Identification of Sugar from Korean Ginseng Saponins by Acid-hydrolysis (인삼(人蔘) Saponin 산가수분해물(酸加水分解物)의 당류(糖類)에 관(關)한 연구(硏究))

  • In order to identify the sugars of saponin originated from Korean ginseng, experimentations were made on the four kinds of ginseng. The results are summarized as follows. 1. The starch content of Ginseng-tail, White-ginseng 3 and 6 years old-whole ginseng were 10.4, 31.5, 8.2, 25.6% and total sugar of its were 37.0, 61.5, 64.5, 62.5% and free sugar were 7.6, 10.5, 11.3, and 10.7% respectively. 2. Saponins were separated from Ginseng-tail, White-ginseng, 3 and 6 years old-whole ginseng by modified SHIBATA method. looms of crude saponin was used for the Thin layer Chromatography (TLC) and thirteen to twelve spots of saponin were isolated by double development $(Solvent:\;CHCL_3\;:\;MeOH\;:\;H_2O=65\;:\;35\;:\;10)$ and by two dimensional development. $(Solvent:\;nBuOH\;:\;HOAC\;:\;H_2O=4\;:\;1\;:\;5)$ The Pattern of spot was not significantly different according to Ginseng sample. 3. Glucose was identified from the acid-hydrolyzate of Ginsen-tail saponin by paper chromatography and isolated the unknown chromatogram seems to be pentose.

Utilization of Persimmon Peel and Its Tannin Extracts for Animal Feeding (가축사양에 있어서 감 과피와 감 과피탄닌 추출물의 이용)

  • Tannins are phenolic compounds that precipitate proteins and composed of a very diverse group of oligomers and polymers. Tannins are potential biological antioxidants, which are widely believed to be an important line of defense against oxidative damage and may participate in the prevention of cancer and cardiovascular disease. Persimmon(Diospyros kaki L.) has been cultivated in East Asia and is a good source of nutritional antioxidant vitamins, carotenoids and tannins. In general persimmon peel was regarded as a waste matter, although based on recent studies, the peel contains more carotenoids and polyphenols than pulp. Several investigation conducted in experimental animals have reported that dietary persimmon fruit and peel effectively lowered the levels of plasma total cholesterol and LDL-cholesterol. We conducted experiments to investigate in vitro antioxidative activities of persimmon peel powder (PP) and its soluble tannin extract (ST) and their dietary effects on productive performances and physiological responses in poultry. The PP and ST exhibited in vitro antioxidative activity in SOD - like activity model. The yolk color and eggshell color were significantly improved by the addition of PP and ST into layer diets. The contents of total cholesterol, triacylglycerol and phospholipid of liver in the groups fed diets containing PP and ST tended to be reduce as compared with those of control. With adding of PP and ST, Haugh unit was increased after 7 and 14 days of storage. In conclusion, PP and ST can be used as valuable feed additives for reducing hepatic lipid contents without harmful effects on overall productive performances and physiological responses in laying hens.

  • 5 bed sand filter, applying biological oxidation, was designed and studied on the treatment of water works. Never using any coagulant agent (drugs), which may cause water pollution in pre-treatment of head water, the auther attempt a high rate filtration by the microorganism (nitrofication bacteria) end plant which populate in multi layer sand beds. The result are as follows : In order to evaluate the oxygen effect on filtration, oxygen was injected in aeration tank attached to each filter tank while filtration, and $NH_3$ was tested as a representaiive ingredient. It was found out that the aeration method was more effective, with over 33% of $NH_3$ removal capacity, than the anerobic and this 5 bed filter showed double removal capacity of $NH_3$ by comparing with conventional sand bed (2 stage bed). According to the examination of two kind of head water, pre-treated with coagulant agent and activated carton, the filtration capacity was affected by the polluted condition of head water, resulting that lower value of pollution and slower velocity of filtration showed more efficiency of $NH_3$ removal. In this experiment $NH_3$ content tested in treated water had a fairly good correlation with others.

