Development and validation of SSR markers in Eleutherococcus senticosus

  • Eleutherococcus senticosus (Siberian ginseng) is an important medicinal tree found in Russian taiga, and northern regions of Korea, Japan, and China. In this study, we analyzed the genome-wide distribution of microsatellites in E. senticosus using developed SSR markers. A total of 711 clones from an SSR-enriched genomic DNA library were sequenced, of which 47 clones (6.6%) were redundant. Of the 664 independent clones, only 12 polymorphic SSR markers were obtained, which also revealed successful amplicons in E. senticosus accessions. Using the developed SSR markers, we estimated genetic diversity and population structure among 131 E. senticosus accessions in Korea and China. The number of alleles ranged from 2 to 11, with an average of 7.4 alleles. The mean values of observed heterozygosity ($H_O$) and expected heterozygosity ($H_E$) were 0.59 and 0.56, respectively. The average polymorphism information content (PIC) was 0.51 in all 131 E. senticosus accessions. E. senticosus accessions in Korea and China showed a close genetic similarity. Significantly low pairwise genetic divergence was observed between the two regions, suggesting a relatively narrow level of genetic basis among E. senticosus accessions. Our results not only provide molecular tools for genetic studies in E. senticosus but are also helpful for conservation and E. senticosus breeding programs.

Phenotypic diversity, major genes and production potential of local chickens and guinea fowl in Tamale, northern Ghana

  • Objective: Our study provides information on phenotypes of local chickens and guinea fowl and their body measures as well as on major genes in local chickens in northern Ghana. Methods: Qualitative and morphometric traits were recorded on 788 local chickens and 394 guinea fowl in urban households in Tamale, Ghana. Results: The results showed considerable variation of color traits and numerous major genes in local chickens, while color variations and related genotypes in guinea fowl were limited. In local chickens, white was preferred for plumage, whereas dark colors were preferred for beak and shanks. More than half of the chickens carried at least one major gene, but the contributions of single gene carriers were low. All calculated allele frequencies were significantly lower than their expected Mendelian allele frequencies. We observed higher mean body weight and larger linear body measures in male as compared to female chickens. In female chickens, we detected a small effect of major genes on body weight and chest circumference. In addition, we found some association between feather type and plumage color. In guinea fowl, seven distinct plumage colors were observed, of which pearl grey pied and pearl grey were the most prevalent. Male pearl grey pied guinea fowl were inferior to pearl grey and white guinea fowl in terms of body weight, body length and chest circumference; their shank length was lower than that of pearl grey fowl. Conclusion: Considerable variation in qualitative traits of local chickens may be indicative of genetic diversity within local chicken populations, but major genes were rare. In contrast, phenotypic and genetic diversity in local guinea fowl is limited. Broader genetic diversity studies and evaluation of trait preferences of local poultry producers are required for the design of appropriate breeding programs.

일본 전통마을의 유지.관리방법 분석 -쯔마고마을과 시라카와마을을 사례로- (An Analysis of Management Methods for Traditional Village in Japan -The Case of Tumago Village and Shiragawa Village-)

  • The traditional village, which is subject for this paper, can be defined as a community having unique combinations of natural, cultural, and social characteristics of that nation, which reflects settlement environments during hundreds of years. Now, in spite of tis potential power of traditional village, national strategies do not find satifsactory directions in Korea. In terms of this concenrs, this paper tries to analyze and diagnose about successful precedents(Tsumage village and Shiragawa village, Japan) with focus on the village management. And this paper aims to explore concrete management systems of Japanese cases, to find clues for practical application, and to suggest several instructive concepts in the light of management system of traditional village management. The analysis is progressed in three viewpoints(village space, village attraction, and village community). And it is extracted that diverse management systems are necessary to secure sustainable traditional village and their way of life, particularly in the face of the pressure of tourism. As a result of exploring of Japanese two cases, representative characteristics, which are found, are as follow; Fist, objective of management is not tourism development but maintenance of sustainable life system. Second, management systems are having not compulsory but spontaneous open decision making process, and in particular, village management is mainly operated by the local government and semipublic institute(inhabitants-oriented0 which have the strongest influential power in village composition units. Finally, village management programs are mostly experience-type and are composed of public law and regulations, diverse operation program, and individual efforts of inhabitants for village management. Because this paper mainly focused on two precedents, there should be more diverse cases. however, extracted conclusions have practical meanings for village management and can be used in re-establishing management concepts of Korean traditional villages.

