Studies on the Effect of Ginseng Extract to Aspergillus Enzyme Activity (인삼추출액이 국균의 효소생산에 미치는 영향에 관한 연구)

  • Effects of the addition of Ginseng extract and it's incubation time on enzyme activity (${\alpha}$-amylase, ${\beta}$-amylase, protease) of Aspergillus app. was investigated. 1. ${\alpha}$-Amylase activity of Asp. oryzae was higher for 48 hours than control, however, the same after 48 hours. ${\beta}$-Amylase activity was stimulated at the concentration of 0.1 to 1.0%, and decreased at the concentration of 3.0 to 5.0%. The acid protease activity was higher than control for 72 hours and in the medium of 120 hours was decreased significantly. The alkaline protease activity was lower than control. However, alkaline protease activity was higher than acid protease activity. 2. ${\alpha}$-Amylase of Asp. niger was increased in proportion to increasing of the extract concentration. ${\beta}$-Amylase was increased at the concentration from 0.5 to 3.0% and it's activity was depressed in proportion to increasing of the extract concentration for 48 hours and on the contrary it was improved in proportion to increasing of the extract concentration for 72 hours except for 5.0% concentration with marked decreasing. Alkaline protease activity was increased at lower concentration (0.1∼0.5%) for 24 to 48 hours and it was depressed at higher concentration (1.0 to 5.0), however after 48 hours incubated, it's activity was decreased in preparation to increasing of the extract concentration.

Effects of Concentration of Inhibitor on the Production of $\alpha$-amylase and Growth of Bacillus amyloliquefaciens (Bacillus amyloliquefaciens 성장과 $\alpha$-amylase 생합성에 미치는 저해제의 영향)

  • In this study, Bucillus amyloliquefaciens was adopted as bacterial source to investigate the concentration of carbon source by adding inhibitors in the batch culture. By adding acetic acid at $10g/\ell$ of initial glucose concentration, maximum dry cell density was obtained with the highest value of /$\3.9gell$ at $1.0g/\ell$ of initial acetic acid concentration. By adding acetic acid al 10g/$\ell$ of initial glucose concentration, maximum ${\alpha}$-amylase production was obtained with 331.55unit/m1 at $2.0g/\ell$ of initial acetic acid concentration. ${\alpha}$-Amylase production was decreased with the increase of initial acetic acrid concentration. By adding acetic acid to the medium, cell growth and ${\alpha}$-amylase production was higher in glucose than in maltose. By adding lactic acid to the medium, cell growth was decreased.

  • Three wethers fitted with silastic catheters for collection of pancreatic juice, and cannulas located in the abomasum and the duodenum were used to investigate the effects of different hay and energy intake on pancreatic exocrine secretion. The wethers were fed Italian ryegrass hay or alfalfa hay at maintenance energy requirement and alfalfa hay ad libitum. High energy intake from alfalfa significantly increased abomasal flow of dry matter and both the concentration and daily secretion of ${\alpha}-amylase $. The high energy intake also tended to increase daily secretion of lipase, trypsin and chymotrypsin through the large volume of pancreatic juice. Compared with Italian ryegrass hay, alfalfa hay at the maintenance decreased abomasal dry matter flow, but increased concentration of ${\alpha}-amylase $ in the pancreatic juice, and tended to increase daily secretion of ${\alpha}-amylase $. The secretion of the other enzymes was not different between the two hays at maintenance intake. These results suggest that the kind of hay could change the concentration of ${\alpha}-amylase $ in the pancreatic juice, and that the intake level of alfalfa hay affects the ${\alpha}-amylase $ concentration and the juice volume secreted from the pancreas.