Isolation of Bacteria Producing a B-Cell-Specific Biological Response Modifier Found in Korean Fermented Soybean Paste

  • In a previous study, a biological response modifier (BRM) specifically enhancing the function of B-cells was isolated from Korean fermented soybean paste (Kfsp), but not from non-fermented soybeans. In this study, we attempted to isolate the bacteria producing the BRM from Kfsp (KfspBRM) by ELISA using anti-KfspBRM and by B-cell proliferation. Five bacteria whose culture supernatants showed the BRM activities were isolated, and one of them was identified as Bacillus licheniformis E1. The bacterial BRM (bBRM) originated from a slime layer of B. licheniformis El had a molecular weight of 1,594 kDa, and contained $33\%\;(w/w)$ of reduced sugar and $4.6\%\;(w/w)$ of protein content. The bBRM appeared to be a glycoprotein that is physically, structurally, and functionally similar to the KfspBRM, suggesting that the isolates including B. licheniformis El may produce the KfspBRM in the fermentation process of soybean paste. The mass production of the BRM by the bacterium may help to study B-cells in immunology, and the enrichment of the BRM in Kfsp may help patients in future who are medically in need of potentiation of B-cell proliferation and antibody production.

Studies on wet polishing characteristic of rice (습식연미 도정특성에 관한 연구)

  • Series of study were conducted to investigated the condition of wet polishing and quality of wet polished rice. The optimal condition for wet polishing was determined to be 0.5% added water after removal bran layer of 95% by normal milling. Its polishing yield was 0.75% lower than normal polishing, and the moisture content of wet and normal polished rice was the same, but the whiteness of wet polished rice was higher by 3.5% than that of normal polished rice and the lightness was very good. The total solid as washing wet polished rice in water and the total solid in residual liquid of cooked wet polished rice was about 0.30 and 2% lower than those of normal polished rice, respectively. The storability of wet polished rice was better than that of normal polished rice.

Effectiveness of Bioremediation on Oil-Contaminated Sand in Intertidal Zone

  • Bioremediation technologies were applied to experimental microcosms, simulating an oil spill in a lower intertidal area. Three treatments (oil only, oil plus nutrients, and oil plus nutrients and microbial inocula) were applied, and each microcosm was repeatedly filled and eluted with seawater every 12 h to simulate tidal cycles. To minimize washing-out of the inoculum by the tidal cycles, microbial cells were primarily immobilized on diatomaceous earth before they were applied to the oiled sand. Oil degradation was monitored by gravimetric measurements, thin layer chromatography/flame ionization detector (TLC/FID) analysis, and gas chromatography (GC) analysis, and the loss of oil content was normalized to sand mass or nor-hopane. When the data were normalized to sand mass, no consistent differences were detected between nutrient-amended and nutrient/inoculum-amended microcosms, although both differed from the oil-only microcosm in respect of oil removal rate by a factor of 4 to 14. However, the data relative to nor-hopane showed a significant treatment difference between the nutrient-amended and nutrient/inoculum-treated microcosms, especially in the early phase of the treatment. The accelerating effect of inoculum treatment has hardly been reported in studies of oil bioremediation in the Tower intertidal area. The inoculum immobilized on diatomaceous earth seemed to be a very effective formulation for retaining microbial cells in association with the sand. Results of this study also suggest that interpretation of the effectiveness of bioremediation could be dependent on the selection of monitoring methods, and consequently the application of various analytical methods in combination could be a solution to overcome the limitations of oil bioremediation monitoring.

