The buffalo (Bubalus bubalis) is an important contributor to milk, meat, power, fuel and leather production in many developing countries. Buffaloes can be categorized into Asian and Mediterranean buffaloes. Asian buffalo includes two subspecies known as Riverine and Swamp types. Riverine (water buffalo) and Swamp buffaloes possess different genetics (50 vs. 48 chromosomes, respectively), morphology (body frame, body weight, horn shape and skin color) and behavior (wallowing in mud or water) and thus, are reared and used for different purposes. Low per head milk yield, poor reproductive performance (seasonal breeding behavior, anestrous, and longer calving interval) and low growth rate in buffaloes have been attributed to insufficient supply of nutrients. In many parts of Asia, where the buffalo is an integral part of the food chain and rural economy, irregular and inadequate availability of quality feedstuffs and their utilization are hampering the performance of this unique animal. Balanced nutrition and better management can enhance buffalo productivity. Many efforts have been made in the last few decades to improve nutrient supply and utilization in buffaloes. Recent research on locally available feed resources such as crop residues, and industrial by-products, dietary addition of micronutrients, use of performance modifiers and use of ruminally protected fat and protein sources have shown significant potential to improve growth, milk yield and reproductive performance of buffaloes. However, a number of issues, including establishment of nutrient requirements for dairy and beef, development of buffalo calf feeding systems, nutritional management of metabolic and reproductive anomalies, and understanding and exploitation of the buffalo gut ecosystem, need to be addressed. Extensive coordinated research and extension efforts are required for improved buffalo nutrition in developing countries.
A cDNA clone encoding phytocystatin was isolated from Brassica rapa seedlings, through rapid amplification of cDNA ends (RACE). This gene (name as Brcpi1; GenBank accession no.: EF079953) had a total length of 881 bp with an open reading frame of 609 bp, and encoded predicted polypeptide of 203 amino acid (aa) residues including a putative N-terminal signal peptide. Other relevant regions found its sequence included the G and PW conserved aa motifs, and the consensus LARFAV sequence for phytocystatins and the reactive site QVVAG. The BrCPI1 protein shared 95, 94, 81, 80 and 78% identity with other CPI proterins isolated from Brassica oleracea (BoCPI-1), Arabidopsis thaliana (AtCY SB), Glycine max (GmCPI), Oryza sativa (OsCYS-2) and Zea may (ZmCPI) at amino acid level, respectively. Southern blot analysis showed that Brcpi1 was a low copy gene. Expression pattern analysis revealed that Brcpi1 was a tissue-specific expressing gene during reproductive growth and strongly expressed at mature seedling stages. Furthermore, overexpression of Brcpi1 in transgenic Arabidopsis was enhanced tolerance to salt and cold stresses. Meanwhile the juvenile seedling of Brcpi1 transgenic plants was not affected by various concentrations ABA in MS medium. Taken together, the results showed that Brcpi1 functioned as a cysteine protease inhibitor and it exhibited a protective agent against diverse types of abiotic stress, which induced this gene in a tissue- and stress-specific manner.
In plants, the fifth step of the plastidial 2-Cmethyl-D-erythritol 4-phosphate (MEP) pathway is catalyzed by 2-C-Methyl-D-erythritol 2,4-cyclodiphosphate synthase (MECP; EC: 4. 6. 1. 12), an enzyme proposed to play a key role in the regulation of isoprenoid biosynthesis. Here we report the isolation and functional characterization of a 823 bp Brassica rapa MECP (Brmecp) cDNA encoding a deduced polypeptide of 230 amino acid residues. Transcription levels of Brmecp were two-fold higher in petal compared to leaves. In addition, Brmecp expression in cabbage seedlings treated with ABA, $H_2O_2$ and drought was higher than control seedlings. These results were consistent with changes in chlorophyll contents in transgenic Arabidopsis. Thus, the Brmecp may contribute to the production of primary (chlorophylls and carotenoids) isoprenoid end-products in chloroplasts.

In order to reduce the problems from the presence of rye crop residues in maize establishment and yield, and also to improve the growth, productivity and nutritive value of rye(Seca1e cereal L.). an experiment was canied out to determine the variety effect of rye on the forage production system and the eft'ect of seeding and harvesting dates on the production and quality of rye. 'This experiment was conducted at the forage testing field of S.N.U., Suweon, from September 1986 to May 1989. Heading date of an early maturing rye variety, Wintermore. was earlier 10 days than that of a late maturing rye variety, Kodiak. A 15-days delay in the seeding dates of early and late maturing varieties of rye tended to delay the heading dates of the rye varieties for 3 and 4 days, respectively. Dry matter and in vitro digestible dry matter yields were markedly increased with earlier seeding date. Before 20 April, the DM and IVDDM yeilds of an early maturing rye variety, Winterrnore. were higher than those of a late maturing rye variety, Kodiak. However, no such a trend was found between the early and late maturing varieties of rye after 27 April. Less than 35% of ADF was recorded until the harvesting dates of 13 and 27 p r i l for early and late maturing rye varieties, respectively, but less than 46% of NDF was maintained until the harvesting drtte of 13 April for botg varieties. Based on the results obtained from this experiment. it may be concluded that the most desirable forage production from corn-rye double cropping system is to advance the seeding time of rye toward the first 10 days of September as well as harvesting time toward the first heading stage with an early maturing rye variety.