Investigation of Genetic Diversity between Wild-caught and Hatchery-reared Rock Bream (Oplegnathus fasciatus) Using Microsatellite DNA Analysis

  • Marine fisheries are important natural resources and must be maintained, especially fish species that are important sources of food. Despite the increase in stocking programs to maintain fisheries with artificially raised fish, the genetic impact stocking has on the wild fry population has not been addressed. Genetic variation in rock bream, Oplegnathus fasciatus, within and between wild-caught parents and the $F_1$ generation produced by them in 1 day was assayed using nine highly variable micro satellite markers. The nine micro satellite loci used in this study displayed diverse polymorphisms, and in total, 98 different alleles were observed over all loci. Differences in genetic variability of the $F_1$ offspring compared to their wild-caught parents (brood stock) were observed in terms of allele frequency, gene diversity, and heterozygosity. Although the $F_1$ generation of rock bream was missing 16% of the micro satellite alleles, no significant reduction was found in mean heterozygosity of the $F_1$ population compared to the brood stock. Eight of nine loci showed significant Hardy-Weinberg equilibrium (HWE) deviations in the $F_1$ population, while the brood stock deviated from HWE at three micro satellite loci (KOF85, KOF360 and KOF374). These deviations showed mostly a deficit of heterozygotes. Our results provide evidence for genetic differences in the $F_1$ hatchery offspring compared to their wild-caught parents and reinforce the need for a series of consecutive egg collections to avoid the loss of genetic variability. This also further underscores the importance of monitoring genetic variability of hatchery populations for the conservation of natural rock bream resources.

목재문화재를 가해하는 막대바구미류(딱정벌레목, 바구미과)에 대한 보고 (Cossonid Weevils (Coleoptera: Curculionidae) Infested on Wood Cultural Properties)

  • 막대바구미류는 대부분이 목재류를 천공가해하기 때문에 목재문화재에 피해를 주는 경우가 있다. 국내 목재류 문화재에서 발생한 막대바구미류를 정리한 결과, 자작나무막대바구미(Hexarthrum brevicorne Wollaston)가 2004년에 전북 전주시에 위치한 사적 제379호인 전주향교에 소장된 완영책판에 심각한 피해를 주었으며, 2007년에는 경남 밀양소재 박물관에 소장중인 반닫이와 2008년에는 서울 소재 박물관에 관복함, 전남 순천소재 사찰에 보관중인 경판에서도 발생되었고, 분비나무막대바구미(Rhyncolus sculpturatus (Waltl))는 2008년 서울 소재 박물관에서 소장된 목제(재)생활도구를 가해하는 것이 확인되었다. 따라서 소중한 국가자산인 문화재를 해충으로부터 보호하기 위해 보존과학분야에서 종합적인 해충 관리기술이 한층 요구되고 있다.

Development of EST-SSRs and Assessment of Genetic Diversity in Little Millet (Panicum sumatrense) Germplasm

  • Little millet (Panicum sumatrense) is well known for its salt and drought stress tolerance and high nutritional value, but very limited knowledge of genetic variation and genomic information is available. In this study, a total of 779 primer pairs were designed from the 22,961 EST sequences of switchgrass (Pancium virgatum), of which 48 EST-SSR markers were developed based on the trials of transferability of these primers in little millet. The EST-SSR amplicons showed reproducible single band polymorphism and produced a total of 160 alleles with an average of 3.3 alleles per locus in 37 accessions of little millet. The average values of expected and observed heterozygosities were 0.266 and 0.123, respectively. The polymorphic information content (PIC) values were observed in range of 0.026 to 0.549 with an average of 0.240. The genetic relatedness among the little millet accessions was evaluated by neighbor-joining dendrogram, which grouped all accessions into two distinct groups. The validation thus demonstrated the utility of the switchgrass EST-SSR markers in assessing genomic relationships in little millet. The findings from this study could be useful for designing strategies for the identification of diverse germplasm for conservation and future molecular breeding programs for little millet.