Studies on the Desalinization in Reclaimed Tide Lads (II) - by the Open Conduit- (간척지 제염에 관한 시험(II) - 개거에 의한 제염효과 시험-)

  • This research was attempted to study on the effects of desalinization by the depth and interval of open conduit in Kang-Hwa polder where is located at the Kil-sang Myun, Kang-Hwa Gun, Kyung-gi Do, and it has been continued for the three years from 1967 to 1969. The results obtained are as follows; 1. The depths of saline expulsion by supplying of irrigation water are approximately 30cm to 50cm under the ground surface, but saline expulsion is hardly done in case of the depth which is deeper than the above mentioned, because the moisture and saline content hardly change in such a condition. 2. The speed of vertical percolation gradually decreases below the 30cm depth, but it is noticed that there is a tendency to make the percolation of the horizental direction from its layer in Kang-Hwa reclaimed tidal land. 3. Comparing experimental treatments-varing depths and intervals of open conduits, the interval of open conduit has a more effect upon the promotion of desalinization and increasing of the rice yields than the depth of it. Therefore, according to the results of experimental data, the optimum depth of open conduit is about 0.9m, the effective interval of it is about 18m. 4. Considering the loss of arable area by the layout of open conduit, the reasonable interval of it could extend to 36m.

A Study on the Soil Stress Distribution in Furrow Slice (역토내(壢土內)의 응력분포(應力分布)에 관(關)한 연구(硏究))

  • In order to identify the stress distribution on the furrow slice a small soil bin instrumented with soil stress meters was designed and constructed. From a series of experiments conducted in the soil bin the following results were obtained. 1) Neither the cutting conditions nor the soil conditions affected the direction of the principal stress. 2) The magnitude of the principal stress increased as the tillage depth increased. However, no effects due to lift angles were shown on the magnitude of the principal stresses. 3) The maximum principal stress increased with increase of the moisture and clay contents in the soil. 4) In the clay soil, the maximum principal stresses were distributed uniformly over the tillage depth. However, as the sand content increased, the maximum principal stresses decreased gradually on the top layer so that the distribution over the tillage depth became a trapezoidal shape.

  • This study is on development of UV-cured water soluble coating composition which have more improved anti-cigar burning to prevent a surface of PVC tile from damage of heat. To make an anti-cigar burning coating solution, thermostable agent that synthesize main materials phosphorus compound, guanidine and ammonium phosphate dibasic used temporary flame retardants, changing their contents from 10 to 30wt% against quantities of resin and compounded. After coating PVC tiles using bar-coating method that can adjust a thickness, we estimated surface properties of coated layer such as anti-cigar burning, adhesive power, chemical resistance, thickness of coating, and so on. Results showed that a coating composition added 30 wt% of phosphorus compound and coated with No.12 bar-coater had the best optimized surface property in anti-cigar burning effect, adhesive power and chemical resistance. Also, we could find anti-cigar burning effect was improved as thermostable agents content and coating thickness increased.

Responses of Benthic Animals in Spatial Distribution to the Sedimentary Environments on the Deep-sea Floor, the Clarion-Clipperton Fracture Zone, Northeastern Pacific Ocean (북동 태평양 심해저 C-C 해역의 퇴적 환경과 대형저서동물 분포와의 관계)

  • Relationships between sedimentary environments and abundance of benthic animals were examined on the deep-sea floor, the Clarion-Clipperton Fracture Zone, in the northeast equatorial Pacific Ocean. Specimens were collected using a box corer at 8 stations by sieving through 0.3 mm mesh screen. Sediments showed finer grain size ranged from 5.63 to $7.97{\varphi}$, 83.1% of mean porosity, 1.81 kPa of mean shear strength and organic carbon content in sediment ranged from 0.97 to $1.87\;mg/cm^3$. Manganese nodules covered on the bottom layer from 4 to 57% of coverages. A total of 26 faunal groups in 6 phyla was sampled and comprised 1,467 individuals. Mean biomass were calibrated to 0.5 gWWt/$0.06\;m^2$. Small-sized animals including foraminiferans and nematods were dominated among the faunal group which comprised 49.1% (892 ind.) and 11.5% (320 ind.), respectively. In SPI-analysis, vertical bio-disturbance marks were not observed except to Beggiatoa-type bacterial mats. As the results of relationship between environments and benthos, abundance of benthic animals, especially nematode, showed only a negative correlation to the coverage of nodules, and any other sedimentary factors analyzed in this study were rarely affected to the spatial distribution of benthic animals.