A fungal strain, Aspergillus terreus strain GA2, isolated from an agricultural field cultivating sweet sorghum, produced feruloyl esterase using maize bran. In order to obtain maximum yields of feruloyl esterase, the solid state fermentation (SSF) conditions for enzyme production were standardized. Effective feruloyl esterase production was observed with maize bran as substrate followed by wheat bran, coconut husk, and rice husk among the tested agro-waste crop residues. Optimum particle size of 0.71-0.3 mm and moisture content of 80% favored enzyme production. Moreover, optimum feruloyl esterase production was observed at pH 6.0 and a temperature of $30^{\circ}C$. Supplementation of potato starch (0.6%) as the carbon source and casein (1%) as the nitrogen source favored enzyme production. Furthermore, the culture produced the enzyme after 7 days of incubation when the C:N ratio was 5. Optimization of the SSF conditions revealed that maximum enzyme activity (1,162 U/gds) was observed after 7 days in a production medium of 80% moisture content and pH 6.0 containing 16 g maize bran [25% (w/v)] of particle size of 0.71-0.3 mm, 0.6% potato starch, 3.0% casein, and 64 ml of formulated basal salt solution. Overall, the enzyme production was enhanced by 3.2-fold as compared with un-optimized conditions.
Comparison of pesticide residue dissipation in/on cucurbitaceous fruit vegetables applied by foliar spraying under greenhouse was carried out to give information on crop grouping for assessment of pesticide risk. Even though the initial deposit of spayed pesticide on the target vegetables were somewhat different from the varieties, the dissipation patterns of the residues gave a similar tendency. In case sub-groupings of cucurbitaceous fruit vegetables are carefully arranged, the residue data among the vegetables are able to be taken for pre-harvest interval (PHI) settings or food safety assessment.

A wide diversity of liquid fertilizers and composts produced from the livestock manure in Korea is commonly applied to agricultural lands as an alternative of chemical fertilizers. However, their effects on the crop production and environmental impacts are still vague. The current study was conducted to understand the effects of the pig manure-based liquid fertilizer on the growth of Zea mays L. and soil properties. Four different liquid fertilizers were treated to each cultivated upland plot located in Gyeong-gi province, Korea while no fertilizer (control A) and a chemical fertilizer (control B) were treated to separate plots for comparison. The liquid fertilizer treatment did not make a significant difference in the fresh weight of Zea mays L. compared to the controls. This is probably due to the nutrient residues carried over from the last year fertilization. Electric conductivity (EC) and organic matter contents in soils were increased right after the liquid fertilizer treatments compared to the controls. However, soil pH was maintained as the same as the level of control A. A long-term effect of the continuous treatment of the manure based liquid fertilizer will be carried out in the successive study.
Gluten is the main functional component of wheat, and is the main source of the viscoelastic properties in a dough. One of the gluten group is glutenin, which is composed of high molecular weight (HMW) and low molecular weight (LMW) subunits. The HMW glutenin subunits (HMW-GS) have been shown to play a crucial role in determining the processing properties of the grain. They are encoded by the Glu-1 loci located on the long arms of homeologous group one chromosomes, with each locus comprising two genes encoding x- and y-type subunits. The presence of certain HMW subunits is positively correlated with good bread-making quality. The highly conserved N- and C- terminal contaning cystein residues which form interand intra-chain disulphide bonds. This inter chain disulphide bonds stabilize the glutenin polymers. In contrast, the repetitive domains that comprise the central part of the HMW-GS are responsible for the elastic properties due to extensive arrays of interchain hydrogen bonds. In this review, we discuss HMW-GS, HMW-GS structure and gluten elasticity, relationship between HMW-GS and bread wheat quality and genetic engineering of the HMW-GS.
BACKGROUND: Biochar has ability to reduce N loss, increase crop yield, and sequestrate carbon in the soil However, there is still limited study concerning the interactive effects of various biochars on NH3 loss and plant growth. This study, therefore, was conducted to investigate the NH4+ adsorption characteristics of biochar derived from rice and maize residues. METHODS AND RESULTS: By-products were pyrolyzed under oxygen-limited conditions at 300-700℃ for 1 hour and used for experiment of NH4+ adsorption in aqueous solution. The adsorption characteristics of biochar were studied using Langmuir isotherm. Biochar yield and hydrogen content decreased with increasing pyrolysis temperatures, whereas pH, EC, and total carbon content increased. The biochar pyrolyzed at lower temperatures was more efficient at NH4+ adsorption than those produced at higher temperatures. In addition, the RL values, indicating equilibrium coefficient were between 0 and 1, confirming that the result was suitable for Langmuir isotherm. CONCLUSION: The maize stalk biochar pyrolyzed at 300℃ was the most efficient to adsorb NH4+ from the aqueous solution. Furthermore, the adsorption results of this experiment were lower than those of other prior studies, which were ascribed to different experimental conditions such as ingredients, and pyrolysis conditions.
The plant uptake of endosulfan, a new persistent organic pollutants from soil environment was investigated on ginseng through the field survey in Korea. The endosulfan residues in soil for this survey were ranged on 0.013-0.136 mg kg-1. The plant uptake factor (PUF) for endosulfan in ginseng was 0.243-1.708 and the highest PUF was found on 3-year-old ginseng. The PUF for endosulfan decreased in the longer cultivation period and it might be caused by the dilution effect of ginseng growth. In addition, the soil organic matter (SOM) content affected on the PUF negatively and Pearson correlation coefficient (r) between SOM and PUF was -0.7812 (p <0.05). Thus, higher SOM would positively affect to reduce the endosulfan residue in ginseng root.