우리나라 대학에서의 지속가능발전교육의 가능성과 과제 - 교육 과정 파트너쉽, 지속가능한 대학 경영을 중심으로 - (Possibilities and Challenges in Education for Sustainable Development in Korean Universities - Focused on Curriculum, Partnership, and Sustainable University Management -)

  • This study aims to address the current status of ESD(Education for Sustainable Development) in Korean universities since UN DESD was launched in 2005, and to explore the possibilities and challenges in them. For this purpose we conducted questionnaire surveys and case studies on members of university communities which were engaged in green campus activities or interested in SD and ESD. Results of questionnaire surveys showed that most of those who answered the questionnaire were well aware of SD, but had comparatively low understanding of ESD. The highest number of respondents answered that since 2005 they had newly opened courses on SD or ESD, or added contents on SD or ESD to existing ones. Ratio of network participation among ESD-related universities was over 30%, and they appeared to have the highest partnership ratio with NGOs. Not many universities had policies for sustainable school management, and 'green space conservation and ecosystem protection' and 'energy and resource saving campaign and monitoring' were most common sustainable environment protection activities. Through case studies on eight universities, it was discovered that ESD programs in universities took various forms such as whole-university approaches, participatory courses and club activities. We suggest that it is needed to make efforts to find out good examples of ESD in Korean universities and share the results with university leaders, professors and staffs for further development of ESD.

일 소음작업장 근로자들의 소음에 대한 인식 및 태도, 예방행위에 관한 연구 (A Study on the Knowledge, Attitude and the Preventive Behavior of Noise of Workers Exposed to the Harmful Noise)

  • This study was to investigate knowledge, attitude and the preventive behaviour of noise of workers exposed to harmful noise and to provide primary data for noise-exposed workers' hearing conservation and the prevention of noise induced hearing loss. Method: The subjects were 104 workers who participated voluntarily in the questionnaire. The instrument used in this study was knowledge, attitude and practice on noise-induced hearing loss tool by Rhee & Yi (1996). The data were collected from August to September, 2002 with a self-administered questionnaire. Data were analyzed by SPSS 10.0 win program for finding frequency, percentage, mean, standard deviation, t-test, and ANOVA. Results: The average age of workers was 35.9 and the current average year of working at the noise place was $5{\sim}$10 years. In the group of previous noise-exposed workers, they showed significant difference in preventive behaviors (t=4.087, p=.048). In the group of current noise-exposed workers, they showed significant difference in recognition and attitude of noise(F=4.707, p=.004). Also, the duration of wearing ear protection equipment(t=2.383, p=.019), the feeling of wearing ear protection equipment(F=3.602, p=.031) and comfortable sense of wearing ear protection (F=3.919, p=.023) was significant in the preventive behaviour of noise. Conclusion: Considering the results above, we can recognize that there are close relations among the knowledge, attitude and the preventive behaviour of noise. Therefore, it is necessary to develop education - programs for noise-exposed workers' better understanding of noise and preventive behaviors

두록의 동결정액의 운동학적 특성과 ESR1 유전자의 SNP(g.35756T>C)와 연관성 분석 (Association with Post-Thawed Semen Motility and Kinematic Characteristics of g.35756 T>C on Estrogen Receptor 1 (ESR1) Gene in Duroc Pigs)

  • Cryopreservation of boar semen is continually researched in reproductive technologies and genetic resource banking in breed conservation. For evaluating the boar semen quality, sperm motility (MOT) is an important parameter because the movement of spermatozoa indicates active metabolism, membrane integrity and fertilizing capacity. Various researches have been trying to improve the quality of semen Post-thawed in boar. Recently, polymorphism (g. 35756 T>C) of Estrogen Receptor 1 (ESR1) gene reported to be significant association with MOT. This study was conducted to evaluate the ESR1 gene as a positional controlling for motility and kinematic characteristics of post-thawed boar semen. To results, The g.35756 T>C SNP of ESR1 was significantly associated with frozen semen motility and kinematic characteristics. The g.35756 T>C SNP was high significantly associated with MOT, VCL, VSL and VAP (p<0.001). The SNP was also significantly associated with ALH (P<0.05). Therefore, we suggest that the g. 35756 T>C polymorphism in the intron 1 region of the porcine ESR1 gene could potentially be applied in frozen semen programs to improve MOT trait, but only after validation in other populations.
