV was significantly higher than the other plots (p<0.05) on May 30. Dense populations of Daphniidae and Culicidae on June 20 were lowest in CTFS compared to no-tillage plots (p<0.05). Furthermore, in principal component analysis (PCA), PC1 explained 44.9% of variance, whereas PC2 explained 26%, for a cumulative total of 70.9% and the PC1 of the PCA separated the samples from NT treatments and CFS (p<0.05).

  • A dispersal experiment on Culex tritaeniorhynchus females by a mark-release-recapture method was carried out at Pohgang city, Korea, in Augst 1974. The results are summarized as follows: 1. Of 23,115 females of C. tritaeniorhynchus released, 120 females were recaptured, giving 0.519% recapture rate and 0.00208 recovery ration. 2. Dispersal rates from the release site were 77.5% in 0-2km, 11.7% in 4-6 km and 5.8% in 6-8km. 3. C. tritaeniorhynchus females flew 7.5km in one day. The average of the daily mean distances was 1.55km, and it showed that the dispersal distance did not correlate with days after release.

Isolation and Activity of Insect Pathogenic Bacillus thuringiensis Strain from Soil (국내 토양으로부터 곤충병원성 세균인 Bacillus thuringiensis 균주의 분리 및 생물검정)

  • Bacillus thuringiensis strains were isolated from the domestic soil and a strain was selected that had a new host range and high toxicity against agriculture insect pest. The 142 samples of soil were sampled from the mountains, paddy fields and patches, in Daejon, Chungnam, Chungbuk and Jeonbuk and used for the investigation. Sixteen B. t strains were isolated from 12 samples among collected samples. There were 11 strains that showed toxical activity on Plutella xylostella (Lepidoptera: Yponomeutidae), 7 steins on Spodoptera litura (Lepidoptera: Noctuidae), 5 strains on Arete coerulea (Lepidoptera: Noctuidae), 5 strains on Culex pipiens pallens (Diptera: Culicidae) among the 16 isolated B. t strains. But there were not any strains that showed activity against Hyphanria cunea (Lepidoptera: Arctiidae) and Sitophilus oryzae (Coleoptera: Rhynchophoridae). And also some of B. thuringiensis strains showed insecticidal activity with 2, 3 or 4 kinds of insects. But there were also 3 strains that did not show any activities to the 6 insects which were used in the experiment. When examined with a phase-contrast microscope, the insecticidal crystal protein produced from 16 selected strains had 13 bipyramidal and 3 spherical shapes. The insecticidal bioactivity of the S. litura showed 100% mortality when there were $1.3{\times}10^{7}\;(cfu/ml)$ of CAB109 isolates.

  • Four mosquito repellents, product A (aerosol), product B (liquid), product C (aerosol) and product D (lotion) were tested for their persistent repellency and quantity of DEET remained with the lapse was analyzed. In shirt term test with human (or 8 hours, product A (aerosol), product B (liquid), product C (aerosol) and product D (lotion) appeared over 95% repellency for eight, five, six and six hours. respectively. In long-term tests with shirt piece for 16 days, product A (aerosol) and product C (aerosol) showed 100% repellency for 10 days. In the assay of quantity of DEET remained in shirt pieces with the lapse of time, two products was decreased to the same pattern.

Comparative Efficacy of Mosquito Repellents against Aedes albopictus (Diptera: Culicidae) (흰줄숲모기에 대한 모기기피제의 효과비교)

  • We investigated the repellency of eight mosquito repellents against Culex pipiens pallens and Aedes albopictus, and the quantity of DEETs were analysed by using gas chromatography. Ae. albopictus showed higher bloodsucking efficacy than Cx. pipiens pallens of the two subjects mouse and human hands, and human hand was more efficacious method for mosquito bloodsucking efficacy than mouse. Among the eight repellents were tested with mouse as subjects for eight hours, product C, G, and H were high repellency than others. In the case of human hands as subject, product C was highly effective than above other products. The DEET quantities of eight repellants varied from 7.31 g to 38.9 g in 100 g contents and we ascertained that there was a correlation between mosquito repellency and the DEET quantities remained after the treatments. It was evidenced from the long term test with shirt piece, in this test the Ae. albopictus attack was increased when the DEET quantity decreased below 40%.