  • The effect of tannic acid on GAs and cyclic-AMP promoted amylase induction in barley aleurone layers was examined. Of a variety of adenine compounds, only cyclic-AMP and ADP showed significant activity, and these activities were promoted by addition of theophylline to the incubation medium. When aleurone layers of barley endosperm tissues were incubated with GAs in the presence of tannic acid, the amylase activity in the incubation medium was reduced. Cyclic-AMP induced amylase activity was also reduced by additiion of tannic acid. The cyclic-AMP response promoted was more sensitive to tannin inhibition than GAs response. The inhibitory effect of tannic acid shwoed reversibility by addition of higher concentration of GAs or cyclic-AMP. The tannic acid effect on GAs response was also recovered by addition of a higher concentration of cyclic-AMP. Experiment with polyacrylamide disc electrophoresis showed different isozyme patterns according to the additions in the incubation medium. Inhibitory effects of decursinol and coumarin was compared with that of tannic acid. They showed different zymogram patterns.

  • In the polyethylene glycol/dextran aqueous two-phase systems, volume ratio was increased and partition coefficient was decreased with the increase of potyethylene glycol molecular weight and concentration. However the volume ratio was decreased and the partition coefficient was increased with the increase of dextran molecular weight. On the other hand, the volume ratio and the partition coefficient were decreased with the increase of dextran concentration. Continuous enzymatic hydrolysis of soluble starch with $\alpha$-amylase which was produced by Bacillus amyloliquefaciens IF0 14141 was investigated in polyethylene glycol/dextran aqueous two-phase systems. Nonreacted soluble starch and $\alpha$-amylase were reused in these systems. $\alpha$-Amylase activity was maintained more than 100 hrs by recycling of $\alpha$-amylase from bottom of settler to reactor.

Effect of Bean Water Concentration and Incubation Time on Amylase Activity and Physicochemical Characteristics of Yukwa Paste (콩물 농도 및 숙성 시간이 Amylase 의 활성과 유과 반죽의 특성에 미치는 영향)

  • ${\alpha}-amylase$ activities of bean water was not significantly influenced by bean water concentrations but they were remarkably influenced by different temperatures and substrates. ${\alpha}-amylase$ activities of bean water on cooked starch were significantly higher than those on raw starch. ${\beta}-amylase$ and glucoamylase activities in 14% bean water were significantly higher than those in 7% bean water. Yukwa paste is glutinous rice flour paste. Bean water was added to Yukwa paste by 0, 7, 14% and incubated 0, 3, 6, 9, 12 hours at $60^{\circ}C$. The pH of Yukwa paste increased with bean water concentration and decreased with the incubation time. The viscosity decreased with bean water concentration and incubation time. The ruducing sugar content of Yukwa paste increased with bean water concentration and incubation time. The changes of reducing sugar content in cooked Yukwa paste were much higher than those in the raw one. ${\alpha},\;{\beta}-amylase$glucoamylase activities of Yukwa paste also increased with bean water concentration, and their activities were much higher on the cooked glutinous rice flour than those on the raw one. The SEM observation on the freeze-dried flour of Yukwa paste showed breakdown of amylopectin structure by addition of bean water in the paste.

Effects of Concentration of Glucose and By-product on the Growth of Bacillus amyloliquefaciens 23350 (B. amyloliquefaciens 세포 성장에 미치는 포도당 및 부산물 농도의 영향에 관한 연구)

  • In this study, an attempt was made to investigate optimum cell growth and products by Bacillus amyloliquefaciens 23350 in batch culture by varing carbon soures. Maximum dry cell density increased with the increase of initial glucose concentration. Maximum dry cell density was obtained with the highest value of 5.2g/l at 30g/l of initial glucose concentration. By adding acetic acid at 20g/l of initial glucose concentration, the cell growth rate decreased with the increase of initial acetic acid concentration. Among the various carbon sources, maximum $\alpha$-amylase production was obtained with 225unit/ml at 10g/1 of initial glucose concentration. Optimum production of $\alpha$-amylase was obtained with 376unit/ml at 2g/l of initial acetic acid concentration and 20g/l of initial glucose concentration. By 10g/1 of initial glucose concentration, both good maximum specific cell growth rate and maximum $\alpha$-amylase production rate were obtained. In view of the results studied optimum production and specific production rate of $\alpha$-amylase, acetic acid was initially added 2~4g/l with 20g/1 of initial glucose concentration in batch culture.

  • $\beta$-Amylase was purified from sweet potato by acetone fractlonatlon, Sephadex A-50 ion exchange chromatography and Sepgadex G-200 gel chromatographyl The higher enzyme concentration was, the higher heat stability of enzyme became. After 1 hour 30 minute. At 6$0^{\circ}C$ in pH 5, enzyme under concentration of 30$\mu$l/ml lost its activity completely and over the concentration of 100$\mu$g/ml remained 25% of activity. The enzyme was stabilized at range of pH 4~10 and pH stability was increased by glycerol. Five moles of NaCl inhibited completely of the enzyme activity. SDS of 0.05% inhibited the enzyme completely after 12 hours at 37$^{\circ}C$ in pH5. One mole guanidine-HCl and 8M urea inhibited the entire enzyme after 13 hours at 37$^{\circ}C$ in pH 5.

  • For the production of ${\alpha}$-amylase cloned from Bacillus licheniformis expressed in E. coli, cultivating factors including the concentrations of glucose, maltose and acetic acid were investigated. The results were as follows: 1) Maximum ${\alpha}$-amylase yield and maximum specific production rate obtained from glucose source were better than those achieved from maltose source. 2) The optimum production yield of ${\alpha}$-amylase was obtained at 1.0ml/$\ell$ or less of initial acetic acid concentration.

Dual Effect of $H_2O_2$ on the Regulation of Cholecystokinin-induced Amylase Release in Rat Pancreatic Acinar Cells

  • [ $H_2O_2$ ], a member of reactive oxygen species (ROS), is known to be involved in the mediation of physiological functions in a variety of cell types. However, little has been known about the physiological role of $H_2O_2$ in exocrine cells. Therefore, in the present study, the effect of $H_2O_2$ on cholecystokinin (CCK)-evoked $Ca^{2+}$ mobilization and amylase release was investigated in rat pancreatic acinar cells. Stimulation of the acinar cells with sulfated octapeptide form of CCK (CCK-8S) induced biphasic increase in amylase release. Addition of $30\;{\mu}M\;H_2O_2$ enhanced amylase release caused by 10 pM CCK-8S, but inhibited the amylase release induced by CCK-8S at concentrations higher than 100 pM. An ROS scavenger, $10\;{\mu}M$ Mn(III)tetrakis(4-benzoic acid)porphyrin chloride, increased amylase release caused by CCK-8S at concentrations higher than 100 pM, although lower concentrations of CCK-8S-induced amylase release was not affected. To examine whether the effect of $H_2O_2$ on CCK-8S-induced amylase release was exerted via modulation of intracellular $Ca^{2+}$ signaling, we measured the changes in intracellular $Ca^{2+}$ concentration $([Ca^{2+}]_i)$ in fura-2 loaded acinar cells. Although $30\;{\mu}M\;H_2O_2$ did not induce any increase in $[Ca^{2+}]_i$ by itself, it increased the frequency and amplitude of $[Ca^{2+}]_i$ oscillations caused by 10 pM CCK-8S. However, $30\;{\mu}M\;H_2O_2$ had little effect on 1 nM CCK-8S-induced increase in $[Ca^{2+}]_i$. ROS scavenger, 1 mM N-acetylcysteine, did not affect $[Ca^{2+}]_i$ changes induced by 10 pM or 1 nM CCK-8S. Therefore, it was concluded that $30\;{\mu}M\;H_2O_2$ enhanced low concentration of CCK-8S-induced amylase release probably by increasing $[Ca^{2+}]_i$ oscillations while it inhibited high concentration of CCK-8S-induced amylase release.