Effects of Heavy Metals on Mortality and Metabolite Changes in Mosquitoes, Culex pipiens pallens (Diptera: Culicidae) (중금속이 빨간집모기(Culex pipiens)의 치사율과 대사물질의 변화에 미치는 영향)

  • To investigate the effect of heavy metals (Cu, Cd, Hg, Pb, Zn) on mosquito Culex pipiens, 50% lethal concentration ($LC_{50}$) concentration, total lipid content, lipid content composition, and total protein content were measured. The results showed that the Hg $LC_{50}$ was $0.45mg\;kg^{-1}$ and the Hg toxicity was higher than the other metals. The results also showed that cadmium (Cd) significantly retarded the growth of mosquito larvae among the tested heavy metals. Six types of lipid bands were isolated from mosquito samples exposed to heavy metals, and five of them were identified (phospholipid, cholesterol, fatty acid, triglyceride, and cholesterol ester). The total lipid content of all treatments decreased compared to the control. In addition, the protein content of the control group ($0.51mg\;ind.^{-1}$) was higher than that of the treated group, and the protein content of the larvae treated with Hg and Cd was very low. In conclusion, heavy metals not only lower the survival rate of mosquitoes, but also affect the protein content and lipids in the mosquito's body, thereby causing growth inhibition. These results indicate that heavy metal contamination will affect the size of mosquito population by inhibiting the survival and growth of mosquitoes, and it is expected that it can be used to measure the degree of contamination using mosquito through subsequent studies.

Monitoring Culicine Mosquitoes (Diptera: Culicidae) as a Vector of Flavivirus in Incheon Metropolitan City and Hwaseong-Si, Gyeonggi-Do, Korea, during 2019

  • The flaviviruses are small single-stranded RNA viruses that are typically transmitted by mosquitoes or tick vectors and are etiological agents of acute zoonotic infections. The viruses are found around the world and account for significant cases of human diseases. We investigated population of culicine mosquitoes in central region of Korean Peninsula, Incheon Metropolitan City and Hwaseong-si. Aedes vexans nipponii was the most frequently collected mosquitoes (56.5%), followed by Ochlerotatus dorsalis (23.6%), Anopheles spp. (10.9%), and Culex pipiens complex (5.9%). In rural regions of Hwaseong, Aedes vexans nipponii was the highest population (62.9%), followed by Ochlerotatus dorsalis (23.9%) and Anopheles spp. (12.0%). In another rural region of Incheon (habitat of migratory birds), Culex pipiens complex was the highest population (31.4%), followed by Ochlerotatus dorsalis (30.5%), and Aedes vexans vexans (27.5%). Culex pipiens complex was the predominant species in the urban region (84.7%). Culicine mosquitoes were identified at the species level, pooled up to 30 mosquitoes each, and tested for flaviviral RNA using the SYBR Green-based RT-PCR and confirmed by cDNA sequencing. Three of the assayed 2,683 pools (989 pools without Anopheles spp.) were positive for Culex flaviviruses, an insect-specific virus, from Culex pipiens pallens collected at the habitats for migratory birds in Incheon. The maximum likelihood estimation (the estimated number) for Culex pipiens pallens positive for Culex flavivirus was 25. Although viruses responsible for mosquito-borne diseases were not identified, we encourage intensified monitoring and long-term surveillance of both vector and viruses in the interest of global public health.

Occurrence of Culex pipiens (Diptera, Culicidae) and Effect of Vent Net Sets for Mosquito Control at Septic Tanks in South-eastern Area of the Korean Peninsula (한반도 동남부 지역 정화조 내의 빨간집모기군(群)(Culex pipiens complex) 발생과 정화조 환기통의 방충망 설치에 의한 방제효과)

  • Culex pipiens complex larvae were found at the septic tanks in Busan and Ulsan cities located in South-eastern area of Korean peninsula. The larvae were collected in 25, 35 and 75 septic tanks out of 3,527, 622 and 1,111 septic tanks which the breeding rates were averages of 0.7%, 5.6% and 6.8% in Busan from January to May, 2003; Busan from February to March, 2004; and Ulsan from January to April, 2005, respectively. The average numbers of the mosquitoes in the tanks were 38.8, 14.9 and 13.7 larvae/dip with a 355-ml dipper. The larval densities were from 25.4 larvae/dip in January to 43.7 larvae/dip in April. 2003 which were not significantly different between them. It was the first report to find mosquito breeding at septic tink in Republic of Korea. Culex pipiens larvae were found in 6 out of 379 septic tanks at complex buildings (over $2,000m^2$ in size) in Ulsan from June to September, 2005, having an average of 1.6% for mosquito breeding rate. The mosquito larvae were collected in 8 and 530 septic tanks out of 210 and 3,729 septic tanks at large (over 300 homes) and small apartments (below 300 homes) with 3.8% and 14.2% of the average breeding rates, respectively At the septic tanks of private houses for below 50 persons, 708 out of 7,178 septic tan had Cx. pipiens larvae and pupae with 9.9% of the average breeding rate. Total average of the mosquito breeding rate was 10.9% of the investigated septic tanks of residential buildings in Ulsan. The number of mosquito larvae in the septic tanks were 15 to 75 larvae/dip. The bent net sets of septic tanks prevented from mosquito breeding to the rate of 93.1%.
